Abstract:An efficient protocol for the synthesis of furans through Rh(III)-catalyzed vinyl C-H activation from acrylic acids and α-diazocarbonyl compounds has been developed. The reaction features broad functional group tolerance and affords a series of furans in moderate to good yields. Moreover, no additives such as copper or silver salts are required. Some control experiments are performed to give insight into the mechanism of this cascade transformation and the decarbonylation process is involved in the formation of the furan product.

Abstract:A rhodium-catalyzed formal [4 + 1]-cycloaddition of pyridotriazoles and aryl propargyl alcohols is reported, providing an effective access to 2-pyridyl-substituted 2,5-dihydrofuran derivatives in moderate to high yields. Mechanistically, the proposed oxonium ylide intermediate in this catalytic alkyne carbocyclization transformation is verified by an interception experiment for the first time.

Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:Alkynyloxiranes have been employed for the first time as effective coupling partners in Cp*Rh-III-catalyzed C-H functionalization reactions. Their annulation with N-aryloxyamides then offers a redox-neutral and efficient synthesis of functionalized 2,3-dihydrobenzofurans bearing an exocyclic E-allylic alcohol and a tetrasubstituted carbon center in good yields with a broad substrate scope. The products can be easily converted to molecules with more complexity, which provides an opportunity for rapid assembly of structurally diverse heterocycles.

Abstract:Ag(I)-catalyzed synthesis of diazo-trisubstituted furans starting from diazo-cumulated allenyl ketones has been investigated. The Rh2(OAc)4-catalyzed reaction of the diazo 2,3,5-trisubstituted furans provided penta-substituted aromatics via cascade intermolecular cyclopropanation/rearrangement/isomerization. The cyclopropanation on the furan ring/rearrangement of cyclopropane moiety has been reported. A reasonable mechanism is proposed.

Abstract:An effective method to synthesize alpha-functionalized furan and pyrrole derivatives was developed using 2-alkoxy-2,3-dihydrofurans as modular precursors. This protocol featured a previously unreported tandem nucleophilic substitution/heteroaromatization reaction. Nucleophiles such as indole, alpha-oxoketene dithioacetal, trimethoxybenzene, and dimethoxynaphthalene can react readily with 2-alkoxy-2,3-dihydrofurans to afford a-functionalized five-membered ring heterocycles in the presence of acid catalysts, such as copper bromide and iron chloride. The mechanism of the reaction was also discussed, in which the first step, nucleophilic substitution, is the key in triggering the succeeding heteroaromatization. This method can also be extended to the synthesis of dihydrothiophenes.

Abstract:Spiro-lactams and polysubstituted pyrroles were synthesized by reactions of furfurylamines with ynones followed by oxidation. Specifically, the protocol involved in situ generation of N-furan-2-ylmethyl-β-enaminones and their subsequent oxidation by ceric ammonium nitrate (6 equiv for spiro-lactam formation, 3 equiv for pyrrole formation). This useful dearomatizing oxidation, which likely proceeds via a free-radical pathway, can be expected to extend the synthetic applications of furan and pyrrole derivatives.

Now, a rhodium-catalysed three-component [2+2+2] cyclotrimerization with excellent chemo-, regio- and stereocontrol is reported that does not require any special operations. The utility of the process is shown and mechanistic studies probe the origins of the selectivity.
