Abstract:Infections of antibiotic-resistant pathogens have caused a series of public health crises across the world. According to the latest published reports, an antibiotic switch has been recognized as a potential strategy to control antibacterial activity for combating this serious drug resistance. Thus, it is anticipated that more effective antibiotic switches should be obtained by further exploring the developed strategies. Here, we report an improved pretreatment strategy using a surfactant (Triton X-100) for constructing an effective supramolecular antibiotic switch based on a poly(fluorene-cophenylene) derivative (PFP) and cucurbit[7]uril (CB[7]), which can regulate the aggregation state of polymers before the supramolecular self-assembly process occurs. Triton X-100 can regulate the aggregation states of conjugated polymers, which is used to successfully realize the reversible control of bactericidal activity of PFP in the dark and under white light irradiation by supramolecular assembly/disassembly between PFP and CB[7]. Specialized antibiotic switches are significantly important to fight pathogenic infections and solve the drug resistance crisis in the future.

Abstract:Macromolecular antimicrobial agents such as cationic polymers and peptides have recently been under an increased level of scrutiny because they can combat multi-drug-resistant microbes. Most of these polymers are non-biodegradable and are designed to mimic the facially amphiphilic structure of peptides so that they may form a secondary structure on interaction with negatively charged microbial membranes. The resulting secondary structure can insert into and disintegrate the cell membrane after recruiting additional polymer molecules. Here, we report the first biodegradable and in vivo applicable antimicrobial polymer nanoparticles synthesized by metal-free organocatalytic ring-opening polymerization of functional cyclic carbonate. We demonstrate that the nanoparticles disrupt microbial walls/membranes selectively and efficiently, thus inhibiting the growth of Gram-positive bacteria, methicillin-resistant Staphylococcus aureus (MRSA) and fungi, without inducing significant haemolysis over a wide range of concentrations. These biodegradable nanoparticles, which can be synthesized in large quantities and at low cost, are promising as antimicrobial drugs, and can be used to treat various infectious diseases such as MRSA-associated infections, which are often linked with high mortality.

Abstract:The problem of bacterial resistance to antibiotics has become a major cause of concern in the modern world, due to the occurrence of multidrug-resistant bacterial strains. A single-component supramolecular antibiotic with different self-assembling morphologies that can effectively regulate antibacterial efficacy and present recyclable antimicrobial activity is proposed. This supramolecular antibiotic consists of a cationic polymer-grafted host-guest-conjugated amphiphilic molecule. With sequential ultrasonic and redox stimuli, its self-assemblies presented reversible morphology transitions from spherical micelles to branched aggregates and finally to dot-like assemblies in aqueous solution. Branched aggregates showed the strongest antibacterial ability for E. coli and S. aureus amongst the three self-assemblies, owing to the special enrichment form and high distribution density of the grafted cationic polymer chains on their surfaces. Fluorogen-activating protein imaging was innovatively employed to deeply investigate the supramolecular antibiotic mechanism. It is anticipated that the design of self-assembling, single-component supramolecular antibiotics may be a successful strategy to fight against the increasingly serious problem of drug-resistant bacteria.

Abstract:The infections and diseases caused by harmful bacteria are regarded as the worldwide health-threatening issue. The most antibiotics-based delivery systems restricted the controlled drug release at specific infection sites due to the stable chemical conjugation, thus largely compromising the treatment effectiveness. Herein, we report the construction of light controlled drug-based supramolecular polymer (LDSP), which is endowed the “switch on” function for realizing the controllable antibiotics released in specific sites. LDSP was first formed based on the host-guest interaction between β-cyclodextrin units grafted hyaluronic acid chain (HA-CD) and azobenzene moiety linked by two enoxacin units (Azo-(Eno)2), and could further self-assemble into light controlled drug-based supramolecular polymer self-assemblies (LDSPSAs). Under the UV light irradiation, the “switch on” function of LDSPSAs could be effectively turned on at the specific infected tissues due to the smartly dissociated host-guest interaction, facilitating the controlled release of the hydrophobic Azo-(Eno)2 to interact with bacteria for efficient antibacterial effect. The biological evaluation demonstrated that LDSPSAs achieved good antibacterial efficiency and well biosafety.

Abstract:Polymeric materials with antibacterial properties hold great promise for combating multidrug-resistant bacteria, which pose a significant threat to public health. However, the synthesis of most antibacterial polymers typically involves complicated and time-consuming procedures. In this study, we demonstrate a simple and efficient strategy for synthesizing functional poly(vinylpyridinium salt)s via pyridinium-yne click polymerization. This click polymerization could proceed with high atom economy under mild conditions without any external catalyst, yielding soluble and thermally stable poly(vinylpyridinium salt)s with satisfactory molecular weights and well-defined structures in excellent yields. Additionally, the incorporation of luminescent units such as fluorene, tetraphenylethylene, and triphenylamine into the polymer backbone confers excellent aggregation-enhanced emission properties upon the resulting polymers, rendering them suitable for bacterial staining. Moreover, the existence of pyridinium salt imparts intrinsic antibacterial activity against multidrug-resistant bacteria to the polymers, enabling them to effectively inhibit wound bacterial infection and significantly expedite the healing process. This work not only provides an efficient method to prepare antibacterial polymers, but also opens up the possibility of various applications of polymers in healthcare and other antibacterial fields.

Abstract:In this work, we fabricated a degradable supramolecular photodynamic polymer (SPP) with an enhanced efficiency of biofilm elimination. The small-molecule photosensitizer, a cationic porphyrin derivative, was grafted to a block polymer backbone (BPB) through a host-guest interaction and metal coordination. The locally enriched cationic photosensitizer in SPP endows a high efficiency to disrupt biofilms due to the generated reactive oxygen species (ROS) around the bacteria under white light illumination, while reducing the cytotoxicity to mammalian cells. After cucurbit[7]uril was added as a competitive agent, the photosensitizer could be disassociated from the BPB and the antibacterial ability was reduced; also the SPP could be further degraded. As a consequence, the supramolecular photodynamic polymer may become a very promising material for biofilm elimination with an enhanced antibacterial efficacy and degradability to fight against drug-resistant bacteria.

Abstract:In this paper, we have designed and synthesized well-defined pegylated-polymers with tertiary amines from readily available commodity monomers 2-(dimethylamino)ethyl methacrylate (DMAEMA) and oligo(ethylene glycol) methyl ether methacrylate (OEGMA, Mn ∼ 475 Da) by reversible addition–fragmentation chain transfer (RAFT) polymerisation. By employing a simple and efficient post-polymerisation functionalization strategy, tertiary amines were quaternized to result in cationic polymers. By the careful selection of the functional halide, X–(CH2)q–R, (where in X=halide; R=the chemical functionality; q=the number of alkyl spacer between the quaternary ammonium group and R), a series of polymers with well-controlled molecular weight, different amphiphilic balance and chemical functionalities (such as alkyl, primary alcohol, primary amine and carboxylic acid) were readily synthesized. The antimicrobial activities of these cationic polymers were determined against Gram-positive bacteria Bacillus subtilis. Minimum inhibitory concentration (MIC), the polymer concentration to completely inhibit the bacterial growth, was found to be dependent both on the nature of functional group and the hydrophobicity of the polymer. Amongst the functional groups, both the alkyl and the alcohol groups were found to be effective, with MIC values in the range of 20–80mg/L. The haemolytic properties of polymers were analyzed against mouse red blood cells. The poymers with a short alkyl or hydroxyl group demonstrated little haemolysis, yet retained strong antimicrobial activity. The overall hydrophobicity of the polymer influenced its haemolytic behavior. These polymers can be promising antimicrobial agents. In addition, the approach proposed in this study to atom-efficient design and synthesis of antimicrobial polymers from the commercially available monomers can also be applied to develop well-defined functional cationic polymers for various biomedical applications.
Supramolecular Assembly of Β-Cyclodextrin-modified Polymer by Electrospinning with Sustained Antibacterial Activity.

Abstract:Supramolecular assembly loading drug as biomedical materials is a research hotspot. Herein, we reported a supramolecular electrospun assembly constructed via the hydrophobic and hydrogen bonding interaction. The obtained results showed that the assembly by supramolecular electrospinning not only increased the interactions of multiple antibacterial active species including antibiotics, cationic polymers, and silver to form a flexible membrane with good mechanical strength but also indicated the dual effects of rapid doxycycline and polyethyleneimine release as well as a sustained Ag release. Interestingly, the assembly showed not only good degradability but also a high bacteriostatic efficacy toward Escherichia coli (E. coli) up to 99.9%. More importantly, the in vivo wound healing assay indicated that the assembly could promote the healing of uninfected, E. coli-infected, and even methicillin-resistant staphylococcus aureus-infected wounds. The current research provides a novel approach to construct a supramolecular assembly by electrospinning mechanically induced strong noncovalent interaction.

Abstract:The increasing number of infections caused by pathogenic bacteria has severely affected human society, for instance, numerous deaths are from Gram-positive methicillin-resistant Staphylococcus aureus (MRSA) each year. In this work, four biodegradable antibacterial polymer materials based on cationic polyaspartamide derivatives with different lengths of side chains are synthesized through the ring-opening polymerization of beta-benzyl-l-aspartate N-carboxy anhydride, followed by an aminolysis reaction and subsequent methylation reaction. The cationic quaternary ammonium groups contribute to the insertion of the catiomers into the negatively charged bacterial membranes, which leads to membranolysis, the leakage of bacterial content, and the death of pathogens. Except for wiping out MRSA readily, the biodegradable polymers possessing alterable antibacterial potency can minimize the possibility of microbial resistance and mitigate drug accumulation by virtue of their cleavable backbone. To manipulate the poor biocompatibility of these polycations, carboxylatopillar[5]arene (CP[5]A) is introduced to the polymeric antibacterial catiomers through the supramolecular host-guest approach to obtain novel antibacterial materials with pH-sensitive characteristics (with CP[5]A departure from cationic quaternary ammonium compounds under acid conditions) and selective targeting of Gram-positive bacteria. Finally, the facile and robust antibacterial system is successfully applied to in vivo MRSA-infected wound healing, providing a significant reference for the construction of advanced antibacterial biomaterials.

Abstract:A novel geminized amphiphilic polymer containing double cationic head groups and hydrophobic tails in each structural unit (denoted as PAGBn), has been prepared in our study. The self-assembly behavior on the solid/liquid interface was investigated by quartz crystal microbalance with dissipation (QCM-D), steady-state fluorescence, dynamic light scattering (DLS) and transmission electron microscope (TEM). A two-regime buildup can be observed, where the first regime is attributed to the rapid adsorption of polymers, while a rearrangements of pre-adsorbed molecules and their aggregation on the surface dominate the second regime, exhibiting a formation of aggregates in large sizes due to the effect of the charge density and hydrophobes on the self-assembly process. It exhibits a drastically enhanced killing efficiency of ∼99.9% against both of S. aureus and E. coli bacteria for PAGBn compared with other traditional single-chained polymers, through the reinforced synergistic effect of electrostatic and hydrophobic interactions, and the formation of large-sized polymer aggregates, encompassing and killing the bacteria more efficiency. Abstract:With the increased prevalence of antibiotic-resistant infections, there is an urgent need for innovative antimicrobial treatments. One such area being actively explored is the use of self-assembling cationic polymers. This relatively new class of materials was inspired by biologically pervasive cationic host defense peptides. The antimicrobial action of both the synthetic polymers and naturally occurring peptides is believed to be complemented by their three-dimensional structure. In an effort to evaluate shape effects on antimicrobial materials, triblock polymers were polymerized from an assembly directing terephthalamide-bisurea core. Simple changes to this core, such as the addition of a methylene spacer, served to direct self-assembly into distinct morphologies-spheres and rods. Computational modeling also demonstrated how subtle core changes could directly alter urea stacking motifs manifesting in unique multidirectional hydrogen-bond networks despite the vast majority of material consisting of poly(lactide) (interior block) and cationic polycarbonates (exterior block). Upon testing the spherical and rod-like morphologies for antimicrobial properties, it was found that both possessed broad-spectrum activity (Gram-negative and Gram-positive bacteria as well as fungi) with minimal hemolysis, although only the rod-like assemblies were effective against Candida albicans.

Abstract:The rise of bacterial resistance to current antibiotics poses a threat to humanity and reinforces the need for new-generation nanomaterials with antibacterial properties and biosafety. Several types of nanomaterials have been shown to hold great potential to combat pathogenic microorganisms. Self-assembly of peptides and proteins, a spontaneous and tunable process, provides a wide range of new routes to construct functional biological nanomaterials with antibacterial properties. In particular, short-peptide-based supramolecular nanomaterials have attained substantial recognition due to their ease of fabrication, favorable physicochemical properties, and structurally diverse functionalities. Here, we present an overview of the recent progress on the design of short peptides, including linear peptides, amphiphilic peptides, and cyclic peptides, for the formation of supramolecular nanostructures as antibacterial agents and their respective therapeutic modes of action. Moreover, supramolecular short peptide composites and biomineralized nanomaterials are discussed, along with their biosafety and antibacterial mechanisms. These nanomaterials hold great promise as antibiotics of the near future due to their biocompatible, biodegradable, and environmentally friendly nature.

Abstract:Supramolecular antimicrobial materials based on synthetic macrocycles have recently aroused enormous interests due to their controllable and effective antibacterial treatment. Herein, a multicharged supramolecular assembly was fabricated employing the moderate host-guest interaction between hexa-adamantane-appended ruthenium polypyridyl (Ru2) and polycationic cyclodextrin (CD-QAS) in water. The positively multicharged feature of supramolecular assembly could remarkably enhance the specific intercalation and accumulation in negatively charged bacteria membrane leading to the physical membrane damage. Subsequently, the assembly could efficiently initiate the significant generation of singlet oxygen (1O2) in situ when irradiated with white light thus exhibiting a highly efficient antibacterial capability. Significantly, antibacterial experiments indicated that Ru2/CD-QAS displayed less effect on suppressing the growth of E. coli only about 25% in the absence of light while they exhibited excellent killing efficiency more than 99% toward E. coli under light irradiation. This work provides a simple approach for constructing supramolecular antimicrobial materials for synergistic photodynamic antibacteria.

Abstract:Herein, a three-armed amphiphilic metallacycle-cored star supramolecular polymer (Por-MOM-PDMAEMA) has been designed and synthesized via highly efficient post-assembly polymerization. This star polymer is further self-assembled into nanoparticles of different sizes depending upon the experimental conditions. The gas-controlled morphology transformation and tunable antibacterial activities of Por-MOM-PDMAEMAis systematically investigated and compared with metallacycle (MOM). The superior antibacterial activity of Por-MOM-PDMAEMA against multidrug-resistant P. aeruginosa implies that the presence of photodynamic photosensitizer (Por) and cationic polymer chain will significantly enhance antibactericidal activity, which is mainly attributed to the synergistic effect of photosensitizer and polymer chain linked in one metallacycle core. By leveraging the unique properties of metallacycle and their dynamic response to gaseous stimuli, the antibacterial properties of the Por-MOM-PDMAEMA can be finely tuned in response to gas triggers.

Abstract:Aiming to address the antimicrobial resistance and environmental burdens because of the widespread indulgence in biocides, a synthetic formula of biocides with controllable antibacterial activities and biodegradable characters are encouraged for the emergence of a novel spectrum of green biocides. In this respect, a polymeric antimicrobial catiomer, with the polymeric backbone sequentially attached with the functional cationic moieties and lipophilic moieties is synthesized. The striking antibacterial potency is achieved for treatment of both amoxicillin (AMO)-resistant Gram-positive S. aureus and Gram-negative E. coli. Moreover, the anionic aggregation-induced emission (AIE) compound is strategically introduced into the aforementioned polymeric antibacterial system, consequently employed as a facile reporter for the utility of bacteria detection based on the electrostatic reaction of anionic species in the bacterial membrane with the cationic polymeric antibacterials to liberate the anionic AIE compound. Ultimately, the proposed system is determined to afford appreciable temporal antibacterial potency by virtue of the readily available cleavage of the ester linkage in the presence of lipase, thereby eliminating the possibility of the antimicrobial resistance and environmental burdens. Hence, the proposed system provided a bottom-up approach for construction of an intriguing green antibacterial molecule.
De Novo Design of Entropy-Driven Polymers Resistant to Bacterial Attachment via Multicomponent Reactions

Abstract:Nonbactericidal polymers that prevent bacterial attachment are important for public health, environmental protection, and avoiding the generation of superbugs. Here, inspired by the physical bactericidal process of carbon nanotubes and graphene derivatives, we develop nonbactericidal polymers resistant to bacterial attachment by using multicomponent reactions (MCRs) to introduce molecular "needles" (rigid aliphatic chains) and molecular "razors" (multicomponent structures) into polymer side chains. Computer simulation reveals the occurrence of spontaneous entropy-driven interactions between the bacterial bilayers and the "needles" and "razors" in polymer structures and provides guidance for the optimization of this type of polymers for enhanced resistibility to bacterial attachment. The blending of the optimized polymer with commercially available polyurethane produces a film with remarkably superior stability of the resistance to bacterial adhesion after wear compared with that of commercial mobile phone shells made by the Sharklet technology. This proof-of-concept study explores entropy-driven polymers resistant to bacterial attachment via a combination of MCRs, computer simulation, and polymer chemistry, paving the way for the de novo design of nonbactericidal polymers to prevent bacterial contamination.

Abstract:Antimicrobial polymer as a new type of antimicrobial agent exhibits the perfect antibacterial activity and application prospects,because of its easy controlling structures and muhiplicative synthesis methods.The ways for synthesis of polymers,several factors impacting on the antibacterial activity and the relationship between antibacterial effect and polymer structures were introduced and discussed to provide a reference for the synthesis of the hishest activity antibacterial polymers.The research status and developing trend are also reviewed.
