Abstract:A novel method related to diffraction tomography is presented for imaging two-dimensional weak-scattering objects embedded in a lossy background with phaseless data.The proposed method requires only intensity measurements.Based on the information of the total field magnitudes the pseudo Fourier transformation of the object function can be determined,which is distributed along a fixed arc in the complex space.After expanding the object function by Fourier series the pseudo Fourier spectrum can be represented by the Fourier coefficients,which can be written in a form of system of equations.By means of regularization the ill-posed matrix can be solved and the object function is reconstructed.Numerical simulation results presented have proven the validity of the proposed method.Since this reconstruction algorithm needs only intensity information to rebuild the geometric and electromagnetic properties of objects embedded in a lossy background,it circumvents the difficulty of direct phase measurement.

Abstract:Diffraction tomography is an inverse scattering technique used to reconstruct the spatial distribution of the material properties of a weakly scattering object. The object is exposed to radiation, typically light or ultrasound, and the scattered waves induced from different incident field angles are recorded. In conventional diffraction tomography, the incident wave is assumed to be a monochromatic plane wave, an unrealistic simplification in practical imaging scenarios. In this article, we extend conventional diffraction tomography by introducing the concept of customized illumination scenarios, with a pronounced emphasis on imaging with focused beams. We present a new forward model that incorporates a generalized incident field and extends the classical Fourier diffraction theorem to the use of this incident field. This yields a new two-step reconstruction process which we comprehensively evaluate through numerical experiments.

Abstract:X-ray diffusive dark-field imaging, which allows spatially unresolved microstructure to be mapped across a sample, is an increasingly popular tool in an array of settings. Here, we present a new algorithm for phase and dark-field computed tomography based on the x-ray Fokker-Planck equation. Needing only a coherent x-ray source, sample, and detector, our propagation-based algorithm can map the sample density and dark-field/diffusion properties of the sample in 3D. Importantly, incorporating dark-field information in the density reconstruction process enables a higher spatial resolution reconstruction than possible with previous propagation-based approaches. Two sample exposures at each projection angle are sufficient for the successful reconstruction of both the sample density and dark-field Fokker-Planck diffusion coefficients. We anticipate that the proposed algorithm may be of benefit in biomedical imaging and industrial settings.

Abstract:Tomography is the three-dimensional reconstruction of an object from images taken at different angles. The term classical tomography is used, when the imaging beam travels in straight lines through the object. This assumption is valid for light with short wavelengths, for example in x-ray tomography. For classical tomography, a commonly used reconstruction method is the filtered back-projection algorithm which yields fast and stable object reconstructions. In the context of single-cell imaging, the back-projection algorithm has been used to investigate the cell structure or to quantify the refractive index distribution within single cells using light from the visible spectrum. Nevertheless, these approaches, commonly summarized as optical projection tomography, do not take into account diffraction. Diffraction tomography with the Rytov approximation resolves this issue. The explicit incorporation of the wave nature of light results in an enhanced reconstruction of the object's refractive index distribution. Here, we present a full literature review of diffraction tomography. We derive the theory starting from the wave equation and discuss its validity with the focus on applications for refractive index tomography. Furthermore, we derive the back-propagation algorithm, the diffraction-tomographic pendant to the back-projection algorithm, and describe its implementation in three dimensions. Finally, we showcase the application of the back-propagation algorithm to computer-generated scattering data. This review unifies the different notations in literature and gives a detailed description of the back-propagation algorithm, serving as a reliable basis for future work in the field of diffraction tomography.

Fast three-dimensional phase retrieval in propagation-based x-ray tomography

Abstract:The following paper describes a method for three-dimensional (3D) reconstruction of multi-material objects based on propagation-based X-ray phase-contrast tomography (PB-CT) with phase retrieval using the homogenous form of the Transport-of-Intensity equation (TIE-Hom). Unlike conventional PB-CT algorithms that perform phase retrieval of individual projections, the described post-reconstruction phase-retrieval method is applied in 3D to a localised region of the CT-reconstructed volume. We demonstrate via numerical simulations the accuracy and noise characteristics of the method under a variety of experimental conditions, comparing it to both conventional absorption tomography and two-dimensional (2D) TIE-Hom phase retrieval applied to projection images. The results indicate that the 3D post-reconstruction method generally achieves a modest improvement in noise suppression over existing PB-CT methods. It is also shown that potentially large computational gains over projection-based phase retrieval for multi-material samples are possible. In particular, constraining phase retrieval to a localised 3D region of interest reduces the overall computational cost and eliminates the need for multiple CT reconstructions and global 2D phase retrieval operations for each material within the sample.

Abstract:A new approach for image reconstruction in THz computed tomography (THz-CT) is presented. Based on a geometrical optics model containing the THz signal amplitude and phase, a novel algorithm for extracting an average phase from the measured THz signals is derived. Applying the algorithm results in a phase-contrast sinogram, which is further used for image reconstruction. For experimental validation, a fast THz time-domain spectrometer (THz-TDS) in transmission geometry is employed, enabling CT measurements within several minutes. Quantitative evaluation of reconstructed 3D printed plastic profiles reveals the potential of our approach in non-destructive testing of plastic profiles.

Abstract:We consider the problem of elastic diffraction tomography, which consists in reconstructing elastic properties (i.e. mass density and elastic Lamé parameters) of a weakly scattering medium from full-field data of scattered waves outside the medium. Elastic diffraction tomography refers to the elastic inverse scattering problem after linearization using a firstorder Born approximation. In this paper, we prove the Fourier diffraction theorem, which relates the 2D Fourier transform of scattered waves with the Fourier transform of the scatterer in the 3D spatial Fourier domain. Elastic wave mode separation is performed, which decomposes a wave into four modes. A new two-step inversion process is developed, providing information on the modes first and secondly on the elastic parameters. Finally, we discuss reconstructions with plane wave excitation experiments for different tomographic setups and with different plane wave excitation frequencies, respectively.

Abstract:We demonstrate a motion-free intensity diffraction tomography technique that enables the direct inversion of 3D phase and absorption from intensity-only measurements for weakly scattering samples. We derive a novel linear forward model featuring slice-wise phase and absorption transfer functions using angled illumination. This new framework facilitates flexible and efficient data acquisition, enabling arbitrary sampling of the illumination angles. The reconstruction algorithm performs 3D synthetic aperture using a robust computation and memory efficient slice-wise deconvolution to achieve resolution up to the incoherent limit. We demonstrate our technique with thick biological samples having both sparse 3D structures and dense cell clusters. We further investigate the limitation of our technique when imaging strongly scattering samples. Imaging performance and the influence of multiple scattering is evaluated using a 3D sample consisting of stacked phase and absorption resolution targets. This computational microscopy system is directly built on a standard commercial microscope with a simple LED array source add-on, and promises broad applications by leveraging the ubiquitous microscopy platforms with minimal hardware modifications.

Abstract:Diffraction tomography is a promising label-free three-dimensional (3D) imaging method for transparent samples due to its low phototoxicity. However, it usually requires coherent illuminations and massive measurements, which will, respectively, introduce speckle noises and time-consuming acquisitions, and thus, limit its applications in multicellular dynamic organisms. To address these problems, here we propose a partially coherent and snapshot diffraction tomography method based on light-field microscopy. By incorporating the illumination optical transfer function into the volumetric differential phase contrast model, we obtain a 3D phase stack from the snapshot four-dimensional phase-space measurements. Then a 3D phase-point-spread function is used for 3D deconvolution to achieve computational optical sectioning and enhance the resolution, and finally, we extract 3D refractive-index information from the 3D phase stack. Experiments on freely moving Caenorhabditis elegans are shown accordingly, demonstrating the great potential of the proposed method for label-free 3D imaging of multicellular organisms in vivo at high speed up to the camera frame rate.

Abstract:Diffraction tomography (DT) is an inversion technique that reconstructs the refractive index distribution of a scattering object. We previously demonstrated that by exploiting the redundant information in the DT data, the scattering object could be exactly reconstructed using measurements taken over the angular range [0, phimin], where pi < phimin < or = 3pi/2. In this paper, we reveal a relationship between the maximum scanning angle and image resolution when a filtered backpropagation (FBPP) reconstruction algorithm is employed for image reconstruction. Based on this observation, we develop short-scan FBPP algorithms that reconstruct a low-pass filtered scattering object from measurements acquired over the angular range [0, phi(c)], where phi(c) < phimin.
Unified Reconstruction Theory for Diffraction Tomography, with Consideration of Noise Control

Abstract:In diffraction tomography, the spatial distribution of the scattering object is reconstructed from the measured scattered data. For a scattering object that is illuminated with plane-wave radiation, under the condition of weak scattering one can invoke the Born (or the Rytov) approximation to linearize the equation for the scattered field (or the scattered phase) and derive a relationship between the scattered field (or the scattered phase) and the distribution of the scattering object. Reconstruction methods such as the Fourier domain interpolation methods and the filtered backpropagation method have been developed previously. However, the underlying relationship among and the noise properties of these methods are not evident. We introduce the concepts of ideal and modified sinograms. Analysis of the relationships between, and the noise properties of the two sinograms reveals infinite classes of methods for image reconstruction in diffraction tomography that include the previously proposed methods as special members. The methods in these classes are mathematically identical, but they respond to noise and numerical errors differently.

Abstract:Phase is not easy to detect directly as intensity, but sometimes it contains the really desired information. The transport-of-intensity equation (TIE) is a powerful tool to retrieve the phase from the intensity. However, by considering the boundary energy exchange and the whole energy conversation in the field of view, the current popular Fast Fourier transform (FFT) based TIE solver can only retrieve the phase under homogeneous Neumann boundary condition. For many applications, the boundary condition could be more complex and general. A novel TIE phase retrieval method is proposed to deal with an optical field under a general boundary condition. In this method, an arbitrarily-shape hard aperture is added in the optical field. In our method, the TIE is solved by using iterative discrete cosine transforms (DCT) method, which contains a phase compensation mechanism to improve the retrieval results. The proposed method is verified in simulation with an arbitrary phase, an arbitrarily-shaped aperture, and non-uniform intensity distribution. Experiment is also carried out to check its feasibility and the method proposed in this work is very easy and straightforward to use in a practical measurement as a flexible phase retrieval tool.

Abstract:A new reconstruction algorithm in a finite form based on the Rytov transform is presented for acoustical diffraction tomography. Applying the Rytov transform to the governing differential wave equation necessarily introduces the so-called generalized scattering. Our analysis shows that the generalized scattered wave is asymptotically equivalent to the physically scattered wave, and also satisfies the Sommerfeld radiation condition in the far field. Using the method of formal parameter expansion, we further find that all other terms in the expansion of the object function vanish except the first- and second-order ones, and thus reach a finite form solution to the diffraction tomography. Our computer simulation confirms the effectiveness of the algorithm in the case of the scattering objects with cylindrical symmetry, also shows its limitations when it applies to the strong scattering.

Abstract:Tomographic diffraction microscopy is a three-dimensional quantitative optical imaging technique in which the sample is numerically reconstructed from tens of holograms recorded under different angles of incidence. We show that combining the measurement of the amplitude, the phase, and the polarization of the field scattered by the sample with an approximate knowledge of the sample permittivity allows reconstruction of spatially complex samples up to 50 nm resolution. This technique should be particularly useful for imaging objects made of known materials. (C) 2016 Optical Society of America

Abstract:While the implementation of single particle coherent diffraction imaging for non-crystalline particles is complicated by current limitations in photon flux, hit rate, and sample delivery a concept of many-particle coherent diffraction imaging offers an alternative way to overcome these difficulties. Here we present a direct, non-iterative approach for the recovery of the diffraction pattern corresponding to a single particle using coherent x-ray data collected from a two-dimensional (2D) disordered system of identical particles, that does not require a priori information about the particles and can be applied to a general case of particles without symmetry. The reconstructed single particle diffraction pattern can be directly used in common iterative phase retrieval algorithms to recover the structure of the particle.

Abstract:Conventional three-dimensional (3D) imaging methods require multiple measurements of the sample in different orientation or scanning. When the sample is probed with coherent waves, a single two-dimensional (2D) intensity measurement is sufficient as it contains all the information of the 3D sample distribution. We show a method that allows reconstruction of 3D sample distribution from a single 2D intensity measurement, at the z-resolution exceeding the classical limit. The method can be practical for radiation-sensitive materials, or where the experimental setup allows only one intensity measurement.

Abstract:The structural complexity and instability of many interference phase microscopy methods are the major obstacles toward high-precision phase measurement. In this vein, improving more efficient configurations as well as proposing methods are the subjects of growing interest. Here, we introduce Fresnel diffraction from a phase step to the realm of quantitative phase imaging. By employing Fresnel diffraction of a divergent (or convergent) beam of light from a plane-parallel phase plate, we provide a viable, simple, and compact platform for three-dimensional imaging of micrometer-sized specimens. The recorded diffraction pattern of the outgoing light from an imaging system in the vicinity of the plate edge can be served as a hologram, which would be analyzed via the Fourier transform method to measure the sample phase information. The period of diffraction fringes is adjustable simply by rotating the plate without the reduction of both the field of view and fringe contrast. The high stability of the presented method is affirmatively confirmed through comparison of the result with that of the conventional Mach–Zehnder based digital holographic method. Quantitative phase measurements on silica microspheres, onion skins, and red blood cells ensure the validity of the method and its ability for monitoring nanometer-scale fluctuations of living cells, particularly in real-time.
