Abstract:The intrinsic optical parameters-absorption coefficient mua, scattering coefficient mus, anisotropy factor g, and effective scattering coefficient mus'--are determined for human red blood cells of hematocrit 42.1% dependent on the shear rate in the wavelength range 250 to 1100 nm. Integrating sphere measurements of light transmittance and reflectance in combination with inverse Monte-Carlo simulation are carried out for different wall shear rates between 0 and 1000 s(-1). Randomly oriented cells show maximal mua, mus, and mus' values. Cell alignment and elongation, as well as the Fahraeus effect at increasing shear rates, lead to an asymptotical decrease of these values. The anisotropy factor shows this behavior only below 600 nm, dependent on absorption; above 600 nm, g is almost independent of shear rate. The decrease of mus' is inversely correlated with the hemoglobin absorption. Compared to randomly oriented cells, aggregation reduces all parameters by a different degree, depending on the hemoglobin absorption. It is possible to evaluate the influence of collective scattering phenomena, the absorption within the cell, and the cell shape.
Modeling of Light Scattering by Biconcave and Deformed Red Blood Cells with the Invariant Imbedding T-matrix Method

Abstract:The invariant imbedding T-matrix method (II-TM) is employed to simulate the optical properties of normal biconcave and deformed red blood cells (RBCs). The phase matrix elements of a RBC model computed with the II-TM are compared with their counterparts computed with the discrete-dipole approximation (DDA) method. As expected, the DDA results approach the II-TM results with an increase in the number of dipoles per incident wavelength. Computationally, the II-TM is faster than the DDA when multiple RBC orientations are considered. For a single orientation, the DDA is comparable with or even faster than the II-TM because the DDA efficiently converges for optically soft particles; however, the DDA method demands significantly more computer memory than the II-TM. After the applicability of the II-TM is numerically confirmed, a comparison is conducted of the optical properties of oxygenated and deoxygenated RBCs and of normal and deformed RBCs. The spectral variations of RBCs' optical properties are investigated in the wavelength range from 0.25 to 1.0 μm. Furthermore, the statistically averaged phase matrix of spheres and biconcave RBCs are compared. Conducted numerical simulations suggest the applicability of the II-TM for the inverse light scattering analysis and radiative transfer simulations in blood.
Optical Properties of Circulating Human Blood in the Wavelength Range 400-2500 nm

Abstract:Knowledge about the optical properties μa,μs, and g of human blood plays an important role for many diagnostic and therapeutic applications in laser medicine and medical diagnostics. They strongly depend on physiological parameters such as oxygen saturation, osmolarity, flow conditions, haematocrit, etc. The integrating sphere technique and inverse Monte Carlo simulations were applied to measure μa,μs, and g of circulating human blood. At 633 nm the optical properties of human blood with a haematocrit of 10% and an oxygen saturation of 98% were found to be 0.210±0.002 mm-1 for μa,77.3±0.5 mm-1 for μs, and 0.994±0.001 for the g factor. An increase of the haematocrit up to 50% lead to a linear increase of absorption and reduced scattering. Variations in osmolarity and wall shear rate led to changes of all three parameters while variations in the oxygen saturation only led to a significant change of the absorption coefficient. A spectrum of all three parameters was measured in the wavelength range 400-2500 nm for oxygenated and deoxygenated blood, showing that blood absorption followed the absorption behavior of haemoglobin and water. The scattering coefficient decreased for wavelengths above 500 nm with approximately λ-1.7; the g factor was higher than 0.9 over the whole wavelength range. © 1999 Society of Photo-Optical Instrumentation Engineers.
Influence of oxygen saturation on the optical scattering properties of human red blood cells in the spectral range 250 to 2,000 nm

Abstract:The intrinsic optical parameters absorption coefficient mu(a), scattering coefficient micros, anisotropy factor g, and effective scattering coefficient micros were determined for human red blood cell (RBC) suspensions of hematocrit 33.2% dependent on the oxygen saturation (SAT O(2)) in the wavelength range 250 to 2,000 nm, including the range above 1,100 nm, about which there are no data available in the literature. Integrating sphere measurements of light transmittance and reflectance in combination with inverse Monte Carlo simulation were carried out for SAT O(2) levels of 100 and 0%. In the wavelength range up to 1,200 nm, the absorption behavior is determined by the hemoglobin absorption. The spectral range above the cells' absorption shows no dependence on SAT O(2) and approximates the absorption of water with values 20 to 30% below the respective values for water. Parameters micros and g are significantly influenced by the SAT O(2)-induced absorption changes. Above 600 nm, micros decreases continuously from values of 85 mm(-1) to values of 30 mm(-1) at 2,000 nm. The anisotropy factor shows a slight decrease with wavelengths above 600 nm. In the spectral regions of 1,450 and 1,900 nm where water has local absorption maxima, g shows a significant decrease down to 0.85, whereas micros increases.

Abstract:An estimation error of the scattering coefficient of hemoglobin in the high absorption wavelength range has been observed in optical property calculations of blood-rich tissues. In this study, the relationship between the accuracy of diffuse reflectance measurement in the integrating sphere and calculated scattering coefficient was evaluated with a system to calculate optical properties combined with an integrating sphere setup and the inverse Monte Carlo simulation. Diffuse reflectance was measured with the integrating sphere using a small incident port diameter and optical properties were calculated. As a result, the estimation error of the scattering coefficient was improved by accurate measurement of diffuse reflectance. In the high absorption wavelength range, the accuracy of diffuse reflectance measurement has an effect on the calculated scattering coefficient.

Abstract:The real part of the refractive index (RI) of aqueous solutions of human hemoglobin is computed from their absorption spectra in the wavelength range $250\,{\rm nm} - 1100\,{\rm nm}$ using the Kramers-Kronig (KK) relations and the corresponding uncertainty analysis is provided. The strong ultraviolet (UV) and infrared absorbance of the water outside this spectral range were taken into account in a previous study employing KK relations. We improve these results by including the concentration dependence of the water absorbance as well as by modeling the deep UV absorbance of hemoglobin's peptide backbone. The two free parameters of the model for the deep UV absorbance are fixed by a global fit.

Abstract:Tissue vasculature is altered when cancer develops. Consequently, noninvasive methods of monitoring blood vessel size, density, and oxygenation would be valuable. Simple spectroscopy employing fiber optic probes to measure backscattering can potentially determine hemoglobin parameters. However, heterogeneity of blood distribution, the dependence of the tissue-volume-sampled on scattering and absorption, and the potential compression of tissue all hinder the accurate determination of hemoglobin parameters. We address each of these issues. A simple derivation of a correction factor for the absorption coefficient, μa, is presented. This correction factor depends not only on the vessel size, as others have shown, but also on the density of blood vessels. Monte Carlo simulations were used to determine the dependence of an effective pathlength of light through tissue which is parameterized as a ninth-order polynomial function of μa. The hemoglobin bands of backscattering spectra of cervical tissue are fit using these expressions to obtain effective blood vessel size and density, tissue hemoglobin concentration, and oxygenation. Hemoglobin concentration and vessel density were found to depend on the pressure applied during in vivo acquisition of the spectra. It is also shown that determined vessel size depends on the blood hemoglobin concentration used.
Determination of multiple optical parameters of red blood cell suspension

Abstract:Objective To develop a measurement method for determination of optical parameters of a red blood cell ( RBC) suspension based on the measurement of spatial scattered light signals without using an integrating sphere.Methods Multiple independent photoelectric sensors and light intensity modulation were used to obtain the measured values of diffuse reflectance,diffuse transmittance and collimated transmittance.The measured data results were imported into a Monte Carlo simulation based RTE to inversely determine the absorption coefficient, scattering coefficient and anisotropy factor of the measured sample using a new perturbation method.Results The described measurement method was applied to determine the optical parameters of a polystyrene microsphere suspension with a mean diameter of 2.6μm,and the results were essen-tially consistent with the calculated optical parameters by Mie code.Then, the RBC suspension was used for testing optical parameters,and the results were basically consistent with the parameters in the literature.Conclusion The system based on the measurement of spatial scattered light signals without using an integrating sphere will provide a quick and accurate approach for quantitative analysis of free hemoglobins and RBC suspensions.

Abstract:Numerical simulations of light scattering by a biconcave shaped human red blood cell (RBC) are carried out using the finite-difference time-domain (FDTD) method. A previously developed FDTD code for the study of light scattering by ice crystals is modified for the current purpose and it is validated against Mie theory using a spherically shaped RBC. Numerical results for the angular distributions of the Mueller scattering matrix elements of an RBC and their dependence on shape, orientation, and wavelength are presented. Also calculated are the scattering and absorption efficiencies. The implication of these results on the possibility of probing RBC shape changes is discussed.

Abstract:Light scattering is a well-established experimental technique, which gains more and more popularity in the biological field because it offers the means for non-invasive imaging and detection. However, the interpretation of light-scattering signals remains challenging due to the complexity of most biological systems. Here, we investigate static and dynamic scattering properties of red blood cells (RBCs) using two mesoscopic hydrodynamics simulation methods-multi-particle collision dynamics and dissipative particle dynamics. Light scattering is studied for various membrane shear elasticities, bending rigidities, and RBC shapes (e.g., biconcave and stomatocyte). Simulation results from the two simulation methods show good agreement, and demonstrate that the static light scattering of a diffusing RBC is not very sensitive to the changes in membrane properties and moderate alterations in cell shapes. We also compute dynamic light scattering of a diffusing RBC, from which dynamic properties of RBCs such as diffusion coefficients can be accessed. In contrast to static light scattering, the dynamic measurements can be employed to differentiate between the biconcave and stomatocytic RBC shapes and generally allow the differentiation based on the membrane properties. Our simulation results can be used for better understanding of light scattering by RBCs and the development of new non-invasive methods for blood-flow monitoring.

Refractive index of solutions of human hemoglobin from the near-infrared to the ultraviolet range: Kramers-Kronig analysis

Abstract:Because direct measurements of the refractive index of hemoglobin over a large wavelength range are challenging, indirect methods deserve particular attention. Among them, the Kramers-Kronig relations are a powerful tool often used to derive the real part of a refractive index from its imaginary part. However, previous attempts to apply the relations to solutions of human hemoglobin have been somewhat controversial, resulting in disagreement between several studies. We show that this controversy can be resolved when careful attention is paid not only to the absorption of hemoglobin but also to the dispersion of the refractive index of the nonabsorbing solvent. We present a Kramers-Kroning analysis taking both contributions into account and compare the results with the data from several studies. Good agreement with experiments is found across the visible and parts of near-infrared and ultraviolet regions. These results reinstate the use of the Kramers-Kronig relations for hemoglobin solutions and provide an additional source of information about their refractive index.

Abstract:Near-infrared technology is an emerging non-invasive technique utilized for various medical applications. Recently, there have been many attempts to utilize NIR technology for the continues monitoring of blood glucose levels through the skin. Different approaches and designs have been proposed for non-invasive blood glucose measurements. Light photons penetrating the skin can undergo multiple scattering events, and the actual optical pathlength becomes larger than the source-to-detector separation (optode spacing) in the reflection-mode configuration. Thus, the differential pathlength factor (DPF) must be incorporated into the modified Beer-Lambert law. The accurate estimation of the DPF values will lead to an accurate quantification of the physiological variations within the tissue. In this work, the aim was to systematically estimate the DPF for human skin for a range of source-to-detector separations and wavelengths. The Monte Carlo (MC) method was utilized to mimic the different layers of human skin with different optical properties and blood and water volume fractions. This work could help improve the accuracy of the near-infrared technique in the measurement of physiological variations within skin tissue.
Broadband dielectric spectroscopy on human blood

Abstract:BACKGROUND: Dielectric spectra of human blood reveal a rich variety of dynamic processes. Achieving a better characterization and understanding of these processes not only is of academic interest but also of high relevance for medical applications as, e.g., the determination of absorption rates of electromagnetic radiation by the human body.METHODS: The dielectric properties of human blood are studied using broadband dielectric spectroscopy, systematically investigating the dependence on temperature and hematocrit value. By covering a frequency range from 1Hz to 40GHz, information on all the typical dispersion regions of biological matter is obtained.RESULTS AND CONCLUSIONS: We find no evidence for a low-frequency relaxation ("α-relaxation") caused, e.g., by counterion diffusion effects as reported for some types of biological matter. The analysis of a strong Maxwell-Wagner relaxation arising from the polarization of the cell membranes in the 1-100MHz region ("β-relaxation") allows for the test of model predictions and the determination of various intrinsic cell properties. In the microwave region beyond 1GHz, the reorientational motion of water molecules in the blood plasma leads to another relaxation feature ("γ-relaxation"). Between β- and γ-relaxations, significant dispersion is observed, which, however, can be explained by a superposition of these relaxation processes and is not due to an additional "δ-relaxation" often found in biological matter.GENERAL SIGNIFICANCE: Our measurements provide dielectric data on human blood of so far unsurpassed precision for a broad parameter range. All data are provided in electronic form to serve as basis for the calculation of the absorption rate of electromagnetic radiation and other medical purposes. Moreover, by investigating an exceptionally broad frequency range, valuable new information on the dynamic processes in blood is obtained.
Effects of Optical Variables in a Single Integrating Sphere System on Estimation of Scattering Properties of Turbid Media

Abstract:This research was aimed at determining the effect of optical variables for a single integrating sphere system, coupled with the inverse adding-doubling method, on estimating the reduced scattering coefficient (mu s') of turbid media. Integrating sphere measurements were performed on a total of 36 liquid samples and two solid samples. The results demonstrated that the system had good accuracy, with average errors for estimating absorption coefficient mu(a) (pure absorption) and mu s' (pure scattering) of liquid samples being 15.0% and 4.7% over 550-900 nm, compared with the collimated transmittance and empirical equation, respectively. The system also had good reproducibility for mu(a) and mu s', with the coefficients of variation lower than 4%. Sensitivity analysis revealed that the detectable values of mu(a) and mu s' were around 0.02 cm(-1)- and 4.5 cm(-1). Entrance and detector port diameters, standard reflectivity and anisotropy factor had a negligible effect on estimating mu s' (<2.2%). However, refractive indices of sample and cuvette, sphere wall reflectivity, beam diameter and its position, and specular reflectance had a relatively large effect on the estimation of mu s' (2.8%-35.3%). Hence these variables should be carefully chosen or accurately measured before optical property measurements are carried out. Application examples of the mu s' estimation for apple skin and flesh further validated the system performance, indicating that appropriate selection of optical variables could improve the estimation of mu s'. Theoretical model for optical oximetry at the capillary level: exploring hemoglobin oxygen saturation through backscattering of single red blood cells

Abstract:Oxygen saturation ( sO 2 ) of red blood cells (RBCs) in capillaries can indirectly assess local tissue oxygenation and metabolic function. For example, the altered retinal oxygenation in diabetic retinopathy and local hypoxia during tumor development in cancer are reflected by abnormal sO 2 of local capillary networks. However, it is far from clear whether accurate label-free optical oximetry (i.e., measuring hemoglobin sO 2 ) is feasible from dispersed RBCs at the single capillary level. The sO 2 -dependent hemoglobin absorption contrast present in optical scattering signal is complicated by geometry-dependent scattering from RBCs. We present a numerical study of backscattering spectra from single RBCs based on the first-order Born approximation, considering practical factors: RBC orientations, size variation, and deformations. We show that the oscillatory spectral behavior of RBC geometries is smoothed by variations in cell size and orientation, resulting in clear sO 2 -dependent spectral contrast. In addition, this spectral contrast persists with different mean cellular hemoglobin content and different deformations of RBCs. This study shows for the first time the feasibility of, and provides a theoretical model for, label-free optical oximetry at the single capillary level using backscattering-based imaging modalities, challenging the popular view that such measurements are impossible at the single capillary level.

Abstract:Non-invasive methods for determining blood hemoglobin (Hb) concentration are urgently needed to avoid the painful and time-consuming process of invasive venous blood sampling. Many such methods rely on assessing the average attenuation of light over a tissue area where hemoglobin is the dominant chromophore, without separating those areas corresponding to vessels and bloodless tissue. In this study, we investigate whether it is possible to determine hemoglobin levels in the blood by assessing the changes in light intensity when passing through large vessels in comparison to adjacent tissues, using this as a Hb level predictor. Using Monte Carlo light transport modeling, we evaluate the accuracy of determining hemoglobin levels via light intensity contrast and vessel widths estimated in the transmittance illumination geometry and estimate the influence of physiologically significant parameters such as vessel depth, dermis vascularization, and melanin content in the epidermis on the blood Hb prediction error. The results show that physiological variations in tissue parameters limit the mean absolute error of this method to ~15 g/L for blood Hb levels varying in the 60–160 g/L range, which finding is also supported by experimental data obtained for volunteers with different total blood Hb levels that have been determined invasively. We believe the application of new approaches to the non-invasive assessment of Hb levels will lead to the creation of reliable and accurate devices that are applicable in point-of-care and clinical practice.

This study presents Monte Carlo calculated $\boldsymbol{\mu} /\boldsymbol{\rho}$, ${\boldsymbol{\mu}}_{\boldsymbol{en}}/\boldsymbol{\rho}$ and ${\boldsymbol{\mu}}_{\boldsymbol{tr}}/\boldsymbol{\rho}$ values of some common tissues and organs found in the human body (namely, adipose tissue, blood, bone-cortical, brain-grey/white matter, breast tissue, eye lens, lung tissue, muscle-skeletal, ovary, soft tissue and testes) as well as water for comparison purposes. The simulation model involves a monoenergetic point source producing a pencil beam where, depending on the parameter under study, particle flux, energy flux or absorbed dose from photon interactions are scored in the range of 10 keV to 20 MeV energy. The simulations were performed using the Monte Carlo package MCNP6.1 and provided $\boldsymbol{\mu} /\boldsymbol{\rho}$, ${\boldsymbol{\mu}}_{\boldsymbol{en}}/\boldsymbol{\rho}$ and ${\boldsymbol{\mu}}_{\boldsymbol{tr}}/\boldsymbol{\rho}$ values. The data produced in this study were compared with theoretical photon attenuation data from the XMUDAT database and demonstrated good agreement. The results, which are based on a simple model geometry and pure elemental compositions, indicate that this approach can be applied to evaluate $\boldsymbol{\mu} /\boldsymbol{\rho}$, ${\boldsymbol{\mu}}_{\boldsymbol{en}}/\boldsymbol{\rho}$ and ${\boldsymbol{\mu}}_{\boldsymbol{tr}}/\boldsymbol{\rho}$ in a broad energy range for any element, compound or mixture.

Abstract:Severe intravascular hemolysis leads to the simultaneous presence of free heme pigments (oxyhemoglobin, methemoglobin, and methemalbumin) and bilirubin in human plasma. Standard spectrophotometric methods used to assess in vivo hemolysis inadequately address this complex analytical situation. Thus, we propose a novel quantification algorithm to ensure the highest analytical specificity. A corresponding second‐derivative fitting algorithm was validated according to the guideline of bioanalytical method validation from the European Medicines Agency using plasma specimens (n=1759) spiked with different concentrations of oxyhemoglobin and methemoglobin. The results were compared to standard spectrophotometric quantification methods described by Harboe, Noe, and Fairbanks. Based on the second‐derivative method, simultaneous quantification of oxyhemoglobin and methemoglobin / methemalbumin in samples with total bilirubin concentrations ≤ 4.9 mg/dL (83.8 μmol/L) provided robust results (inaccuracy ≤ 20 %, imprecision ≤ 16 %). Analyzing UV/VIS spectra of plasma from patients with confirmed severe intravascular hemolysis evidenced an underestimation of up to 33 % for the combined free heme pigment content. The employed second‐derivative algorithm allows for automated and highly specific quantification of the free heme pigment content in diluted human plasma, which cannot be realized with standard spectrophotometric evaluation methods.
