Effects of water stress and nutrient deficiency on ethylene synthesis in harvested broccoli

Abstract:Ethylene plays an important role in broccoli floret yellowing and senescence. The effects of water stress and nutrient deficiency during broccoli storage on ethylene production rate, 1-aminocyclopropane-1-carboxylic acid (ACC) contents, malonyl ACC (MACC) amounts, 1-aminocyclopropane-1-carboxylic acid oxidase (ACO) activity and the expression of two genes encoding key enzymes involved in ethylene synthesis were studied in this work. We found that water deficit stress promotes ethylene production in the three stem tissues as well as in florets with an accumulation of ACC and MACC in florets. The nutrient deficiency alone or together with water deficit stress should be considered as the key factor affecting ACC contents. In addition, our data show that nutrient deficiency has a major role in the MACC accumulation in the second stem layer of broccoli. The ACO activity peaks were delayed by both water stress and nutrient deficiency, concomitant with the observed increase in both ethylene synthesis and ACC levels. During broccoli postharvest storage, BoACS1 gene showed relatively high levels. BoACS1 transcript levels only slightly changed in control florets during storage while the ACC contents increased. There were no significant differences in the expression of BoACO3, BoACO1 and BoACO2 genes during broccoli senescence, in good accordance with the recorded ACO activity. In contrast, BoACS1 gene displayed relatively high transcript levels. Water deficit stress induced ethylene production due to the increase in BoACO3 transcription.
