Abstract:An alternative route has been developed for the construction of multisubstituted pyrrole derivatives from readily available, doubly activated cyclopropanes and anilines using an iron-mediated oxidation domino reaction (i.e., sequential ring-opening, cyclization, and dehydrogenation reactions). This reaction uses readily available reactants and is tolerant of a broad range of substrates, with the desired products being formed in good to excellent yields.

Abstract:The unique reactivity and synthetic applications of oxime acetate derivatives with N, O and S substituents at the α‐position are disclosed for the first time, which leads to 2‐substituted fulleropyrrolidines and mono‐ and disubstituted 1‐fulleropyrrolines via copper‐catalyzed redox‐neutral N‐heteroannulation reactions with C60. This transformation is operationally simple and has a broad substrate scope and good functional group tolerance. Theoretical calculations at the level of B3LYP/6‐31G(d) were performed to elucidate the chemoselectivity of the reaction.

Abstract:Radical chemistry is synthetically useful but can be plagued by the non-intuitive reaction course and indiscriminate reactivity profile. Herein, dynamic radical effect is revealed as a conceptual logic for the predictive achievement of radical reaction selectivity. The reversible bonding association/dissociation of two radicals serves as a synthetic handle for directing one radical to the target reaction recourse, without the participation of the other radical. A Mn catalytic protocol has been developed for cycloalkene ring expansion synthesis of azaheterocycles. An initial azidyl radical addition to alkene and subsequent reversible O2 occupation of C-radical site prevents further radical coupling and steers the reaction toward the intramolecular rearrangement pathway. A broad substrate scope has been established for the synthesis of pyridine and isoquinoline derivatives. This new radical synthetic perspective promises as an important guiding principle for empowering radical-based chemical transformations.

Abstract:Herein, we disclose a general and practical iminoarylation of alkenes by nickel-catalyzed reductive cross-coupling of unsaturated oxime esters with readily available aryl halides, providing an expedient approach for constructing pyrroline derivatives. The absence of organometallic reagents enables the reaction to occur under mild conditions with a broad substrate scope and good functional group tolerance. Moreover, other C-based electrophiles, including alkenyl, alkynyl and alkyl halides, or pseudohalides, were also competent substrates for this reaction.

Abstract:A Cu‐catalyzed oxidative radical [2+2+1] annulation reaction of phenol‐linked 1,7‐enynes with α‐active methylene nitrile is described, affording cyclopenta[c]chromene derivatives bearing several functional groups in excellent yields. The wide substrate range, good group tolerance, and easy amplification experiments indicate the practicability of this synthesis strategy. This process provides an alternative convenient route for the synthesis of cyclopenta[c]chromene derivatives.

Abstract:An efficient copper-catalyzed tandem oxidation/alpha-formylation reaction of 2,3-dihydro-1H-pyrroles in an oxygen atmosphere has been developed. The new reaction is conducted in dimethyl sulfoxide (DMSO), which serves as both the carbonyl carbon source and solvent, in the presence of trifluoroacetic acid to directly generate structurally diverse alpha-formylpyrrole derivatives in one pot in moderate to good yields. Sensitive aldehyde groups remain intact in the presence of an oxidant, trifluoroacetic acid and a transition metal salt. A possible mechanism is presented.

Abstract:Pyridines and cyclopropanes are important structural units in chemistry. Herein, we introduce a photoredox-catalyzed approach for the ring opening and 1,3-oxypyridylation of aryl cyclopropanes using 4-cyanopyridines and carboxylic acids. This sequential process involves single-electron oxidation of the aryl cyclopropane, leading to nucleophilic ring opening and radical pyridylation at the benzylic position. The redox-neutral reaction exhibits high regioselectivity under mild reaction conditions, offering a broad substrate scope and wide applicability.

Abstract:We developed a novel protocol for the preparation of functionalized chromeno[2,3-c]pyrrol-9(1H)-one derivatives (CMPOs) 3 & 3' from 3-formylchromones with proline derivatives. This strategy involved an unprecedented cascade reaction entailing simple heating of a mixture of substrates 1 with 2 and toluene, followed by oxidation with oxygen and catalysis by CuBr. As a result, a series of CMPOs 3 & 3' were produced via cleavage of two bonds (2 C−C) and formation of three bonds (1 C−N and 2 C−C bonds) in a single step. This protocol can be used to synthesize functionalized CMPOs for combinatorial and parallel syntheses of CMPOs in a one-pot reaction.

Abstract:A variety of functionalized 2‐pyrrolidinones bearing N‐trifluoromethylphenyl moieties were prepared in moderate to good yields with high diastereoselectivity through copper(II)‐catalyst and base co‐catalyzed formal [4+1] cycloaddition of cinnamaldehyde nitrones with 1‐ethynylnaphthalen‐2‐ols in the presence of m‐CPBA as the oxidant. The triple bond of 1‐ethynylnaphthalen‐2‐ol served as one‐carbon synthon for the pyrrolidinone ring formation. Mechanistic studies revealed that m‐CPBA played an important role to interrupt the intramolecular O‐cyclization to afford the 2‐pyrrolidinones. Moreover, the reaction could be easily performed at gram scales and the obtained products were selectively converted into diverse pyrroline scaffolds.

Abstract:A variety of dihydropyrrole-functionalized phenanthridines were efficiently synthesized by the metal-free, radical cascade cyclization reaction of 2-isocyanobiphenyls with γ,δ-unsaturated oxime esters. The C-N/C-C/C-C bonds were formed via the oil bath method in a one-pot procedure with broad substrate applicability. The radical process was supported by kinetic isotope effect studies and radical inhibition studies.

Abstract:An four-component cascade cyclization reaction between readily available 2,4-pentanediones, primary amines and sodium sulfinates is reported. Highly functionalized 5-alkylidene-3-pyrrolin-2-ones were efficiently synthesized in moderate to excellent yields under mild conditions. This regioselective cascade cyclization was achieved by direct transformation of C(sp3)−H bonds at two weak nucleophilic sites in the enaminones structure, and two strong nucleophilic sites of the structure were not involved.

Abstract:We have rationally designed a new class of alkyne-tethered oximes and applied them in an unprecedented iron-catalyzed radical relay protocol for the rapid assembly of a wide array of structurally new and interesting fused pyridines. This method shows broad substrate scope and good functional-group tolerance and enabled the synthesis of several biologically active molecules. Furthermore, the fused pyridines could be diversely functionalized through various simple transformations, such as cyclization, C-H alkylation, and a click reaction. DFT calculation studies indicate that the reactions involve cascade 1,5-hydrogen atom transfer, 5-exo-dig radical addition, and cyclization processes. Moreover, preliminary biological investigations suggest that some of the fused pyridines exhibit good anti-inflammatory activity by restoring the imbalance of inflammatory homeostasis of macrophages in a lipopolysaccharide-induced model.
