Abstract:Previous studies have shown that the application of soil amendments is efficient in reducing acidity and heavy metal bioavailability in mine soils. However, it remains a challenge for environmentalists to predict accurately and control economically the re-acidification in re-vegetated mine soils. In this study, net acid generation (NAG) test and bioassay technique were employed to assess the effectiveness of the amendments [including lime, N-P-K (nitrogen, phosphorous and potassium) fertilizer, phosphate and river sediment] on re-acidification and heavy metal immobilization in an extremely acid (pH < 3) mine soil. Our results suggested that NAG test was a rapid and accurate approach to assess the effectiveness of the amendments on re-acidification potential of the mine soil. Interestingly, it was found that phosphate and river sediment played quite specific roles in preventing the re-acidification in the mine soil. In addition, the results also indicated that the addition of 25 t ha(-1) lime combined with river sediment (30%) might be an economical method to successfully control the acidification and re-acidification in the extremely acid mine soil, allowing the re-establishment of the plants. Collectively, our results implied that the combined use of NAG test and bioassay assessment was effective in evaluating a reclamation strategy for extremely acidic mine soils.
Effect of pH on anoxic sulfide oxidizing reactor performance.

Abstract:The effects of pH on the performance of anoxic sulfide oxidizing (ASO) reactor were evaluated. Performance was investigated under various operational conditions at influent pH range of 4–11. At the influent pH of 7–7.5 during loading tests and HRT tests, the sulfide oxidation was partial. In general, the amount of sulfate formed decreased with the increasing sulfide and nitrite loadings. The bacterial communities in ASO reactors were more sensitive to acidic pH compared with alkaline pH, as nitrite and sulfide removal rates dropped significantly when exposed to acidic pH 3. High dissolved bisulfide ions, nitrite and excess of sulfate (>300mg/L) might have inhibited the sulfide oxidation under highly acidic and alkaline conditions in the ASO reactor. Based on sulfide and nitrite removal efficiencies, the ASO reactor can be operated in a wide range of pH, i.e. 5–11.

Abstract:Acid mine drainage (AMD) is a major environmental challenge for the mining industry in northern climates. Laboratory-scale experiments were conducted to test various simple and complex carbon sources available in the Yukon as electron donors for sulfate reduction to allow the subsequent removal of Cd and Zn from a Yukon AMD. The 1 L capacity bioreactors were monitored biweekly at 5 °C. After 162 days, a diminution of both total organic carbon and sulfate concentrations was observed in all bioreactors. A long adaptation period was necessary before consumption of the carbon source started, which might be due to the cold temperature. Simple organic sources of carbon (methanol and ethylene glycol) and complex organic sources of carbon (potato oil, brewery residue, peat, and straw) were used to support SRB growth. Methanol and ethylene glycol led to a diminution of sulfate concentrations of 71.2 and 36.9%, respectively, while the decrease of sulfate concentrations was limited to 13.8 and 5.3% when using peat and straw, respectively.

Iron Kinetics and Evolution of Microbial Populations in Low-pH, Ferrous Iron-Oxidizing Bioreactors

Abstract:Iron-rich, acidic wastewaters are commonplace pollutants associated with metal and coal mining. Continuous-flow bioreactors were commissioned and tested for their capacities to oxidize ferrous iron in synthetic and actual acid mine drainage waters using (initially) pure cultures of the recently described acidophilic, iron-oxidizing heterotrophic bacterium Acidithrix ferrooxidans grown in the presence of glucose and yeast extract. The bioreactors became rapidly colonized by this bacterium, which formed macroscopic streamer growths in the flowing waters. Over 97% of ferrous iron in pH 2.0-2.2 synthetic mine water was oxidized (at up to 225 mg L(-1) h(-1)) at dilution rates (D) of 0.6 h(-1). Rates of iron oxidation decreased with pH but were still significant, with influent liquors as low as pH 1.37. When fed with actual mine water, >90% of ferrous iron was oxidized at D values of 0.4 h(-1), and microbial communities within the bioreactors changed over time, with Atx. ferrooxidans becoming increasingly displaced by the autotrophic iron-oxidizing acidophiles Ferrovum myxofaciens, Acidithiobacillus ferrivorans, and Leptospirillum ferrooxidans (which were all indigenous to the mine water), although this did not have a negative impact on net ferrous-iron oxidation. The results confirmed the potential of using a heterotrophic acidophile to facilitate the rapid commissioning of iron-oxidizing bioreactors and illustrated how microbial communities within them can evolve without compromising the performances of the bioreactors.
Removal of Heavy Metals in Sewage Sludge and Losses of Nutrition During Bioleaching

Abstract:Nutrition(TN,TP,TK and organic materials) loss is one of problems in bioleaching process of heavy metals from sewage sludge,because of the acid environment.The removal of Zn and Cu from sludge from the Sibao wastewater treatment plant in Hangzhou and losses of nutrition after bioleaching were studied to supply reference to application of the bioleaching for heavy metal control.Two 10%(V/V) different inoculants,iron-oxidizing bacteria Thiobacillus ferrooxidans(T.fe) and sulfur-oxidizing bacteria Thiobacillus thiooxidans(T.th.),were used in the batch experiment.The results demonstrated that T.fe could faster start bioleaching.The removal rate of Zn reached 97.8% after 2 d and kept the high removal rate until the end of the experiment.The efficiency of Cu removing reached 37% at the 4th day and increased persistently.Both of bioleaching rates of Zn and Cu by T.th had high level after 6 d.The bioleaching of Zn and Cu in sludge were affected by pH.For Zn,high removal rates occurred at pH 3.0~4.0,while better Cu bioleaching rate occurred at pH<3.0.When a large number of heavy metals were removed from sludge,the losses of nutrition(TN,TP,TK and organic materials) concurred.Especially,the loss of TP reached 70%.The losses of nutrition in the system with T.fe were more than these in the system with T.th.
Effects of Loading Rate and Hydraulic Residence Time on Anoxic Sulfide Biooxidation

Abstract:The optimal operation conditions in an anoxic sulfide oxidizing (ASO) bioreactor were investigated. The maximal removal rates for sulfide and nitrate were found to be 4.18 kg/(m 3 ·d) and 1.73 kg/(m 3 ·d), respectively. The volumetrical volumetri loading rates (LRs) observed through decreasing hydraulic retention time (HRT) at fixed substrate concentration are higher than those by increasing substrate concentration at fixed HRT. The sulfide oxidation in ASO reactor was partially producing both sulfate and sulfur; but the amount of sulfate produced was approximately one third that of sulfur. The process was able to tolerate high sulfide concentration, as the sulfide removal percentage always remained near 99% when influent concentration was up to 580 mg/L. It tolerated relatively lower nitrate concentration because the removal percentage dropped to 85% when influent concentration was increased above 110 mg/L. The process can tolerate shorter HRT but careful operation is needed. Nitrate conversion was more sensitive to HRT than sulfide conversion since the process performance deteriorated abruptly when HRT was decreased from 3.12 h to 2.88 h. In order to avoid nitrite accumulation in the reactor, the influent sulfide and nitrate concentrations should be kept at 280 mg/L and 67.5 mg/L respectively. Present biotechnology is useful for removing sulfides from sewers and crude oil.

Abstract:Treatment of acid mine drainage (AMD) highly rich in sulfate and multiple metal elements has been investigated in a continuous flow column experiment using organic and inorganic reactive media. Treatment substrates that composed of spent mushroom compost (SMC), limestone, activated sludge and woodchips were incorporated into bacterial sulfate reduction (BSR) treatment for AMD. SMC greatly assisted the removals of sulfate and metals and acted as essential carbon source for sulfate-reducing bacteria (SRB). Alkalinity produced by dissolution of limestone and metabolism of SRB has provided acidity neutralization capacity for AMD where pH was maintained at neutral state, thus aiding the removal of sulfate. Fe, Pb, Cu, Zn and Al were effectively removed (87–100%); however, Mn was not successfully removed despite initial Mn reduction during early phase due to interference with Fe. The first half of the treatment was an essential phase for removal of most metals where contaminants were primarily removed by the BSR in addition to carbonate dissolution function. The importance of BSR in the presence of organic materials was also supported by metal fraction analysis that primary metal accumulation occurs mainly through metal adsorption onto the organic matter, e.g., as sulfides and onto Fe/Mn oxides surfaces.

Sulfidogenic fluidized-bed bioreactor kinetics for co-treatment of hospital wastewater and acid mine drainage

Abstract:A passive co-treatment of acid mine drainage and hospital wastewater previously demonstrated a promising bioremediation viable approach for both toxic streams. The study of inhibition kinetics and microbial communities is essential to understand better the diverse species and the reaction mechanisms within the system. The kinetics and microbiology diversity in the sulfidogenic fluidized-bed reactor (at 30 °C) for co-treatment of hospital wastewater and metal-containing acidic water were examined. The alkalinity from organic oxidation raised the pH of the effluent from 2.3 to 6.1-8.2. Michaelis-Menten modeling yielded (K m =7.3 mg/l, V max = 0.12 mg/l min-1) in the batch bioreactor treatment using sulfate-reducing bacteria. For COD oxidation, the dissolved sulfide inhibition constant (Ki) was 3.6 mg/l, and the Ki value for H2S was 9 mg/l. The dominant species in the treatment process belong to the Proteobacteria group (especially Deltaproteobacteria). The ibuprofen and diclofenac compounds achieved the highest removal rates in the bioreactor of 58.6% and 52.3%, respectively; while, ketoprofen and naproxen of 41.9% and 46.6%, respectively. The findings in COD kinetics, sulfate-reducing bacteria abundance, and selected pharmaceutical concentration reduction provide insight into this co-treatment process's capability.

Abstract:The filling substrate is one of key factors influencing effectiveness of sulfate reducing packed-bed bioreactor (SRPB) treating acid mine drainage (AMD). The effects of four substrates (i.e. quartz sand, steel residue, biochar, and peanut shell) on remediation performance and sulfur transformation of SRPB treating AMD was studied. The results showed that steel residue and biochar improved sulfate reduction efficiency (61% and 49%) compared to quartz sand (32%), whereas peanut shell inhibited sulfate reduction efficiency (19%), attributed to its decomposition process leading to a severe accumulation of acetic acid. More amounts of sulfides generated in steel residue bioreactor were converted into acid volatile sulfide and elemental sulfur, resulting in a significant decrease in dissolved sulfide in the effluent. Metals (Fe, Al, Zn, Cd and Cr) except for Mn were effectively immobilized in the bioreactors, particularly for Al and Cd. Sulfate reducing bacteria and sulfide oxidizing bacteria lived symbiotically in all bioreactors which exhibited similar heterogeneity in microbial distribution and function, i.e. bacterial sulfate reduction mainly occurring in bottom-middle layers and photoautotrophic sulfide oxidation in upper layer close to outlet. The microbial response mechanism to various substrate environments was revealed through co-occurrence networks analysis. This study suggests that attention should be paid to the inhibitory effect of acetic acid accumulation on sulfate reduction when using sole lignocellulosic waste (peanut shell), and steel residue and biochar could be utilized as filling substances to promote sulfate reduction.

Abstract:The bioleaching technology has been considered as a promising green technology for remediation of contaminated sediments in recent years. Bioleaching technology was generally conducted in the batch bioreactor; however, the continuous bioreactor should be developed for the application of bioleaching technology in the future. The purposes of this study were to establish a continuous bioleaching process, and to evaluate the effects of sulfur dosage on the efficiency of metal removal during this continuous bioleaching process. The obtained results show that the pH decrease, sulfate production and metal removal efficiency all increased with increasing sulfur dosage in the continuous bioleaching process due to high substrate concentration for sulfur-oxidizing bacteria. After 30 days of operation time, the maximum solubilization efficiencies for Zn, Ni, Cu and Cr were found to be 78%, 90%, 88% and 68%, respectively, at 5% of sulfur dosage. After the bioleaching process, heavy metals bound in the carbonates, Fe-Mn oxides and organics/sulfides in the sediment were effectively removed and the potential ecological and toxic risks of treated sediment were greatly reduced. The results of bacterial community analyses demonstrated that this continuous bioleaching process were dominated by several acidophilic sulfur-oxidizing bacteria; S. thermosulfidooxidans, At. thiooxidans/At. ferrooxidans, S. thermotolerans and At. albertensis, whereas the percentage of less-acidophilic sulfur-oxidizing bacteria (T. thioparus and T. cuprina) was lower than 15% of total bacteria. In addition, the cell numbers of sulfur-oxidizing bacteria increased as the sulfur dosage was increased in the continuous bioleaching process.

Abstract:Interactions between metals and anaerobic ammonium oxidizing consortia substantially affect the quality of wastewater treatment plant effluent. In this study, we conducted acid-base titrations to ascertain the surface reactivity and proton adsorptive capacity of anammox consortia. A combination of titration data modeling and infrared spectroscopy suggested the presence of carboxyl, amine, and hydroxyl groups. Cd adsorption experiments demonstrate that 1 g of dry biomass could bind an equivalent of 7.12 x 10(-6) mol/L of Cd. Density functional theory calculations further reveal that carboxyl and hydroxyl groups are able to form stable Cd complexes. Furthermore, considerable carboxyl and hydroxyl groups promote bacterial aggregation, and thus solid-liquid separation. The results of this study highlight the potential role of anammox consortia in adsorbing metal cations, and thus help to improve the understanding of the universally significant contribution of anammox consortia at the detoxification of metal cations in wastewater treatment systems. Dynamics of Microbial Communities during the Removal of Copper and Zinc in a Sulfate-Reducing Bioreactor with a Limestone Pre-Column System

Abstract:Biological treatment using sulfate-reducing bacteria (SRB) is a promising approach to remediate acid rock drainage (ARD). Our purpose was to assess the performance of a sequential system consisting of a limestone bed filter followed by a sulfate-reducing bioreactor treating synthetic ARD for 375 days and to evaluate changes in microbial composition. The treatment system was effective in increasing the pH of the ARD from 2.7 to 7.5 and removed total Cu(II) and Zn(II) concentrations by up to 99.8% and 99.9%, respectively. The presence of sulfate in ARD promoted sulfidogenesis and changed the diversity and structure of the microbial communities. Methansarcina spp. was the most abundant amplicon sequence variant (ASV); however, methane production was not detected. Biodiversity indexes decreased over time with the bioreactor operation, whereas SRB abundance remained stable. Desulfobacteraceae, Desulfocurvus, Desulfobulbaceae and Desulfovibrio became more abundant, while Desulfuromonadales, Desulfotomaculum and Desulfobacca decreased. Geobacter and Syntrophobacter were enriched with bioreactor operation time. At the beginning, ASVs with relative abundance <2% represented 65% of the microbial community and 21% at the end of the study period. Thus, the results show that the microbial community gradually lost diversity while the treatment system was highly efficient in remediating ARD.

Bioremediation of acid mine drainage using sulfate-reducing wetland bioreactor: Filling substrates influence, sulfide oxidation and microbial community

Abstract:Two sulfate-reducing wetland bioreactors (SRB-1 filled with lignocellulosic wastes and SRB-2 with river sand) were applied for synthetic acid mine drainage treatment with bio-waste fermentation liquid as electron donor, and the influence of filling substrates on sulfate reduction, sulfur transformation and microbial community was studied. The presence of lignocellulosic wastes (mixture of cow manure, bark, sawdust, peanut shell and straw) in SRB-1 promoted sulfate reduction efficiency (68.9%), sulfate reduction rate (42.1 ± 11 mg S/(L·d)), dissolved sulfide production rate (27.4 ± 7 mg S/(L·d)), and particularly caused high conversion ratio of sulfate reduction into dissolved sulfide (66.4%). In comparison, the relatively low sulfate reduction efficiency (42.9%), sulfate reduction rate (27.0 ± 10 mg S/(L·d)), dissolved sulfide production rate (5.6 ± 3 mg S/(L·d)) and low dissolved sulfide conversion efficiency (21.2%) occurred in SRB-2. Mixed organic substrates including easily assimilated electron donors (in manure) and lignocellulosic matter were effective to promote quick start and long-term microbial sulfate reduction. More than 98% of produced dissolved sulfide was oxidized dominantly by photoautotrophic green sulfur bacteria (genera Chlorobium and Chlorobaculum), of which 64.6% and 54.5% was converted into elemental sulfur for SRB-1 and SRB-2. The oxidation of sulfide into elemental sulfur for potential recovery rather than sulfate is preferred. Diverse sulfate reducing bacteria and sulfide oxidizing bacteria co-existed in the treatment system, which led to a sustainable sulfur transformation. High metal removal efficiency for Fe (99.6%, 92.5%), Cd (99.9%, 99.9%), Zn (99.4%, 98.5%), Cu (94.5%, 94.6%) except for Mn (9.3%, 3.6%) was achieved, and effluent pH increased to 6.5-7.7 and 6.7-7.7 for SRB-1 and SRB-2, respectively. Microbial community was regulated by filling substrates. Synergism between lignocellulosic decomposing bacteria and sulfate reducing bacteria played a vital role in lignocellulosic bioreactor treating AMD, in addition to fermentation liquid serving as effective electron donor.

Abstract:Freshwater lakes are often polluted with various heavy metals in the Anthropocene. The iron-oxidizing microorganisms and their mineralized products can coprecipitate with many heavy metals, including Al, Zn, Cu, Cd, and Cr. As such, microbial iron oxidation can exert a profound impact on environmental remediation. The environmental pH is a key determinant regulating microbial growth and mineralization and then influences the structure of the final mineralized products of anaerobic iron-oxidizing bacteria. Freshwater lakes, in general, are neutral-pH environments. Understanding the effects of varying pH on the mineralization of iron-oxidizing bacteria under neutrophilic conditions could aid in finding out the optimal pH values that promote the coprecipitation of heavy metals. Here, two typical neutrophilic Fe(II)-oxidizing bacteria, the nitrate-reducing Acidovorax sp. strain BoFeN1 and the anoxygenic phototrophic Rhodobacter ferrooxidans strain SW2, were selected for studying how their growth and mineralization response to slight changes in circumneutral pH. By employing focused ion beam/scanning electron microscopy (FIB–SEM) and transmission electron microscopy (TEM), we examined the interplay between pH changes and anaerobic iron-oxidizing bacteria and observed that pH can significantly impact the microbial mineralization process and vice versa. Further, pH-dependent changes in the structure of mineralized products of bacterial iron oxidation were observed. Our study could provide mechanical insights into how to manipulate microbial iron oxidation for facilitating remediation of heavy metals in the environment.

Abstract:This study used six identical reactors to investigate the effects of heavy metals on anaerobic digestion sludge. The addition of 1-50 mg L-1 of Zn(II), Cd(II), Pb(II), Hg(II), and a mixture of four metals had a slight impact on COD removal. The removal efficiency decreased slightly to 92.9%, 92.0%, 92.7%, 91.8%, and 93.7% compared with 95.8% in control when 50 mg L-1 of heavy metals were added. The biogas production also varied slightly when exposed to 1-50 mg L-1 of heavy metals, with average production rates of 0.271, 0.244, 0.263, 0.269, 0.255, and 0.261 L d-1, respectively. Extracellular polymeric substance increased to protect microorganisms from the toxicity of heavy metals, and dehydrogenase activity increased from 0.42 to 1.20, 0.44, 0.51, 0.71, and 0.59 EU g-1 SS, respectively. The relative abundance of Methanothrix decreased from 91.2% to 88.5%, 87.6%, 85.7%, and 87.0% when Cd(II), Pb(II), Hg(II), and the mixture of heavy metals were added. Among the heavy metals, Zn(II) had the least impact on microbial activity but the most severe inhibition on biogas production, while the mixture of heavy metals showed relatively slight inhibition due to an antagonistic effect. Anaerobic digestion sludge exhibited good tolerance to heavy metals and could be a potential treatment option for heavy metal-containing wastewater. Environmental implication: Anaerobic digestion has a promising application prospect for treating organic wastewater. Heavy metal pollution exists in many industrial wastewaters. However, few studies have comprehensively compared the effects of Zn(II), Cd(II), Pb(II), Hg(II), and the mixture of heavy metals on anaerobic digestion, especially in the treatment of relatively low-strength wastewater. The main objectives of this study were to investigate the effects of heavy metals on COD removal, biogas production, sludge properties, and microbial characteristics of anaerobic digestion sludge. The results have proved that the anaerobic digestion process can be used as a pretreatment unit for treating heavy metal-containing wastewater.
Treatment of Metal‐contaminated Water and Vertical Distribution of Metal Precipitates in an Upflow Anaerobic Bioreactor

Abstract:Abstract A lab‐scale upflow anaerobic bioreactor filled with granular sludge and cow manure was operated for 140 days to determine the mechanism of metal removal and the vertical distribution of metal precipitates. Heavy metal ions were removed in the order of Cu2+, Cd2+, Zn2+, Fe2+ and Mn2+ with respect to the height in the reactor. The solid phase analysis showed that the heavy metals were mostly precipitated in the form of metal sulfides by sulfate reduction. The contents of metal precipitates in the reactor were as follows: (i) Cd and Zn were highest in the bottom, (ii) Fe was highest at the low‐middle layer, and (iii) Mn was increased with the height in the reactor. The vertical distribution of metal sulfides in the reactor was directly related to the solubility product (Ksp). Results obtained in this study suggest a feasibility of the application to separate precipitation of metal ions and recovery of valuable metals from metal‐containing wastewater

Abstract:Persistence of ammonium nitrogen and heavy metals in wastewater still remains a challenge, and many wastewater treatment plants face the challenge of removing nitrogen under heavy metal stresses. There is no preferred method for the biological treatment of wastewater containing nitrogen and heavy metals with the possible exception of the anaerobic ammonium oxidation (anammox), since it has shown promise for removing nitrogen under heavy metal stresses. This article reviews the recent research results of the nitrogen-removal performance and mechanism by the anammox process under heavy metal stresses, mainly discussing the enhancing and inhibition effects of heavy metals on the performance of the Anammox reactor. The influencing mechanism of heavy metals on the microbial community and extracellular polymeric substances is also presented, and examples are given for explanation. The main problems of the present research are pointed out, and it is proposed that unifying the metal ion concentrations of inhibiting or promoting anammox activity is necessary for the development and industrial application of the anammox process. The information of this review can offer a great possibility for achieving desired nitrogen removal in wastewater treatment under heavy metal stresses and with significant energy savings.
Stabilization and Management of Sulfate-Reducing Bioreactor Residues After Acid Mine Drainage Treatment

Abstract:Passive and semi-passive sulfate-reducing bioreactors (SRBRs) are attracting increasing attention worldwide for the treatment of contaminated mine effluents. This study focuses on the management of post-treatment residues from these bioreactors. The residues of two bioreactors with different medium compositions were studied after they were mixed with different alkaline amendments (25% biomass ash, 30% aluminum red mud, 10% lime kiln dust, or 15% hydroxyapatite). Weathering cell tests showed that aluminum red mud and Bedford lime kiln dust efficiently maintain the leachate at neutral pH (≥ 7.0) and immobilize some metals (Al, Cd, Cr, Cu, Fe, Mn, Ni, Pb, and Zn) in the post-treatment residues with average dissolved concentrations < 0.5 mg/L for all toxic metals, except for Mn, which are less than 3.2 mg/L. Column tests were also performed on the unamended post-treatment residues under unsaturated, saturated, and immersed conditions. The storage of the post-treatment SRBR residues under saturated and immersed conditions preserved the reducing conditions of the media and the neutrality of the leachate pH and also limited the leaching of metals retained by the bioreactor media.

Refining habitat selection for sulfate-reducing bacteria: Evaluating suitability and adaptability for sulfate-metal wastewater treatment during anaerobic-to-aerobic transitions

Abstract:Inoculating sulfate-reducing bacteria (SRB) habitats offers an eco-friendly method for treating sulfate-metal laden wastewater, characterized by high sulfate levels, low pH, and elevated heavy metals. This study optimizes source habitat selection of SRB by evaluating groundwater, sewage sludge, and lake sediment, focusing on their suitability and adaptability to aerobic-anaerobic transitions in industrial settings. Sewage sludge, with its slightly acidic pH, reducing environment, and high nutrient levels (Total organic carbon: 207.53 g kg-1, Total nitrogen: 47.12 g kg-1), provides robust SRB potential, as supported by its highest diversity index. However, heavy metals and polycyclic aromatic hydrocarbons pose application challenges. All habitats effectively reduced metal concentrations anaerobically, with Cu removal reaching 95%-99%, and groundwater achieved the highest chemical oxygen demand reduction (63.6%) aerobically. Sludge and sediment showed high biomass and extracellular polymeric substances (EPS) accumulation, while groundwater's nucleic acid-rich EPS enhanced metal immobilization, resulting in stable residual metal forms but with potential remobilization under oxidative conditions. Microbial analysis revealed that Proteobacteria and Firmicutes were key players during transitions, with the highest SRB abundance in groundwater. SRB composition varied across habitats, with Sedimentibacter (13.04%), Desulfovibrio (6.33%), and Desulfomonile (8.1%) dominating in groundwater, sludge, and sediment, respectively, during the anaerobic stage. Functional analysis highlighted sludge's persistence in sulfate reduction under aerobic conditions, while groundwater's limited nitrogen cycle involvement indicated broader biogeochemical limitations. Collectively, these findings highlight strengths and limitations of each habitat as SRB inoculum source, emphasizing the importance of tailored anaerobic-to-aerobic strategies for effective wastewater management.

Abstract:Sulfate-rich mine water must be treated before it is released into natural water bodies. We tested ethanol as substrate in bioreactors designed for biological sulfate removal from mine water containing up to 9 g L−1 sulfate, using granular sludge from an industrial waste water treatment plant as inoculum. The pH, redox potential, and sulfate and sulfide concentrations were measured twice a week over a maximum of 171 days. The microbial communities in the bioreactors were characterized by qPCR and high throughput amplicon sequencing. The pH in the bioreactors fluctuated between 5.0 and 7.7 with the highest amount of up to 50% sulfate removed measured around pH 6. Dissimilatory sulfate reducing bacteria (SRB) constituted only between 1% and 15% of the bacterial communities. Predicted bacterial metagenomes indicated a high prevalence of assimilatory sulfate reduction proceeding to formation of l-cystein and acetate, assimilatory and dissimilatory nitrate reduction, denitrification, and oxidation of ethanol to acetaldehyde with further conversion to ethanolamine, but not to acetate. Despite efforts to maintain optimal conditions for biological sulfate reduction in the bioreactors, only a small part of the microorganisms were SRB. The microbial communities were highly diverse, containing bacteria, archaea, and fungi, all of which affected the overall microbial processes in the bioreactors. While it is important to monitor specific physicochemical parameters in bioreactors, molecular assessment of the microbial communities may serve as a tool to identify biological factors affecting bioreactor functions and to optimize physicochemical attributes for ideal bioreactor performance.
