Abstract:Although combined chemotherapy (Chemo), photothermal (PTT) and photodynamic (PDT) in cancer therapy has drawn significant attention due to its superior anticancer ability, the required high intensity of irradiation results in serious photo-toxicity to healthy neighboring cells, and thus limits its biomedical applications. Herein, we developed an ultralow-intensity near infrared (NIR) light synchronously activated collaborative Chemo/PTT/PDT nanoplatform. The nanoplatform is composed of a highly emissive upconversion (UC) core, chlorin e6 (Ce6) photosensitizer and the anticancer drug doxorubicin hydrochloride (DOX) co-loaded in a mesoporous silica (MS) shell, and polyethylene glycol-modified graphene (PGO) acts as both the photothermal reagent and smart switch for promoted drug release. Upon 808 nm NIR light exposure with ultralow intensity (0.25 W cm-2), which is below the maximum permissible exposure (MPE, 0.33 W cm-2) for skin, the mild hyperpyrexia of PGO induced both cancer cell irreversible death for PTT and greatly promoted drug release for enhanced Chemo. On the other hand, the upconverted 660 nm light from UC activated Ce6 to generate reactive oxygen species for PDT, while the upconverted 540 nm light from UC could be employed for visualizing the treatment process. The in vitro and in vivo anticancer experiments demonstrate that the ultralow-intensity NIR light synchronously activated Chemo/PTT/PDT nanoplatform exhibits remarkable therapeutic efficacy with minimal photodamage.

Abstract:Tumor microenvironment (TME)-activatable phototheranostics is highly desirable in cancer management but still remains challenging for clinical applications owing to the lack of multifunctional theranostic agents and the limited tissue penetration depth. Reported here is an "all-in-one" phototheranostic platform based on near-infrared II (NIR-II) dual-plasmonic Au@Cu2- x Se core-shell nanocrystals (dpGCS NCs) for combined photoacoustic (PA)/photothermal (PT) imaging-guided chemodynamic therapy (CDT)/photocatalytic therapy (PCT)/photothermal therapy (PTT) all triggered by a single NIR-II laser. The dpGCS NCs feature excellent NIR-II plasmonic and PT properties, which guarantee their capabilities of NIR-II PA and PT imaging for real-time visual observation of tumor size and location during cancer treatment. Additionally, the TME-activated in situ •OH production via dpGCS NC-catalyzed Fenton-like reaction is further enhanced by the NIR-II irradiation, while photoexcited plasmonic hole-induced formation of extra •OH is also evidenced for PCT. Both in vitro and in vivo experiments confirm remarkable therapeutic efficacy of the present phototheranostic platform under NIR-II laser through the CDT/PCT/PTT trimodal combination therapy, achieving complete inhibition of tumor growth in tumor-bearing mice after administration of dpGCS NCs plus a single NIR-II laser irradiation. This work provides a distinctive paradigm for the development of NIR-II phototheranostic platforms for imaging-guided cancer therapy using a single laser.

Abstract:The phototheranostics in the second near-infrared window (NIR-II) have proven to be promising for the precise cancer theranostics. However, the non-responsive and "always on" imaging mode lacks the selectivity, leading to the poor diagnosis specificity. Herein, a tumor microenvironment (TME) activated NIR-II phototheranostic nanoplatform (Ag2 S-Fe(III)-DBZ Pdots, AFD NPs) is designed based on the principle of Förster resonance energy transfer (FRET). The AFD NPs are fabricated through self-assembly of Ag2 S QDs (NIR-II fluorescence probe) and ultra-small semiconductor polymer dots (DBZ Pdots, NIR-II fluorescence quencher) utilizing Fe(III) as coordination nodes. In normal tissues, the AFD NPs maintain in "off" state, due to the FRET between Ag2 S QDs and DBZ Pdots. However, the NIR-II fluorescence signal of AFD NPs can be rapidly "turn on" by the overexpressed GSH in tumor tissues, achieving a superior tumor-to-normal tissue (T/NT) signal ratio. Moreover, the released Pdots and reduced Fe(II) ions provide NIR-II photothermal therapy (PTT) and chemodynamic therapy (CDT), respectively. The GSH depletion and NIR-II PTT effect further aggravate CDT mediated oxidative damage toward tumors, achieving the synergistic anti-tumor therapeutic effect. The work provides a promising strategy for the development of TME activated NIR-II phototheranostic nanoprobes.

Abstract:Although research on photodynamic therapy (PDT) of malignant tumor has made considerable progress in recent years, it is a remaining challenge to extend PDT to the second near-infrared window (NIR-II) along with real-time and accurate NIR-II fluorescence imaging to determine drug enrichment status and achieve high treatment efficacy. In this work, lanthanide nanoparticles (Ln NPs)-based nanoplatform (LCR) equipped with photosensitizer Chlorin e6 (Ce6) and targeting molecular NH2-PEG1000-cRGDfK are developed, which can achieve NIR-II photodynamic therapy (PDT) and NIR-II fluorescence imaging by dual channel excitation. Under 808 nm excitation, Nd3+ in the outer layer can absorb the energy and transfer inward to emit strong NIR-II emissions (1064 and 1525 nm). Due to the low background noise of NIR-II light and the targeting effect of NH2-PEG1000-cRGDfK, LCR can recognize tiny tumor tissue (≈3 mm) and monitor drug distribution in vivo. Under 1530 nm excitation, internal Er3+ can be self-sensitized, generating intense upconversion emission (662 nm) that can effectively activate Ce6 for in vivo PDT due to the deep tissue penetration of NIR-II light. This study provides a paradigm of theranostic nanoplatform for both real-time fluorescence imaging and PDT of orthotopic breast tumor in NIR-II window.

Abstract:Despite the fact that photodynamic therapy (PDT) has promising clinical prospects, its therapeutic efficacy is still restricted by the hypoxic tumor microenvironment (TME). Therefore, how to suitably employ tumor hypoxia is vital for optimizing PDT. Herein we developed a hypoxia-activated prodrug for combination therapy with PDT. A hypoxia-responsive camptothecin (CPT)-based prodrug and a near-infrared (NIR) BODIPY photosensitizer were coloaded within a β-cyclodextrin-based polymer through supramolecular host–guest recognition to obtain nanoparticles named CPT-BODIPY-PNs. After being internalized by the tumor, the oxygen in the tumor tissues was rapidly consumed by the photosensitizer moiety to generate reactive oxygen species under NIR irradiation. The prodrug was efficiently activated under this intensified hypoxic condition to release the chemotherapeutic CPT drug. This system makes the best use of hypoxia TME to achieve effectively combined PDT and chemotherapy, offering an approach for cancer treatment.

Abstract:The strategy of synergetic chemodynamic treatment (CDT) and photothermal therapy (PTT) to improve the anticancer effect has been a hot spot in the field of biomedicine. Herein, a high-performance near-infrared II (NIR-II) fluorescence imaging-guided molecular phototheranostic platform () was designed via a convenient strategy. can serve as a PTT agent to kill cancer cells under near-infrared laser (808 nm) irradiation and a CDT agent to convert the endogenously less reactive H 2 O 2 into the harmful •OH, simultaneously deplete the glutathione (GSH) to achieve an amplified CDT as well. Notably, PTT-induced hyperthermia can further enhance the CDT effect, achieving a synergistic PTT/CDT therapy. Due to the enhanced permeability and retention (EPR) effect and the high GSH microenvironment of the tumor, exhibited much higher tumor accumulation revealed by NIR-II fluorescence imaging. More importantly, efficiently damages tumorous tissues without inducing side effects to normal tissues under 808 nm laser irradiation. This work has proposed an alternative PTT-CDT combined therapy platform based on a rational design of disintegrable NIR-II fluorescence imaging-guided molecular for effective tumor treatment.

Abstract:Phototheranostic with highly integrated functions is an attractive platform for cancer management. It re-mains challenging to develop a facile phototheranostic platform with complementary bimodal imaging and combinational therapy capacity. Herein, the small-molecule cyanine IR780 loaded liposomes have been harnessed as a nanoplatform to simultaneously realize photoacoustic (PA)/the second near-infrared window (NIR-II) fluorescence imaging and image-guided surgery/adjuvant photothermal therapy (PTT). This nanoplatform exhibits attractive properties like uniform controllable size, stable dispersibility, NIR-II fluorescence emission, photothermal conversion, and biocompatibility. Benefiting from the complemen-tary PA/NIR-II fluorescence bimodal imaging, this nanoplatform was successfully applied in precise vas-culature delineation and tumor diagnosis. Interestingly, the tumor was clearly detected by NIR-II fluores-cence imaging with the highest tumor-to-normal-tissue ratio up to 12.69, while signal interference from the liver was significantly reduced, due to the difference in the elimination rate of the nanoplatform in the liver and tumor. Under the precise guidance of the image, the tumor was accurately resected, and the simulated residual lesion after surgery was completely ablated by adjuvant PTT. This combined ther-apy showed improved antitumor efficacy over the individual surgery or PTT. This work develops a facile phototheranostic nanoplatform with great significance in accurately diagnosing and effectively treating tumors using simple NIR light irradiation. Abstract:Despite tremendous advances in gas therapy,there are major concerns about the inevitable concentration of toxicity and the ability to perform real-time tracking of drug delivery.Second near-infrared(NIR-Ⅱ)window absorbing nanoplatforms hold great promise for precision medicine because of their excellent tissue penetration of light and non-invasive nature.In this study,we engineered an NIR-Ⅱ laser-activated theranostic agent(named CP-bF@PEG)that was composed of amphiphilic polymers(Pluronic F127,with polyethylene glycol,PEG,moieties)coated with an NIR-Ⅱ-absorbing conjugated polymer(PTTBBT,CP)and nitric oxide(NO)donor(benzofuroxan,bF),which served as an NIR-Ⅱ photothermal inducer and NO nanogenerator.Under deep tissue penetration of NIR-Ⅱ laser irradiation,CP-bF@PEG was found to possess fluorescence imaging ability to accurately identify tumor and excellent photothermal effect.Moreover,CP-bF@PEG could generate NO via glutathione activation in the tumor microenvironment in a controllable manner.This NIR-Ⅱ-absorbing polymer for high-contrast NIR-Ⅱ fluorescence imaging-guided precision photothermal therapy achieved synergistic effects with NO therapy,as evidenced by pronounced tumor therapeutic efficacy and few side effects.This nanotheranostic agent is a highly promising candidate for high-contrast NIR-Ⅱ imaging-guided precision photothermal therapy combined with gas therapy against cancer.
Endogenous oxygen generating multifunctional theranostic nanoplatform for enhanced photodynamic-photothermal therapy and multimodal imaging

Abstract:Phototherapy, including photothermal therapy (PTT) and photodynamic therapy (PDT), has been considered as a noninvasive option for cancer therapy. However, insufficient penetration depth, tumor hypoxia, and a single treatment method severely limit the effectiveness of treatment. Methods: In this study, a multifunctional theranostic nanoplatform has been fabricated based on Au/Ag-MnO2 hollow nanospheres (AAM HNSs). The Au/Ag alloy HNSs were first synthesized by galvanic replacement reaction and then the MnO2 nanoparticles were deposited on the Au/Ag alloy HNSs by the reaction between Ag and permanganate (KMnO4), finally obtained the AAM HNSs. Then, SH-PEG was modified on the surface of AAM HNSs by the interaction of sulfhydryl and Au/Ag alloy, which improved the dispersibility and biocompatibility of the HNS. Next, the PDT photosensitizer Ce6 was loaded into AAM HNSs, benefiting from the hollow interior of the structure, and the AAM-Ce6 HNSs were obtained. Results: The AAM HNSs exhibit broad absorption at the near infrared (NIR) biological window and remarkable photothermal conversion ability in the NIR-II window. The MnO2 nanoparticles can catalyze endogenous H2O2 to generate O2 and enhance the therapeutic effect of PDT on tumor tissue. Simultaneously, MnO2 nanoparticles intelligently respond to the tumor microenvironment and degrade to release massive Mn2+ ions, which introduce magnetic resonance imaging (MRI) properties. When AAM-Ce6 HNSs are loaded with Ce6, the AAM-Ce6 HNSs can be used for triple-modal imaging (fluorescence/photoacoustic/magnetic resonance imaging, FL/PAI/MRI) guided combination tumor phototherapy (PTT/PDT). Conclusion: This multifunctional nanoplatform shows synergistic therapeutic efficacy better than any single therapy by achieving multimodal imaging guided cancer combination phototherapy, which are promising for the diagnosis and treatment of cancer.

Abstract:Cancer phototheranostics in the second near-infrared window (NIR-II, 1000−1700 nm) has low tissue scattering and high permissive laser exposure. However, most of the few available theranostic agents in this spectral region suffer from thermal instability and poor programmability of absorption profiles. Here, we proposed a programmable method for the plasmonic modulation of gold nanotheranostics in the NIR-II window. By adjusting the sequences and concentrations of DNA templates, the localized surface plasmon resonance (LSPR) of gold theranostics successfully redshifts to the NIR-II window. The as-prepared gold nanodumbbells (AuNDs) possess good thermal stability, strong photoacoustic (PA) signals as well as high photothermal conversion efficiency (84.9 %). After conjugation with nucleolin-targeted DNA aptamer AS1411, the obtained targeted gold nanotheranostics (denoted as Ap-AuND) could perform effective NIR-II PA imaging-guided PTT for subcutaneous 4T1 tumors. Notably, the Ap-AuND-mediated NIR-II PTT significantly suppressed tumor growth and stimulated potent host antitumor immunity, resulting in significant inhibition of both distant tumor growth and whole-body spreading of tumor cells when combined with an anti-PDL1 antibody.

Multifunctional Nanoplatform for Single NIR Laser-Regulated Efficient PDT/PTT/Chemotherapy

Abstract:The combination of phototherapy and chemotherapy with superior advantages is a promising strategy for cancer therapy. However, combination therapy is generally regulated by two different wavelengths of light or other stimuli, which results in complex operations and inevitable systemic side effects, even affecting therapeutic efficacy. Herein, we design a signal NIR light-regulated nanoplatform via the self-assembly process of reactive oxygen species (ROS)-sensitive prodrug (DTD), human serum albumin (HSA), and IR780 for combined photothermal/photodynamic therapy and chemotherapy. Upon 808 nm laser irradiation, IR780 in nanoparticles generates abundant ROS and a significant photothermal effect to achieve photothermal/photodynamic therapy. Meanwhile, the generating ROS further cleans up the thioketal link to release DOX for chemotherapy. Hence, signal NIR light can effectively control the process of combination therapy. In vivo and in vitro experiment results demonstrate that the multifunctional nanoparticles exhibit excellent antitumor efficacy via the combination of phototherapy and chemotherapy controlled by a signal NIR laser. Overall, the signal NIR light-regulated nanoparticles with combination therapy performance provide a versatile platform for enhancing antitumor efficacy.

Abstract:Mild phototherapy with heat shock protein 90 (HSP90) inhibitors can be employed for tumor therapy, but suffers from non-exhaustive therapeutic effects and uncontrollable release. Herein, we present NIR-responsive and dual sensitized nanoparticles for mild tumor phototherapy. These nanoparticles were based on two perylene monoimide and a diamino anthraquinone (2PMI-AQ) chromophore as an inner hydrophobic core. To explore the sensitized function, 2PMI-AQ was modified with hydrophilic thermosensitive polymers, resulting in stable nanoparticles (NP1) via spontaneous self-assembly. The as-prepared NP1 possess a robust nanostructure and enable combined photothermal therapy (PTT) and photodynamic therapy (PDT). Besides, outside thermosensitive polymers of NP1 allow loading and photothermal release of HSP90 inhibitors. Under NIR laser irradiation, photothermal/photodynamic effects and photothermally controlled release of HSP90 inhibitors are realized simultaneously. The inhibitors inactivate the cell guardian protein HSP90 to attenuate the cytoprotection, which enhances both PTT and PDT. We thus achieve mild tumor phototherapy in vivo by using NIR-absorbing nanoparticles with dual sensitized function induced by low-power irradiation.

Abstract:A near‐infrared (NIR)‐triggered nanoplatform with controlled release of an heat shock proteins (HSPs) inhibitor for synergistic mild photothermal and enhanced Fenton nanocatalysis is developed. Upon NIR irradiation, gambogic acid is released and inhibits the expression of HSP90. Notably, the nanoplatform simultaneously relieves the tumor hypoxia and enhances the efficacy of the nanocatalytic therapy in an acid tumor microenvironment. Mild photothermal therapy (PTT, 50 °C). However, its therapeutic efficacy is limited owing to the hypoxic tumor environment and tumor thermoresistance owing to the overproduction of heat shock proteins (HSPs). Herein, a near‐infrared (NIR)‐triggered theranostic nanoplatform (GA‐PB@MONs@LA) is designed for synergistic mild PTT and enhanced Fenton nanocatalytic therapy against hypoxic tumors. The nanoplatform is fabricated by the confined formation of Prussian blue (PB) nanoparticles in mesoporous organosilica nanoparticles (MONs), followed by the loading of gambogic acid (GA), an HSP90 inhibitor, and coating with thermo‐sensitive lauric acid (LA). Upon NIR irradiation, the photothermal effect (44 °C) of PB not only induces apoptosis of tumor cells but also triggers the on‐demand release of GA, inhibiting the production of HSP90. Moreover, the delivered heat simultaneously enhances the catalase‐like and Fenton activity of PB@MONs@LA in an acidic tumor microenvironment, relieving the tumor hypoxia and promoting the generation of highly toxic •OH. In addition, the nanoplatform enables magnetic resonance/photoacoustic dual‐modal imaging. Thus, this study describes a distinctive paradigm for the development of NIR‐triggered theranostic nanoplatforms for enhanced cancer therapy.

Tumor Microenvironment Stimuli‐Responsive Single‐NIR‐Laser Activated Synergistic Phototherapy for Hypoxic Cancer by Perylene Functionalized Dual‐Targeted Upconversion Nanoparticles

Abstract:An intelligent dual‐targeted nanosystem is designed based on upconversion nanoparticles coated with the TA/Fe3+ coordination film through a one‐step assembly method. This not only can realize acidic tumor microenvironment responsive photosensitizer (perylene probe (PC4)) and Fe3+ release, but also simultaneously catalyzes the overexpressed H2O2 to generate O2 for highly efficient single near‐infrared laser‐triggered four‐modal imaging‐guided photothermal therapy/photodynamic therapy synergistic therapy against hypoxic cancer. Although synergistic therapy has shown great promise for effective treatment of cancer, the unsatisfactory therapeutic efficacy of photothermal therapy/photodynamic therapy is resulted from the absorption wavelength mismatch, tumor hypoxia, photosensitizer leakage, and inability in intelligent on‐demand activation. Herein, based on the characteristics of tumor microenvironment (TME), such as the slight acidity, hypoxia, and overexpression of H2O2, a TME stimuli‐responsive and dual‐targeted composite nanoplatform (UCTTD‐PC4) is strategically explored by coating a tannic acid (TA)/Fe3+ nanofilm with good biocompatibility onto the upconversion nanoparticles in an ultrafast, green and simple way. The pH‐responsive feature of UCTTD‐PC4 remains stable during the blood circulation, while rapidly releases Fe3+ in the slightly acidic tumor cells, which results in catalyzing H2O2 to produce O2 and overcoming the tumor hypoxia. Notably, the emission spectrum of the UCTTD perfectly matches the absorption spectrum of the photosensitizer (perylene probe (PC4)) to achieve the enhanced therapeutic effect triggered by a single laser. This study provides a new strategy for the rational design and development of the safe and efficient single near‐infrared laser‐triggered synergistic treatment platform for hypoxic cancer under the guidance of multimodal imaging.

Abstract:Multifunctional nanoplatforms for imaging‐guided synergistic antitumor treatment are highly desirable in biomedical applications. However, anticancer treatment is largely affected by the pre‐existing hypoxic tumor microenvironment (TME), which not only causes the resistance of the tumors to photodynamic therapy (PDT), but also promotes tumorigenesis and tumor progression. Here, a continuous O2 self‐enriched nanoplatform is constructed for multimodal imaging‐guided synergistic phototherapy based on octahedral gold nanoshells (GNSs), which are constructed by a more facile and straightforward one‐step method using platinum (Pt) nanozyme‐decorated metal–organic frameworks (MOF) as the inner template. The Pt‐decorated MOF@GNSs (PtMGs) are further functionalized with human serum albumin‐chelated gadolinium (HSA‐Gd, HGd) and loaded with indocyanine green (ICG) (ICG‐PtMGs@HGd) to achieve a synergistic PDT/PTT effect and fluorescence (FL)/multispectral optoacoustic tomography (MSOT)/X‐ray computed tomography (CT)/magnetic resonance (MR) imaging. The Pt‐decorated nanoplatform endows remarkable catalase‐like behavior and facilitates the continuous decomposition of the endogenous H2O2 into O2 to enhance the PDT effect under hypoxic TME. HSA modification enhances the biocompatibility and tumor‐targeting ability of the nanocomposites. This TME‐responsive and O2 self‐supplement nanoparticle holds great potential as a multifunctional theranostic nanoplatform for the multimodal imaging‐guided synergistic phototherapy of solid tumors.

Abstract:Traditional combination therapy of photodynamic therapy (PDT) and photothermal therapy (PTT) is limited in the field of clinical cancer therapy due to activation by light with separate wavelengths, insufficient O2 supply, antioxidant ability of glutathione (GSH) in tumor cell, and low penetration depth of light. Here, a multifunctional nanoplatform composed of MoO3-x nanosheets, Ag nanocubes, and MnO2 nanoparticles was developed to overcome these drawbacks. For this nanoplatform, hyperthermia and reactive oxygen species (ROS) were simultaneously generated under single 808 nm near-infrared (NIR) light irradiation. Once this nanoplatform accumulated in the tumor region, GSH was depleted by MnO2 and intracellular H2O2 was catalyzed by MnO2 to produce O2 to relieve hypoxia. Ultrasound (US) imaging confirmed in-situ O2 generation. Magnetic resonance (MR) imaging, photoacoustic (PA) imaging, and fluorescence imaging were used to monitor in vivo biodistribution of nanomaterials. This provides a paradigm to rationally design a single NIR laser induced multimodal imaging-guided efficient PDT/PTT cancer strategy.

Activatable NIR‐II Plasmonic Nanotheranostics for Efficient Photoacoustic Imaging and Photothermal Cancer Therapy

Abstract:Precise manipulation of optical properties through the structure‐evolution of plasmonic nanoparticles is of great interest in biomedical fields including bioimaging and phototherapy. However, previous success has been limited to fixed assembled structures or visible–NIR‐I absorption. Here, an activatable NIR‐II plasmonic theranostics system based on silica‐encapsulated self‐assembled gold nanochains (AuNCs@SiO2) for accurate tumor diagnosis and effective treatment is reported. This transformable chain structure breaks through the traditional molecular imaging window, whose absorption can be redshifted from the visible to the NIR‐II region owing to the fusion between adjacent gold nanoparticles in the restricted local space of AuNCs@SiO2 triggered by the high H2O2 level in the tumor microenvironment (TME), leading to the generation of a new string‐like structure with strong NIR‐II absorption, which is further confirmed by finite‐difference‐time‐domain (FDTD) simulation. With the TME‐activated characteristics, AuNCs@SiO2 exhibits excellent properties for photoacoustic imaging and a high photothermal conversion efficiency of 82.2% at 1064 nm leading to severe cell death and remarkable tumor growth inhibition in vivo. These prominent intelligent TME‐responsive features of AuNCs@SiO2 may open up a new avenue to explore optical regulated nano‐platform for intelligent, accurate, and noninvasive theranostics in NIR‐II window.

Abstract:Second near-infrared (NIR-II) phototheranostics, operating in the second near-infrared region (>1000 nm), offer significant advantages for tumor diagnosis and therapy, making them highly promising in biomedical research. In this study, we successfully synthesized a heptamethine cyanine molecule fluorophore (IRCS-1060), which exhibits excitation and emission spectra in the NIR-II region. The BIT NPs (BSA@IRCS-1060@TPZ Nanoparticles) were fabricated by integrating the heptamethine cyanine molecule fluorophore (IRCS-1060) with bovine serum albumin (BSA) and the hypoxic-activated prodrug tirapazamine (TPZ). Under the irradiation of NIR laser, the BIT NPs can provide NIR-II optical imaging (fluorescence imaging (FI) and photoacoustic imaging (PAI)) guided photothermal/photodynamic (PTT/PDT) therapy. Moreover, PDT exacerbated the hypoxic environment of the tumor microenvironment, subsequently activating the TPZ and initiating a cascade chemotherapy process. The combination of NIR-II PTT/PDT and chemotherapy could effectively suppress the growth of tumors with negligible side effects. This work presents a highly promising 1064 nm laser excitation phototheranostic agent for spatiotemporally specific diagnosis and combination therapy of tumors.
High‐Performance NIR‐II Fluorescent Type I/II Photosensitizer Enabling Augmented Mild Photothermal Therapy of Tumors by Disrupting Heat Shock Proteins

Abstract:NIR‐II fluorescent photosensitizers as phototheranostic agents hold considerable promise in the application of mild photothermal therapy (MPTT) for tumors, as the reactive oxygen species generated during photodynamic therapy can effectively disrupt heat shock proteins. Nevertheless, the exclusive utilization of these photosensitizers to significantly augment the MPTT efficacy has rarely been substantiated, primarily due to their insufficient photodynamic performance. Herein, we present the utilization of high‐performance NIR‐II fluorescent type I/II photosensitizer (AS21:4) as a simple but effective nanoplatform derived from molecule AS2 to enhance the MPTT efficacy of tumors without any additional therapeutic components. By taking advantage of heavy atom effect, AS21:4 as a type I/II photosensitizer demonstrates superior efficacy in producing 1O2 (ΦΔ = 12.4%) and O2•− among currently available NIR‐II fluorescent photosensitizers with absorption exceeding 800 nm. In vitro and in vivo findings demonstrate that the 1O2 and O2•− generated from AS21:4 induce a substantial reduction in the expression of HSP90, thereby improving the MPTT efficacy. The remarkable phototheranostic performance, substantial tumor accumulation, and prolonged tumor retention of AS21:4, establish it as a simple but superior phototheranostic agent for NIR‐II fluorescence imaging‐guided MPTT of tumors.
