Abstract:The goal of this work is to develop non-destructive techniq ues that can conveniently, rapidly and accurately assess FCV tobacco leaf biochemis try status at the maturity stage at leaf levels by selecting optimal hyperspectr al method for estimation of biochemistry concentration and biophysical parameter . The data of reflectance and absorption spectra of fresh tobacco leaf were meas ured with ASD Fieldspec FR2500 at the maturity stage, and the parameters of biop hysical and biochemistry of the leaf were collected by normal method. The correl ation between spectral parameters consisting of single band reflect ance, logarithm of reciprocal of reflectance, transmittance and absorption and 1 00 existing spectral indices selected in the visible and near-infrared spectrum (350~1650 nm), with biochemistry concentration was analyzed by using linear, pow er and exponential fitting. Optimal method was selected based on the strongest s trength of determination coefficient. The result indicated that optimal optical methods for chlorophyll a, chlorophyll b, carotenoids and potassium concentratio n of leaf were respectively Gitelson and Merzlyak2(GM2)(R2=0.81), Photo chemical Reflectance Index 2(PRI2)(R2=0.80), Gitelson and Merzlyak2(GM 2)(R2=0.83), the starting position of reflectance gallery at 1420 nm(λs  1420)(R2=0.67) using linear fitting. Water-solubility total sugar , specific leaf weight(SLW), total nitrogen, nicotine were respectively first de rivative of reflectance at 532 nm using linear fitting(R2=0.54), transmitt ance at 1423 nm using linear fitting(R2=0.45), first derivative of log arit hm of reciprocal of reflectance (δlog(1/R)) at 666 nm using power fitti ng(R2=0.44), second derivative of logarithm of reciprocal of reflectance (δδlog(1/R)) at 1135 nm using power fitting(R2=0.20). It is feasible to predict matter concentration of tobacco leaf by optimal optical method for spectral parameters.

Abstract:It is shown that Fourier transform infrared emission spectroscopy can be used to analyze carotenoids and chlorophyll in whole leaves of cultivated tobacco Nicotiana tabacum L and its two modifications L 1 and L 2 . The results show that information on the structure and component composition of plant tissue pigments can be obtained in vivo without the negative effect of water and autofluorescence.

Abstract:Possible use of integral characteristics of slow stage of chlorophyll fluorescence induction (CFI) for monitoring of physiological state of plants in phytocenoses was studied in the leaves of wheat Triticum aestivum L. and radish Raphanus sativus var . radicula Pers ., cv. Virovskii belyi, grown in artificial-light culture. Investigation concerned the behavior of Т 0.5 (half-time of fluorescence intensity decrease from its peak to a steady-state level), R fd (fluorescence decrease ratio also known as vitality index), parameter (maximum possible average rate of induction transient), and (average efficiency of PSA in the induction period) during leaf ontogenesis. Plant material was the discs cut from uniformly illuminated wheat leaves of different age from the top (sixth) storey of plants and the disks from the second (in the order of emergence) true leaves of radish at the age from 3 to 24 days. The plants were grown in controlled-climate chambers in hydroponic culture on claydite at irradiance of 100 W/m 2 of photosynthetically active radiation (PAR). CFI curves were recorded using a single-beam unit. Values of parameter Т 0.5 slightly changed during leaves' ontogenesis up to the time of senescence (the age of 26 days). Relative changes in the values of CFI parameters (Δ rel., %) at the age from 2 to 24 days were 28% for and 30% for Т 0.5 . For and R fd , the values were 67 and 70%, respectively. Upon more advanced ageing and comparison of parameters for 24- and 26-day-old leaves, more pronounced changes were observed for Т 0.5 (by 37.6%) and (by 36.0%) and less pronounced for (by 23.0%) and R fd (by 13.0%). Thus, the pattern of parameter Т 0.5 behavior observed in the course of wheat leaves' ontogenesis distinguished it from other fluorescence parameters of CFI slow stage (R fd , , and ); this gives grounds for a potential use of Т 0.5 as one of the simplest integral indicators of physiological state and pathophysiological (irreversible) changes occurring in plant leaves within a wide age interval and upon senescence. Unambiguous interpretation of the data obtained by means of parameter Т 0.5 under stress conditions for estimation of irreversibility of the observed changes in PSA requires more detailed investigations.

Abstract:The application of near-infrared (NIR) spectroscopy for in-line monitoring of extraction process of scutellarein from Erigeron breviscapus (vant.) Hand-Mazz was investigated. For NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2mm pathlength flow cell were utilized to collect spectra in real-time. High performance liquid chromatography (HPLC) was used as a reference method to determine scutellarein in extract solution. Partial least squares regression (PLSR) calibration model of Savitzky–Golay smoothing NIR spectra in the 5450–10,000cm−1 region gave satisfactory predictive results for scutellarein. The results showed that the correlation coefficients of calibration and cross validation were 0.9967 and 0.9811, respectively, and the root mean square error of calibration and cross validation were 0.044 and 0.105, respectively. Furthermore, both the moving block standard deviation (MBSD) method and conformity test were used to identify the end point of extraction process, providing real-time data and instant feedback about the extraction course. The results obtained in this study indicated that the NIR spectroscopy technique provides an efficient and environmentally friendly approach for fast determination of scutellarein and end point control of extraction process.
Retrieving the in vivo Scopoletin Fluorescence Excitation Band Allows the Non-invasive Investigation of the Plant–Pathogen Early Events in Tobacco Leaves

Abstract:In this study, we developed and applied a new spectroscopic fluorescence method for the in vivo detection of the early events in the interaction between tobacco ( Nicotiana tabacum L.) plants and pathogenic bacteria. The leaf disks were infiltrated with a bacterial suspension in sterile physiological solution (SPS), or with SPS alone as control. The virulent Pseudomonas syringae pv. tabaci strain ATCC 11528, its non-pathogenic ΔhrpA mutant, and the avirulent P. syringae pv. tomato strain DC3000 were used. At different post-infiltration time–points, the in vivo fluorescence spectra on leaf disks were acquired by a fiber bundle-spectrofluorimeter. The excitation spectra of the leaf blue emission at 460 nm, which is mainly due to the accumulation of coumarins following a bacterial infiltration, were processed by using a two-bands Gaussian fitting that enabled us to isolate the scopoletin (SCT) contribution. The pH-dependent fluorescence of SCT and scopolin (SCL), as determined by in vitro data and their intracellular localization, as determined by confocal microscopy, suggested the use of the longer wavelength excitation band at 385 nm of 460 nm emission (F 385_460 ) to follow the metabolic evolution of SCT during the plant–bacteria interaction. It was found to be directly correlated ( R 2 = 0.84) to the leaf SCT content, but not to that of SCL, determined by HPLC analysis. The technique applied to the time-course monitoring of the bacteria–plant interaction clearly showed that the amount and the timing of SCT accumulation, estimated by F 385_460 , was correlated with the resistance to the pathogen. As expected, this host defense response was delayed after P. syringae pv. tabaci ATCC 11528 infiltration, in comparison to P. syringae pv. tomato DC3000. Furthermore, no significant increase of F 385_460 (SCT) was observed when using the non-pathogenic ΔhrpA mutant of P. syringae pv. tabaci ATCC 11528, which lacks a functional Type Three Secretion System (TTSS). Our study showed the reliability of the developed fluorimetric method for a rapid and non-invasive monitoring of bacteria-induced first events related to the metabolite-based defense response in tobacco leaves. This technique could allow a fast selection of pathogen-resistant cultivars, as well as the on-site early diagnosis of tobacco plant diseases by using suitable fluorescence sensors.

Choice of Reference Surfaces to assess Plant Health through leaf scale temperature monitoring

Abstract:We use infrared thermal imaging to remotely monitor the temperature of the leaves and plant which in turn is an indicative of their health and stress; in particular water stress. A series of experiments were conducted using Fluke TiX580 thermal imager on tomato plants to correlate the leaf temperature to different stages such as healthy, dying/wilted, and completely dead/dry. The leaf temperature was compared to a series of paper-based reference surface temperatures; these are of different colors and some are dry while some are wet. All the reference surfaces were insulated at the bottom ensuring the heat interaction between their top surfaces and ambient only. The surfaces were kept sufficiently far from one another. The healthy leaf temperature was found to be close to that of white dry and black wet reference surfaces whereas the wilted and dying leaves temperature were observed to range between yellow and red signifying that this temperature range can better predict the onset of water stress in the leaves. The completely dead/dry leaves were observed to range between green and blue dry surfaces, respectively. However, most of the dead leaf temperature data was found to be accumulated closer to the green surface signifying green dry surface can better indicate a dead leaf condition. The dying leaves were observed to exhibit 8-10 degree centigrade higher temperatures as compared to the healthy leaves in similar ambient conditions. Temperature-based health assessment provides us with a timely intervention to prevent leaf death compared to the optical monitoring since IR images revealed elevated leaf temperature 2-3 days before the optical unhealthiness (appearance of yellowness on the leaf) was noticed.

Learning from the pregnane X receptor: how to fight one aggressor with two strategies

Abstract:Cholestasis is a common feature of human liver diseases. Potentially toxic bile salts accumulate in the cholestatic liver and promote hepatocyte damage by apoptosis and necrosis, thereby contributing to disease progression. The glycine and taurine conjugates of chenodeoxycholate (GCDC, TCDC) are the predominant dihydroxy bile salts in cholestatic patients and have been held responsible for cholestasis-associated liver injury (1). Other potentially toxic bile salts like deoxycholate (DC) are quantitatively less relevant in cholestatic liver disease (2). In the past decade, potential mechanisms by which bile salts induce cell injury and apoptosis in hepatocytes, have been investigated extensively. A better understanding of these mechanisms might offer new therapeutic options to ameliorate the detrimental effects of longstanding cholestasis, as seen in patients with chronic cholestatic disorders such as primary biliary cirrhosis (PBC) or primary sclerosing cholangitis (PSC). Glycine conjugate of chenodeoxycholate effectively induces hepatocyte apoptosis at low micromolar concentrations by a Fas death receptor-dependent process that is independent of the Fas ligand, but requires oligomerization of Fas by increasing cell surface trafficking of this death receptor (3). GCDC-induced generation of reactive oxygen species followed by epidermal growth factor receptor-dependent tyrosine phosphorylation of Fas appear to be required for GCDC-induced Fas signalling (4). GCDC can also directly induce the intrinsic pathway of apoptosis by a Fas-independent mechanism, which might represent an escape mechanism, when the faster extrinsic pathway is not available (5). Inhibition of bile salt-induced apoptosis has been shown to reduce liver injury and fibrosis in animal models of cholestasis, but clinical applications in patients are still lacking. The therapeutic bile salt ursodeoxycholate (UDC) exerts anti-apoptotic properties (6). However, these may play a minor role for the therapeutic effect of UDC when compared with anticholestatic properties at the level of hepatocytes and cholangiocytes and its bile detoxifying properties (7). In this issue of Liver International, Zucchini-Pascal et al. (8) used both DC and GCDC to induce apoptosis in primary rat hepatocytes. Consistent with previous work, DC induced apoptosis (at concentrationso 400 mM) shifting to necrosis at concentrations exceeding 400 mM. They then used an activator of the pregnane X receptor (PXR) to reduce bile salt-induced apoptosis. The work of Zucchini-Pascal and colleagues raises two questions: (i) How does PXR affect hepatocyte injury and apoptosis? (ii) Do the findings have potential implications for future therapeutic strategies in cholestatic disorders? The pregnane X receptor ( NR1I2) is a ligand-activated member of the nuclear receptor superfamily and has a key role in regulating the expression of many genes involved in detoxification and metabolism of bile salts as well as other endoand xenobiotics, including biotransformation phase 1 enzymes CYP3A4 and CYP7A1, biotransformation phase 2 enzymes SULT2A1, UGT1A3 and UGT1A4 and biotransformation phase 3 transporters MDR1, MRP2 and MRP3. Livers of PXR knockout mice are more vulnerable than wild-type mice both after bile duct ligation and cholate feeding, possibly because of impaired biotransformation (9–11). The potent PXR ligand 5-pregnen-3b-ol-20-one-16a-carbonitrile reduced (litho-)cholate-induced liver injury in wild-type mice, but not in PXR knockout mice (11, 12). PXR also seems to have anti-fibrogenic effects, presumably by inhibiting NF-kB (13). In the present issue, Zucchini-Pascal et al. (8) demonstrate that the PXR activator clotrimazole reduced apoptosis induced by DC and GCDC in primary rat hepatocytes. Because PXR agonists stimulate bile salttransporting/detoxifying enzymes leading to reduced serum levels of bile salts in healthy as well as bile ductligated mice (14), this might be considered a sufficient explanation for the anti-apoptotic properties of clotrimazole. However, PXR agonists appear to increase hepatocyte survival by additionally upregulating the anti-apoptotic Bcl-2 proteins Bcl-2 and Bcl-xL (15). Upregulation of anti-apoptotic Bcl-2 proteins might also be responsible for the observed reduced activation of the effector caspases-3, -6 and -7 and the initiator caspase-9. These caspases are all downstream of the mitochondrial loop, which is essential to induce apoptosis in type II cells such as hepatocytes. They are, therefore, less active when cytochrome C release is reduced by Bcl-2 or Bcl-xL (16). It is difficult to distinguish the direct inhibitory effects on caspases from the indirect effects modulated by the Bcl-2 protein family. Likewise, reduced cleavage of PARP and other substrates of effector caspases by clotrimazole observed in the present study could solely be explained by the upregulation of anti-apoptotic Bcl-2 proteins. Abstract:Near-infrared (NIR) excited Fourier transform (FT) Raman spectroscopy has been applied for in situ analysis of carotenoids in living plant samples. Pelargonium x hortorum leaf has been mapped using a Raman mapping technique to illustrate heterogeneous distribution of carotenoids. Mapping has also been employed for visualization of carotenoid changes induced by abiotic and biotic stress. In a tomato (Lycopersicon esculentum Mill.) fruit, inhibition of lycopene biosynthesis and accumulation of beta-carotene are demonstrated in tissue affected by sunscald physiological disorder. Raman map of diseased sugarbeet (Beta vulgaris L.) leaf shows a local carotenoid decline at infection site while the carotenoid accumulation is evident in parsley (Petroselinum crispum Mill. Nym.) as a response to Septoria petroselini infestation. Additionally, occurrence of lutein, beta-carotene and capsanthin, and changes in their relative content during bell pepper (Capsicum annum L.) fruit ripening are described by single Raman spectra. Based on these examples, the potential application of NIR-FT-Raman spectroscopy for a non-destructive analysis of carotenoids in various living plant tissues of the size ranging from about 0.01 mm2 to 35 cm2 is discussed.

Abstract:Leaf aging is a significant process during herbaceous plant senescence, which is influenced by various internal and external factors. During leaf aging, chlorophyll catabolism is one of the most important metabolism pathways and results in leaf yellowing. Understanding the underlying mechanism is important for the regulation of senescence in tobacco leaf. However, there are few studies on explaining tobacco leaf senescence from the proteomics level. Here, photosynthesis experiments, cell ultrastructure, and proteomics were used to study tobacco leaves of different growth stages. We applied iTRAQ-based quantitative proteomics and parallel reaction monitoring (PRM) to determine the accumulation of proteins in aging tobacco leaves. Overall, we screened 4747 proteins. The result of KEGG pathways analysis showed that differently expressed proteins (DEPs) were involved in four pathways: metabolic pathways, biosynthesis of secondary metabolites, microbial metabolism in diverse environments, and starch and sucrose metabolism. This would be first report based on iTRAQ-PRM technique, in which we identified proteins related to photosynthesis showed a differently expressed during senescence stage in flue-cured tobacco plants.

Abstract:By using imaging techniques, plant physiological parameters can be assessed without contact with the plant and in a non-destructive way. During plant-pathogen infection, the physiological state of the infected tissue is altered, such as changes in photosynthesis, transpiration, stomatal conductance, accumulation of Salicylic acid (SA) and even cell death. In this study, the different temperature distribution between the leaves infected by tobacco mosaic virus strain-TMV-U1 and the noninfected leaves was visualized by digital infrared thermal imaging with the microscopic observations of the different structure within different species tomatoes. Results show a presymptomatic decrease in leaf temperature about 0.5-1.3 °C lower than the healthy leaves. The temperature difference allowed the discrimination between the infected and healthy leaves before the appearance of visible necrosis on leaves.

Abstract:Plant hormones are important in the control of physiological and developmental processes including seed germination, senescence, flowering, stomatal aperture, and ultimately the overall growth and yield of plants. Many currently available methods to quantify such growth regulators quickly and accurately require extensive sample purification using complex analytic techniques. Herein we used ultra-performance liquid chromatography-high-resolution mass spectrometry (UHPLC-HRMS) to create and validate the prediction of hormone concentrations made using attenuated total reflection Fourier-transform infrared (ATR-FTIR) spectral profiles of both freeze-dried ground leaf tissue and extracted xylem sap of Japanese knotweed (Reynoutria japonica) plants grown under different environmental conditions. In addition to these predictions made with partial least squares regression, further analysis of spectral data was performed using chemometric techniques, including principal component analysis, linear discriminant analysis, and support vector machines (SVM). Plants grown in different environments had sufficiently different biochemical profiles, including plant hormonal compounds, to allow successful differentiation by ATR-FTIR spectroscopy coupled with SVM. ATR-FTIR spectral biomarkers highlighted a range of biomolecules responsible for the differing spectral signatures between growth environments, such as triacylglycerol, proteins and amino acids, tannins, pectin, polysaccharides such as starch and cellulose, DNA and RNA. Using partial least squares regression, we show the potential for accurate prediction of plant hormone concentrations from ATR-FTIR spectral profiles, calibrated with hormonal data quantified by UHPLC-HRMS. The application of ATR-FTIR spectroscopy and chemometrics offers accurate prediction of hormone concentrations in plant samples, with advantages over existing approaches.

Detection of transgenic tomato leaf with LeETR1 antisense gene by near-infrared spectroscopy

Abstract:The increasingly widespread use of Fourier transform near-infrared spectroscopy (FT-NIR) and its reasonable costs indicate that it could be a suitable technique for on-line process monitoring and quality control. In this study, 208 tomato leaves (106 transgenic leaves produced by introducing the antisense ethylene receptor gene LeERT1, and 102 of their conventional parents) were used for diffuse reflectance measurement. Discriminant analysis (DA) based on principal component analysis (PCA) in the region of 900-2500 nm was used to discriminate the samples. The results show that the DA method led to a good classification. The percentage of correctly classified transgenic and non-transgenic samples during the calibration process was 98.7% and 100%, respectively. For the validation process, 96% of the non-transgenic samples were correctly classified and no transgenic sample was misclassified, as there was 100% correctness. This indicates that NIR spectroscopy combined with chemometrics techniques could be used as a rapid and reliable method to differentiate transgenic tomato leaves from non-transgenic leaves. This method avoids time-consuming, costly, and laborious chemical and sensory analyses.

Abstract:Drought is a climatic event that considerably impacts plant growth, reproduction and productivity. Toona sinensis is a tree species with high economic, edible and medicinal value, and has drought resistance. Thus, the objective of this study was to dynamically monitor the physiological indicators of T. sinensis in real time to ensure the selection of drought-resistant varieties of T. sinensis . In this study, we used near-infrared spectroscopy as a high-throughput method along with five preprocessing methods combined with four variable selection approaches to establish a cross-validated partial least squares regression model to establish the relationship between the near infrared reflectance spectroscopy (NIRS) spectrum and physiological characteristics (i.e., chlorophyll content and nitrogen content) of T. sinensis leaves. We also tested optimal model prediction for the dynamic changes in T. sinensis chlorophyll and nitrogen content under five separate watering regimes to mimic non-destructive and dynamic detection of plant leaf physiological changes. Among them, the accuracy of the chlorophyll content prediction model was as high as 72%, with root mean square error (RMSE) of 0.25, and the RPD index above 2.26. Ideal nitrogen content prediction model should have R 2 of 0.63, with RMSE of 0.87, and the RPD index of 1.12. The results showed that the PLSR model has a good prediction effect. Overall, under diverse drought stress treatments, the chlorophyll content of T. sinensis leaves showed a decreasing trend over time. Furthermore, the chlorophyll content was the most stable under the 75% field capacity treatment. However, the nitrogen content of the plant leaves was found to have a different and variable trend, with the greatest drop in content under the 10% field capacity treatment. Comprehensive investigation of tobacco leaves during natural early senescence via multi-platform metabolomics analyses

Abstract:Abstract Senescence is the final stage of leaf growth and development. Many different physiological activities occur during this process. A comprehensive metabolomics analysis of tobacco middle leaves at 5 different developmental stages was implemented through multi-platform methods based on liquid chromatography, capillary electrophoresis and gas chromatography coupled with mass spectrometry. In total, 412 metabolites were identified, including pigments, sterols, lipids, amino acids, polyamines, sugars and secondary metabolites. Dramatic metabolic changes were observed. Firstly, membrane degradation and chlorophyll down-regulation occurred after the 50% flower bud stage. Levels of major membrane lipids decreased, including those of the glycolipids in chloroplast thylakoids and phospholipids in membrane envelopes. Clear decreases in free sterols and acylated sterol glucosides were detected along with the accumulation of sterol esters. The accumulation of alkaloids was found. The amino acid levels were significantly decreased, particularly those of N-rich amino acids (glutamine and asparagine), thus reflecting N translocation. Subsequently, the antioxidant system was activated. Sugar alcohols and polyphenols accumulated when the lower leaves turned yellow. These results comprehensively revealed the metabolic changes that occur during tobacco leaf development and senescence under natural conditions.

Abstract:The objectives of this study were to characterize leaf reflectance spectra of tomato leaves damaged by leaf miner and to determine those leaf reflectance wavelengths that were most responsive to plant damage caused by the pest. Near infrared (NIR) Spectral characteristics of single tomato leaves at various levels of infestation by the leaf miner, were measured and analyzed using a spectrometer. Tomato leaf damage was classified into five scales, i.e., 0 (no damage), 1 (light damaged), 2 (10-25% damaged), 3 (more than 25% damaged), and 4 (severe damaged), based on the scale of infestation displayed on the surfaces of plant parts. Spectral parameter such as reflectance sensitivity was used to find the optimal wavelengths to determining and evaluating the damage level. Results showed that there were significant differences in reflectance among infestations at wavelengths of 1450nm and 1900 nm particularly. The determining coefficients (R2) for a linear relationship were 0.98 and 0.91 for the spectral-infestation levels relations. Thus, both of these wavelengths were good indicators of leaf senescence caused by the leaf miner.

Abstract:<p>Tobacco (<em>Nicotiana tabacum</em>) is extensively cultivated all over the world for its economic value. During curing and storage, senescence occurs, which is associated with physiological and biochemical changes in postharvest plant organs. However, the molecular mechanisms involved in accelerated senescence due to high temperatures in tobacco leaves during curing need further elaboration. We studied molecular mechanisms of senescence in tobacco leaves exposed to high temperature during curing (Fresh, 38 °C and 42 °C), revealed by isobaric tags for relative and absolute quantification (iTRAQ) for the proteomic profiles of cultivar Bi'na1. In total, 8903 proteins were identified, and 2034 (1150 up-regulated and 1074 down-regulated) differentially abundant proteins (DAPs) were obtained from tobacco leaf samples. These DAPs were mainly involved in posttranslational modification, protein turnover, energy production and conversion. Sugar- and energy-related metabolic biological processes and pathways might be critical regulators of tobacco leaves exposed to high temperature during senescence. High-temperature stress accelerated tobacco leaf senescence mainly by down-regulating photosynthesis-related pathways and degrading cellular constituents to maintain cell viability and nutrient recycling. Our findings provide a valuable inventory of novel proteins involved in senescence physiology and elucidate the protein regulatory network in postharvest organs exposed to high temperatures during flue-curing.</p>

Abstract:Assessment of plant traits (phenotyping) is central to modern advanced techniques of plant sciences and accelerated breeding of crop plants, including fruit crops, for improving productivity and stress resilience. Hyperspectral reflectance imaging is an emerging method allowing to capture a vast amount of the structural, biochemical, and phenological information about plants. The advent of low-cost hyperspectrometers made this method affordable for a broad community of plant scientists. However, extraction of sensible information from reflectance images is hindered by the complexity of plant optical properties, especially when they are measured in the field. We propose using reflectance indices (Plant Senescence Reflectance Index, PSRI; Anthocyanin Reflectance Index, ARI; and spectral deconvolution) previously developed for remote sensing of vegetation and point-based reflectometers to infer the spatially resolved information on plant development and biochemical composition using lettuce ( Lactuca sativa L.) leaves and ripening apple ( Malus × domestica Borkh.) fruit as the model. Specifically, the proposed approach enables capturing data on distribution of chlorophylls and primary carotenoids as well as secondary carotenoids (both linked with fruit ripening and leaf senescence during plant development) as well as the information on spatial distribution of anthocyanins (known as stress pigments) over the plant surface. We argue that the proposed approach would enrich the phenotype assessments made on the base of reflectance image analysis with valuable information on plant physiological condition, stress acclimation state, and the progression of the plant development.

Abstract:Rapid assessment of physiological status is a precondition for addressing biological stress in trees so that they may recover. Environmental stress can cause water deficit in plants, while terahertz (THz) spectroscopy is sensitive to changes in aqueous solutions within organisms. This has given the THz sensor a competitive edge for evaluating plant phenotypes, especially under similar environmental stress, if there are existing differences in the corresponding THz information. In this study, we utilized THz technology in association with traditional weighing methods to explore physiological changes in citrus leaves under different temperature, duration, and stress treatment conditions. It was found that the higher the temperature and the longer the exposure duration, the more severe the reduction in the relative absorption coefficient. There was a positive correlation between the trends and the increase in the ion permeability of cells. In addition, based on the effective medium theory, THz spectral information can be transformed into information on free water and bound water in the leaves. Under different treatment conditions, water content shows different trends and degrees of change on the time scale, and accuracy was verified by traditional weighing methods. These findings revealed that characteristics of THz information can serve as a simple and clear indicator for judging a plant's physiological status.
