Abstract:Bacterial infections have become major threats to public health all over the world. With the emergence of antibiotic resistance, it is urgent to develop novel antimicrobial materials to efficiently overcome drug resistance with high bactericidal activity. In this work, UiO-66-NH-CO-MoS2 nanocomposites (UNMS NCs) are constructed through the amidation reaction. The UNMS NCs are positively charged which is beneficial for capturing and restricting bacteria. Significantly, UNMS NCs possess a synergistic bactericidal efficiency based on near-infrared irradiation (808 nm) regulated combination of photothermal, photodynamic, and peroxidase-like enzymatic activities. Both the photodynamic property and nanozymatic activity of UNMS NCs can lead to the generation of reactive oxygen species. The UNMS NCs show high catalytic activity in a wide pH range and exhibit excellent antibacterial ability against ampicillin-resistant Escherichia coli and methicillin-resistant Staphylococcus aureus with negligible cytotoxicity. Interestingly, due to the 808 nm irradiation-induced hyperthermia in the presence of UNMS NCs, the glutathione oxidation process can be accelerated, resulting in bacterial death more easily. Mice wound models are established to further manifest that UNMS NCs can promote wound healing with good biosafety in living systems.

Abstract:Infections of antibiotic-resistant pathogens have caused a series of public health crises across the world. According to the latest published reports, an antibiotic switch has been recognized as a potential strategy to control antibacterial activity for combating this serious drug resistance. Thus, it is anticipated that more effective antibiotic switches should be obtained by further exploring the developed strategies. Here, we report an improved pretreatment strategy using a surfactant (Triton X-100) for constructing an effective supramolecular antibiotic switch based on a poly(fluorene-cophenylene) derivative (PFP) and cucurbit[7]uril (CB[7]), which can regulate the aggregation state of polymers before the supramolecular self-assembly process occurs. Triton X-100 can regulate the aggregation states of conjugated polymers, which is used to successfully realize the reversible control of bactericidal activity of PFP in the dark and under white light irradiation by supramolecular assembly/disassembly between PFP and CB[7]. Specialized antibiotic switches are significantly important to fight pathogenic infections and solve the drug resistance crisis in the future.

Abstract:Macromolecular antimicrobial agents such as cationic polymers and peptides have recently been under an increased level of scrutiny because they can combat multi-drug-resistant microbes. Most of these polymers are non-biodegradable and are designed to mimic the facially amphiphilic structure of peptides so that they may form a secondary structure on interaction with negatively charged microbial membranes. The resulting secondary structure can insert into and disintegrate the cell membrane after recruiting additional polymer molecules. Here, we report the first biodegradable and in vivo applicable antimicrobial polymer nanoparticles synthesized by metal-free organocatalytic ring-opening polymerization of functional cyclic carbonate. We demonstrate that the nanoparticles disrupt microbial walls/membranes selectively and efficiently, thus inhibiting the growth of Gram-positive bacteria, methicillin-resistant Staphylococcus aureus (MRSA) and fungi, without inducing significant haemolysis over a wide range of concentrations. These biodegradable nanoparticles, which can be synthesized in large quantities and at low cost, are promising as antimicrobial drugs, and can be used to treat various infectious diseases such as MRSA-associated infections, which are often linked with high mortality.

Abstract:A single layer zwitterionic polymer brush grafted surface with pH-responsive charge reversal capacity was prepared in a simple and widely used strategy. The amide bond of the zwitterionic polymer will break under acidic condition and undergo a charge reversal, resulting the surface to convert from an antifouling zwitterionic surface to a bactericidal cationic surface. The surface we constructed can successfully resist the attachment of initial bacteria at physiological conditions and kill bacteria on demand to defend bacterial infection, showing an attractive application in implantable medical devices.

Abstract:Infections caused by drug-resistant pathogens have posed a series of public health crises across the world. To address this challenging issue, development of new antibiotics is one of the most effective methods, however it always lags behind the spreading of antibiotic resistance unfortunately. In this work we illustrate an innovative supramolecular strategy to design germicide switch for decelerating the emergence of drug-resistant bacteria. The germicide switches are built up between commercial germicides and cucurbit[7]uril (CB[7]) through host-guest interactions, and their bactericidal activity is switched off and on reversibly by assembly and dis-assembly processes of germicide/CB[7] complex. Structure-property relation exhibits that the interaction between active sites of germicides and CB[7] plays a decisive role in the antimicrobial activity of these germicide switches. By turning antibacterial activity on when needed and turning it off when not needed, the germicide switch will not continue to produce pressure on bacteria, thus decelerating the emergence of drug-resistant bacteria. This work offers a proof-of-concept approach to efficiently prevent the emergence of drug-resistant bacteria by controllable and reversible release and concealment of active sites of germicides by supramolecular strategy.
An Enzymatic Antibiotic Adjuvant Modulates the Infectious Microenvironment to Overcome Antimicrobial Resistance of Pathogens

Abstract:The emergence and evolution of antimicrobial resistance (AMR) pose a significant challenge to the current arsenal to fight infection. Antibiotic adjuvants represent an appealing tactic for tackling the AMR of pathogens, however, their practical applications are greatly constrained by the harsh infectious microenvironment. Herein, it is found that silver nanoclusters (Ag NCs) can possess tunable enzymatic activities to modulate infectious microenvironments. Based on this finding, an enzymatic nanoadjuvant (EnzNA) self-assembled from Ag NCs, which is inert under neutral physiological conditions but can readily disassemble into isolated Ag NCs exhibiting biofilm destructive oxidase-mimetic activity in the acidic biofilm microenvironment, is developed. Once internalized into the neutral cytoplasm of bacteria, Ag NCs switch to reveal the thiol oxidase-mimetic activity to suppress ribosomal biogenesis for AMR reversal and evolution inhibition of pathogens. Consequently, EnzNAs revitalize various existing antibiotics against methicillin-resistant Staphylococcus aureus, and potentiate the antibiotic efficacy against biofilm-mediated skin infection and lethal lung infection in mice. These findings highlight the capability of enzyme-mimetic nanomaterials to modulate the infectious microenvironment and potentiate antibiotics, providing a paradigm shift for anti-infection therapy.

Abstract:Antibiotic resistance is a significant global health concern that requires novel approaches to tackle the serious crisis. In this study, we designed an intelligent supramolecular antibacterial agent using Dequalinium chloride (DQ), cucurbit[7]urils (CB[7]), and maleic anhydride-cyclohexylamine hydrochloride (ADA-MA) based on supramolecular chemistry. The antibacterial activity of DQ was regulated by CB[7] and ADA-MA, which responded to the pH value of the infection microenvironment. The acidic infection microenvironment triggered the release of DQ from CB[7] and restored its antibacterial activity, while the neutral physiological environment maintained the stability of DQ@2CB[7] and turned off its antibacterial activity. This process provided a simple and effective strategy for regulating the biological activity of DQ on demand. It could "turn on" and "turn off" its antibacterial activity according to the needs of the infection microenvironment, thus delaying the generation of resistant bacteria and reducing the toxicity of the drug under physiological conditions.

Abstract:Achieving versatile dispersion of nanoparticles in a broad range of solvents (e.g., water, oil, and biofluids) without repeatedly recourse to chemical modifications are desirable in optoelectronic devices, self-assembly, sensing, and biomedical fields. However, such a target is limited by the strategies used to decorate nanoparticle’s surface properties, leading to a narrow range of solvents for existing nanoparticles. Here we report a concept to break the nanoparticle’s dispersible limit via electrochemically anchoring surface ligands capable of sensing the surrounding liquid medium and rotating to adapt to it, immediately forming stable dispersions in a wide range of solvents (polar and nonpolar, biofluids, etc.). Moreover, the smart nanoparticles can be continuously electrodeposited in the electrolyte, overcoming the electrode surface-confined low throughput limitation of conventional electrodeposition methods. The anomalous dispersive property of the smart Ag nanoparticles enables them to resist bacteria secreted species-induced aggregation and the structural similarity of the surface ligands to that of the bacterial membrane assists them to enter the bacteria, leading to high antibacterial activity. The simple but massive fabrication process and the enhanced dispersion properties offer great application opportunities to the smart nanoparticles in diverse fields.

Engineering the novel azobenzene-based molecular photoswitches for suppressing bacterial infection through dynamic regulation of biofilm formation

Abstract:Background: Bacterial biofilm is a strong fortress for bacteria to resist harsh external environments, which can enhance their tolerance and exacerbate the drug/pesticide resistance risk. Currently, photopharmacology provides an advanced approach via precise spatiotemporal control for regulating biological activities by light-controlling the molecular configurations, thereby having enormous potential in the development of drug/pesticides. Results: To further expand the photopharmacology application for discovering new antibiofilm agents, we prepared a series of light-controlled azo-active molecules and explored their photo isomerization, fatigue resistance, and anti-biofilm performance. Furthermore, their mechanisms of inhibiting biofilm formation were systematically investigated. Overall, designed azo-derivative A11 featured excellent anti-Xoo activity with an half-maximal effective concentration (EC50) value of 5.45 μg mL-1, and the EC50 value could be further elevated to 2.19 μg mL-1 after ultraviolet irradiation (converted as cis-configuration). The photo-switching behavior showed that A11 had outstanding anti-fatigue properties. An in-depth analysis of the action mechanism showed that A11 could effectively inhibit biofilm formation and the expression of relevant virulence factors. This performance could be dynamically regulated via loading with private light-switch property. Conclusion: In this work, designed light-controlled azo molecules provide a new model for resisting bacterial infection via dynamic regulation of bacterial biofilm formation. © 2024 Society of Chemical Industry.
Surface Charge Switchable Antibiotics Based Small Molecular Micelles Combined with Photodynamic Therapy for Antibacterial and Antibiofilm Applications

Abstract:The appearance of multidrug-resistant bacteria and the formation of bacterial biofilms have necessitated the development of alternative antimicrobial therapeutics. Antibiotics embedded in or conjugated with inert nanocarriers (nano-antibiotics) show many therapeutic advantages over free drugs, but the inert carrier materials are the major component (generally more than 90%) in nano-antibiotics, causing low drug loading contents and limited therapeutic output. Herein, we fabricate nanocarriers by directly using antibiotics as building blocks to minimize use of inert materials, substantially increase the drug loading content, and suppress premature burst release. Firstly, we conjugated betaine carboxylate with ciprofloxacin (CIP) through ester bond to form the amphiphilic conjugates (CIP-CB), which was self-assembled into micelles (CIP-CBMs) in aqueous solutions with CIP loading contents as high as 65.4 wt.% and the micelles showed pH induced surface charge reversal properties. Secondly, a model photosensitizer (PS), i.e., 5, 10, 15, 20-tetraphenylporphyrin (TPP), was encapsulated in CIP-CBMs, resulting infectious targeted photodynamic (PDT)/antibiotics combined nanomedicines (denoted as TPP@CIP-CBMs). After accumulation at infectious sites or at deep bacterial biofilms, the ester bond between betaine carboxylate and CIP undergoes hydrolysis to release free TPP and CIP, resulting in an excellent antibacterial and antibiofilm activity in vitro and in vivo.
Facile Assembly of Multifunctional Antibacterial Nanoplatform Leveraging Synergistic Sensitization between Silver Nanostructure and Vancomycin

Abstract:The emergence of antibiotic-resistant bacterial strains renders the conventional antibiotic therapy less efficient. The integration of two distinct bactericides into one compact platform provides a promising strategy to realize a combinational antimicrobial therapy. In this work, an efficient chemo-Ag nanohybrid antibacterial platform was facilely developed based on the integration of vancomycin-carrying polydopamine with silver nanoparticles (). The as-synthesized antibacterial nanohybrid inherited the intrinsic properties of both bactericides to achieve a synergistic antibacterial performance against both Staphylococcus aureus and Escherichia coli strains by attacking bacteria from two distinct fronts. Through this combinational therapy, the efficiency of antibiotic against S. aureus was significantly improved by reducing drug dosage with less opportunity for imparting drug resistance. In addition, this antibacterial nanohybrid, with innate photothermal properties, could achieve auxiliary hyperthermia-assisted bacterial inactivation in the meantime. Furthermore, the outstanding in vivo bacteria-killing activity and wound-healing acceleration were demonstrated in a S. aureus-infected mouse skin defect model. Taken together, this bactericidal nanohybrid could achieve sustained antibiotic release and wipe out bacteria more effectively in a synergistic way, thus reducing the emergence of antibiotic resistance. This work holds great potential to advance the development of novel antibacterial agents and combinational strategies as a promising supplement of antibiotics in the near future.

Dynamic covalent nano-networks comprising antibiotics and polyphenols orchestrate bacterial drug resistance reversal and inflammation alleviation

Abstract:New antimicrobial strategies are urgently needed to meet the challenges posed by the emergence of drug-resistant bacteria and bacterial biofilms. This work reports the facile synthesis of antimicrobial dynamic covalent nano-networks (aDCNs) composing antibiotics bearing multiple primary amines, polyphenols, and a cross-linker acylphenylboronic acid. Mechanistically, the iminoboronate bond drives the formation of aDCNs, facilitates their stability, and renders them highly responsive to stimuli, such as low pH and high H2O2 levels. Besides, the representative A1B1C1 networks, composed of polymyxin B1(A1), 2-formylphenylboronic acid (B1), and quercetin (C1), inhibit biofilm formation of drug-resistant Escherichia coli, eliminate the mature biofilms, alleviate macrophage inflammation, and minimize the side effects of free polymyxins. Excellent bacterial eradication and inflammation amelioration efficiency of A1B1C1 networks are also observed in a peritoneal infection model. The facile synthesis, excellent antimicrobial performance, and biocompatibility of these aDCNs potentiate them as a much-needed alternative in current antimicrobial pipelines.

Abstract:Drug delivery systems capable of sequential multistage drug release are urgently needed for antibacterial applications. Herein, we report a molecular switch‐integrated, photo‐responsive nanoplatform (namely HAVH) based on hollow mesoporous silica nanospheres (HMSN) loaded with Ag NPs, vancomycin (Van), and hemin for bacteria elimination and abscess therapy. Upon near‐infrared (NIR) light irradiation, the molecular switch, hemin, can effuse from the mesopores of HMSN, triggering the release of pre‐loaded Ag+ and Van, which enables photothermal‐modulated drug release and synergistic photothermal‐chemo therapy. The HAVH_NIR irreversibly disrupts the bacterial cell membrane, facilitating the penetration of Ag+ and Van. We find that these compounds restrain the transcription and translation of ribosomes and lead to rapid bacterial death. Furthermore, hemin can effectively inhibit excessive inflammatory responses associated with the treatment, promoting accelerated wound healing in a murine abscess model. This work presents a new strategy for antibacterial drug delivery with high controllability and extendibility, which may benefit the development of smart multifunctional nanomedicine for diseases not limited to bacterial infections. Abstract:Natural enzymes are expected to be used in biomedical fields due to their extensive antibacterial activity, good biocompatibility and resistance to bacteria. Herein, we constructed a multi-enzyme protein assembly with ultrathin single protein sheet structure by the strategy of interface assembly and developed an efficient and intelligent antibacterial system based on the combination of multi-enzymes. The multi-enzyme protein assembly not only displayed extremely high antibacterial activity, but can also be turned on and off through the denaturant induced folding and unfolding of natural enzymes, and this process can be realized for several reversible cycles. Such an intelligent antibacterial material provides the possibility for its application in biomedical and other fields.

Abstract:Constructing safe and effective antibacterial surfaces has continuously received great attention, especially in healthcare-related fields. Bioinspired mechano-bactericidal nanostructure surfaces could serve as a promising strategy to reduce surface bacterial contamination while avoiding the development of antibiotic resistance. Although effective, these nanostructure surfaces are prone to be contaminated by the accumulation of dead bacteria, inevitably compromising their long-term antibacterial activity. Herein, a bioinspired nanopillar surface with both mechano-bactericidal and releasing actions is developed, via grafting zwitterionic polymer (poly(sulfobetaine methacrylate) (PSBMA)) on ZnO nanopillars. Under dry conditions, this nanopillar surface displays remarkable mechano-bactericidal activity, because the collapsed zwitterionic polymer layer makes no essential influence on nanopillar structure. Once being incubated with aqueous solution, the surface could readily detach the killed bacteria and debris, owing to the swelling of the zwitterionic layer. Consequentially, the surface antibacterial performances can be rapidly and controllably switched between mechano-bactericidal action and bacteria-releasing activity, guaranteeing a long-lasting antibacterial performance. Notably, these collaborative antibacterial behaviors are solely based on physical actions, avoiding the risk of triggering bacteria resistance. The resultant nanopillar surface also enjoys the advantages of substrate-independency and good biocompatibility, offering potential antibacterial applications for biomedical devices and hospital surfaces.

Abstract:Antibacterial materials are often bothered by the problems of drug-resistance-induction and adhesion-to-invalidation, coming from the exposure of reusable bactericidal materials before or after utilization. Herein, an efficient and easily transformable supramolecular switching surface for antifouling and bactericidal was successfully fabricated, through introducing a negatively charged macrocyclic host S6-corona[3]arene[3]pyridazine (S6-CAP) to a contact-killing surface constructed by a positively charged amphiphilic bactericide. The S6-CAP was able to fully switch off the bactericidal activity and make the surfaces antifouling. After switched on by simply washing off much of S6-CAP, the bactericidal activity was even better than the original contact-killing surfaces.

Abstract:With the emergence of drug-resistant bacteria, conventional antibiotics are becoming increasingly ineffective for the treatment of bacterial infections. Nanomaterial-modified antibiotics, denoted as "nanoantibiotics", can usually circumvent most of the shortcomings of conventional antibiotics, thus improving antibacterial activities. Here, we developed triclosan-based supramolecular hydrogel nanoantibiotics by conjugating small molecule antibiotic triclosan (TCS) to self-assembling peptides. The resultant nanoantibiotics presented many beneficial characteristics: (i) a stable three-dimensional nanofiber structure; (ii) increased TCS solubility by 850-fold; (iii) acid-responsive TCS release; (iv) favorable biocompatibility. In consequence, the nanoantibiotics showed potent in vitro broad-spectrum antibacterial activities against both Gram-positive and Gram-negative bacteria based on the cooperative effect of antibiotic TCS and the nanostructure-induced bacterial membrane disruption. Furthermore, the TCS-based supramolecular hydrogel nanoantibiotics exhibited enhanced antibacterial activities with low side effects, according to the in vivo antibacterial evaluation at the macro and micro level. Therefore, the simple and effective hydrogel nanoantibiotics developed here hold great potential for the treatment of intractable bacterial infections.

Abstract:Heterostructure nanozymes as antibiotic‐free antimicrobial agents exhibit great potential for multidrug‐resistant (MDR) bacterial strains elimination. However, realization of heterostructure antimicrobials with enhanced interfacial interaction for synergistically amplified antibacterial therapy is still a great challenge. Herein, oxygen‐vacancy‐enriched glucose modified MoOx (G‐MoOx) is exploited as a reducing agent to spontaneously reduce Ag (I) into Ag (0) that in situ grows onto the surface of G‐MoOx. The resultant Ag doped G‐MoOx (Ag/G‐MoOx) heterostructure displays augmenting photothermal effect and NIR‐enhanced oxidase‐like activity after introducing Ag nanoparticles. What's more, NIR hyperthermia accelerate Ag+ ions release from Ag nanoparticles. Introduction of Ag greatly enhances antimicrobial activities of Ag/G‐MoOx against MDR bacteria, especially the hybrid loading with 1 wt% Ag NPs exhibiting antibacterial efficacy up to 99.99% against Methicillin‐resistant Staphylococcus aureus (MRSA, 1×106 CFU mL−1).
