Abstract:The postharvest senescence accompanied by yellowing limited the shelf-life of broccoli. In this study, we developed a novel W/O/W double emulsion co-delivering brassinolide and cinnamon essential oil and applied it to broccoli for preservation. Results showed that double emulsion prepared by whey protein concentrate-high methoxyl pectin (1:3) exhibited best storage stability with largest particle size (581.30 nm), lowest PDI (0.23) and zeta potential (-40.31 mV). This double emulsion also exhibited highest encapsulation efficiency of brassinolide (92%) and cinnamon essential oil (88%). The broccoli coated with double emulsion maintained higher chlorophyll contents and activities of chlorophyllase and magnesium-dechelatase were reduced by 9% and 24%, respectively. The energy metabolic enzymes (SDH, CCO, H+-ATPase, Ca2+-ATPase) were also activated, inducing higher level of ATP and energy charge. These results demonstrated W/O/W double emulsion co-delivering brassinolide and cinnamon essential delayed the senescence of broccoli via regulating chlorophyll degradation and energy metabolism.
Effect of 1-methylcyclopropene on shelf life, visual quality, antioxidant enzymes and health-promoting compounds in broccoli florets

Abstract:The effect of 1-methylcyclopropene (1-MCP) on quality, antioxidant enzymes and glucosinolate contents in broccoli (Brassica oleracea var. italica) florets was investigated in the present study. Broccoli florets were treated with air (control) and 2.5μl/l 1-MCP for 6h at 20°C, and were then stored at 20°C for 5days. 1-MCP treatment markedly extended shelf life, reduced postharvest deterioration, retarded chlorophyll degradation and inhibited the increase of malondialdehyde amount and activities of polyphenol oxidase and lipoxygenase in florets. The activities of superoxide dismutase, peroxidase and catalase in florets treated with 1-MCP were higher than those in control florets. 1-MCP treatment reduced the rate of decrease of total carotenoids, ascorbic acid and glucosinolates in florets when compared to those in the control. These results indicated that 1-MCP treatment could be a good candidate for extending shelf life, maintaining visual quality and reducing loss of health-promoting compounds, particularly glucosinolates in broccoli.

Abstract:Broccoli has been described as one of the most important vegetables worldwide, and it is a rich source of vitamin C and anti-carcinogenic phytochemicals, such as glucosinolates. The edible part of broccoli is floret head, which is high in respiration rate, and easy to be senescent and yellowing. As a kind of highly perishable products, broccoli heads usually undergo losses in nutrients and quality during postharvest period. The researches on the influence of different postharvest handlings in functional components and quality of broccoli are significant to postharvest physiology, but also have an important guiding role for the broccoli industry. Based on recent investigations, the effects of different postharvest handlings, including cooling storage, atmosphere control, modified atmosphere packaging, 1-methylcyclopropene (1-MCP) and light treatment on functional components and quality of broccoli were reviewed, and the related mechanisms were verified by elucidating the biosynthesis and metabolism pathway of glucosinolates. The common postharvest handling methods of broccoli products were summarized and evaluated, and future researches as well as their potential in broccoli industry were prospected.

Abstract:Broccoli florets were treated with 1-methylcyclopropene (1-MCP) at concentrations of 0-5.0 μL/L for 6 h at 20 C followed by storage at 20 ℃. 1-MCP markedly extended the storage life and delayed the degradation of chlorophyll. The beneficial effects were dependent upon concentrations, and 2.5 μL/L 1-MCP was the most effective one among all the treatments, the contents of vitamin C and protein in this treatment were always higher than those in the control during storage at 20 C. SOD and CAT activities were higher while POD activity was lower in the treatment than in the control.

Abstract:The effect of 1-MCP on postharvest storage of broccoli was studied. In all four varieties tested, the shelf life was extended, degradation of chlorophyll was delayed and the weight loss was reduced under 20℃ and 85%-90% relative humidity storage condition. This indicated that 1-MCP was generally effective in broccoli postharvest storage. Especially, even if the treatment was delayed 0-9 hour, the shelf life of broccoli still can be extended, and the senescence can be delayed, though the effect was reduced with the delay of the treatment.

Abstract:Effect of melatonin treatment on visual quality and contents of health-promoting compounds of broccoli florets under room temperature was investigated in the present study. Broccoli florets were treated with 1 μM melatonin and then stored at room temperature. Results showed that melatonin treatment could delay the post-harvest senescence of broccoli, and performed well in maintaining higher levels of antioxidants, such as carotenoids, vitamin C and total phenols, as well as higher antioxidant capacity than the control. Besides, 1 μM melatonin treatment sustained higher content of glucosinolates, and also resulted in increased percentage of the most potent anticarcinogenic profile, glucoraphanin. Further analysis revealed that 1 μM melatonin strongly induced the expression of glucosinolate biosynthesis-related genes BoMYB28, BoMYB34, BoCYP79F1, and BoCYP79B2, as well as BoTGG1, a gene involved in glucosinolate hydrolysis. In conclusion, post-harvest treatment with 1 μM melatonin is potential in maintaining visual quality and health-promoting properties of broccoli florets.

Abstract:In this study, we tested the exogenous application of phytosulfokine alpha (PSK alpha) for delaying the yellowing of broccoli florets during cold storage. Our results showed that the lower yellowing in broccoli florets treated with 150 nM PSK alpha was probably due to the higher endogenous accumulation of PSK alpha, leading to the endogenous accumulation of guanosine 3', 5'-cyclic monophosphate (cGMP). Besides, broccoli florets treated with 150 nM PSK alpha exhibited a higher accumulation of phenols and flavonoids by triggering gene expression and activities of phenylalanine ammonia-lyase (PAL) and chalcone synthase (CHS). Moreover, the higher expression of L-galactotno-1,4-lactone dehydrogenase (GLDH) gene and the lower expression of ascorbic acid oxidase (AAO) gene in broccoli florets treated with 150 nM PSK alpha may be the reasons for the higher accumulation of ascorbic acid. In conclusion, the exogenous application of PSK alpha is a promising strategy in delaying the yellowing and preserving the nutritional quality of broccoli florets during cold storage.

Abstract:During postharvest life, broccoli suffers from florets yellowing, which reduces its economic and nutritional value. We investigated the mechanisms employed by exogenous phytosulfokine alpha (PSK alpha) applying at 150 nM for delaying senescence in broccoli florets stored at 4 degrees C for 28 days. The results showed that the SnRK1 gene expression was higher in broccoli florets treated with 150 nM PSK alpha, which may warrant sufficient intracellular availability of ATP, resulted from a higher activity of H+-ATPase, Ca2+-ATPase, succinate dehydrogenase (SDH), and cytochrome c oxidase (CCO). In addition, avoiding H2O2 accumulation by higher expression of alternative oxidase (AOX) and uncoupling protein (UCP1) genes may arise from higher SnRK1 gene expression. Additionally, lower mitochondrial permeability transition pore (mPTP) opening may avoid ROS accumulation in broccoli florets treated with 150 nM PSK alpha. Taken together, our results assume that exogenous PSK alpha application as signaling bioactive peptide could be considered as a promising procedure for delaying senescence of broccoli florets during cold storage.

Abstract:Broccoli (Brassica oleracea var. italica) florets were packaged in polyethylene bags with no holes (M0), two microholes (M1), and four macroholes (M2), and then stored at 4 or 20°C. The effects of modified atmosphere packaging (MAP) treatments on visual quality and glucosinolate contents were determined by comparing with non-wrapped florets. The results showed that MAP treatments, especially with M0 and M1, extended the shelf life and reduced the postharvest deterioration of broccoli florets stored at 4 and 20°C. All three MAP treatments reduced the decreasing concentration rates of individual, total aliphatic and indole glucosinolates in broccoli florets when compared to those in the control, with M0 being the most significant, followed by M1 and M2 during 23 days of storage at 4°C or 5 days of storage at 20°C. Broccoli florets with M0 treatment maintained the visual quality and glucosinolate contents for 13 days at 4°C and 3 days at 20°C.

Abstract:Asparagus is a highly perishable vegetable which deteriorated rapidly after harvesting.Treated with mixture of 6-benzylaminopurine(6-BA)and CaCl2 or NaClO,asparagus' shelf life and quality were improved.Quality parameters including chlorophyll,shear force,lignin,cellulose content,PAL,POD,ASA-POD and CAT activity were studied.The results showed that the mixture additives significantly decreased the loss of the chlorophyll,maintained good shear force,increased CAT and POD activity,inhibited PAL peak,but no significant effect on lignin,cellulose content and ASA-POD activity.

Abstract:Brassica vegetables are rich in glucosinolates, a group of secondary metabolites, which is related to flavor, quality, resistance and anticancer activity. In addition, they also play a vital role in plant defense against pathogens and herbivores. In this review, we briefly elucidated the biosynthetic and degradation pathway of glucosinolate, and highlighted the effects of different postharvest treatments, such as cooling storage, controlled atmosphere storage, modified atmosphere packaging, 1-methylcyclopropene (1-MCP) treatment, as well as processing and cooking methods on compositions and contents of glucosinolate profiles and nutritional quality in Brassica vegetables. Particularly, from the perspective of dynamic equilibrium of glucosinolate metabolism, we analyzed the mechanism of regulating glucosinolate components and contents, and also discussed how to maintain the contents of favorable glucosinolate profiles and nutritional quality in Brassica vegetable products during postharvest period.

Abstract:The effects of H2S on yellowing and energy metabolism of broccoli florets treated with hydrogen sulfide (H2S) or DL-propargylglycine (PAG) were investigated after four days of storage at 20 degrees C. Our study showed that H2S treatment enhanced endogenous H2S content by 28.19% and 49.78% in comparison to the control and PAG-treated group respectively. This result might be related to the increase of L-cysteine desulfhydrase (LCD) and D-cysteine desulfhydrase (DCD) activities. Meanwhile, H2S treatment can maintain chlorophyll content at 0.329 g kg(-1), whereas the control at 0.298 g kg(-1) and PAG-treated group at 0.275 g kg(-1). This led to an alleviation of the yellowing in broccoli florets. In addition, high endogenous H2S content also activated the key enzymes, involved in energy metabolism, including ATPases, cytochrome C oxidase (CCO), succinate dehydrogenase (SDH), glucokinase, fructokinase, glucose-6-phosphate dehydrogenase (G6PDH) and 6-phosphogluconate dehydrogenase (6PGDH). As a result, significantly higher energy charge was observed in H2S-treated group (p < 0.05). These results suggest that H2S can effectively inhibit the yellowing and maintain high energy charge, therefore prolong the shelf life of postharvest broccoli.

Introduction Folic acid (FA) is a critical metabolite in all living organisms and an important nutritional component of broccoli. Few studies have been conducted on the impact of an exogenous application of FA on the postharvest physiology of fruits and vegetables during storage. In this regard, the mechanism by which an exogenous application of FA extends the postharvest quality of broccoli is unclear. Objective This study utilized a multicomponent analysis to investigate how an exogenous application of FA effects the postharvest quality of broccoli. Methods Broccoli was soaked in 5 mg L -1 FA for 10 min and the effect of the treatment on the appearance and nutritional quality of broccoli was evaluated. These data were combined with transcriptomic, metabolomic, and DNA methylation data to provide insight into the potential mechanism by which FA delays senescence. Results The FA treatment inhibited the yellowing of broccoli during storage. CHH methylation was identified as the main type of methylation that occurs in broccoli and the FA treatment was found to inhibit DNA methylation, promote the accumulation of endogenous FA and chlorophyl, and inhibit ethylene biosynthesis in stored broccoli. The FA treatment also prevented the formation of off-odors by inhibiting the degradation of glucosinolate. Conclusions FA treatment inhibited the loss of nutrients during the storage of broccoli, delayed its yellowing, and inhibited the generation of off-odors. Our study provides deeper insight into the mechanism by which the postharvest application of FA delays postharvest senescence in broccoli and provides the foundation for further studies of postharvest metabolism in broccoli. Abstract:Due to its high concentration of bioactive components, broccoli heads have a lot of potential as a functional food. Broccoli is perceived as a tasty, wholesome and healthful food product. This product nevertheless has a limited shelf life and quality retention issues, especially in tropical temperature zones. The hurdles techniques are starting to become more acceptable in the sector of food preservation since they affect moderate levels. This study evaluates the impact of hurdles techniques and various storage conditions (ambient at 25 °C, cold at 4 °C, and frozen at -18 °C) on broccolis cut characteristics. For the hurdles experiment, broccoli cuts were soaked in four solutions (the 1st: boiling water, the 2nd: metabisulphite solution (150 ppm), the 3rd: sodium chloride (4%), citric acid (1%), sodium benzoate (200 ppm) and the 4th: 1% chitosan) followed by packaging using two approaches (aerobic and vacuum). Sensory attributes, browning index, ascorbic acid, total phenolic, total flavonoids and antioxidant activity % (by DPPH, ABTS and FRAP) of hurdled and un-hurdled broccoli cuts during storage were investigated. Results demonstrated that using hurdle technology (different soaking solutions, packaging methods and storage conditions) had significant impact on the quality properties of the broccoli cuts. Additionally, vacuum packaging demonstrated the highest protection against decreasing the values of ascorbic acid, total phenolic, total flavonoid, and antioxidant components.

Foliar application of salicylic acid and calcium chloride delays the loss of chlorophyll and preserves the quality of broccoli during storage

Abstract:Consumer awareness of broccoli's unusual color, rich flavor, and concentration of desired phytochemicals has led to a steady increase in consumption in recent years. However, its short shelf-life, which is linked with quick discoloration and degeneration after harvest, limits industrial production and marketing. The effect of pre-harvest salicylic acid (SA) and calcium chloride (Ca) and their combination on the post-harvest quality of broccoli during storage (5 ± 1°C) was explored in this study. The foliar spray treatments reduced weight loss of broccoli head during storage. At the end of storage, Ca (2%) alone and in combination with SA (0.01%) significantly maintained the chlorophyll concentration rather than control. The total phenols, flavonoid, and antioxidant capacity of the Ca (2%) + SA (0.01%) treated samples was significantly greater than the control. SA (0.01%) alone or in conjunction with Ca (2%), showed higher catalase (CAT) activity; however, Ca (1%), alone or in combination with SA (0.01%), showed higher peroxidase (POD) activity. Generally, the marketability of the treated broccoli was significantly greater than the control at the end of storage. Based on these findings, we believe Ca (2%) + SA (0.01%) improves the antioxidant system, delays chlorophyll degradation, and extends the shelf life of broccoli heads stored at 5 ± 1°C. It is proposed that the green color, marketability, and nutrient content of broccoli during postharvest handling and storage can be retained longer by foliar application of this treatment. PRACTICAL APPLICATIONS: Broccoli (Brassica oleracea L. Italaia) is a widely-consumed floral green vegetable due to its high content of nutrients and bioactive compounds. However, after harvest, florets rapidly senesce and suffer from yellowing which affects the quality of broccoli. The senescence of post-harvest broccoli is characterized by fresh weight loss, chlorophyll degradation, and a significant reduction in nutritional content. Therefore, preventing the decline in the quality of harvested broccoli is essential to maintain its economic and nutritional value. The results of this study showed that pre-harvest foliar application of Ca (2%) + SA (0.01%) with delayed weight loss, chlorophyll degradation, preservation of antioxidant compounds, and increased enzyme activity has a positive effect in maintaining broccoli heads quality during cold storage.

Abstract:The effects of cooling and controlled atmosphere (CA) treatments on glucoraphanin content and induction of quinone reductase (QR) activity in broccoli florets were investigated. The results showed that both the glucoraphanin content and QR activity of broccoli florets increased during the initial 6 day of storage, and then decreased markedly at different temperatures of 0, 5, and 10°C. Immediate cooling of broccoli florets at 0 and 5°C could preserve the glucoraphanin content and QR activity for 12 days. Keeping broccoli florets at 20°C for 6h before cooling at 5°C had little effects on shelf-life, glucoraphanin content and QR activity. CA treatments with elevated CO2 (21% O2+10% CO2, and 21% O2+20% CO2) and an air treatment (21% O2) were found to increase the glucoraphanin content and the QR activity over the first 5 days of storage at 5°C, while CA treatments with reduced O2 concentrations (1% O2, 1% O2+10% CO2) led to a steady decrease of glucoraphanin content and the QR activity during 20 days of storage at 5°C. The highest content of glucoraphanin and QR activity was found in florets stored under 21% O2+10% CO2 at 5°C. These conditions were able to maintain the visual quality, glucoraphanin content and QR activity of the broccoli florets for 20 days.

Abstract:The effect of a combination of 25 mg L−1 kojic acid and 10 g L−1 calcium chloride (KA + CaCl2) on the postharvest senescence of broccoli was investigated, along with potential modes of action. Results indicated that the KA + CaCl2 treatment of broccoli florets maintained their sensory evaluation scores, inhibited yellowing of florets, and generally prolonged shelf-life. The KA + CaCl2 treatment decreased the rate of respiration, ethylene production, and inhibited the degradation of chlorophyll. Furthermore, the relative gene expression and enzyme activity of ascorbate peroxidase, peroxidase, and catalase were elevated, and the expression of genes (BoCHL1, BoCHL2, BoCHL3, BoPAO, and BoPPH) involved in chlorophyll degradation were down-regulated. KA + CaCl2 also maintained the production of the characteristic volatiles produced by broccoli florets as measured with the use of an electronic nose. Results of the present study provide new insights into the ability of KA + CaCl2 to inhibit the postharvest senescence of broccoli florets and extend their shelf-life.

Effect of Modified Atomosphere on Postharvest Storage of Kiwifruit and the Mechanisms Involved

Abstract:Postharves softening of kiwifruit was retarded in modified atmosphere (7.4% CO 2 and 9.8% O 2) at 20℃, the storage life being doubled. The treatment inhibited the formation of ethylene, retarded the degradation of starch and decreased the activity of amylase. Water soluble pectin (WSP) content increased while covalently bound pectin (CBP) content decreased during storage, and the treatment inhibited these changes; Ionically bound pectin (IBP) content remained unchanged during storage, and the treatment caused a transient increase during later stage. The esterity of WSP, IBP and total pectin decreased while that of CBP increased during storage, and the treatment also inhibited these changes. The activity of pectinesterase (PE) decreased during storage, and being less in treated fruits than in control fruits. The mechanisms for retardation of fruit softening in modified atmopsphere were discussed.

Abstract:The effects of long-term freezer storage and different defrosting methods on the retention of glucosinolates, vitamin C, and total phenols in frozen broccoli florets were investigated in the present study. Frozen broccoli florets were stored in a freezer at −20 °C for 165 days or subjected to defrosting by three different house-hold thawing methods (water, air, and refrigerator defrosting). Results showed that all glucosinolates were well preserved, while vitamin C and total phenols were reduced by less than 12% and 19% of the control, respectively, during the storage. Besides, refrigerator and air defrosting were better than water defrosting in glucosinolates retention, and refrigerator defrosting was the best in vitamin C preservation. No difference was observed in reserving phenolic compounds among the three methods. In conclusion, long-term freezer storage is an excellent way to preserve broccoli florets, and refrigerator defrosting is the best way to maintain the nutritional compounds in frozen broccoli florets.

Abstract:The purpose of this study was to investigate the impact of high‑oxygen-modified atmospheric packaging (HOMAP) on aroma changes in fresh-cut broccoli during storage and to explore its regulatory mechanisms. The results showed that HOMAP reduced the levels of undesirable aroma substances hexanoic acid, isobutyric acid, cyclopentanone and increased glucosinolate accumulation by inhibiting the expression of arogenate/prephenate dehydratase (ADT), bifunctional aspartate aminotransferase and glutamate/aspartate-prephenate aminotransferase (PAT), thiosulfate/3-mercaptopyruvate Transferase (TST) to reduce the odor of fresh-cut broccoli. HOMAP inhibited the expression of respiratory metabolism related genes 6-phosphate fructokinase 1 (PFK), pyruvate kinase (PK), and NADH-ubiquinone oxidoreductase chain 6 (ND6). In HOMAP group, the low expression of phospholipase C (PLC), phospholipase A1 (PLA1), linoleate 9S-lipoxygenase 1 (LOX1) related to lipid metabolism and the high expression of naringenin 3-dioxygenase (F3H), trans-4-Hydroxycinnamate (C4H), glutaredoxin 3 (GRX3), and thioredoxin 1 (TrX1) in the antioxidant system maintained membrane stability while reducing the occurrence of membrane lipid peroxidation.
