Abstract:Fractures are the most common large-organ, traumatic injury in humans. The fracture healing stage includes the inflammatory stage (0-5d), cartilage callus stage (5-14d) and hard callus stage (14-21d). All mice underwent open tibial fracture surgery and were treated with saline, Glu or SCII for 21d. Calluses were harvested 5d, 10d and 21d after fracture. Compared with the model group, SCII significantly decreased TNF-alpha and increased aggrecan serum levels by 5d. H&E results showed that fibrous calluses were already formed in the SCII group and that chondrocytes had begun to proliferate. By 10d, the chondrocytes in the SCII group became hypertrophic and mineralized, and the serum TGF-beta and Col-I alpha levels were significantly increased, which indicated that the mice with SCII treatment rapidly passed the cartilage repair period and new bone formation was accelerated. Skeletal muscle repaired bones through muscle paracrine factors. IGF-1 and irisin are the two major secretory cytokines. The results showed that the content of muscle homogenate IGF-1 in the SCII group reached the peak at 10d, followed by the up-regulation of Ihh, Patched, Gli1 and Col10 alpha in the callus through the bone surface receptor IGF-1R. Besides, SCII also significantly elevated the muscle irisin level (10 and 21d), and then increased Wnt10b, LRP5, beta-catenin and Runx2 expression in the callus by receptor alpha V beta 5. These results suggest that SCII can accelerate the process of endochondral osteogenesis and promote fracture healing through activating the Ihh/PThrp and Wnt/beta-catenin pathways by regulating muscle paracrine factors. To our knowledge, this is the first study to investigate the effect of marine-derived collagen on fracture healing. This study may provide a theoretical basis for the high-value application of the laryngeal cartilage of squid in the future.
Squid Cartilage Type II Collagen Accelerates Osteoporotic Fractures Healing Through Regulating Homocysteine Mediated EGF and FBN1 Signals

Abstract:Osteoporosis fracture (OPF) is a serious social public health concern in humans. Type II collagen from squid cartilage (SCII) has been proven to promote the healing process in normal fracture mice, however, the effect and mechanism of SCII to improve osteoporosis fracture remain unknown. In this study, type II collagen was obtained from squid laryngeal cartilage, and the effects of SCII on OPF were investigated in mice that experienced bilateral ovariectomy and open tibial fracture surgery after oral administration. The results showed that SCII supplementation significantly decreased the serum TNF-alpha, increased aggrecan levels on the 5 d, and elevated the serum TGF-beta and Col10 alpha levels on 13 d after fracture. Notably, H&E staining exhibited an obvious chondrocyte proliferation on 5 d, and a specific promotion of chondrocyte hypertrophic differentiation and mineralization in the SCII group on 13 d. These findings suggested that SCII promoted osteoporosis fracture healing by enhancing the proliferation and hypertrophy of chondrocyte. Mechanically, SCII supplementation significantly reduced homocysteine levels through down-regulating TMAO production and bile acid levels and activated the EGF/AKT/ CylinD1 and TGF beta/Smad1/5/8 pathways, thereby recovering the delay of chondrocyte proliferation and hypertrophy in OPF mice. This study investigated the effects of SCII on osteoporosis fracture healing for the first time and provided a new theoretical basis and options for the high-value application of squid cartilage.
Inhibition of Ss-Catenin Signaling in Chondrocytes Induces Delayed Fracture Healing in Mice

Abstract:Appropriate and controlled chondrogenesis and endochondral ossification play fundamental roles in the fracture healing cascade, a regenerative process involved in highly coordinated biological events, including the Wnt/beta-catenin signaling pathway. To examine the role and importance of this pathway in chondrocytes, we studied bone repair of closed tibias fractures in Col2a1-ICAT transgenic mice, in which the Wnt/beta-catenin signaling pathway is specially inhibited in chondrocytes. Radiological, histological, and histomorphometric analyses at 7, 9, 12, 14, 21, and 28 days after fracture demonstrated the bone repairs were retarded in Col2a1-ICAT transgenic mice, due to reduced and delayed cartilage formation, chondrocyte hypertrophy, and bone generation. In addition, at 5 weeks, Col2a1-ICAT transgenic mice exhibited a weak mechanical tolerance to four-point bending. Furthermore, quantitative-PCR analysis revealed that the expression of genes associated specifically with cartilage extracellular matrix formation (collagen II, collagen X, and mmp13), bone remodeling (alp, collagen I, and osteocalcin), and vascular extravagation (vegf), and transcriptional activators involved in cartilage generation and ossification (sox9 and runx2) was decreased and delayed in the fracture sites of Col2a1-ICAT transgenic mice during healing. Collectively, these results suggest that Wnt/beta-catenin signaling is critical for fracture healing, especially with respect to chondrogenesis and endochondral ossification. Thus, our study provides insight into the possible mechanisms of and therapeutic targets for improving normal facture repair and the healing of non-union fractures. Abstract:In growing children, growth plate cartilage has limited self-repair ability upon fracture injury always leading to limb growth arrest. Interestingly, one type of fracture injuries within the growth plate achieve amazing self-healing, however, the mechanism is unclear. Using this type of fracture mouse model, we discovered the activation of Hedgehog (Hh) signaling in the injured growth plate, which could activate chondrocytes in growth plate and promote cartilage repair. Primary cilia are the central transduction mediator of Hh signaling. Notably, ciliary Hh-Smo-Gli signaling pathways were enriched in the growth plate during development. Moreover, chondrocytes in resting and proliferating zone were dynamically ciliated during growth plate repair. Furthermore, conditional deletion of the ciliary core gene Ift140 in cartilage disrupted cilia-mediated Hh signaling in growth plate. More importantly, activating ciliary Hh signaling by Smoothened agonist (SAG) significantly accelerated growth plate repair after injury. In sum, primary cilia mediate Hh signaling induced the activation of stem/progenitor chondrocytes and growth plate repair after fracture injury.

Abstract:Fractures are frequent and severe musculoskeletal injuries. This study aimed to investigate the function of tenascin-C (TNC) in regulating chondrogenic during fracture healing and elucidate the underlying molecular mechanisms. A well-established femur fracture model in male C57BL/6J mice was used to transect the middle diaphysis of the femur. To identify the essential role of TNC, shTNC lentiviruses or TNC protein were administered in the animal model. Micro-CT analysis, histologic analysis, immunostaining assays, and gene expression analysis were employed to investigate the effect of TNC during fracture healing. An in vitro mesenchymal stem cell culture system was developed to investigate the role and molecular mechanism of TNC in regulating chondrogenesis. TNC expression was induced at the inflammatory phase and peaked at the cartilaginous callus phase during fracture healing. Knockdown of TNC expression in callus results in decreased callus formation and impaired fracture healing. Conversely, administration of exogenous TNC promoted chondrogenic differentiation, cartilage template formation and ultimately improved fracture healing. Both the Hedgehog and Hippo signaling pathways were found to be involved in the pro-chondrogenic function of TNC. Our observations demonstrate that TNC is a crucial factor responsible for endochondral ossification in fracture healing and provide a potential therapeutic strategy for promoting fracture healing.

Preparation of Novel Sea Cucumber Intestinal Peptides to Promote Tibial Fracture Healing in Mice by Inducing Differentiation of Hypertrophic Chondrocytes to the Osteoblast Lineage

Abstract:Scope: Hypertrophic chondrocytes have a decisive regulatory role in the process of fracture healing, and the fate of hypertrophic chondrocytes is not only apoptosis. However, the mechanism of sea cucumber (Stichopus japonicus) intestinal peptide (SCIP) on fracture promotion is still unclear. This study aims to investigate the effect of sea cucumber intestinal peptide on the differentiation fate of hypertrophic chondrocytes in a mouse tibial fracture model. Methods and results: Mice are subjected to open fractures of the right tibia to establish a tibial fracture model. The results exhibit that the SCIP intervention significantly promotes the mineralization of cartilage callus, decreases the expression of the hypertrophic chondrocyte marker Col X, and increases the expression of the osteoblast marker Col I. Mechanically, SCIP promotes tibial fracture healing by promoting histone acetylation and inhibiting histone methylation, thereby upregulating pluripotent transcription factors induced the differentiation of hypertrophic chondrocytes to the osteoblast lineage in a manner distinct from classical endochondral ossification. Conclusion: This study is the first to report that SCIP can promote tibial fracture healing in mice by inducing the differentiation of hypertrophic chondrocytes to the osteoblast lineage. SCIP may be considered raw material for developing nutraceuticals to promote fracture healing.
Type II Collagen from Cartilage of Acipenser baerii Promotes Wound Healing in Human Dermal Fibroblasts and in Mouse Skin

Abstract:Type II collagen is an important component of cartilage; however, little is known about its effect on skin wound healing. In this study, type II collagen was extracted from the cartilage of Acipenser baerii and its effect on in vitro and in vivo wound healing was compared to type I collagen derived from tilapia skin. Sturgeon cartilage collagen (SCC) was composed of α1 chains and with a thermal denaturation (Td) at 22.5 and melting temperature (Tm) at 72.5 °C. Coating SCC potentiated proliferation, migration, and invasion of human dermal fibroblast adult (HDFa) cells. Furthermore, SCC upregulated the gene expression of extracellular matrix (ECM) components (col Iα1, col IIIα1, elastin, and Has2) and epithelial-mesenchymal transition (EMT) molecules (N-cadherin, Snail, and MMP-1) in HDFa. Pretreatment with Akt and mitogen-activated protein kinase (MAPK) inhibitors significantly attenuated the HDFa invasion caused by SCC. In mice, the application of SCC on dorsal wounds effectively facilitated wound healing as evidenced by 40-59% wound contraction, whereas the untreated wounds were 18%. We observed that SCC reduced inflammation, promoted granulation, tissue formation, and ECM deposition, as well as re-epithelialization in skin wounds. In addition, SCC markedly upregulated the production of growth factors in the dermis, and dermal and subcutaneous white adipose tissue; in contrast, the administration of tilapia skin collagen (TSC) characterized by typical type I collagen was mainly expressed in the epidermis. Collectively, these findings indicate SCC accelerated wound healing by targeting fibroblast in vitro and in vivo.

Abstract:Totoaba macdonaldi-meat collagen peptides (TMCP)with molecular weight of 837 Da obtained from Mexican farmed Totoaba used combinative enzyme to investigate its effects on wound healing in mice.40 healthy male ICR mice were underwent surgical full-thickness incision with a diameter of 8 mm and were divided into 4 groups,which were normal control group and 0.66,1.33,2.0 g/kg.bw TMCP groups.TMCP were orally administered by gavage to mice in TMCP group from post-operation,and the control mice were given vehicle only for 14 d.The healing of the wound were observed every 2 d.Necropsy was performed on postoperative 7 and 14 d.The wound tissues were excised for HE staining,Masson staining,hydroxyproline measurement and immunohistochemical analysis.According to results,TMCP-M and TMCP-H had highly significant positive effect on all indexes (p ＜ 0.01).TMCP-L could highly significantly increase the healing rate,collagen content,VEGF and bFGF expression on 14 d(p ＜ 0.01).TMCP groups could promote collagen producing and tissue remodeling.TMCP had potential to promote wound healing and could be used in high-quality oral healthy products,that would provide a theoretical basis for high -efficient utilization.
FN1 Promotes Chondrocyte Differentiation and Collagen Production Via TGF-β/PI3K/Akt Pathway in Mice with Femoral Fracture.

Abstract:Fibronectin (FN) functions as a potent stimulator of osteogenic differentiation, and bone fracture healing. In FN family, FN1 acts as an interactive protein gene product to mediate chondrocyte adhesion. However, its effect on fracture healing remains elusive. Therefore, we aimed to investigate the involvement of FN1 in fracture healing. Hard callus formations were found at fracture site with thicker periosteum in lateral cortical bone area outside the fracture site in model mice. The decreased number of osteogenic cells in the middle of the callus region and increased extracellular matrix were suggestive of successful induction. Immunoblotting and RT-qPCR revealed that expression of FN1 was increased in tissues of fracture mice. As displayed by Safranin-fast green staining hematoxylin-eosin staining, the overexpression of FN1 at fracture site promoted osteoid formation and chondrocyte differentiation. The stimulating role of FN1 in collagen production was evidenced by increased levels of Col2, Col1, ColX, Osteonectin, and Osteocalcin and enhanced BMD, BV, BV/TV and Tb.Th values verified by immunoblotting and immunohistochemical staining. Additionally, the upregulation of FN1 contributed to promoted TGF-β, c-Caspase-9/t-Caspase-9 ratio and NF-κB p65 protein expression as well as lowered p-PI3K/PI3K and p-AKT/AKT ratios, implying the positive correlation between FN1 and the TGF-β/PI3K/Akt signaling pathway. The key findings of the present study provided evidence indicating that overexpression of FN1 contributes to fracture healing by activation of the TGF-β/PI3K/Akt signaling pathway.

Abstract:ABSTRACT Fractures heal predominantly through the process of endochondral ossification. The classic model of endochondral ossification holds that chondrocytes mature to hypertrophy, undergo apoptosis and new bone forms by invading osteoprogenitors. However, recent data demonstrate that chondrocytes transdifferentiate to osteoblasts in the growth plate and during regeneration, yet the mechanism(s) regulating this process remain unknown. Here, we show a spatially-dependent phenotypic overlap between hypertrophic chondrocytes and osteoblasts at the chondro-osseous border in the fracture callus, in a region we define as the transition zone (TZ). Hypertrophic chondrocytes in the TZ activate expression of the pluripotency factors [Sox2, Oct4 (Pou5f1), Nanog], and conditional knock-out of Sox2 during fracture healing results in reduction of the fracture callus and a delay in conversion of cartilage to bone. The signal(s) triggering expression of the pluripotency genes are unknown, but we demonstrate that endothelial cell conditioned medium upregulates these genes in ex vivo fracture cultures, supporting histological evidence that transdifferentiation occurs adjacent to the vasculature. Elucidating the cellular and molecular mechanisms underlying fracture repair is important for understanding why some fractures fail to heal and for developing novel therapeutic interventions.

Bio-inspired Mineralization Collagen Induce Fibrocartilage Regeneration after Tendon-Bone Injury by Activating Gli1+Dkk3+ Progenitor Cells

Abstract:Summary A fibrocartilaginous connection between the tendon and bone, plays a critical role in transferring force from muscle to bone to enable joint movement. However, due to the high mechanical stress it experiences, the enthesis is vulnerable to injury and incapable of regenerating. The spatial relationship and functional basis of the principal components of the fibrocartilage - mineral and collagen - have not been clearly elucidated, which is a significant remaining gap in reconstructing complex architectures for promoting interface tissue regeneration. Here, using three-dimensional electron tomography imaging and high-resolution two-dimensional electron microscopy, we discover that mineral particles form a continuous cross-fibrillar phase within the fibrocartilage region. By developing a “floating mineralization” system, we fabricate a three-layer hydrogel that mimics the hierarchical nano- to micro-scale structure of tendon-bone interface (TBI). The middle layer is noteworthy for its resemblance to the nanostructure of fibrocartilage and its superior ability to induce mineralized fibrochondrogenesis in vitro . Based on motor function analysis, imaging diagnosis, histological staining, immunofluorescence staining, and biomechanics performance, we demonstrate that in situ transplantation of the gradient hydrogel achieved tendon-fibrocartilage-bone synchronous regeneration and result in 68% maximum mechanical recovery at 8-week postoperation. Single-cell RNA sequencing analysis reveals that a unique atlas of in situ stem/progenitor cells is generated during the TBI healing in vivo . Notably, the bio-inspired hydrogel microenvironment drived endogenous Gli1 + Dkk3 + progenitor cells, playing a key role in TBI regeneration. Therefore, we have successfully decoded and reconstructed the nanostructure of fibrocartilage, which has great potential in TBI regeneration.
Mechanically Conditioned Multilayered Angle-Ply Collagen Scaffolds Promote Annulus Fibrosus Regeneration

Abstract:Annulus fibrosus (AF) injury, as a result of intervertebral disk degeneration (IVDD) or herniation, is very difficult to heal due to its avascular characteristics. Tissue engineering strategies have become increasingly promising to treat IVDD. However, in light of the fact that AF displays an anisotropic microstructure that results in gradient mechanical properties, fabrication of tissue engineered AF remains challenging. In this study, we fabricated micropatterned collagen scaffolds with angle-ply structure mimicking the microstructural features of AF. Following that, we cultured bone marrow mesenchymal stromal cells (BMSCs) in the scaffolds and applied cyclic stretching to them to mechanically stimulate the cells. We found that aligned cells under mechanical loading showed elevated expression of matrix anabolism-related genes and proteins. Further studies indicated that Caveolin-1 (CAV1) might mediate the nuclear translocation of Yes-associated protein (YAP) in response to external mechanical cues of microgroove-patterned membranes and mechanical loading. Micropatterned collagen membranes, with cells aligning at +/- 30 degrees along the longitudinal dimension, were rolled up together to form an angle-ply and multilayered tissue. After being implanted into the caudal vertebra in a rat model for 4 weeks, biomimetic tissues showed improved restoration of the disk. In general, our biomimetic collagen membranes exhibit great potential in terms of AF regeneration, with AF-like heterogeneous microstructures that satisfy the biomechanical property of the native disk.
Squid Type II Collagen As a Novel Biomaterial: Isolation, Characterization, Immunogenicity and Relieving Effect on Degenerative Osteoarthritis Via Inhibiting STAT1 Signaling in Pro-Inflammatory Macrophages.

Abstract:Collagen from marine organisms has a broad prospect in biomedical field, yet the knowledge on marine-derived type II collagen is rare. Herein, a novel type II collagen was successfully isolated from squid cartilage for the first time. After being characterized, the immunogenicity of squid type II collagen (SCII) was evaluated and compared with that of bovine type II collagen (BCII). Then investigations were further conducted for the impacts of SCII on pro-inflammatory macrophages and macrophage chemotaxis. The degenerative osteoarthritis (OA) -relieving effects of SCII were explored using OA rat model in vivo. Our results demonstrated that the isolated SCII maintained triple-superhelical structure of native collagen with high purity. Different from BCII, SCII presented no immunogenicity since it neither induced abnormal proliferation of lymphocytes in vitro nor changed the basic levels of IgM, IgG, anti-type II collagen IgG and CD4+/CD8+ lymphocytes ratio in vivo. Additionally, SCII also exerted prominent anti-inflammatory effects. SCII significantly reduced the production of pro-inflammatory cytokines by enhancing the activity of TCPTP and subsequently prompting the dephosphorylation of p-STAT1 in pro-inflammatory macrophages. Besides, it indirectly prevented hypertrophic changes of chondrocytes, and markedly impeded chemotaxis of macrophages. Moreover, inflammation condition in OA rats was significantly alleviated under treatment with SCII. These data suggested that the newly developed SCII could not only avoid the immunogenic risks of collagen derived from terrestrial animals, but more importantly, provide new choice for the control and treatment of OA.

Abstract:Osteogenesis Imperfecta (OI) is a genetic bone dysplasia characterized by bone deformities and fractures caused by low bone mass and impaired bone quality. OI is a genetically heterogeneous disorder that most commonly arises from dominant mutations in genes encoding type‐I collagen (COL1A1 and COL1A2). In addition, OI is recessively inherited with the majority of cases resulting from mutations in prolyl‐3‐hydroxylation complex members, which includes Cartilage Associated Protein (CRTAP). OI patients are at an increased risk of fracture throughout their lifetimes. However, non‐union or delayed healing has been reported in 24% of fractures and 52% of osteotomies. Additionally, re‐fractures typically go unreported making the frequency of re‐fractures in OI patients unknown. Thus, there is an unmet need to better understand the mechanisms by which OI affects fracture healing. Using an open tibial fracture model, our study demonstrates delayed healing in both Col1a2G610c/+ and Crtap–/– OI mouse models (dominant and recessive OI, respectively) that is associated with reduced callus size and predicted strength. Callus cartilage distribution and chondrocyte maturation were altered in OI suggesting accelerated cartilage differentiation. Importantly, we determined that healed fractured tibia in female OI mice are biomechanically weaker when compared to the contralateral unfractured bone suggesting that abnormal OI fracture healing OI may prime future re‐fracture at the same location. We have previously shown up‐regulated TGF‐β signaling in OI and we confirm this in the context of fracture healing. Interestingly, treatment of Crtap–/– mice with the anti‐TGF‐β antibody, 1D11 resulted in further reduced callus size and predicted strength, highlighting the importance of investigating dose response in treatment strategies. These data provide valuable insight into the effect of the extracellular matrix (ECM) on fracture healing, a poorly understood mechanism, and support the need for prevention of primary fractures to decrease incidence of refracture and deformity in OI patients.This article is protected by copyright. Cartilage Repair in Degenerative Osteoarthritis Mediated by Squid Type II Collagen Via Immunomodulating Activation of M2 Macrophages, Inhibiting Apoptosis and Hypertrophy of Chondrocytes.

Abstract:Cartilage lesions in degenerative osteoarthritis (OA) are involved with pathological microenvironmental alterations induced by inflammatory macrophages, and apoptotic and/or hypertrophic chondrocytes. However, current non-operative therapies for cartilage repair in OA can rarely achieve long-term and satisfactory outcomes. This study aims to evaluate a newly developed squid type II collagen (SCII) for repairing OA-induced cartilage lesions. Our in vitro data show that SCII induces M2 polarization of macrophages, and activates macrophages to express pro-chondrogenic genes (TGF-β and IGF), which greatly improves the microenvironment around chondrocytes to produce type II collagen and glycosaminoglycan. In addition, glycine in SCII activates glycine receptors on inflammatory chondrocytes to decrease intracellular calcium concentration, leading to effective inhibition of chondrocyte apoptosis and hypertrophy. The in vitro effects of SCII are further confirmed in vivo. In a rat model of OA, SCII increases the ratio of M2 macrophages, elevates the levels of pro-chondrogenic cytokines (TGF-β1 and TGF-β3) in synovial fluid, and inhibits chondrocyte apoptosis and MMP13 production. Our findings show that SCII immunomodulates M2 activation of macrophages to skew the local OA microenvironment towards a pro-chondrogenic atmosphere, and promotes cartilage repair under inflammatory condition. It shows great potential for SCII to be a novel biomaterial for cartilage repair in OA.
The Experimental Study on the Gene Expession ofType- I Collagen in the Repair of Mandibular Defect in Rat

Abstract:To detect the gene expression of extracellar matrix type I collagen caused by biomaterial and the influence of type I collagen gene expression to the healing pattems in different time points, single hydroxyapatit and hydroxyapatit combined with TGF- β 1 were implanted in the mandibular defect model and single defect was used for control. Methods:In this experiment, mandibular defects were established in SD rats(n = 68), divided into 3 groups. Hydroxyapatit (HA) combined with TGF - β 1 as a bone graft substitute was tested by slot blot hybridiztion to observe type I collagen mRNA and the distribution of Ⅰ collagen protein under sirius red stain-poplarizing light. Results: The protein and mRNA of I collagen existed statistical difference (P ＜ 0.05)in 3 groups. Conclusion: During the bone defect healing, there existed the I collagen mRNA increasing gradually. The study show TGF - β 1 promoting the bone healing process by the path of their corresponding collagen protein.
Type II Collagen Sponges Facilitate Tendon Stem/Progenitor Cells to Adopt More Chondrogenic Phenotypes and Promote the Regeneration of Fibrocartilage-Like Tissues in a Rabbit Partial Patellectomy Model.

Abstract:OBJECTIVE:Fibrocartilage transition zone (FC) is difficult to regenerate after surgical re-attachment of tendon to bone. Here, we investigated whether type II collagen-sponges (CII-sponges) facilitated tendon stem/progenitor cells (TSPCs) to adopt chondrogenic phenotypes and further observed if this material could increase the FC areas in bone-tendon junction (BTJ) injury model.METHODS:CII-sponges were made as we previously described. The appearance and pore structure of CII-sponges were photographed by camera and microscopies. The viability, proliferation, and differentiation of TSPCs were examined by LIVE/DEAD assay, alamarBlue, and PKH67 in vitro tracking. Subsequently, TSPCs were seeded in CII-sponges, Matrigel or monolayer, and induced under chondrogenic medium for 7 or 14 days before being harvested for qPCR or being transplanted into nude mice to examine the chondrogenesis of TSPCs. Lastly, partial patellectomy (PP) was applied to establish the BTJ injury model. CII-sponges were interposed between the patellar fragment and tendon, and histological examination was used to assess the FC regeneration at BTJ after surgery at 8 weeks.RESULTS:CII-sponges were like sponges with interconnected pores. TSPCs could adhere, proliferate, and differentiate in this CII-sponge up to 14 days at least. Both qPCR and immunostaining data showed that compared with TSPCs cultured in monolayer or Matrigel, cells in CII-sponges group adopted more chondrogenic phenotypes with an overall increase of chondrocyte-related genes and proteins. Furthermore, in PP injured model, much more new formed cartilage-like tissues could be observed in CII-sponges group, evidenced by a large amount of positive proteoglycan expression and typical oval or round chondrocytes in this area.CONCLUSION:Our study showed that CII-sponges facilitated the TSPCs to differentiate toward chondrocytes and increased the area of FCs, which suggests that CII-sponges are meaningful for the reconstruction of FC at bone tendon junction. However, the link between the two phenomena requires further research and validation.
The Comprehensive Role of Mechanosensitive Piezo1 in Fracture Healing and Delayed Union

Abstract:Background: The biomechanical environment plays a dominant role in the fracture healing, and Piezo1 is regarded as the major mechanosensor in bone homeostasis. However, the role of Piezo1 in fracture healing is not delineated yet.Methods: Using mid-femur fracture model and clinical samples, we examined the expression pattern of Piezo (Piezo1 and Piezo2) during the healing phases by singal cell RNA-seq, RT-PCR and western blot analyses. Knockdown of Piezo1 by shRNA, as well as specific agonist were used to elucidate the role and regulatory mechanism of Piezo1 during fracture healing. The tail suspension experiment was used to evaluate the therapeutic effect of activated Piezo1.Findings: We find that Piezo1 is mainly expressed in osteoblastic lineage cells and endothelial cells during fracture repair. Moreover, Piezo1 expression is upregulated during fracture repair in human and mice. Downregulation of Piezo1 in callus leads to impaired fracture healing. Furthermore, administration of Piezo1 channel specific agonist accelerates fracture healing through the stimulation of angiogenesis and osteogenesis, along with bone remodeling in late stage. Mechanistically, activation of Piezo1 promotes expression of Yes-associated protein (YAP) and its nuclear localization, which in turn increases the expression and nuclear localization of β-catenin. Moreover, our results showed that YAP and β-catenin could interact to form a transcription complex in the nucleus, which upregulates both BMP2 and VEGFA. The PI3K/AKT pathway promotes fracture healing through its crosstalk with Wnt/β-catenin

Abstract:<p>PI3K/AKT is one of the key pathways that regulate cell behaviors including apoptosis, proliferation, and differentiation. Although previous studies have demonstrated that this pathway is a crucial regulator of osteoblasts, the role of PI3K/AKT in fracture healing remains unclear. It is well known that the Wnt/β-catenin pathway plays an essential role in bone regeneration. However, whether there exists crosstalk between Wnt/β-catenin and PI3K/AKT in regulating osteoblasts and bone repair has not been reported. To address these issues, we establish a stabilized fracture model in mice and show that PI3K inhibitor LY294002 substantially inhibits the bone healing process, suggesting that PI3K/AKT promotes fracture repair. More importantly, we report that PI3K/AKT increases phosphorylation of GSK-3β at Ser9 and phosphorylation of β-catenin at Ser552 in fracture callus and murine osteoblastic MC3T3-E1 cells, both of which lead to β-catenin stabilization, nuclear translocation, as well as β-catenin-mediated TCF-dependent transcription, suggesting that β-catenin is activated downstream of PI3K/AKT. Furthermore, we show that ICG001, the inhibitor of β-catenin transcriptional activity, attenuates PI3K/AKT-induced osteoblast proliferation, differentiation, and mineralization, indicating that the PI3K/AKT/β-catenin axis is functional in regulating osteoblasts. Notably, the PI3K/AKT pathway is also activated by Wnt3a and is involved in Wnt3a-induced osteoblast proliferation and differentiation. Hence, our results reveal the existence of a Wnt/PI3K/AKT/β-catenin signaling nexus in osteoblasts, highlighting complex crosstalk between PI3K/AKT and Wnt/β-catenin pathways that are critically implicated in fracture healing.</p>

Abstract:The high-value-added compounds of sea cucumber (Stichopus japonicus) intestine have not been exploited in sea cucumber processing. Here, low molecular weight peptides from the sea cucumber intestine (SCIP) were pre-pared using papain, and the effect of SCIP on promoting cartilage callus formation and their underlying mechanisms were studied. The results exhibited that the SCIP supplement markedly increased the area of the cartilage callus and ameliorated the early inflammatory environment at the fracture site by modulating the levels of inflammatory factors. Furthermore, SCIP promoted the polarization of M1 to M2 macrophages and finally promoted the differentiation of mesenchymal stem cells into chondrocytes at the fracture site through M2-like macrophage-mediated TGF-family factors and secretion of exosomes. This is the first report that SCIP can pro-mote cartilage callus formation in mice with tibial fractures by promoting the polarization of M1 to M2 mac-rophages. SCIP might be considered raw material for developing nutraceutical additives.
