Abstract:Perilla (Perilla frutescens, PF) is an annual labiaceae herb that can be used as vegetable, seasoner, and herb, which mainly includes red PF (P. frutescens var. crispa) and green PF (P. frutescens var. frutescens). Red and green Perilla is mainly used for medicine and food, respectively. In order to explore the differences between these two Perilla cultivars and the effects of origin on them, we studied the components of 130 Perilla samples from different origins by ultra-high performance liquid chromatography quadrupole-time-of-flight mass spectrometry combined with partial least squares discriminant analysis and orthogonal partial least squares discriminant analysis. In this study, 57 potential compounds were analyzed, mainly including organic acids, flavonoids, terpenoids, and anthocyanins. In different varieties of Perilla, anthocyanins were found only in red PF. In addition, the content of four flavonoids, two organic acids, and one coumarin in red PF is much higher than that in green PF. Among the same variety of Perilla, the contents of three metabolites were higher in the north of China for red Perilla, while five metabolites were higher in the south of China for green Perilla. Overall, this research provided a basis for distinguishing different varieties and sources of Perilla through the differential metabolites of Perilla.
Non-destructive measurement of antioxidant activity and water content in chili powder (Capsicum annuum L.) using near-infrared spectroscopy

Abstract:Chili (Capsicum annuum L.) is a potential source of antioxidants due to its high vitamin and lutein contents. The information on antioxidant activity and water content of chili are needed by the chili-based industries which utilise chili's natural antioxidant content for various purposes in health, food, or herbal products. These information are often obtained through destructive testing which damages the tested materials, and also requires a relatively long analysis time. Therefore, the present work aimed to non-destructively measure the antioxidant activity and water content of chili powder using near-infrared spectroscopy (NIS) combined with partial least square (PLS) analysis. A total of 84 samples of chili powder from four different colour groups were used in the present work. Approximately 70% of the samples were used for calibration, while the rest were used for validation. Samples' reflectance data were acquired using NIRFlex N500 Fiber Optic Solids Cells at wavelengths of 1000 - 2500 nm. The antioxidants activity and water content were also analysed destructively for comparison/reference data. PLS was then performed on the spectrum and reference using the Unscrambler X version 11.0 software. There were also three pre-treatments used to improve the spectrum, i.e., multiplicative scatter correction, first derivative Savitzky Golay, and de-trending. The best calibration model was successfully obtained when the antioxidant activity spectrum was processed using the de-trending pre-treatment with six PLS factors. The coefficient of determination (R2) and residual predictive deviation (RPD) values for this were 0.83 and 2.60, respectively. Additionally, the best calibration model was successfully obtained when the water content spectrum was processed using the first derivative Savitzky Golay pre-treatment with five PLS factors. The R2 and RPD values for this were 0.96 and 2.25, respectively. Both models were observed to be accurate, and have the potential to be used for measuring the antioxidant activity and water content of chili powder samples.

Identification and quantitation of phenolic compounds from the seed and pomace of Perilla frutescens using HPLC/PDA and HPLC-ESI/QTOF/MS/MS.

Abstract:INTRODUCTION:Perilla frutescens (L.) Britt., an essential traditional Asian crop and Chinese medicine, potentially exerts anti-oxidation effects through its phenolic compounds. These compounds have already been reported in perilla seed, however, little is reported in Perilla pomace, the primary waste during oil production of Perilla seed. OBJECTIVE:To investigate major phenolic compounds in perilla seeds and pomaces in order to check whether the pomace could be an alternative resource to the seed for nutritional and medical purposes. METHODS:Compounds in extracts of perilla seeds and pomaces were separated by high-performance liquid chromatography and detected by photodiode array, and by electrospray ionisation with quadrupole time-of-flight tandem mass spectrometry. Herb-markers selected by principal components analysis were then quantified in both seeds and pomaces. Moreover, a fingerprinting approach and multiple discriminant analysis were applied to screen the phenolic markers in 22 samples. RESULTS:Ten phenols were tentatively identified, among which four (rosmarinic acid, luteolin, apigenin and rosmarinic acid-3-O-glucoside) were selected as herb-markers. Perilla seeds and pomaces showed similar phenol profiles, however, the pomaces contained almost two times the amount of the four herb-markers than the seeds. CONCLUSION:The results indicated perilla pomace is a promising alternative source of phenolic compounds that could be recovered and potentially used as natural anti-oxidants.

Abstract:A method based on capillary electrophoresis with electrochemical detection has been developed to analyze flavonoids and phenolic acids in Perilla frutescens L. for the first time. Catechin, ferulic acid, apigenin, luteolin, rosmarinic acid, and caffeic acid are major important active ingredients in the plant. Operated in a wall-jet configuration, a 300 mu m diameter carbon-disk electrode was used as the working electrode, which exhibits a good response at 0.90 V (versus saturated calomel electrode) for the analytes. Under the optimum conditions, the analytes were baseline separated within 20 min in a 100 mmol/L borax buffer (pH 8.7). Notably, excellent linearity was obtained over 3 orders of magnitude with detection limits (S/N = 3) ranging from 2 x 10(-7) to 1 x 10(-6) g/mL for all analytes. This proposed method has been successfully applied to monitor the flavonoids and phenolic acids contents in the leaves and seeds of P. frutescens L. at different growth stages with relatively simple extraction procedures, and the assay results were satisfactory.
Human papillomavirus vaccination and the primary prevention of cancer: implications for survivors of childhood cancer.

Abstract:Effective vaccination is now available to prevent human papillomavirus (HPV), the most common sexually transmitted infection and the cause of cervical cancer, the second most common cancer among women worldwide. HPV vaccine uptake is particularly important for females surviving cancer, who are at high risk for HPV-related complication due to the direct and indirect effects of cancer therapy. Thus, Version 3.0 of the Children's Oncology Group Long-Term Follow-Up Guidelines for Survivors of Childhood, Adolescent and Young Adult Cancer recommends HPV vaccination for all eligible females surviving childhood cancer. Because this vaccine was only FDA approved in 2006, little is known about the complexity of vaccination uptake among those surviving childhood cancer. This chapter describes HPV vaccination and its usefulness in survivors of childhood cancer, provides a rationale for describing survivors as being at increased risk for HPV-related complication, identifies factors that are predictive of HPV vaccination, and discusses the utilization of these predictors in designing strategies to promote adherence to the HPV vaccination recommendations among survivors.

Abstract:The flavonoids from Perilla leaves were extracted using flash extraction assisted by ultrasonic extraction with ethanol. Subsequently, macroporous resin was employed for the isolation and purification of these flavonoids, followed by an investigation into their antioxidant activity. The process conditions for the extraction of flavonoids from Perilla leaves were designed and optimized using a one-way experiment combined with a response surface methodology. The optimal extraction conditions were determined as follows: the liquid–solid ratio was 20:1, ethanol volume fraction of 60%, ultrasound temperature of 60 °C, ultrasound time of 10 min and flash evaporation time of 60 s. The optimal extraction rate of flavonoids is 9.8 mg/g. In terms of separation and purification, a high-performance macroporous resin (HPD450 resin) with high purification efficiency was selected through static analysis and adsorption experiments. The optimal enrichment conditions were as follows: loading concentration of 0.06 mg/mL, optimal loading concentration of 20 mL, elution concentration of 70% and 76 mL, providing a reference for the further development and utilization of Perilla leaf flavonoids.

On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.
Drying the leaves of Perilla frutescens increases their content of anticancer nutraceuticals

Abstract:A regular intake of plant-derived bioactive agents has gained popularity because of the health benefits. Fresh leafy greens, however, normally have a low concentration of such bioactive agents. In this study, we found that drying markedly affected the accumulation of secondary metabolites and that dried leaves of Perilla frutescens L. (perilla) contained more anticancer flavonoids than fresh leaves. Drying is a major method of food preparation, particularly for plant-based foods, but the quality of the bioactive agents contained in the fresh and dried leaves of perilla has received only scant attention. Quantitative analysis of the concentrations of perillaldehyde, rosmarinic acid, apigenin, luteolin, 4-hydroxyphenyllactic acid, and 4-coumaric acid, some of which are known as nutraceuticals, revealed that the effect of drying significantly increased apigenin (28-fold) and luteolin (86-fold), but decreased rosmarinic acid in all leaf stages. We examined the positive effect on flavonoid levels on perilla leaves and confirmed that, by comparison with fresh perilla leaves, the dried leaves contained greater concentrations of anticancer flavonoids regardless of variety, form, or manner of cultivation. This indicates that drying can significantly increase the level of flavonoids in perilla leaves without a loss of flavor. Therefore, drying is a simple and effective method to improve the concentrations of bioactive agents, which increases the intake of beneficial substances derived from herbs and edible plants. This finding serves as a method for the supply of raw plant materials rich in bioactive agents that are suitable for labeling as edible nutraceuticals.

Abstract:Perillae Folium (PF), the leaf of Perilla frutescens (L.) Britt is extensively used as a culinary vegetable and medicinal herb in many countries. Purple PF (P. frutescens var. arguta) and green PF (P. frutescens var. frutescens) are two of the main varieties. In this study, UHPLC-Q-TOF/MS assisted with feature-based molecular networking (FBMN) was applied for chemical characterization. In total, 82 metabolites, mainly phenolic acid derivatives were identified from PF. With the help of FBMN, five organic acid glucosides together with three N-phenylpropenoyl-L-amino acids (NPAs) were identified in PF for the first time. Multivariate statistical methods were utilized for comprehensive comparison of purple and green PF profiles. 12 compounds with their relative contents varied significantly between purple and green PF were screened out. Overall, the present study offers valuable insights for chemical elucidation of PF which would be helpful for comprehensive utilization of PF resource.

Abstract:Dendrobium officinale (D. officinale), often used as a dual-use plant with herbal medicine and food applications, has attracted considerable attention for health-benefiting components and wide economic value. The antioxidant ability of D. officinale is of great significance to ensure its health care value and safeguard consumers' interests. However, the common analytical methods for evaluating the antioxidant ability of D. officinale are time-consuming, laborious, and costly. In this study, near-infrared (NIR) spectroscopy and chemometrics were employed to establish a rapid and accurate method for the determination of 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) scavenging capacity, 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging capacity, and ferric reducing antioxidant power (FRAP) in D. officinale. The quantitative models were developed based on the partial least squares (PLS) algorithm. Two wavelength selection methods, namely the genetic algorithm (GA) and competitive adaptive reweighted sampling (CARS) method, were used for model optimization. The CARS-PLS models exhibited superior predictive performance compared to other PLS models. The root mean square errors of cross-validation (RMSECVs) for ABTS, FRAP, and DPPH were 0.44%, 2.64 μmol/L, and 2.06%, respectively. The results demonstrated the potential application of NIR spectroscopy combined with the CARS-PLS model for the rapid prediction of antioxidant activity in D. officinale. This method can serve as an alternative to conventional analytical methods for efficiently quantifying the antioxidant properties in D. officinale.
Rapid Quantitative Determination of Multi-Indicators in Lonicerae Japonicae Flos by Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a set of quantitative calibration models based on near-infrared (NIR) spectroscopy and chemometrics methods for rapid determination of chlorogenic acid and luteoloside content in Lonicerae Japonicae Flos at the same time, which is useful for the quality control process. METHODS The PLSR algorithm was finally used to build NIR quantitative models between the spectra and the content of chlorogenic acid and luteoloside determined by the reference methods. The validation set was used to identify the predictive performance of the established models. RESULTS For PLSR models, the correlation coefficients of calibration and cross-validation were both >0.90, and the root mean square errors of cross validation (RMSECV) of two indicators were 0.469 and 0.012 4, respectively. External validation proved that the root mean square errors of prediction (RMSEP) were 0.478 and 0.010 5, which were close to RMSEC values, respectively. The ratio of RMSECV to RMSEP was close to 1, and the relative standard errors (RSEP) for chlorogenic acid and luteoloside content were 10.19% and 14.76%. This established method was initiatively proved to be accurate and stable. CONCLUSION NIR can be used for the rapid determination of chlorogenic acid and luteoloside content in Lonicerae Japonicae Flos at the same time, which is beneficial and crucial for the quality control of this raw material in TCM pharmaceutical factory.

Abstract:The global demand for medicinal plants is increasing. The quality of plants grown outdoors, however, is difficult to control. Myriad environmental factors influence plant growth and directly impact biosynthetic pathways, thus affecting the secondary metabolism of bioactive compounds. Plant factories use artificial lighting to increase the quality of medicinal plants and stabilize production. Photosynthetic photon flux density (PPFD) and electrical conductivity (EC) of nutrient solutions are two important factors that substantially influence perilla (Perilla frutescens, Labiatae) plant growth and quality. To identify suitable levels of PPFD and EC for perilla plants grown in a plant factory, the growth, photosynthesis, and accumulation of secondary metabolites in red and green perilla plants were measured at PPFD values of 100, 200, and 300 μmol m-2 s-1 in nutrient solutions with EC values of 1.0, 2.0, and 3.0 dS m-1. The results showed significant interactive effects between PPFD and EC for both the fresh and dry weights of green perilla, but not for red perilla. The fresh and dry weights of shoots and leafy areas were affected more by EC than by PPFD in green perilla, whereas they were affected more by PPFD than by EC in red perilla. Leaf net photosynthetic rates were increased as PPFD increased in both perilla varieties, regardless of EC. The perillaldehyde concentration (mg g-1) in red perilla was unaffected by the treatments, but accumulation in plants (mg per plant) was significantly enhanced as the weight of dry leaves increased. Perillaldehyde concentrations in green perilla showed significant differences between combinations of the highest PPFD with the highest EC and the lowest PPFD with the lowest EC. Rosmarinic acid concentration (mg g-1) was increased in a combination of low EC and high PPFD conditions. Optimal cultivation conditions of red and green perilla in plant factory will be discussed in terms of plant growth and contents of medicinal ingredients.

Abstract:Perillae Folium (PF), the leaf of Perilla frutescens (L.) Britt, is extensively used as culinary vegetable in many countries. It can be divided into two major varietal forms based on leaf color variation, including purple PF (Perilla frutescens var. arguta) and green PF (P. frutescens var. frutescens). The aroma of purple and green PF is discrepant. To figure out the divergence of chemical composition in purple and green PF, gas chromatography–tandem mass spectrometry (GC-MS) was applied to analyze compounds in purple and green PF. A total of 54 compounds were identified and relatively quantified. Multivariate statistical methods, including principal component analysis (PCA), orthogonal partial least-squares discrimination analysis (OPLS-DA) and clustering analysis (CA), were used to screen the chemical markers for discrimination of purple and green PF. Seven compounds that accumulated discrepantly in green and purple PF were characterized as chemical markers for the discrimination of the purple and green PF. Among these 7 marker compounds, limonene, shisool and perillaldehyde that from the same branch of the terpenoid biosynthetic pathway were with relatively higher contents in purple PF, while perilla ketone, isoegomaketone, tocopheryl and squalene on other branch pathways were higher in green PF. The results of the present study are expected to provide theoretical support for the development and utilization of PF resources.

A TLC-direct Bioassay Method for Detection of Anti-Lipid Peroxidation Constituents from Fruits of Perilla Frutescens

Abstract:A practical TLC-direct bioassay method was established for screening and evaluating antioxidant activity. The method was based on series of reactions. Irradiation of linoleic acid with UV 254 nm produced lipid peroxides. Upon heating, the peroxides were converted into malondialdehyde, which combined with thiobarbituric acid to a red pigment with a maximum absorption at 535 nm. Following a TLC separation, substance with anti-lipid peroxidation activity showed white spots/zones against red background on the TLC plate. The proposed method was established by the dot blot test, and validated with respect to selectivity, sensitivity, linearity, precision, and recovery after optimizing key experimental parameters. In an exemplary screening for antioxidant constituents from Perilla fruits, two active zones were discovered and tentatively characterized as salviaflaside and rosmarinic acid by TLC-interface-ESI-MS, respectively. Further verification by HPTLC-bioassay-guidedisolation was carried out and in addition to salviaflaside and rosmarinic acid, also luteolin 5-O-& beta;-D-glucopyranoside and 3 & PRIME;-dehydroxyl-rosmarinic acid-3-O-& beta;-D-glucoside were discovered to show prominent anti-lipid peroxidation activity.

Abstract:Perilla frutescens was an annual herb considered as one of the "One Root of Medicine and Food", and it was used widely for food and medical treatment. Its main active ingredients were volatile organic compounds (VOCs), but it was easily affected during drying and storage. The leaves, seeds and stems had shown differences in therapeutic effects, but the underlying reason remained unclear. In the present work, headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) and headspace solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) were utilized to effectively and comprehensively analyze VOCs of fresh and dried P. frutescens between the leaves, stems and seeds. Meanwhile, chemometric analysis was applied to compare and identify characteristic volatile markers. As a result, 60 VOCs were identified by HS-GC-IMS and 115 VOCs were identified by HS-SPME-GC–MS from P. frutescen . 25 potential volatile markers were selected based on a combination model of orthogonal partial least squares discriminant analysis (OPLS-DA) and random forest (RF). The models could be used to analyze the variation between fresh and dried P. frutescens and distinguish the differences in different parts effectively and comprehensively. It was the first research regarding the method development of HS-GC-IMS and HS-SPME-GC–MS that comprehensively analyzes the VOCs characterization of fresh and dried P. frutescens in different parts and the findings obtained would evaluate the quality and provide a reference for further exploration of the edible and medicinal effects of P. frutescens . Abstract:Fine particulate matter (PM2.5) exposure worsens chronic respiratory diseases through oxidative stress and inflammation. Perilla frutescens (L.) has potential respiratory protective properties, but the impact of growth stages on its beneficial metabolites is unclear. We aimed to evaluate how different growth stages affect phenolic acids, flavonoids, and polycosanols in perilla seeds and flowers and their efficacy in countering PM2.5-induced damage. Perilla seeds and flowers from five varieties at 10, 20, 30, and 40 days post-flowering were analyzed for metabolite content. Their antioxidant, anti-inflammatory, and respiratory protective effects were tested in RPMI 2650 cells. Our findings indicated that perilla flowers contained higher levels of functional components than seeds and exhibited significant variation with maturation. Phenolic acids of perilla flowers were highest at the early stages of maturation after flowering. However, individual flavones of perilla flowers were the highest at the late maturation stages after flowering. Extracts from perilla flowers harvested 20 days after flowering exhibited significant respiratory protection, effectively inhibiting inflammatory cytokines, mucus secretion, and oxidative stress markers. In conclusion, the flower parts of perilla, particularly those harvested 20 days after flowering, are useful materials for obtaining phenolic compounds, including rosmarinic acid, with high antioxidant and respiratory enhancement effects.

Abstract:Ethnopharmacology relevanceChrysanthemum morifolium Ramat. (Flos Chrysanthemi, FC) the most economically significant “food and drug dual-use” plants, with positive effects on relieving eye fatigue, and reduce internal heat, shows significant activities, such as anti-inflammatory, antioxidant, and neuroprotective, as well as alleviating diabetes effects.Aim of the studyThis study was undertaken to a screening of natural antioxidants in five kinds of medicinal FC and development of an integrated quality control method based on the antioxidant activity.Materials and methodsA novel quality control method for FC was established by combining the on-line HPLC-DPPH, ESI-MS, and NIR spectra analysis. Firstly, the on-line HPLC-DPPH-MS system was employed to identify the antioxidants in FC extracts. Then, the relationship between the NIR spectra and antioxidant activities of FC samples was calibrated to evaluate the total antioxidant capacity of FC rapidly.ResultsThe established antioxidant activity-fingerprints contain both chemical information and antioxidant activity characteristics of FC. A total of 16 antioxidants were identified by on-line HPLC-ESI-MS analysis. The results of heat map analysis and cluster analysis showed that the classification method based on antioxidants in FC can be used to identify different cultivars of FC. The optimal pretreatment of the NIR spectra was determined to be row center (RC) 1st der + multiple-scatter correction (MSC) with an optimal LV value of 11. The developed spectral-antioxidant activity model had the excellent predictive ability and was successfully used to evaluate new batches of FC samples, where Rcal = 0.9445 and Rval = 0.8821.ConclusionsThis comprehensive strategy may prove to be a powerful technique for the rapid screening, identification, and activity prediction of antioxidants, which could be used for the quality control of FC, and can serve as reference for design of quality control of other herbs and foods samples.

Abstract:Perilla (Perilla frutescens L.) is a time-honored herbal plant with widespread applications in both medicine and culinary practices around the world. Profiling the essential organs and tissues with medicinal significance on a global scale offers valuable insights for enhancing the yield of desirable compounds in Perilla and other medicinal plants. In the present study, genome-wide RNA-sequencing (RNA-seq) and assessing the global spectrum of metabolites were carried out in the two major organs/tissues of stem (PfST) and leaf (PfLE) in Perilla. The results showed a total of 18,490 transcripts as the DEGs (differentially expressed genes) and 144 metabolites as the DAMs (differentially accumulated metabolites) through the comparative profiling of PfST vs PfLE, and all the DEGs and DAMs exhibited tissue-specific trends. An association analysis between the transcriptomics and metabolomics revealed 14 significantly enriched pathways for both DEGs and DAMs, among which the pathways of Glycine, serine and threonine metabolism (ko00260), Glyoxylate and dicarboxylate metabolism (ko00630), and Glucagon signaling pathway (ko04922) involved relatively more DEGs and DAMs. The results of qRT-PCR assays of 18 selected DEGs confirmed the distinct tissue-specific characteristics of all identified DEGs between PfST and PfLE. Notably, all eight genes associated with the flavonoid biosynthesis/metabolism pathways exhibited significantly elevated expression levels in PfLE compared to PfST. This observation suggests a heightened accumulation of metabolites related to flavonoids in Perilla leaves. The findings of this study offer a comprehensive overview of the organs and tissues in Perilla that have medicinal significance.

Abstract:The effects of three extraction methods on the yields, physicochemical properties, stabilities and antioxidant activities of anthocyanins from perilla frutescens (L.) Britt. were investigated and compared. Three extraction methods were applied to extract perilla leaves anthocyanins (PLAs), which include ultrasonic-assisted water extraction (UWE), ultrasonic-assisted ethanol extraction (UEE) and ultrasonic-assisted aqueous two-phase extraction (UATPE). The yields, color quality, stabilities and antioxidant activities of three anthocyanins were significantly different. More specifically, UATPE-PLAs had the maximum total anthocyanins content (18.37 ± 0.91 mg/g) and the best color valence (30.01 ± 0.08). After storage at 4 °C for 15 days, the retention rate of the UATPE-PLAs (95.52 ± 1.29%) was higher than UEE-PLAs (89.73 ± 1.34%) and UWE-PLAs (62.22 ± 3.85%). And the UATPE-PLAs were stable at pH 3.7, while the UWE-PLAs and the UEE-PLAs were stable at pH 2.5. What's more, UATPE-PLAs displayed higher DPPH radical scavenging (98.21 ± 0.04%), reduction power (IC 50 = 0.58 mg Trolox equivalent/mL), hydroxyl radical scavenging ability (89.45 ± 0.01%) and anti-lipid peroxidation ability (87.72 ± 0.03%).
