Abstract:Objective To establish a method for the separation of circulating tumor cells in the peripheral blood based on the magnetic actived cell sorting(MACS) technique and evaluate its clinical value. Methods Monoclonal antibodies to the gastrointestinal carcinoma cell membrane epitopes MG7, CEA, QY and MMP9 were conjugated to immunomagnetic beads with CNBr\|actived Sepharose 4B. Using these immunomagnetic beads the carcinoma cells were separated from the peripheral blood and portal venous blood. Results In 29 of 59(49.15%) samples from peripheral blood and 28 of 31(90.32%) samples from portal venous the carcinoma cells were determined using the magnetic cell sorting (MACS). We have found that there was no relationship between the samples of separated tumor cells and TNM stage. Conclusions MACS appears to be an efficient technique to detect disseminated tumor cells. It can separate carcinoma cells from the peripheral blood with a few carcinoma cells in gastrointestial carcinoma patient. It will contribute to studying the etiology, development, metastasis, and multidrug resistance of tumor.
CD34+ stem cell recovery after positive selection of "overloaded" immunomagnetic columns

Abstract:Techniques for CD34+ cell enrichment of hematopoietic progenitor cells in grafts destined for transplantation of certain patients with the aim of lowering the amount of infused T lymphocytes and subsequently decreasing the risk of graft versus host disease (GVHD) have been well developed. Adaptations of these techniques should be useful for isolation of other phenotypically defined stem cells. However, a major limitation of techniques now available consists of the number of total nucleated cells or phenotypically defined stem cells that can be processed in a single procedure. Here, we show that recommended levels are much lower than the levels of cells that can be effectively processed by immunomagnetic sorting. Twenty-nine procedures were performed using the Clini- MACS (Miltenyi Biotec) device, which is recommended for processing <6x10(10 )total nucleated cells or <6x10(8) CD34+ cells. Procedures were divided in groups according to their total cellular or CD34+ cell content. We achieved a median CD34+ cell recovery of 68.60% with a median purity of 98.56%, regardless of the loading dose when samples possessing 2-10x10(10) total nucleated cells and 0.8-12.5x10(8) CD34+ cells were applied to a single column. The median levels of CD3+ cells and CD19+ cells in the final product were depleted by 5 logs and 3.8 logs, respectively; no differences were noted when the initial loading dose was increased. Moreover, we found no correlation between the total number nucleated or CD34+ cells loaded and the resultant CD34+ cell recovery. In conclusion, levels of both total nucleated cells and CD34+ can be processed in a single procedure with satisfactory and similar CD34+ cell recovery when these columns are loaded with up to two times as many cells as recommended.

Abstract:A real-time, compact architecture is presented for translation-invariant windowed nonlinear discrete operators represented in computational mathematical morphology. The architecture enables output values to be computed in a fixed number of operations and thus can be pipelined. Memory requirements for an operator are proportional to its basis size. An operator is implemented by three steps: 1) each component of a vector observation is used as an index into a table of bit vectors; 2) all retrieved bit vectors are "ANDed" together; and 3) the position of the first nonzero bit is used as an index to a table of output values. Computational mathematical morphology is described, the new architecture is illustrated through examples, and formal proofs are given. A modification of the basic architecture provides for increasing operators.

Abstract:Objective Separating & purifying CD34+ cell from umibilical cord blood is a hinge of stem celi transfer and in-vitro culture MNC were isolated from umbilical cord blood by using denshy grodlent centrifugation(Ficoll-Hypaque)or 3 g/dl gelatin matural sedimentation methods,then separating CD34+ cell from MNC by MACS.Results show,the number of MNC and CD34+ cell were(5.76±0.67)×107/ml and(5.53±1.16)×106/ml respectively by 3g/dl gelstin natural sedimentation.which was better than that from Ficoll-Hypaque method.So,3g/dl gelatin natural sedimentation is an optimal method for separating MNC.collaboration with MACs can separate and purify CD34+ surccessfully.

Abstract:Objective To investigate Density Gradient Centrifugation and Magnetic Activated Cell Sorting combined separation of fetal nucleated red blood cells(FNRBCs) from cord blood. Methods 5 cases of cord blood FNRBCs separated by Density Gradient Centrifugation and Magnetic Activated Cell Sorting in turn. Using Fluorescence Activated Cell Sorting (FACS) technique and cell staining to conducted purity analysis.Results 1. FACS detectes the purity of CD71+ FNRBCs from cord blood separated by CD71 monoclonal antibody beads can reach to 81% while only 23.2% before the election. 2. Pre-sorting of cord blood smear shows a large number of mature red blood cells and contains little FNRBCs.After Density gradient centrifugation,most cells were mononuclear cells included lymphocytes,monocytes and FNRBCs and little red cells.CD71+ cells separated by CD71 monoclonal antibody beads are most FNRBCs which have basophilic cytoplasm.Conclusion Density Gradient Centrifugation and Magnetic Activated Cell Sorting combined separation can increase the sorting purity of FNRBCs from cord blood.

Abstract:A method is described for the preparation of human peripheral blood mononuclear cell (PBMC) suspensions containing highly enriched natural killer (NK) cell cytotoxicity. The technique involved the negative selection of OKM1+ cells by the selective removal of nylon wool nonadherent PBMC which are reactive with the Leu-1 monoclonal antibody. The Leu-1+ cells are removed by subsequent rosette formation with anti-mouse IgG coated bovine erythrocytes. The resultant OKM1+ cell suspension had a greater number of large granular lymphocytes, K562 target binding effector cells, and lytic activity than concomitantly prepared fractions of Percoll gradients.
Are CD133 and CD271 Useful in Positive Selection to Enrich Umbilical Cord Blood Mesenchymal Stem Cells

Abstract:It is well accepted that umbilical cord blood has been a source for mesenchymal stem cells. However, there was less success in culturing these cells by using traditional method. Lots of studies showed that CD271 (low affinity nerve growth factor receptor LNGFR) and CD133 could be used to positive sort of bone marrow-derived mesenchymal stem cells (MSC) in recent years. The present study was designed to explore whether the method of positive sorting of umbilical cord blood-derived MSC (UCB-MSC) was effective by using CD271 and CD133 immunomagnetic beads. The ability of forming CFU-F and differentiation capacity of the four kinds of cells, namely CD271+, CD271-, CD133+ and CD133- were analysed. The results indicated that the purities of immunomagnetically selected CD271+ and CD133+ cells were (87.58±0.48)% and (89.89±8.10)% respectively. More than 99% of CD271+ and CD133+ cells expressed CD34 and CD45, the markers of hematopoietic stem cells. CFU-F assay showed that CD271+ and CD133+ cells could not form any CFU-F and only few single fibroblast-like cells could be found in the dishes. However, part of the negative samples (27%) could form CFU-F. Phenotype of cells (passage 3) isolated by the two methods was the same, that was CD34-, CD45-, CD14-, CD29+, CD44+, CD73+, CD105+ and CD90+. They both had osteogenic and adipogenic differentiation potential. It is concluded that nearly all the CD271 and CD133 positive cells are hematopoietic cells, MSC can be effectively collected by negative selection with immunomagnetic beads against CD271 and CD133.

Abstract:Background aims Cord blood mononuclear cells (CBMCs) comprise a variety of single-nucleated cells found in the cord blood, mainly consisting of monocytes and lymphocytes. They also include a smaller proportion of other cell types, such as hematopoietic stem and progenitor cells (HSPCs) and mesenchymal stromal cells (MSCs). CBMCs are vital for acquiring hematopoietic stem and progenitor cells, MSCs, and other immune cells, like natural killer cells. These cells are essential for supporting subsequent research and clinical applications. Although automated equipment for CBMC enrichment has shown promise, the high cost of these machines and the expense of disposable consumables limit their routine use. Furthermore, limited information is available on manual strategies for isolating CBMCs from cryopreserved cord blood. Therefore, we aimed to optimize the dilution buffer and refine the isolation procedure for CBMCs. Methods We enhanced the CBMC recovery rate from cryopreserved cord blood using an optimized dilution buffer and a modified isolation procedure. Results We achieved average recovery rates of 42.4 % and 54.3 % for CBMCs and CD34+ cells, respectively. Notably, all reagents used in the isolation procedure were of GMP-grade or pharmaceutical preparations, underscoring the potential clinical benefits of our strategy. Discussion We devised an optimized protocol suitable for routine research and clinical applications for enhanced recovery of CBMCs from cryopreserved cord blood units using an optimized dilution buffer and a modified isolation procedure.

Abstract:Immunomagnetic colloids have the properties of solutions and, therefore, offer distinct advantages in their ability to bind to cells as compared to larger magnetic microspheres. B cells and T cells have been isolated with higher degrees of purity following incubation with monoclonal antibodies (MoAbs) and a goat anti-mouse Ig ferrofluid. This was demonstrated both with mixtures of cell lines and lines titrated into normal bone marrow. In all cases, low non-specific binding was observed. Maximal cell capture was obtained without washing out excess MoAb, resulting in cell separations that could be completed in under 30 min. The system permits not only the magnetic capture of cells onto pins in a high gradient separator, but also their recovery. Cells separated in this way appear to be highly viable and it is possible to manipulate them further as the immunocolloid is sufficiently small for it not to interfere with tests such as indirect immunofluorescence. The advantages and disadvantages of immunocolloids versus larger magnetic microspheres are discussed.

Abstract:Background: This study aimed to compare the IMG counting by an auto hematology analyzer with the flow cytometric enumeration of CD34+ cells. Methods: All data from 124 samples submitted to the hematology laboratory for CD34+ cell counting in 2019 and 2020 were retrospectively evaluated. Whole blood samples were taken into EDTA tubes and assayed within 2 hours. The numbers and percentages of WBC and IMG were determined by using Mindray BC6200 hematology analyzer, while the same were determined for CD45 and CD34+ cells by using Beckman Coulter Navios flow cytometer. The performance of the new method was determined by the receiver operating characteristic (ROC) analysis. Results: Out of the 124 samples, 94 were collected from healthy individuals and 30 were collected from patients. A significant correlation was found between IMG and CD34+ cell counts in all patients (r = 0.71, p = 0.000) at the cutoff values of > 20/µL and > 50/µL. The Bland-Altman analysis of all patients showed that there was an agreement between the two methods. When the cutoff value of 20/µL for CD34+ cells was used in the ROC analysis, the sensitivity and specificity were calculated as 100 (96.1 - 100) and 96.77 (83.3 - 99.9), respectively, for the IMG count of > 900/µL. Conclusions: An IMG count of 900/µL can be used as the cutoff value in the analysis with the Mindray BC6200. The IMG counting cannot replace the flow cytometric CD34+ cell enumeration but can be used in the selection of the samples for stem cell enumeration by flow cytometry.

Abstract:While the single-platform flow cytometric CD34+ cell counting method is the preferred choice to predict the yield of mobilized peripheral blood stem cells, most flow cytometers lack the ability of hematology counter analyzers to perform volumetric counting. However, one of the problems using reference microbeads is the vanishing counting bead phenomenon. This phenomenon results in a drop in microbeads concentration and reduces the total and relative number of beads in calibration procedures. In the last years, flow cytometers including a volumetric system to quantify cells have been developed and may represent a promising alternative to enumerate CD34+ cells avoiding the use of beads. In this study we have used a direct true volumetric counting of CD34+ cells under continuous flow pump to overcome potential drawbacks with impact in rare cell analysis. To confirm this hypothesis, we have compared the results of CD34+ cell enumeration using non-volumetric vs. volumetric systems with FC500 (Beckman Coulter) and Attune NxT (ThermoFisher) flow cytometers, respectively, in mobilized peripheral blood samples. No statistically significant differences were observed between measurements of CD34+ cells using beads, when the FC500 and Attune NxT absolute counting values were compared, or when CD34+ counts were compared on the Attune NxT, either using or not using beads. Linear regressions to study the relationship between volumetric and non-volumetric CD34+ counts confirmed the accuracy of each method. Bland-Altman test showed agreement between both methods. Our data showed that CD34 + cell enumeration using a volumetric system is comparable with current counting systems. This method represents an alternative with the advantage of the simplification of sample preparation and the reduction of the analysis subjectivity.

High gradient magnetic cell separation with MACS.

Abstract:A flexible, fast and simple magnetic cell sorting system for separation of large numbers of cells according to specific cell surface markers was developed and tested. Cells stained sequentially with biotinylated antibodies, fluorochrome-conjugated avidin, and superparamagnetic biotinylated-microparticles (about 100 nm diameter) are separated on high gradient magnetic (HGM) columns. Unlabelled cells pass through the column, while labelled cells are retained. The retained cells can be easily eluted. More than 10(9) cells can be processed in about 15 min. Enrichment rates of more than 100-fold and depletion rates of several 1,000-fold can be achieved. The simultaneous tagging of cells with fluorochromes and very small, invisible magnetic beads makes this system an ideal complement to flow cytometry. Light scatter and fluorescent parameters of the cells are not changed by the bound particles. Magnetically separated cells can be analysed by fluorescence microscopy or flow cytometry or sorted by fluorescence-activated cell sorting without further treatment. Magnetic tagging and separation does not affect cell viability and proliferation.

Isolating Human Peripheral Blood Mononuclear Cells from Buffy Coats via High Throughput Immunomagnetic Bead Separation

Abstract:Peripheral blood mononuclear cells (PBMCs) are a heterogeneous population of monocytes and lymphocytes. Cryopreserved PBMCs have stable viability in long-term storage, making them an ideal cell type for many downstream research purposes, including flow cytometry, immunoassays, and genome sequencing. Typically, PBMCs are isolated via density gradient centrifugation, however, it is a low-throughput workflow that is difficult and costly to scale. This article presents a high-throughput workflow using a magnetic bead-based PBMC isolation method that is quick to implement. Total cell concentration, viability, and population distribution with PBMCs obtained using density gradient isolation were compared, and cell viability and proportion of cell types were comparable for both techniques. Isolated PBMCs demonstrated over 70% viability up to 9 days after blood collection, although yield decreased by half after 5 days compared to PBMCs processed within 24 h of collection. In summary, this article describes a PBMC protocol that utilizes a bead-based approach to adapt to a high throughput workflow and demonstrates that both manual and automated bead-based methods can increase processing capacity and provide flexibility for various budgets.
Magnetic Separation of Cell-Secreted Vesicles with Tailored Magnetic Particles and Downstream Applications

Abstract:The analysis of the receptors on the surface of the cell-secreted vesicles provides valuable information of the cell signature and may also offer diagnosis and/or prognosis of a wide range of diseases, including cancer.Due to their low concentration, conventional procedures for extracellular vesicle (EV) detection usually require relatively large sample volumes, involving preliminary purification or preconcentration steps from complex specimens. Here, we describe the separation and preconcentration in magnetic particles of extracellular vesicles obtained from cell culture supernatants from MCF7, MDA-MB-231, and SKBR3 breast cancer cell lines, human fetal osteoblastic cell line (hFOB), and human neuroblastoma SH-SY5Y cell line, as well as exosomes from human serum. The first approach involves the covalent immobilization for the exosomes directly on micro (4.5 μm)-sized magnetic particles. The second approach is based on tailored magnetic particles modified with antibodies for further immunomagnetic separation of the exosomes. In these instances, micro (4.5 μm)-sized magnetic particles are modified with different commercial antibodies against selected receptors, including the general tetraspanins CD9, CD63, and CD81 and the specific receptors (CD24, CD44, CD54, CD326, CD340, and CD171). The magnetic separation can be easily coupled with downstream characterization and quantification methods, including molecular biology techniques such as immunoassays, confocal microscopy, or flow cytometry.

Abstract:OBJECTIVE:To explore the biological characteristics of mesenchymal stem cells (MSC) derived from umbilical cord blood (UCB) and their supporting capacities in ex vivo expansion of hematopoietic stem/progenitor cells (HSPCs).METHODS:Low-density mononuclear cells (MNCs) from UCB were cultured in IMDM containing 20% FBS to form confluent adherent cells through 15 passages. Some cytokines in the conditioned medium were determined with ELISA. UCB-derived adherent cells were displayed with antibodies and analyzed with flow cytometry. The supporting capacity of UCB-derived adherent cells for ex vivo expansion of CD34(+) cells was assayed by co-culture in a two step culture. UCB-derived adherent cells were induced for chondrogenic differentiation with chondrogenic medium, and the induced cells were analyzed for the type II pro-collagen gene expression with RT-PCR.RESULTS:The mean number of adherent fibroblast like colonies derived from UCB was (3.5 +/- 0.7)/10(6) MNCs. UCB-derived MSCs could survive for at least 15 passages of expansion. In their undifferentiated status, UCB-derived MSCs were CD13(+), CD29(+), CD90(+), CD105(+), CD166(+), SH2(+), SH3(+), SH4(+), CD45(-), CD34(-), and CD14(-). Stem cell factor (SCF), interleukin 6 (IL-6) and tumor necrosis factor alpha (TNF-alpha) could be detected in the supernatant of the cultures. The MSCs cultured in chondrogenic media could differentiate into chondrogenic cells and express type II pro-collagen mRNA. UCB-derived MSCs could support the proliferation and differentiation of UCB CD34(+) cells in vitro.CONCLUSION:UCB-derived MSCs are similar to those derived from adult bone marrow and can support the proliferation of hematopoietic stem/progenitor cells.

Abstract:Abstract Background Accurate measurement of CD34-positive hematopoietic stem cells (HSCs) is critical for successful peripheral blood stem cell transplantation (PBSCT). We have developed a new image-based cell counter, Microscanner C3 (MSC3, Biozentech, South Korea) as an alternative method for enumeration of CD34-positive HSCs and evaluated its performance. Methods 100 μl of apheresis product was stained with CD45-BB515 and CD34-PE antibodies (Becton Dickinson, USA) at room temperature in the dark. Red blood cell lysis with 500 μl of OptiLyze C solution (Beckman Coulter, USA) was followed by centrifugation. After the removal of supernatants, the sample was resuspended with 500 μL PBS, which then infused to filter paper imbedded BZ-1 microchip (Biozentech, South Korea). Thirty-fields of the loaded microchip was auto-scanned by MSC3 and its dedicated software analyzed the images and yielded CD34-positive cell fraction out of CD45-positive cells as percentage. Results of thirty-six samples were compared to that of FacsLyric (Beckton Dickinson, USA) flow cytometry and accuracy and imprecision (pentaplicate measurements for three samples), repeatability (two experimenters performed triplicate measurements for three samples each), linearity (triplicate measurements for five levels), limit of detection (hexaplicate measurements for five levels) and specificity (using whole blood specimen from three donors with monoclonal gammopathy, thrombocytosis or monocytosis) was evaluated. Results CV% values of inaccuracy, imprecision and repeatability did not exceed 15%. The method was found to be linear across the measuring range. LOD was evaluated to be 7 cells/μL and no background interference was detected. The difference between the measured fraction of MSC3 and FacsLyric was within allowable limit, with an R-value greater than 0.98. Conclusion We developed a reliable method for quantifying CD34-positive HSCs based on image analysis. The limitation involved in MSC3 applications in CD34-positive HSCs is that, like dual-platform flow cytometry, the results should be combined with the results of conventional hematology analyzer in order to define absolute HSC numbers. However, with its simpler and faster procedures, MSC3 exhibited a great potential to be used as an alternative to conventional flow cytometry in CD34-positive HSCs enumeration and other applications as well.
