Abstract:Integrating multifunctional nanostructures capable of radiotherapy and photothermal ablation is an emerging alternative in killing cancer cells. In this work, we report a novel plasmonic heterostructure formed by decorating AuPt nanoparticles (NPs) onto the surfaces of CuS nanosheets ( NSs) as a highly effective nanotheranostic toward dual-modal photoacoustic/computed tomography imaging and enhanced synergistic radiophotothermal therapy. These heterostructures can confer higher photothermal conversion efficiency via the local electromagnetic enhancement as well as a greater radiation dose deposition in the form of glutathione depletion and reactive oxygen species generation. As a result, the depth of tissue penetration is improved, and hypoxia of the tumor microenvironment is alleviated. With synergistic enhancement in the efficacy of photothermal ablation and radiotherapy, the tumor can be eliminated without later recurrence. Abstract:Aim: Polyethylene glycol modified mesoporous silica-coated bismuth nanohybrids (Bi@mSiO2-PEG) are fabricated for chemothermotherapy and multimodal imaging. Materials & methods: The Bi@mSiO2-PEG are synthesized by coating mesoporous SiO2 onto metallic Bi cores, followed by PEG modification. Their cytotoxicity, photothermal effect, drug loading, antitumor effect and imaging abilities are evaluated. Results: The nanohybrids show good biocompatibility, strong near-infrared absorbance, high photothermal conversion efficiency (∼36.6%), prominent infrared thermal imaging and photothermal killing efficacy on cancer cells. Utilizing the nanohybrids as potent drug carriers, a synergistic antitumor effect through chemothermotherapy is realized. Thanks to the superhigh x-ray attenuation coefficient and strong photothermal ability, high-contrast photoacoustic and x-ray computed tomography imaging are achieved. Conclusion: These results reveal great potentials of the Bi@mSiO2-PEG for precise and efficient anticancer treatments.
Nanoparticles of Chlorin Dimer with Enhanced Absorbance for Photoacoustic Imaging and Phototherapy

Abstract:Nanoparticle assembled from organic molecules is a versatile platform to integrate various functionalities for theranostics. In this work, nanoparticles are constructed from chlorin dimers that are synthesized by reducing porphyrin molecules. Chlorin dimers can assemble into nanoscale aggregates in the absence of surfactants or other auxiliary agent. The resulting nanoparticles of chlorin dimer exhibit much higher absorbance than the porphyrin counterparts, resulting in enhanced photodynamic and photothermal activity upon irradiation. The forming nanoparticles can be effectively endocytosed by the tumor cells, inducing apoptosis under irradiation. Tumor growth on mice model is inhibited by the photodynamic and photothermal treatment in vivo. Furthermore, this nanoparticle can be used for photoacoustic imaging. It is believed that the integrated imaging and phototherapeutic capability in one nanoparticle is beneficial for future cancer diagnosis, therapy, and molecular imaging. Robust organic nanoparticles are developed for simultaneous photodynamic and photothermal therapy as well as photoacoustic imaging. The formed nanoparticles possess enhanced absorbance, which results in better efficacy of phototherapy in tumor inhibition. This result can eventually be very helpful in design of simple and ideal nanoparticle formulations for cancer patients.

Abstract:Optical theranostic nanoagents that seamlessly and synergistically integrate light-generated signals with photothermal or photodynamic therapy can provide opportunities for cost-effective precision medicine, while the potential for clinical translation requires them to have good biocompatibility and high imaging/therapy performance. We herein report an intraparticle molecular orbital engineering approach to simultaneously enhance photoacoustic brightness and photothermal therapy efficacy of semiconducting polymer nanoparticles (SPNs) for in vivo imaging and treatment of cancer. The theranostic SPNs have a binary optical component nanostructure, wherein a near-infrared absorbing semiconducting polymer and an ultrasmall carbon dot (fullerene) interact with each other to induce photoinduced electron transfer upon light irradiation. Such an intraparticle optoelectronic interaction augments heat generation and consequently enhances the photoacoustic signal and maximum photothermal temperature of SPNs by 2.6- and 1.3-fold, respectively. With the use of the amplified SPN as the theranostic nanoagent, it permits enhanced photoacoustic imaging and photothermal ablation of tumor in living mice. Our study thus not only introduces a category of purely organic optical theranostics but also highlights a molecular guideline to amplify the effectiveness of light-intensive imaging and therapeutic nanosystems.

Abstract:<p>Multi‐modality imaging‐guided cancer therapy is considered as a powerful theranostic platform enabling simultaneous precise diagnosis and treatment of cancer. However, recently reported multifunctional systems with multiple components and sophisticate structures remain major obstacles for further clinical translation. In this work, a single‐photomolecular theranostic nanoplatform is fabricated via a facile nanoprecipitation strategy. By encapsulating a semiconductor oligomer (IT‐S) into an amphiphilic lipid, water‐dispersible IT‐S nanoparticles (IT‐S NPs) are prepared. The obtained IT‐S NPs have a very simple construction and possess ultra‐stable near‐infrared (NIR) fluorescence (FL)/photoacoustic (PA) dual‐modal imaging and high photothermal conversion efficiency of 72.3%. Accurate spatiotemporal distribution profiles of IT‐S NPs are successfully visualized by NIR FL/PA dual‐modal imaging. With the comprehensive in vivo imaging information provided by IT‐S NPs, tumor photothermal ablation is readily realized under precise manipulation of laser irradiation, which greatly improves the therapeutic efficacy without any obvious side effects. Therefore, the IT‐S NPs allow high tumor therapeutic efficacy under the precise guidance of FL/PA imaging techniques and thus hold great potential as an effective theranostic platform for future clinical applications.</p>

Abstract:The core of the figure represents a mechanistic diagram of the localized surface plasmon resonance effect under second near‐infrared window laser irradiation, followed by an outer circle listing several representative plasmonic nanomaterials. The right half of the outermost circle lists four photoacoustic imaging applications, and the left half indicates the four photothermal therapy aspects of the plasmonic nanomaterials. Photoacoustic imaging (PAI) and imaging‐guided photothermal therapy (PTT) in the second near‐infrared window (NIR‐II, 1000–1700 nm) have received increasing attention owing to their advantages of greater penetration depth and higher signal‐to‐noise ratio. Plasmonic nanomaterials with tunable optical properties and strong light absorption provide an alternative to dye molecules, showing great prospects for phototheranostic applications. In this review, the research progress in principally modulating the optical properties of plasmonic nanomaterials, especially affecting parameters such as size, morphology, and surface chemical modification, is introduced. The commonly used plasmonic nanomaterials in the NIR‐II window, including noble metals, semiconductors, and heterostructures, are then summarized. In addition, the biomedical applications of these NIR‐II plasmonic nanomaterials for PAI and PTT in phototheranostics are highlighted. Finally, the perspectives and challenges for advancing plasmonic nanomaterials for practical use and clinical translation are discussed.

Abstract:Nanotheranostic agents that integrate diagnosis and treatment are promising for precision medicine, but they encounter some obstacles such as penetration depth and efficiency. In this study, novel carbon nitride-rose bengal nanoparticles (CN-RB NPs) with a graphite carbon nitride skeleton were synthesized by one-step thermal copolymerization. The enhanced absorption in the near-infrared-II region (NIR-II) endows CN-RB NPs with an excellent photothermal effect under 1064 nm laser irradiation, as well as an obvious photoacoustic signal for imaging in vivo . Interestingly, due to the introduced iodine element, CN-RB NPs exhibit enhanced radiation therapy, indicating that CN-RB NPs can achieve ideal therapeutic outcome through collaborative photothermal/radiation therapy under the guidance of NIR-II photoacoustic imaging. Moreover, CN-RB NPs demonstrate minimal side effects and long-term biological stability after 14 days. Therefore, the proposed new multifunctional nano-platform CN-RB NPs hold great potential in the application of deep therapeutics.

Abstract:The integrated functionalities of multiple diagnostic and therapeutic modalities in a single phototheranostic nanoplatform have been validated as a significant breakthrough toward cancer theranostics. Nevertheless, most reported nanoplatforms are constructed by using complex components through difficult fabrication processes, and often need different excitation wavelengths. Herein, a novel small-molecule dye, DPP-BDT, with absorption in the NIR-I window and fluorescence emission in the NIR-II region was designed and synthesized, and it presented excellent photodynamic and photothermal performance. Based on single component DPP-BDT, multifunctional phototheranostics were rationally and successfully constructed. From in vitro and in vivo experiments, such nanotheranostics showed excellent tumor destruction abilities under a single wavelength of laser irradiation, benefiting from the NIR-II fluorescence/photoacoustic dual-modal imaging guided photodynamic/photothermal combination therapy.

Abstract:Combination of photothermal therapy (PTT) and photodynamic therapy (PDT) has become a promising cancer treatment in recent years. However, their applications are limited by complex synthetic protocols and low efficacy. Hence, optimizing experimental approach and improving the efficiency of phototherapy is the current research focus. In this work, various pyrolysis temperatures and sizes of zeolitic imidazolate framework-8 (ZIF-8) derived carbon nanoparticles (ZCNs) are obtained by a simple direct pyrolysis of the ZIF-8 nanoparticles. Meanwhile, the ZCNs can be used as photothermal agents and photosensitizers to produce heat and reactive oxygen species simultaneously upon near-infrared laser irradiation. Moreover, it is observed that the phototherapy effects and photoacoustic (PA) signal of ZCNs could be enhanced with the increase in the nanoparticle size. Subsequently, guided by PA imaging, the therapeutic effect of ZCNs is investigated on a small animal model, where tumors are entirely eliminated with minimal side effect, demonstrating the high efficacy of the larger size of ZCNs through combination of PTT and PDT. Therefore, it is expected that the ZCN is a simple and highly effective phototherapeutic platform for oncotherapy, and the concept of size-dependent enhanced behavior of phototherapy and PA imaging will be very useful in the development of nanomaterials for cancer therapy.

Abstract:Synergy of light and sound, such as photoacoustic imaging, has demonstrated promising potentials in advancing the state-of-the-art of biomedical imaging by ultrasonically detecting the absorption of light, no matter the light is diffusive or not. As a result, optical contrast can be revealed with ultrasound or sub-ultrasound spatial resolution, which can be exploited to map sensitively the early tissue changes associated with the onset and development of early-stage diseases. The sources of photoacoustic emissions, however, could be very complicated within living biological tissue, as many tissue constituents may absorb light and generate heat to emit the ultrasonic waves, which considerably reduces the contrast between the target and the background. Therefore, exogenous agents, such as nanoparticles, that have absorption spectrum distinctive from that of background tissues have been widely used in the field to enhance the photoacoustic detection contrast and sensitivity at selected optical wavelengths. This talk summarizes our recent efforts in this direction that have explored various nanoparticles for more robust and more sensitive diagnosis and treatment of early-stage liver cancer based on photoacoustic imaging and photothermal effect. While the studies are based on merely small animal models, they may inspire further explorations towards preclinical and clinical trials.

Ultrathin Two‐Dimensional Plasmonic PtAg Nanosheets for Broadband Phototheranostics in Both NIR‐I and NIR‐II Biowindows

Abstract:<p>Broadband near-infrared (NIR) photothermal and photoacoustic agents covering from the first NIR (NIR-I) to the second NIR (NIR-II) biowindow are of great significance for imaging and therapy of cancers. In this work, ultrathin two-dimensional plasmonic PtAg nanosheets are discovered with strong broadband light absorption from NIR-I to NIR-II biowindow, which exhibit outstanding photothermal and photoacoustic effects under both 785 and 1064 nm lasers. Photothermal conversion efficiencies (PCEs) of PtAg nanosheets reach 19.2% under 785 nm laser and 45.7% under 1064 nm laser. The PCE under 1064 nm laser is higher than those of most reported inorganic NIR-II photothermal nanoagents. After functionalization with folic acid modified thiol-poly(ethylene glycol) (SH-PEG-FA), PtAg nanosheets endowed with good biocompatibility and 4T1 tumor-targeted function give high performances for photoacoustic imaging (PAI) and photothermal therapy (PTT) in vivo under both 785 and 1064 nm lasers. The effective ablation of tumors in mice can be realized without side effects and tumor metastasis by PAI-guided PTT of PtAg nanosheets under 785 or 1064 nm laser. The results demonstrate that the prepared PtAg nanosheets with ultrathin thickness and small size can serve as a promising phototheranostic nanoplatform for PAI-guided PTT of tumors in both NIR-I and NIR-II biowindows.</p>

Abstract:Developing an effective theranostic nanoplatform remains a great challenge for cancer diagnosis and treatment. Here, BiOI@Bi2S3@BSA (bovine serum albumin) semiconductor heterojunction nanoparticles (SHNPs) for triple-combination radio/photodynamic/photothermal cancer therapy and multimodal computed tomography/photoacoustic (CT/PA) bioimaging are reported. On the one hand, SHNPs possess strong X-ray attenuation capability since they contain high-Z elements, and thus they are anticipated to be a very competent candidate as radio-sensitizing materials for radiotherapy enhancement. On the other hand, as a semiconductor, the as-prepared SHNPs offer an extra approach for reactive oxygen species generation based on electron-hole pair under the irradiation of X-ray through the photodynamic therapy process. This X-ray excited photodynamic therapy obviously has better penetration depth in bio-tissue. What's more, the SHNPs also possess well photothermal conversion efficiency for photothermal therapy, because Bi2S3 is a thin band semiconductor with strong near-infrared absorption that can cause local overheat. In vivo tumor ablation studies show that synergistic radio/photodynamic/photothermal therapy achieves more significant therapeutic effect than any single treatment. In addition, with the strong X-ray attenuation and high near-infrared absorption, the as-obtained SHNPs can also be applied as a multimodal contrast agent in CT/PA imaging.

Abstract:Phototherapy has garnered considerable interest for its potential to revolutionize conventional cancer treatment. Organic materials with near-infrared II (NIR-II, 1000-1700 nm) fluorescence and photothermal effects are key for precise tumor diagnosis and treatment, yet optimizing their output for higher resolution and reduced photodamage remains a challenge. Herein, a multifunctional NIR-II photosensitizer (LSC) has been developed using the aggregation-induced emission (AIE) technology. The utilization of Thieno[3,2-b]thiophene as an electron-rich and bulky donor/acceptor bridge has allowed for the elongation of conjugation length and distortion of the AIE main chain. This strategic modification effectively enhances the electron push-pull effect, endowing the LSC with a Stokes shift of over 400 nm and AIE characteristics. We have successfully built-up stable nanoparticles called FA-LSC NPs using a nano-precipitation method. These nanoparticles exhibit high NIR-II fluorescent brightness (ε × QY = 1064 M-1 cm-1) and photothermal conversion efficiency (41%). Furthermore, the biocompatible FA-LSC NPs demonstrate effective tumor accumulation and exceptional photothermal therapeutic efficacy both in vitro and in vivo. These nanoparticles were applied to fluorescence-photothermal dual-mode imaging-guided photothermal ablation in a Hela tumor xenograft mouse model, resulting in favorable photothermal clearance outcomes.

Abstract:Engineering design of plasmonic nanomaterials as on-demand theranostic nanoagents with imaging, drug carrier, and photothermal therapy (PTT) functions have profound impact on treatment of cancer. Here, a facile 'one-pot' template-free hydrothermal route was firstly developed for synthesis of plasmonic oxygen deficiency molybdenum oxide hollow nanospheres functionalized by poly(ethylene glycol) (PEG-MoO(3-x) HNSs). The as-prepared PEG-MoO(3-x) HNSs not only have good biocompatibility but also exhibit obvious localized surface plasmon resonance (LSPR) absorption in the near-infrared (NIR) region. Especially, due to its intrinsic mesoporous properties and effective photothermal conversion efficiency upon 808-nm NIR laser irradiation, the PEG-MoO(3-x) HNSs can be applied as a pH/NIR laser dual-responsive camptothecin (CPT) drug delivery nanoplatform for chemotherapy as well as PTT to cancer cells. A remarkably improved synergistic therapeutic effect to pancreatic (PANC-1) tumor-bearing mice was obtained compared to the result of chemotherapy or PTT alone. Apart from its application for drug delivery, the PEG-MoO(3-x) HNSs can also be employed as an effective contrast nanoagent for photoacoustic (PAT) imaging because of its high NIR absorption, making it promising as a theranostic nanoagent for PAT imaging-guided chemo-photothermal combinational cancer therapy in the nanomedicine field.

Abstract:Precise manipulation of optical properties through the structure‐evolution of plasmonic nanoparticles is of great interest in biomedical fields including bioimaging and phototherapy. However, previous success has been limited to fixed assembled structures or visible–NIR‐I absorption. Here, an activatable NIR‐II plasmonic theranostics system based on silica‐encapsulated self‐assembled gold nanochains (AuNCs@SiO2) for accurate tumor diagnosis and effective treatment is reported. This transformable chain structure breaks through the traditional molecular imaging window, whose absorption can be redshifted from the visible to the NIR‐II region owing to the fusion between adjacent gold nanoparticles in the restricted local space of AuNCs@SiO2 triggered by the high H2O2 level in the tumor microenvironment (TME), leading to the generation of a new string‐like structure with strong NIR‐II absorption, which is further confirmed by finite‐difference‐time‐domain (FDTD) simulation. With the TME‐activated characteristics, AuNCs@SiO2 exhibits excellent properties for photoacoustic imaging and a high photothermal conversion efficiency of 82.2% at 1064 nm leading to severe cell death and remarkable tumor growth inhibition in vivo. These prominent intelligent TME‐responsive features of AuNCs@SiO2 may open up a new avenue to explore optical regulated nano‐platform for intelligent, accurate, and noninvasive theranostics in NIR‐II window.

Abstract:As a key branch of the cross-discipline biophotonics, phototherapy, including photodynamic therapy (PDT), and photothermal therapy (PTT), is promising in biomedicine and visible light-driving PDT has been applied to clinical treatment. However, extensive applications of phototherapy are limited by the hypoxic microenvironment, laser penetration depth, and potential complexity for combined PDT/PTT. Thus, NIR-responsive oxygen self-supply nanocomposites functionalized with photosensitizers for achieving simultaneous in-depth PDT/PTT are urgently required. Herein, a multifunctional platform has been fabricated by co-immobilizing monodispersed ultrasmall Pd nanoclusters and a photosensitizer 5,10,15,20-Tetrakis (4-Aminophenyl)-21H,23H Porphyrin (Thp) on the surface of Ti 3 C 2 T x MXene nanosheets, generating the Pd-Thp-Ti 3 C 2 T x nanocomposite. Material characterization demonstrated that Pd nanoclusters and Thp were well-distributed on the MXene surface while MXene maintained its photothermal conversion efficiency and broad absorption. In this nanoplatform, irradiated by the single 808 nm laser, Pd selectively catalyzed the decomposition of H 2 O 2 to O 2 , and O 2 was continuously supplied to Thp for enhanced NIR-driving PDT. The in vivo fluorescence and photothermal imaging demonstrated the pronounced accumulation of nanocomposites in the tumor site. Both in vitro and in vivo results clearly demonstrated the nanocomposite had good biocompatibility, and that the synergistic PTT and enhanced PDT made apoptosis of the tumor cell achievable. This work not only proves this Pd-Thp-Ti 3 C 2 T x nanocomposite serves a promising solution for tumor hypoxia by inducing apoptosis of tumor cells with synergistic PTT and PDT, but also broadens the application of promising optical materials in biomedical field.

Abstract:Organic nanomaterials have made great progress in photothermal cancer cells therapy. However, it is still a challenge to synthesize self-assembled organic materials with good water dispersity and biocompatibility. Here, we designed near-infrared squaric acid derivatived nanoparticles as a versatile platform for tumor cells ablation. Such self-assembled organic nanaoparticles showed high photothermal conversion and strong PA signals, enabling the successful thermal abaltion of cancer cells.
