Methods and Applications in Fluorescence

Abstract:Silver is widely used in medical materials, photography, electronics and other industries as a precious metal. The large-scale industrial production of silver-containing products and liquid waste emissions aggravate the environmental pollution. Silver ion is one of the most toxic metal ions, causing pollution to the environment and damage to public health. Therefore, the efficient and sensitive detection of Ag+ in the water environment is extremely important. Sulfur-doped carbon nitride nanosheets (SCN Ns) were prepared by melamine and thiourea via high-temperature calcination. The morphology, chemical composition and surface functional groups of the SCN Ns were characterized by SEM, TEM, XRD, XPS, and FT-IR. The fluorescence of SCN Ns was gradually quenched as the Ag+ concentration increased. The detection limit for Ag+ was as low as 0.28 nM. The quenching mechanism mainly is attributed to static quenching. In this paper, SCN Ns were used as the fluorescent probe for detecting Ag+. SCN Ns have successfully detected Ag+ in different environmental aqueous samples and cells. Finally, SCN Ns were further applied to the visual quantitative detection of intracellular Ag+.

Abstract:Water-soluble cadmium telluride quantum dots capped with mercaptosuccinic acid (MSA-CdTe QDs) were prepared for the selective and sensitive fluorescence probing of silver(I) ion. The fluorescence intensity of the MSA-CdTe QDs could be quenched only in the presence of Ag+, and the relative fluorescence intensity was decreased as a linear function of Ag+ concentration. Moreover, the proposed sensor was free from common interfering metal ions. The MSA-CdTe QDs were 3.0 nm in particle size and the maximum photoluminescence wavelength was 558 nm. Better sensitivity was obtained with 4.5 x 10(-4) mol L-1 MSA-CdTe QDs in 200 mmol L-1 phosphate buffer (pH 9.0) for a reaction time of 30 min. A linear response between the relative fluorescence intensity, and the concentration of Ag+ was obtained over the range 0.4 to 8 mmol L-1, with a relative standard deviation of 3.6% (n = 5) for 1.5 mmol L-1 and a detection limit of 54 nmol L-1. The proposed method was applied for the determination of Ag+ in two fixer samples at the mmol L-1 level. The spike recovery at a spiked level of 15 mmol L-1 varying from 89.4% to 92.3%, and good agreement between the results by the proposed and AAS methods proved the accuracy of the proposed method.
Ratiometric fluorescent probe based on dual-emissive carbon dots for determination of silver ions

Abstract:In this work, a one-step solvothermal method using O-phenylenediamine and DL-malic acid was used to obtain blue/yellow-emitting carbon dots (B/Y-CDs). The B/Y-CDs have two independent fluorescence emission peaks centered at 453 nm and 584 nm at a single excitation wavelength. In the presence of silver ions (Ag + ), the fluorescence at 453 nm was quenched and the fluorescence at 584 nm was enhanced by dynamic quenching and redox reaction mechanisms, respectively. The "Double-Response-Reverse" ratiometric fluorescent probes constructed on the basis of the B/Y-CDs showed a good linear relationship with Ag + in the range of 1-90 μM. In addition, the feasibility of this detection strategy for the detection of Ag + in real water samples was verified, and the probe showed satisfactory recoveries, which could be applied to the quantitative analysis of Ag + in real environmental water samples. This work provided a new strategy for the identification and detection of Ag + , enriching the ways of detecting Ag + .

Abstract:Fluorescent nitrogen and sulfur co-doped carbon dots (NSCDs) were synthesized by a one-step pyrolysis strategy using casein as carbon, nitrogen and sulfur sources, and characterized by UVevis spectrum, fluorescent spectrum, X-ray photoelectron spectroscopy (XPS) and FT-IR, etc. The synthesized NSCDs displayed a blue emission under ultraviolet illumination with a quantum yield of 31.8%, and a good aqueous solubility, photostability and biocompatibility. It was found that the fluorescence intensity of NSCDs could be selectively quenched by Hg2+, so NSCDs was used as an effective probe for the detection of Hg2+. The linear range and the limit of detection (LOD) of the fluorescent sensor based on NSCDs for the detection of Hg2+ were 0.01-0.25 mu M and 6.5 mu M, respectively. Spiked water samples were detected by the sensor with the recovery of 95.4-106.3% and relative standard deviation (RSD) of 3.6-8.6%. It was also observed that the quenched NSCDs-Hg2+ system could be restored by the addition of biothiols such as L-cysteine (Lcy), homocysteine (Hcy) and glutathione (GSH), thus NSCDs-Hg2+ system was employed as a fluorescent sensor for the detection of biothiols. The linear range and LOD of the NSCDs-Hg2+ system were 1-10 mM and 23.6 nM for Lcy, 0.2-2.5 mM and 12.3 nM for Hcy, and 0.1-2.0 mM and 16.8 nM for GSH, respectively. The NSCDs-Hg2+ system was applied for the detection of biothiols in serum samples with satisfied results. In addition, the study in vitro imaging HeLa cells revealed that the synthesized NSCDs could be used as effective fluorescent probes in cellular imaging without noticeable cytotoxicity. Abstract:Green fluorescent nitrogen-rich quantum dots (N-dots) have been synthesized under mild conditions (50 °C in methanol) and applied as a turn-off chemosensor for the rapid and selective detection of Hg2+ and Ag+ in aqueous solutions. Distinguishing between Hg2+ and Ag+ was also successfully achieved simply through the addition of ethylene diamine tetraacetic acid (EDTA). These N-dots-Hg and N-dots-Ag could also be used as turn-on chemosensors for biothiols through their binding competition for Hg2+ and Ag+ with N-dots.

Abstract:Ag+, extensively used in our daily life, has caused serious harm to human health and ecological environment, so highly selective detection and accurate quantification of Ag+ are critical. Herein, we report a novel water-soluble dual-element composite doped carbon quantum dots, N,S-CDs, then constructed them into fluorescent probes for Ag+ detection. Carbon source ammonium citrate (AC) could achieve nitrogen doping without additional nitrogen source, inorganic sulfur source sodium thiosulfate was used for rapid sulfur doping, the N,S-CDs emitting dazzling blue fluorescence were obtained through a rapid hydrothermal process. The synthesized N,S-CDs had an average particle size of 3.25 nm, with high quantum yield (QY = 36.8%) and excitation-independent fluorescence properties. Under optimized conditions, the fluorescent probe constructed by N,S-CDs could detect Ag+ with high sensitivity and fast response, showing a good piecewise linear relationship (0.1–700 μM). In addition, the fluorescent probe exhibited remarkable anti-interference ability and reusability, which is practical and feasible, and has broad application prospects in biological and environmental sample detection.

A Novel Method for the Detection of Silver Ions with Carbon Dots: Excellent Selectivity, Fast Response, Low Detection Limit and Good Applicability

Abstract:With the extensive application of silver containing materials in industry, the issue of release of silver ions has attracted increasing attention due to toxic effect of silver ions on the human health and environment. Traditionally, silver ions can be detected by atomic absorption spectroscopy, atomic emission spectrometry etc., which needed expensive and elaborate instruments and were high energy consumption. Therefore, economic and convenient methods are required for the effective determination of silver ions. Herein, two types of carbon dots (CDs-1 and CDs-2) have been prepared through a solvothermal synthesis method by using sucrose or citric acid and tris(hydroxymethyl)aminomethane as precursor, respectively. Then, CDs-1 were developed for the detection of silver ions through a colorimetric method. The detection mechanism was based on the reduction of silver ions and the formation of silver nanoparticles, which possess unique surface plasma resonance properties. CDs-1 could both act as reductants and stabilizers during the reaction for determination of silver ions. This CDs-1 based detection system could quantitatively determine silver ions in 3 min with a limit of detection (LOD) of 26 nmol/L (2.81 ppb). Interestingly, this strategy could also realize the detection of silver ions by CDs-2. Furthermore, the sensing assay is applicable to detecting silver ions in lake water and silver sulfadiazine cream samples. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:In this paper, nitrogen-doped carbon quantum dots (N-CQDs) were synthesized by a solvothermal method using 1,2,4-triaminobenzene as a carbon precursor. The surface of the synthesized N-CQDs was modified with amino functional groups. The results indicated that N-CQDs had various N-related functional groups and chemical bonds and were amorphous in structure. At the same time, the quantum yield of N-CQDs was 5.11%, and the average lifetime of fluorescence decay was 5.79 ns. The synthesized N-CQDs showed good selectivity for and sensitivity to Ag+. A linear relationship between N-CQDs detection efficiency and Ag+ concentration was observed for concentration ranges of Ag+ corresponding to 0–10 μM and 10–30 μM. In addition, N-CQDs were used for the detection of trace Ag+ in food packaging material. The silver ion content of the sample determined by the N-CQDs detection method was 1.442 mg/L, with a relative error of 6.24% with respect to flame atomic absorption spectrometry, according to which the Ag+ content was 1.352 mg/L. This indicates that the N-CQDs detection method is reliable. Therefore, the N-CQDs prepared in this paper can detect Ag+ rapidly, simply, and sensitively and are expected to be a promising tool for the detection of trace Ag+ in food packaging materials.

Abstract:Fluorescent sensing methods, because of their simple, cheap, fast, high selectivity and sensitivity , are widely used in the detection of cationic , anionic and other chemicals. Anthraquinone derivatives have many pharmacological activities, rigid planar structure and large ring conjugate system. They have moderate fluorescence emission wavelengths, good optical stabilities and high luminous efficiencies. In this work, anthraquinone derivatives were employed as fluorophore to prepare new fluorescent probes. Based on the mechanism of intramolecular charge transfer (ICT), a series of symmetric bis-1 ,2 ,3-triazole hydrazine derivatives K1 and K2 , which were confirmed by H-1 NMR and C-13 NMR, were designed and synthesized. The spectral properties were also investigated. The structure and theoretical spectrum of K1 were calculated using Gauss View 9. 0 quantization program and time-dependent density functional theory. The results showed that exhibited good selectivity to Ag+ in a mixed solution of DMSO/HEPES (1:7, V/V) . Binding analysis using the method of continuous variations (Job's plot) established that the stoichiometry of K1-Ag+ complex was estimated to be 1:1. The detection limit of K2 for Ag+ was 21. 2 mu mol/L (S/N= 3) . A complexation mechanism of K1-Ag+ was proposed, which was consistent with the calculated results. The fluorescence of K1 was increased dramatically in the presence of Ag+, and it remained nearly constant in the range of pH 6. 0-8.0. Other metal ions , such as Al3+, Ba2+, Cd2+, Co2+, Cr3+, Cu2+, Hg2+, Fe3+, K+, Mn2+, Na+, NH4+, Ni2+, Pb2+ and Zn2+ induced negligible fluorescence increasing for under the same conditions. The synthesized fluorescent probe K1 could be applied to image intracelluar Ag+ in living HepG2 cells , indicating that it was a useful tool in bioanalytical applications.

Abstract:To detect silver ions conveniently and rapidly in vitro and in vivo, a selective fluorescent probe with a wide measurement range, AgP, was developed. This probe exhibits a bright green fluorescence with an emission wavelength of 532 nm under 390 nm excitation, and its detection of Ag+ is stable in the pH range of 4–10 with a quench-type fluorescence response. Specially, the probe and its easily prepared test strips can be directly used for the colorimetric detection of Ag+ in aqueous samples with simple and convenient characteristics, the color change can be observed within a few seconds. The recovery rate of AgP detection water samples was between 117.6% and 98.3%, and the relative standard deviation (RSD) was between 0.41% and 2.06%. AgP is also suitable for in vivo imaging of Ag+ in the classical model plant, Arabidopsis thaliana, and 50 μM Ag+ can completely quench its fluorescence, which will provide a new detection tool for studying the distribution of Ag+ in the environment and live plants.

Abstract:In this report, red-emissive carbon dots (C-dots) were facilely prepared from o-phenylenediamine via microwave-assisted hydrothermal treatment. The C-dots demonstrated excitation wavelength-independent emission with maximums at 621 nm that could be effectively quenched by Ag+ via static quenching. This phenomenon was exploited to establish a sensitive fluorescence assay with a low detection limit (0.37 μM) and wide linear range (0-50 μM). In addition, this assay demonstrated excellent selectivity toward Ag+, free from the interference of 16 commonly seen metal ions. Most importantly, the assay demonstrated high reliability toward samples in deionized water, mineral water, lake water, and serum, which could indicate potential applications for Ag+ monitoring in complicated natural and biological environments.

Abstract:Herein, we report a plasmonic metal nanoparticle-involved sensor for cyanide ion based on the inner filter effect by using photoluminescent carbon dots as the signal reporter. With commercial bee pollen as the carbon resource, we synthesized photoluminescent nitrogen-doped carbon dots by a one-pot hydrothermal process, and their fluorescence quantum yield reached as high as 10.2% ± 0.5%. Fluorescence measurements indicated that the fluorescence of the carbon dots was insusceptible to the presence of many environmentally ordinary ions. Thanks to this “inert” property, we then developed a turn-on fluorescent sensor for cyanide ion in an inner filter effect manner by using carbon dots as the fluorophore and gold or silver nanoparticle as the light absorber. This detection technique is expected to be used for other metal nanoparticles-carbon dots ensemble fluorescent assays.

Abstract:In this study, fluorescent copper nanoclusters (Cu NCs) have been efficiently synthesized in a novel strategy via sonochemical method, using N-acetyl-L-cysteine (NAC) as reducing and stabilizing reagent. The preparation of Cu NCs could complete within 15 min due to the ultrasonic radiation treatment. The obtained NAC-Cu NCs exhibited red fluorescence at λem 630 nm. Silver ions could significantly induce fluorescence quenching of NAC-Cu NCs. Subsequently, a detection method for Ag+ with high response sensitivity and selectivity was constructed. The proposed method displayed a good linear relationship in the Ag+ concentration range of 1.64×10-10-1.16×10-8 M. The limit of detection (LOD) was as low as 7.76×10-11 M (signal/noise = 3). In addition, the practical application was further evaluated with environmental water samples, and the tested results were compared with those obtained by the inductively coupled plasma mass spectrometry (ICP-MS) method. The satisfactory recoveries (98.50%-101.6%) showed that the fluorescent detection method had potential application in environmental monitoring field.

Abstract:This study reports the successful preparation of nitrogen-doped carbon quantum dots (N-CQDs) with a quantum yield of 15 % utilizing the hydrothermal method. Citric acid was employed as the carbon source, while 1,3-Diamino-2-propanol was utilized as the dopant. It was discovered that the emission of synthesized N-CQDs was dependent on concentration and excitation. By altering the N-CQDs concentration, the fluorescence emission peak was shifted from 453 to 574 nm, causing the emission to transition from blue to orange. The presence of multiple fluorescence emission centers within N-CQDs was confirmed through two-dimensional fluorescence matrix scanning. As the concentration of N-CQDs increased, the distance between these centers decreased, ultimately leading to aggregation. The energy or electrons were transferred as a result of the surface functional groups' interaction, which ultimately causes the fluorescence emission peak to shift to the red. Furthermore, an increase in concentration results in the formation of larger aggregates and larger sp 2 carbon domains, ultimately resulting in longer wavelengths. Because of the convenience of use, high sensitivity, and rapid response of the fluorescent probe detection approach, multi-color N-CQDs with tunable concentration were used for the identification of Fe 3+ , with a 10–200 μM linear detection range and limits of detection (LOD) of 0.56 μM.

Abstract:Herein, a novel nitrogen-doped carbon dots (N-CDs) nanosensor for detecting Ag+ and glutathione (GSH) has been designed. Interestingly, Ag+ could significantly improve the fluorescence (FL) intensity of the N-CDs we prepared, which is different from most other metal ions with FL quenching effects on N-CDs. The sensing mechanism can be attributed to the oxidation characteristics of Ag+ and the formation of silver nanoparticles (AgNPs). In the reaction, the N-CDs acted as the reducing agent and stabilizer. Under the optimal conditions, the N-CDs could detect Ag+ with a nice linear relationship of R2 = 0.992 and the limit of detection was 0.091 μM. Furthermore, the FL of N-CDs solutions could return to nearly original intensity after introducing glutathione (GSH), which allowed the N-CDs to be applied for GSH detection sensitively and selectively. In addition, the N-CDs were also successfully applied to cellular multicolor imaging of Hela cells. The results reported here pave the way for N-CDs-based fluorescent "off-on-off" sensor in environment, bioassay and bioimaging.

Abstract:A ratiometric fluorometric assay for silver(I) is described. The method makes use of a dually emitting quantum dot hybrid, which is composed of (a) blue-fluorescent silicon- and nitrogen-doped carbon quantum dots (CQDs), and (b) of red-emitting CdTe quantum dots (QDs) capped with N-acetyl-L-cysteine. The red-emitting CdTe QDs undergo strong and specific quenching by Ag(I), whereas the blue-emitting N,Si-CQDs are not quenched. The two kinds of QDs are mixed and used as a ratiometric fluorescent probe. A linear relationship is found between the log of intensities [(I608/I441)0/(I608/I441)] and the concentration of Ag(I) in the range from 5.0–1000 nM, and the limit of detection (at S/N = 3) is 1.7 nM. Possible interferents (including 17 general metal ions, 12 anions and fulvic acid) do not interfere with the determination. The assay was successfully used for the determination of Ag(I) in surface water and wastewater samples. The fluorescence quenching mechanism of the ratiometric assay system was also discussed in detailed.
