Abstract:OBJECTIVE:To induce differentiation of CD(34)+ cells from varying sources into epithelial cells for potential application of blood vessel prostheses.METHODS:CD(34)+ cells were isolated from canine peripheral blood and bone marrow or human umbilical cord blood by an immune magnetic cell sorting system. The isolated CD(34)+ cells were induced to differentiate into endothelial cells in a liquid culture with VEGF. Endothelial cells were evaluated by immunocytochemistry and transmission electron microscopy. CD(34)+ cells were seeded on PTFE prostheses, which were harvested and observed by electron microscopy.RESULTS:The average isolated CD(34)+ cells from peripheral blood, bone marrow and umbilical cord blood were (26.30+/-2.42)%,(41.84 +/-3.65)%and (74.62+/-4.46)%, respectively. The number of CD(34)+ cells increased with the culture duration and reached to the highest level at the 14th d. Immunostaining showed positive signal for CD31 in endothelial cells and positive for factor VIII in cell culture. Transmission electron microscopy found Weibel-Palade bodies in the cytoplasm. Scanning electron microscopy demonstrated a single- layer of flat cells in the innermost layer of PTFE prostheses, and the cells were not of tropism.CONCLUSION:CD(34)+ cells isolated from peripheral blood, bone marrow and umbilical cord blood can be induced into endothelial cells; bone marrow and umbilical cord blood can be used as the sources of seeding cells for endothelialization of prostheses.
E-Selectin Mediated Adhesion and Migration of Endothelial Colony Forming Cells is Enhanced by SDF-1α/CXCR4

Abstract:OBJECTIVE:Endothelial-colony forming cells (ECFCs) can be readily expanded from human umbilical cord blood and can facilitate repair of endothelial injury. E-selectin and SDF-1α are produced following endothelial injury and can regulate endothelial progenitor homing. Mechanisms of vascular repair specific to the mode of injury have not been well described in homogenous cell populations such as ECFCs and are needed for development of more effective vascular repair strategies.METHODS AND RESULTS:Lipopolysaccharide (LPS)-induced endotoxic injury to mature human umbilical vein endothelial cells (HUVEC) was compared with hypoxic and radiation injury. E-selectin expression in HUVEC cells is markedly increased (208-fold) following LPS-induced injury and facilitates increased ECFC adhesion and migration function in vitro. SDF-1α expression remains unchanged in LPS-treated HUVEC cells but increases more than 2 fold in fibroblasts undergoing similar endotoxic injury. SDF-1α induces expression of E-selectin ligands on ECFCs and facilitates greater E-selectin-mediated adhesion and migration of ECFCs in a CXCR4-dependent manner. Induction of E-selectin expression in HUVECs following hypoxic or radiation injury is negligible, however, while SDF-1α is increased markedly following hypoxia, highlighting injury-specific synergism between mediators of vascular repair.CONCLUSION:E-selectin mediates adhesion and migration of ECFCs following endotoxic endothelial injury. SDF-1α augments E-selectin mediated ECFC adhesion and migration in a CXCR4-dependent manner.
Biological Properties of Endothelial Progenitor Cells Derived from Human Peripheral Blood Mononuclear Cells

Abstract:Objective To develope the methods of isolating and culturing endothelial progenitor cells(EPCs)from adult peripheral blood mononuclear cells,and to characterize the biological properties of EPCs.Methods Mononuclear cells were isolated by density-gradient centrifugation and incubated in fibronectin-coated dishes with endothelial medium in the presence of vascular endothelial growth factor.After cultivation,cells were sequential replanted by allowing cells to adhere to tissue culture dishes during two successive 2-houre incubations in order to remove nonadherent cells.Medium was changed every second day,and the number of early colonies was counted at 7 days after plating.Furthermore,the samples were divided into 2 aliquots and cells from one aliquot were cultivated continually until the late colonies generated.At the same time,another aliquot was tested when the early colonies can be observed after 7 days cultivation.Flow cytometry analysis was used to evaluate cells surface antigen expression and the concentration of vascular endothelial growth factor(VEGF)in cultivation medium was determined with ELISA method,and the capacity of vascular genesis was assessed by plating cells onto collagen gels.Results Early colonies were identified as spindle like sprouting cells radiating from a central core of round cells.When replan-ted,the early colonies failed to form second colony,and were devoid of the capacity of in vitro vascular genesis.Furthermore,cells derived from the early colonies expressed mainly CD14 and CD45,and concentration of VEGF in conditioned medium originated from early colonies increased obviously.In contrasted to early colonies,the late colonies did not emerge until 21 to 28 days after cultivation and exhibited typical endothelial cells properties.Remarkably,cells originated from late colonies had the ability to form second endothelial colonies after replanted and generated tube-like structures when seeded onto collagen gels.In addition to the clearly morphological differences,cells derived from late clones showed increased CD146(P0.01)expression and reduced CD45 and CD14 expression(P0.001).Conclusion Under endothelial cultivating conditions,human peripheral blood mononuclear cells can generate early and late colonies.The majority of cells derived from early colonies belongs to monocyte linage and can secrete VEGF,but failed differentiate into endothelial cells.Alternatively,cells originated from late colonies exhibit morphologically and biologically characteristics of EPCs.
Acquiring and Identification of Endothelial Progenitor Cells from Human Peripheral Blood Mononuclear Cells by Sequential Adherence

Abstract:Aim:To explore the ways of isolation and cultivation for endothelial progenitor cell (EPC) from adult peripheral blood mononuclear cells and study the biological properties of EPC.Methods:Mononuclear cells were isolated by density-gradient centrifugation and incubated on fibronectin-coated dishes in endothelial medium supplemented with vascular endothelial growth factor. After that, the cells were divided into 2 groups of A and B. Cells from group A were cultured with common method and group B cells were handled with sequential adherence method. In group A, non-adherent cells were discarded with washing after 4 days cultivation and medium was changed every other days. At the same time, group B cells were sequential planted by allowing cells to adhere to tissue culture dishes during two successive 2-hour incubations in order to remove non-adherent cells. In both groups, the early clones were counted at 7 days and cultivation continued until the late clones generated. Flow cytometry analysis was used to evaluate cells surface antigen expression and cells binding Ulex europaeus agglutinin (UEA)-1 of the early and late clones in group B was detected by direct fluorescence staining.Results: There were no differences about the early clone numbers between two groups, but the counts of late clones were obviously increased and CD3 expression was also decreased in group B (P0.05).With clearly morphological differences, the late clones occurred at 21 to 28 days after cultivation and the expression of CD45 and CD14 on cells derived from late clones were significantly reduced but CD146 expression increased clearly compared with those from early clones(P0.01 or 0.001).When replanted, only the late clones generated second endothelia like clones.Conclusion: Cells derived from late clones exhibit morphological and biological characteristics of EPC, and the counts of late clones increase by using of sequential adherence method.

Results Early clones failed to form second clone and were devoid of the capacity of in vitro and in vivo vascular genesis.Furthermore,cells derived from the early clones expressed mainly CD14 and CD45.In contrast to early clones,the late clones emerged until 21 to 28 days after cultivation and exhibited typically endothelial cells properties.Remarkably,cells originated from late clones had the ability to form second endothelial clones after replanted and generated tube-like structures when seeded onto collagen gels or transplanted into nude mice.In addition to the clearly morphological and functional differences,cells derived from late clones expressed obviously increased CD146(P<0.01) and reduced CD45 and CD14(P<0.001). Comparison of biological properties between human peripheral blood derived endothelial progenitor cells and mature vessel wall endothelial cells

Abstract:AIM: To explore whether there are some similar or different properties between endothelial progenitor cells (EPCs) derived from human peripheral blood and mature vessel wall endothelial cells in antigen expression, cell morphology, proliferation potential, and in vitro and in vivo vasculogenesis. METHODS: Mononuclear cells were isolated by density-gradient centrifugation and incubated onto fibronectin-coated dishes in endothelial me-dium in the presence of vascular endothelial growth factor. After cultivation, cells were sequentially replanted by allowing cells to adhere to tissue culture dishes during two successive 2-h incubations in order to remove nonadherent cells. Medium was changed every other day and the cultivation was made continually until the late clones generated. At the same time, human umbilical vein endothelial cells (HUVECs) were cultured and same biological properties were compared with EPCs. Flow cytometry analysis was used to evaluate cell surface antigen expression and cells binding ulex europaeus agglutinin (UEA)-1 and up-taking DiI-labeled acetylated low density lipoprotein (DiI-Ac-LDL) were detected by direct fluorescence staining. Moreover, cell proliferation potential was estimated by calculating the number of population doubling times and the capacities of in vitro and in vivo vascular genesis were assessed by plating cell onto collagen gels and implanting the cellularized gel into nude mice. RESULTS: EPCs emerged until 21 to 28 d after cultivation and exhibited coble-stone like typical endothelial cells. When compared with HUVECs, EPCs expressed higher levels of CD36 and KDR (P<0.01), but there were no significant differences between the 2 kinds of cells in CD146 expression, cells binding UEA-1, and up-taking DiI-Ac-LDL. During the 100-d in vitro cultivation, EPCs and HUVECs reached 46 and 25 divisions respectively. Remarkably, only cells originated from EPCs had the ability to generate tube-like structures when seeded onto collagen gels or transplanted into nude mice. CONCLUSION: EPCs derived from human peripheral blood retain the complete biological properties of stem/progenitor cells in spite of exhibiting the phenotypic and morphologic characteristics of mature endothelial cells.

Abstract:The discovery of endothelial progenitor cells has revolutionized our understanding of postnatal blood vessel formation, with endothelial colony-forming cells (ECFCs) emerging as key players in vasculogenesis. Among various ECFC sources, cord blood-derived ECFCs (CB-ECFCs) are of particular interest due to their superior proliferative and clonogenic potential and their ability to promote vascular network formation. Human embryonic stem cell-derived endothelial cells (hESC-ECs) have also shown potential in regenerative medicine, though their vasculogenic efficacy remains unclear compared to CB- and adult blood-derived ECFCs (AB-ECFCs). This study aimed to directly compare the angiogenic and vasculogenic capabilities of CB-ECFCs, AB-ECFCs, and hESC-ECs in vitro and in vivo. Our results demonstrated that CB-ECFCs had a significantly higher proliferation rate than both AB-ECFCs and hESC-ECs ( p < 0.01). In tube formation assays, CB-ECFCs exhibited superior ability to form capillary-like structures compared to hESC-ECs ( p < 0.0001) and AB-ECFCs ( p < 0.01). In vivo, CB-ECFCs significantly improved blood flow recovery in ischemic tissue ( p < 0.01), outperforming both AB-ECFCs and hESC-ECs, with no significant recovery observed in the latter two groups. These findings suggest that CB-ECFCs represent a more effective cell source for therapeutic angiogenesis, while further optimization is needed to enhance the efficacy of hESC-ECs for clinical applications. Future research should explore the molecular mechanisms underlying the superior regenerative potential of CB-ECFCs and focus on improving the stability and functionality of stem cell-derived ECs for therapeutic use.

Abstract:Endothelial colony-forming cells (ECFC) are currently considered as a promising cell population for the pre-endothelialization or pre-vascularization of tissue-engineered constructs, including small-diameter biodegradable vascular grafts. However, the extent of heterogeneity between ECFC and mature vascular endothelial cells (EC) is unclear. Here, we performed a transcriptome-wide study to compare gene expression profiles of ECFC, human coronary artery endothelial cells (HCAEC), and human umbilical vein endothelial cells (HUVEC). Characterization of the abovementioned cell populations was carried out by immunophenotyping, tube formation assay, and evaluation of proliferation capability while global gene expression profiling was conducted by means of RNA-seq. ECFC were similar to HUVEC in terms of immunophenotype (CD31+vWF+KDR+CD146+CD34-CD133-CD45-CD90-) and tube formation activity yet had expectedly higher proliferative potential. HCAEC and HUVEC were generally similar to ECFC with regards to their global gene expression profile; nevertheless, ECFC overexpressed specific markers of all endothelial lineages (NRP2, NOTCH4, LYVE1), in particular lymphatic EC (LYVE1), and had upregulated extracellular matrix and basement membrane genes (COL1A1, COL1A2, COL4A1, COL4A2). Proteomic profiling for endothelial lineage markers and angiogenic molecules generally confirmed RNA-seq results, indicating ECFC as an intermediate population between HCAEC and HUVEC. Therefore, gene expression profile and behavior of ECFC suggest their potential to be applied for a pre-endothelialization of bioartificial vascular grafts, whereas in terms of endothelial hierarchy they differ from HCAEC and HUVEC, having a transitional phenotype.

Abstract:Abstract Abstract 2337 The nucleosome remodeling and deacetylase (NuRD) complex is involved in gene repression and normal hematopoiesis. In embryonic stem (ES) cells, NuRD complex directly maintains the self-renewal from lineage commitment. Murine models with defects in NuRD complex present with increased hematopoietic stem/progenitor cells (HSPCs). Several transcriptional factors (TFs), such as SALL4, FOG1, BCL11A, BCL11B, ZNF521 use a conservative 12 amino acid domain to recruit NuRD components for targeted gene repression. In this study, we plan to test the hypothesis that by blocking the interaction between NuRD and its TFs, we can phenocopy its knockdown effects on human HSPC. Human cord blood (CB) CD34+ cells were purified, and treated with peptide with the NuRD interacting sequences (wt-pep) or its controls mutant peptide (mut-pep) or scramble peptide (scr-pep). In vitro and in vivo assays were performed to evaluate whether wt-pep treatment can maintain or expand human cord blood HSPCs, which were further characterized as CD34+CD90+ and CD34+CD38-CD90+ sub-population with known repopulating potential after ex vivo culture. First we observed wt-pep treated CB CD34+ (1×104) cultures with cytokines (SCF, Flt3 ligand, IL-3 and IL-6) yielded 10.6 times greater numbers of CD34+CD90+ cells (Fig 1) and 19.3 times greater numbers of CD34+CD38-CD90+ as compared to the cultures containing cytokines with scr-pep after 6 days of culture (p<0.05). Colony formation assay demonstrated wt-pep treated cells gave mainly rise to large colonies, while scr-pep treated cells generated high proportion of small colonies, 8 days after plating (p<0.05). Treatment of CB CD34+ cells with wt-pep resulted in an increase in total colony-forming cell (CFC) content compared to other control groups, 14 days after plating (p<0.005). Then, to determine the most contributed to populations, Lin-CD34+CD38+CD90±, Lin-CD34+CD38-CD90± populations were fractionally sorted and treated with wt-pep. While the expansion rate of Lin-CD34+CD38+CD90± were too low to evaluate, wt-pep treated Lin-CD34+CD38-CD90+ and Lin-CD34+CD38-CD90- cells gave rise to 79.3% and 52.7% of CD34+CD90+ cells, respectively. In addition, Lin-CD34+CD38-CD90 + cells gave rise to much more CD34+CD38- population in wt-peptide treated cells as compared with scr-peptide treated cells (44.0%, 16.2% respectively) and almost all of these cells co-expressed CD90. These data suggested that both Lin-CD34+CD38-CD90± cells treated with wt-peptide could give rise to CD34+CD90+ as well as CD34+CD38-CD90+ cells. In addition, CD34+CD38-CD90+ cells seemed more contributed than CD34+CD38-CD90- cells for expansion of CD34+38-CD90+ cells. Based on in vitro experiment results, we investigated immunodeficient (SCID) repopulating activity (SRA) to determine efficacy of wt-pep to HSPCs in vivo. We observed that mice transplanted the progeny of 1×104 CB CD34+ cells treated with wt-pep (n=5) had higher degree of human cell chimerism (0.38%) than that with scr-peptide (0%) (n=5) at 4 weeks in peripheral blood. The 8 weeks chimerism of wt-peptide treated progeny (13.6%; n=5) were greater than scr-peptide treated progeny (1.88%; n=5) (Fig 2; p < 0.005) and also greater than unmanipulated CD34+ cells (1×104) (0.09%; n=3) (Fig 2; p < 0.05). In addition, these mice (n=5) demonstrated that multilineage differentiation. We further analyzed transcription levels of several HSPC related genes, which were compared between CB CD34+ cells treated with wt-peptide and scramble peptide. Significantly higher transcript levels were detected for HoxB4 (p < 0.05), GATA 2 (p < 0.05) and Evi1 (p < 0.05) and lower levels were detected for PTEN (p < 0.05) by real time quantitative RT-PCR in wt-peptide expanded cells as compared with scr-pep cultures. Unlike embryonic stem cells, human HSPC can not be maintained ex vivo in a culture condition. The limit in source cells has greatly hampered the field of HSPC research. Optimizing ex vivo HSPC expansion culture condition not only can facilitate basic research on these cells, but also can be translated into clinical applications such as enhancement of CB engraftment and gene therapy. Our study demonstrates a novel peptide approach for ex vivo expansion of a unique CD34+CD90+ and CD34+CD38-CD90+ population with increased engraftment potential. Cocultivation of Umbilical Cord Blood CD34+cells with Retro-Transduced Hmscs Leads to Effective Amplification of Long-Term Culture-Initiating Cells

Abstract:AIM: To establish a novel coculture system for ex vivo expansion of umbilical cord blood(UCB) hematopoietic progenitors using thrombopoietin (TPO)/Flt-3 ligand (FL)-transduced human marrow-derived mesenchymal stem cells (tfhMSCs) as feeder.METHODS: UCB CD34(+) cells were isolated and cultured using four culture systems in serum-containing or serum-free medium. Suitable aliquots of cultured cells were used to monitor cell production, clonogenic activity, and long-term culture-initiating culture (LTC-IC) output. Finally, the severe-combined immunodeficient (SCID) mouse-repopulating cell (SRC) assay was performed to confirm ability of the cultured cells to reconstitute long-term hematopoiesis.RESULTS: There were no significant differences in the number of total nucleated cells among different culture systems in serum-containing medium during 21-d culture. However, on d 14, the outputs of CD34(+) cells, CFU-C and CFU-GEMM in tfhMSCs coculture system were significantly enhanced. LTC-IC assay demonstrated that the tfhMSCs coculture system had the most powerful activity. The severe-combined immunodeficient (SCID) mouse repopulating cell (SRC) assay confirmed extensive ability of the expanded cells to reconstitute long-term hematopoiesis. Furthermore, PCR analysis demonstrated the presence of human hematopoietic cells in the bone marrow and peripheral blood cells of NOD/SCID mice.CONCLUSION: The TPO/FL-transduced hMSCs, in combination with additive cytokines, can effectively expand hematopoietic progenitors from UCB in vitro and the tfhMSCs coculture system may be a suitable system for ex vivo manipulation of primitive progenitor cells under contact culture conditions. Abstract:Objective:To investigate the methods of isolation and cultivation from adult peripheral blood mononuclear cells for the differentiation into endothelial progenitor cells(EPCs) and study the biological properties of EPCs.Method: Mononuclear cells were isolated by density-gradient centrifugation and incubated onto fibronectin-coated dishes in endothelial medium in the presence of vascular endothelial growth factor.After cultivation,cells were sequential replanted by allowing cells to adhere to tissue culture dishes during two successive 2-houre incubations in order to remove nonadherent cells.Medium was changed every other days,and the number of early clones was counted at 7 days after plating.Furthermore,every samples were divided into 2 aliquots and cells from one aliquot were cultivated continually until the late clones generated.At the same time,another aliquot was handled when the early clones can be observed after 7 days cultivation.Flow cytometry analysis was used to evaluate cells surface antigen expression and cells binding ulex europaeus agglutinin(UEA)-1 and up-taking DiI-labeled acetylated low density lipoprotein(DiI-Ac-LDL) were detected by direct fluorescence staining.The capacity of in vitro vascular genesis was assessed by plating cells onto collagen gels.Result:Early clones were identified as spindle like sprouting cells radiating from a central core of round cells.When replanted,the early clones failed to form second clone,and were devoid of the capacity of in vitro vascular genesis.Furthermore,cells derived from the early clones expressed mainly CD14 and CD45.In contrasted to early clones,the late clones emerged until 21 to 28 days after cultivation and exhibited typically endothelial cells properties.Remarkably,cells originated from late clones had the ability to form second endothelial clones after replanted and generated tube-like structures when seeded onto collagen gels.In addition to the clearly morphological differences,cells derived from late clones expressed obviously increased CD146(P0.01) and reduced CD45 and CD14(P0.001).There were no differences about cell binding UEA-1 and up-taking DiI-Ac-LDL between early and late clones.Conclusion:Under endothelial cultivating conditions,human peripheral blood mononuclear cells can generate early and late clones.Cells derived from early clones are relevant to monocyte linage and have not the ability to differentiate into endothelial cells.Alternatively,cells originated from late clones exhibit morphologically and biologically characteristics of EPCs.
Isolation of endothelial progenitor cells from cord blood with CD133 immunomagnetic sorting

Abstract:AIM: To isolate, purify and differentiate endothelial progenitor cells from cord blood in vitro and to study their biological characteristics. METHODS: CD133~+ cells were selected from fresh cord blood mononuclear cells (MNC) by magnetic activated cell-sorting system (MACS). EPC was studied by flow cytometry, immunocytochemistry and immunofluorescence staining. Isolated cells were cultured in IMDM medium supplemented with or without VEGF, bFGF, SCF. RESULTS: The percentage of CD133~+ cells of cord blood MNC was (1.41±1.14)%, and purity was 75%-85% (FACS method). CD133~+ cells were grown on fibronectin-coated chamber slides in the presence of VEGF, bFGF, SCF. Within 1-2 hours of culture cells became adherent. On day 7-10, the adherent cells displayed a typical cobblestone morphology. After 14 days of culture, the adherent cells revealed a heterogeneous cell population, comprising small-sized round cells, spindle-like cells and formed tube-like structure. Weibel-Palade bodies were shown on the transmission electron microscopy photomicrographs. Compared with the (original,) cell markers CD133 and CD34 decreased significantly (77.0%±3.3% to 1.6%±2.2% and 93.1%±4.7% to 37.4%±4.9%, P0.05), while Flk-1 increased significantly (from 22.3%±3.3% to 94.3%±4.1%, P0.05) after 14 days of culture with VEGF, bFGF, SCF. The vWF was strongly expressed (77.9%±3.3%) on the 14th day later. CONCLUSION: Vascular endothelial progenitor cells were isolated from cord blood with specific expression of CD133/CD34/Flk-1. With the stimulation of the growth factors, seven-ten days after culture EPCs could be turned to endothelial cells.

Abstract:Regeneration of injured tissues requires effective therapeutic strategies supporting vasculogenesis. The lack of instantly available autologous cell sources and immunogenicity of allogeneic endothelial (progenitor) cells limits clinical progress. Based on the immunosuppressive potency of mesenchymal stem/progenitor cells (MSCs), we investigated whether crosstalk between endothelial colony-forming progenitor cells (ECFCs) and MSCs during vasculogenesis could lower allogeneic T cell responses against ECFCs allowing long-term engraftment in vivo. Immunodeficient mice received subcutaneous grafts containing human ECFCs alone, or pairs of human ECFCs/MSCs from the same umbilical cord (UC) to study vasculogenesis in the presence of human leukocyte antigen (HLA)-mismatched human peripheral blood mononuclear cells (PBMCs). In vitro, cell surface marker changes due to interferon gamma (IFNγ) stimulation during ECFC/MSC coculture were determined and further effects on allostimulated T cell proliferation and cytotoxic lysis were measured. IFNγ-induced HLA-DR expression on ECFCs and MSCs, but both cell types had significantly less HLA-DR in cocultures. ECFC-induced T cell proliferation was abolished after MSC coculture as a result of HLA-DR downregulation and indolamin-2,3-dioxygenase activation. Additionally, allospecific CD8+ T cell-mediated lysis of ECFCs was reduced in cocultures. ECFC/MSC coapplication in immunodeficient mice not only promoted the generation of improved blood vessel architecture after 6 weeks, but also reduced intragraft immune cell infiltration and endothelial HLA-DR expression following PBMC reconstitution. Crosstalk between UC-derived ECFCs and MSCs after combined transplantation can lower the risk of ECFC rejection, thus enabling their coapplication for therapeutic vasculogenesis. Adherent Cell Layer from Human Umbilical Cord Blood and Its Support to Ex Vivo Expansion of Hematopoietic Stem/progenitor Cells

Abstract:To investigate the feature of adherent cells from human umbilical cord blood (HUCB) and their supporting effects on ex vivo expansion of hematopoietic stem/progenitor cells (HSPCs), the morphology of the HUCB-derived adherent cells was observed and their characteristics was identified by Flow cytometry. The HUCB-derived adherent cells were used as adherent cell feeder layer to support ex vivo expansion of HSPCs. Co-culture system was established based on the feeder layer to expand the mononuclear cells (MNC) and CD34~+ cells, and the effects of feeder layer upon ex vivo expansion were observed. GEMM-CFC formations from starting cells and expanded cells were assayed. Results showed that the HUCB-derived adherent cells had typical,fibroblastic shape. CD14 was negative (3.8%), but CD13, CD166 and CD29 were positive (51.8%, 35.6%, 16.5, respectively). MNC in the culture system with adherent cell feeder layer had an expansion of 2.09-fold higher than that in the culture system without adherent cell feeder layer. CD34~+ cell in the culture system with adherent cell feeder layer had an expansion of 2.94-fold higher than that in the culture system without adherent cell feeder layer. Expanded cells in the culture system with adherent cell feeder layer had higher potentials of GEMM-CFC formation than those in the culture system without adherent cell feeder layer. HUCB-derived adherent cells showed the characteristics of mesenchymal stem cells (MSCs) cells from bone marrow. Under appropriate conditions, the adherent cells from HUCB could form the feeder layer to support long-term hematopoiesis and provide the supporting stromal cells for the ex vivo expansion of HSPCs.
Study the Culture Condition and Biological Characteristics of Endothelial Progenitor Cells from Rat Marrow in Vitro

Abstract:To elucidate a simple method of isolating endothelial progenitor cells (EPCs) from rat marrow monocytes (MNCS) and observe the endothelial cell specific expression profile during proliferation and differentiation in vitro. MNCS were isolated from rat marrow using ficoll density gradient centrifugation, induced, proliferated and differentiated in EGM-2 culture medium. The EPCs were identified by immunofluorescent staining (CD34inverted commasVWF diplmark and FLK1) at 4dinverted commas7dinverted commas10dinverted commas14dinverted commas21d. The proliferation of EPCs was evaluated by MTT, and endothelial cell specific markers (VWFinverted commasFLK1) were determined with western blotting and real-time PCR at 1dinverted commas4dinverted commas7d, 10dinverted commas14dinverted commas 21d. During culturing process, the cells became spindle shaped and displayed endothelium-like cobble-stone morphology with outgrowth at 4d and 7d. Result of MTT indicated that the number of EPCs increased to peak at 10d (P < 0.05). Percentage of the cells that were positive for CD34/VWF was 90% at 7d. Expression for VWF at 1d was not found with western blotting, and however gradually increased at 4d and reached to the top during 10d to 14d. The results with real-time PCR indicated that the expression of VWF and FLK1 mRNA within non-induced cells was not found, gradually increased at 4d and reached to top at 14d, the low-expression at 1d was significantly different from the other ones (P < 0.01). (These data demonstrated that EPCs could be isolated from rat marrow monocytes (MNCS) and cultured in vitro. The EPCs could be used as seeding cells in vascular tissue engineering.

Abstract:Most circulating endothelial cells are apoptotic, but rare circulating endothelial colony-forming cells (C-ECFCs), also known as blood outgrowth endothelial cells, with proliferative and vasculogenic activity can be cultured; however, the origin and naive function of these C-ECFCs remains obscure. Herein, detailed lineage tracing revealed murine C-ECFCs emerged in the early postnatal period, displayed high vasculogenic potential with enriched frequency of clonal proliferative cells compared with tissue-resident ECFCs, and were not committed to or derived from the BM hematopoietic system but from tissue-resident ECFCs. In humans, C-ECFCs were present in the CD34 bright cord blood mononuclear subset, possessed proliferative potential and in vivo vasculogenic function in a naive or cultured state, and displayed a single cell transcriptome sharing some umbilical venous endothelial cell features, such as a higher protein C receptor and extracellular matrix gene expression. This study provides an advance for the field by identifying the origin, naive function, and antigens to prospectively isolate C-ECFCs for translational studies.

Abstract:Objective: To study the changes of biological characteristics of CD34+/ CD133+/VEGFR-3+ lymphatic endothelial progenitor cells(EPCs) isolated from umbilical cord blood after induction with VEGF-C and investigate the mechanisms of EPCs differentiation toward lymphatic endothelial cells. Methods: Mononuclear cells were isolated from umbilical cord blo od using density centrifugation with Percoll solution. CD34+/CD133+/ VEGFR-3+ cells were sorted with FACS. Differentiation of the cells was induced with VEGF-C. The ultrastructural changes of the cells were viewed under scanning and transmission electron microscopes. Changes in expression of markers of the cells were viewed under confocal laser scanning microscope. Results: Lymphatic EPCs isolated from umbilical co rd blood expressed CD34, CD133 and VEGFR-3. At 7 day after induction with VEGF-C, CD34+/ CD133+/VEGFR-3+ cells were shuttle-like, there were a lamellipodium, numerous filopodia and many short microvilli. Caveolae were observed. Mitochondrion and rough endoplasmic reticulum were rich in cytoplasm. At 14 day after induction, the cells demonstrated appearance of the endothelial cell and expressed lymphatic endothelial specific makers LYVE-1 and 5′-nucleotidase, the expression of CD133 disappeared. Weibel-Palade body was observed in the cytoplasm of the cell. Conclusion: There are CD34+/CD133+/VEGFR-3+ lymphatic EPCs in human umbilical cord blood. The cells may differentiate into lymphatic endothelial cells through VEGF-C/VEGFR-3 signaling pathway after induction with VEGF-C.

Abstract:AIM: Endothelial progenitor cells(EPCs) are a group of stem cells/progenitor cells,which exist in postnatal body and can be of specially homing to the foci of angiogenesis and then differentiate into endothelial cells.This investigation was to study the method for culturing endothelia progenitor cells(EPCs) in vitro,and to observe its feasibility and condition formed vessel-like structure.METHODS: The cells were isolated from born marrow,peripheral blood,umbilical cord blood or spleen in different laboratories.The EPCs derived from human umbilical cord blood and rabbit peripheral blood were cultured in vitro through adhesion selection and were differentiated into endothelial cells under the induction of special cytokines.The expression of CD34,VEGFR-2,AC133 and VE-cadherin were detected by fluorescence-activated cell sorting.The endothelial cell lineage was confirmed by DiI-ac-LDL up-taking and Ⅷ factor immunocytochemistry.RESULTS: The EPCs derived from human umbilical cord blood and rabbit peripheral blood were cultured in vitro successfully,forming cord-like and tube-like structure.The EPCs derived from rabbit peripheral blood differentiated more mature and formed vessel-like structure.CONCLUSION: The EPCs derived from human umbilical cord blood and rabbit peripheral blood formed vessel-like structure in vitro.EPCs may be a potential resource of vessel tissue engineering.
