Abstract:Cold atmospheric plasma (CAP) is a novel technology which is widely used in the biomedical field and has developed quickly over the past few years, especially in cancer therapy. Compared to traditional anti-cancer approaches, CAP treatment shows a selective anti-cancer mode which makes CAP a promising anti-cancer treatment modality. However, the underlying mechanism of this selective cytotoxicity is not fully elaborated. In this study, we demonstrated that A375 human melanoma cells were more sensitive to CAP treatment than HaCaT cells and compared the differences in metabolites of these two cell lines after CAP treatment. Using ultra-high performance liquid tandem chromatography quadrupole time of flight mass spectrometry (UHPLC-QTOF-MS) approach, we identified 361 changes (metabolites) in HaCaT cells and 1531 changes in A375 cells. Further research using KEGG pathway analysis found that purine metabolism and Pantothenate and CoA biosynthesis were highly correlated with changes of HaCaT cells and A375 cells after CAP treatment. Our results shed light on the mechanism of selective cytotoxicity in the metabolites' aspect.
In vitro study of cold atmospheric plasma-activated liquids inhibits malignant melanoma by affecting macrophage polarization through the ROS/JAK2/STAT1 pathway

Abstract:Melanoma is a prevalent malignant skin tumor known for its high invasive ability and a high rate of metastasis, making clinical treatment exceptionally challenging. Tumor-associated macrophages (TAMs) are the most abundant immune cells in the tumor microenvironment and play a crucial role in tumor survival and development. Cold atmospheric plasma (CAP) is an emerging tool for tumor treatment that has garnered attention from scholars due to its interaction with non-tumor cells in the tumor microenvironment. Here, we used the macrophage lines THP-1 and RAW264.7, as well as the melanoma cell lines A375 and MV3, as research subjects to investigate the effect of plasma-activated liquid (PAL) on macrophage differentiation and its inhibitory effect on melanoma cell proliferation. We confirmed that the killing effect of PAL on melanoma cells was selective. Using flow cytometry and PCR, we discovered that PAL can influence macrophage differentiation. Through in vitro cell coculture, we demonstrated that PAL-treated macrophages can significantly impede tumor cell development and progression, and the effect is more potent than that of PAL directly targeting tumor cells. Furthermore, we have proposed the hypothesis that PAL promotes the differentiation of macrophages into the M1 type through the ROS/JAK2/STAT1 pathway. To test the hypothesis, we employed catalase and fludarabine to block different sites of the pathway. The results were then validated through Western Blot, qPCR and ELISA. This study illustrates that PAL therapy is an effective tumor immunotherapy and expands the scope of tumor immunotherapy. Furthermore, these findings establish a theoretical foundation for potential clinical applications of PAL.

Abstract:Atmospheric pressure cold plasma that contains numerous act ive components including electrons, ions, free radicals, reacti ve molecules and photons has been extensively researched as a promising anticancer treatment technology in recent years 1 . Melanoma is the most aggressive form of skin cancer with a high prevalence, a median overall survival of less than 2 year s, and resistance to current modalities of cancer therapy 2 . Plas ma can inactivate melanoma caner cells effectively, but the m echanism of the interaction between plasma and cells is not y et fully understood and needs to be investigated further. In this work, an improved atmospheric pressure plasma jet (A PPJ) with three electrodes in helium was utilized for treatmen t of murine melanoma caner cells cultured in vitro. After bein g exposed to APPJ with different time durations, the cell viab ility, transmission electron microscopy (TEM) images, intrac ellular superoxide dismutase (SOD), malondialdehyde (MD A), reactive oxygen species (ROS) and cell cycle distribution of melanoma cells were measured, respectively. It was show ed that, compared with the control cells, the cell viability and SOD activity decreased, the degree of cell damage, MDA an d ROS content increased in a time-dependent manner with 10, 30, and 60 s of plasma treatment. Furthermore, the APPJ ma y reduced cell viability of melanoma cells via inducing G1/S and G2/M cell cycle arrest. It was concluded that oxidative st ress in melanoma cells was mainly induced by the reactive ox ygen and nitrogen species (ROS and RNS) in the plasma jet, which were detected by optical emission spectroscopy.

Abstract:Cold atmospheric plasma was shown to induce cell apoptosis in numerous tumor cells. Recently, some other biological effects, such as induction of membrane permeation and suppression of migration, were discovered by plasma treatment in some types of tumor cells. In this study, we investigated the biological effects of plasma treatment on multiple myeloma cells. We detected the detachment of adherent myeloma cells by plasma, and the detachment area was correlated with higher density of hydroxyl radical in the gas phase of the plasma. Meanwhile, plasma could promote myeloma differentiation by up-regulating Blimp-1 and XBP-1 expression. The migration ability was suppressed by plasma treatment through decreasing of MMP-2 and MMP-9 secretion. In addition, plasma could increase bortezomib sensitivity and induce myeloma cell apoptosis. Taking together, combination with plasma treatment may enhance current chemotherapy and probably improve the outcomes.

Abstract:Cold atmospheric pressure plasma (CAP) is widely used in research for biomedical applications and is a promising therapy to selectively eradicate cancer cells. However, there is no unity for the species that cause this different effect. The aim of this article was to further determine the key reactive species causing differences in viability between melanoma cells (B16) and fibroblasts (L929). In this article, the cell viability of two cell lines exposed directly to the CAP was tested. The result shows CAP exhibit different cytotoxicity against the viability of two cell lines. The proliferation of B16 cells is significantly inhibited. By using superoxide anions scavengers and catalase, long-lived reactive oxygen species (ROS) generated by CAP, such as hydrogen peroxide (H2O2), were found to have stronger cytotoxicity to B16. Besides, the B16 cells were significantly inhibited after simultaneously scavenging free radicals and hydrogen peroxide, while it has the opposite effect on L929 cells. In order to explore whether there may be other long-lived species, such as protein oxides, which have important effects on cells, CAP-treated bovine serum albumin (BSA) was applied in cultured cells. We observed the inhibition effect of CAP-treated BSA on the growth of B16 but proliferation versus L929 cells. The content of peroxide in CAP-treated BSA was measured by xylenol orange method. The rate of peroxide reduction in B16 cells is faster than that of L929 cells. In addition, after adding certain enzymes to cells, the effects of CAP on intracellular ROS generation were examined by liquid chromatography/mass spectrometry (LC/MS). There is a significant difference in the content of O-2(center dot-) and H2O(2) in the two cells.

Abstract:Abstract Skin color is derived from epidermal melanocytes that contain specialized organelles in which melanin is formed. The formation of melanin is a well-orchestrated process, and reactive oxygen species (ROS) play a role in numerous enzymatic conversions, such as the reactions catalyzed by tyrosinase and tyrosine hydroxylase. Currently, there is ample evidence that cold plasma exerts biological effects on cells through the impact of ROS and reactive nitrogen species (RNS). Modulation of melanin biosynthesis by cold plasma has not yet been investigated. This study investigated melanin biosynthesis of human melanoma cell lines with different endogenous melanin contents (SK-Mel 28, G-361, FM-55-P and MNT-1) in response to cold plasma-derived reactive species. Initially, the distribution of melanosomes, via immunofluorescence, and the influence of microphthalmia-associated transcription factor (MiTF), as a key transcription factor, was investigated. In our experimental setup, all of the tested cell lines had an elevated melanin content after exposure to cold plasma. These promising results suggest a novel potential application of cold plasma for the regulation of melanogenesis and may be a useful tool for influencing skin color in the future.

Abstract:Reports show that atmospheric pressure cold plasma could induce cancer cell apoptosis in several minutes. However, up to now, only little has been known about the mechanism of plasma inducing cancer cell apoptosis. In this paper, an atmospheric pressure plasma plume is used to treat human hepatocellular carcinoma (HepG2) cell. Experimental results show that plasma can modulate the intracellular reactive oxygen species (ROS), and nitric oxide (NO) concentration. The relationship between the cell apoptosis and the concentrations of ROS, and NO is analysis. The results indicate that, firstly, the plasma generates NO, which increase the NO concentration of the cells outer environment. Secondly, the intracellular NO concentration is increased due to the NO diffusion from the medium. Thirdly, the intracellular ROS concentration is increased due to the increase of the intracellular NO concentration. Finally, the increase of the intracellular NO, and ROS concentrations result in the HepG2 cell apoptosis.

Abstract:Cold atmospheric pressure plasma is widely used in research for biomedical applications and is a promising therapy to selectively eradicate cancer cells. However, fundamental information related to the plasma modification of biomolecules in aqueous solution remains elusive. In this work, we studied the chemical basis of tryptophan (Trp) oxidation using an Ar plasma jet and investigated the effects of plasma-treated tryptophan (PTT) on cell viability. The results show that the main product of the Trp of plasma induced oxidation is a mixture of hydroxyl derivatives and hydroperoxides in aqueous form. The products result primarily from the hydroxyl radical (·OH) attacking the Trp, which can be explained by the interaction with both the aromatic and the pyrrole rings. We observed that the PTT has a different proliferation effect between the growth of the B16 melanoma cells and the L929 fibroblast cells. The experimental results indicated that the effect of the PTT is dose-dependent in the B16 cells, ranging from cell proliferation to cytotoxic damage with apoptosis. Furthermore, we examined the intracellular changes in hydrogen peroxide (H 2 O 2 ) and superoxide radicals ( O_2^·̅ ) following the Trp-derived treatment using the fluorescence probe method. Rigorous identification of the reactive oxygen species (ROS) produced by PTT in cells as O_2^·̅ and H 2 O 2 helped establish the cellular source of ROS. It is shown that the intracellular H 2 O 2 might originate from the activation of NADPH. These results suggested that the amino acid and protein hydroperoxides may play a crucial role in the action of plasma on the biologic target.
Cold Atmospheric Plasma Induces Apoptosis of Melanoma Cells Via Sestrin2‐mediated Nitric Oxide Synthase Signaling

Abstract:Cold atmospheric plasma (CAP) represents a promising therapy for selectively cancer killing. However, the mechanism of CAP‐induced cancer cell death remains unclear. Here, we identified the tumor necrosis factor‐family members, especially Fas, and overloaded intracellular nitric oxide participated in CAP induced apoptosis in A375 and A875 melanoma cell lines, which was known as extrinsic apoptosis pathway. This progress was mediated by antagonistic protein of reactive oxygen species, Sestrin2. The over expression of Sestrin2 induced by plasma treatment resulted in phosphorylation of p38 mitogen‐activated protein kinase (MAPK), followed by increased expression of nitric oxide synthase (iNOS), Fas and Fas ligand. Depletion of Sestrin2 reduced iNOS and Fas expression, which was associated with reduction of plasma‐induced apoptosis. In contrast, inhibition of iNOS activity and phosphorylation of p38 did not alter Sestrin2 expression in plasma‐treated melanoma cells. Taken together, cold atmospheric plasma increases Sestrin2 expression and further activates downstream iNOS, Fas and p38 MAPK signaling to induce apoptosis of melanoma cell lines. These findings suggest a previously unrecognized mechanism in melanoma cells response to cold atmospheric plasma therapy.

Abstract:Background Despite new progress of chemotherapy in multiple myeloma (MM) clinical treatment, MM is still a refractory disease and new technology is needed to improve the outcomes and prolong the survival. Cold atmospheric plasma is a rapidly developed technology in recent years, which has been widely applied in biomedicine. Although plasma could efficiently inactivate various tumor cells, the effects of plasma on tumor cell metabolism have not been studied yet. Methods In this study, we investigated the metabolite profiling of He plasma treatment on myeloma tumor cells by gas-chromatography time-of-flight (GC-TOF) mass-spectrometry. Meanwhile, by bioinformatic analysis such as GO and KEGG analysis we try to figure out the metabolism pathway that was significantly affected by gas plasma treatment. Results By GC-TOF mass-spectrometry, 573 signals were detected and evaluated using PCA and OPLS-DA. By KEGG analysis we listed all the differential metabolites and further classified into different metabolic pathways. The results showed that beta-alanine metabolism pathway was the most significant change after He gas plasma treatment in myeloma cells. Besides, propanoate metabolism and linoleic acid metabolism should also be concerned during gas plasma treatment of cancer cells. Conclusions Cold atmospheric plasma treatment could significantly alter the metabolite profiling of myeloma tumor cells, among which, the beta-alanine metabolism pathway is the most susceptible to He gas plasma treatment.

Abstract:The biological characteristics of three-dimensional (3D) tumors cannot be adequately represented through two-dimensional (2D) cell cultures. Here, to acquire more precise information about plasma-tumor interactions, the plasma-induced cytotoxicity was tested in both 2D cell culture and 3D tumor-spheroid culture. The results indicated that the toxic effects were markedly reduced in 3D cancer cells compared with 2D cells. We found that reactive oxygen and nitrogen species can penetrate through the entire depth of 3D tumor spheroid, but local apoptosis was observed only on the outermost layer. The permeation depth of plasma-generated reactive species was then tracked in 3D tumor spheroids. We hypothesized that plasma-induced apoptosis within a tumor depends on the "deep" delivery of H2O2, NO2-, and NO3- rather than O-2(-), OH center dot, and ONOOH/ONOO-.
Low Temperature Plasma Activated Liquid Induces Melanoma Apoptosis in Vivo and in Vitro

Abstract:In this study, the inhibiting effect and underlying mechanism of plasma activated Liquid (PAL) on B16 melanoma cell are proved. The helium atmospheric-pressure plasma jet (APPJ) is used to generate the plasma-activated saline and medium (PAS and PAM). Then the PAS is injected subcutaneously to treat B16-tumor bearing mice in vivo and the PAM is used to treat cancer cells in vitro. The weight of tumor nodules, cryosections, cell viability, apoptosis, DNA damage, p53 pathway-related protein and the concentration of H 2 O 2 in PAM are detected. The in vivo results show that PAS can inhibit tumor growth and induce apoptosis. The in vitro results show the concentration of H 2 O 2 in PAM increases as APPJ irradiation time is longer, while the cell viability decreases as APPJ treatment time increases. The flow cytometry results indicate that, compared with the control group, all the PAM treatment groups show a higher apoptotic rate. The single cell gel assay and an 8-OHdG assay display that the PAM can induce DNA damage. And the western blotting assay indicate that the PAM increases the expressions of p53, p53-induced gene 8 protein, PIG3, cytoplasmic cytochrome c, cleaved-caspase-9/3. However, CAT can alleviate all these changes effectively. These outcomes suggest that plasma can exert an apoptotic effect on B16 cells mainly through activating the p53/PIGs/caspase pathway.

Abstract:Summary form only given. In recent years, atmospheric pressure nonequilibrium plasmas (APNPs) have been used in many biomedical applications such as bacteria, yeasts, fungi and algae inactivation, surface treatment for cell sheet engineering to controlled cell cultivation, promote blood coagulation and root canal of teeth sterilization. The APNPs can operate at room temperature and APNPs can form numerous short-lived but highly active chemical particles, such as reactive oxygen species, hydroxyl radical (OH), and other excited species. These active chemical particles are essential for various biological processes in cells and human body, reactive oxygen species are involved in initiation, progression and metastasis of cancers and nitric oxide (NO) can induce apoptosis, necrosis or protect cells from death, depending on the cell type and the amount, duration, and site of NO production. Therefore, it is important to study the impact of plasma on cancer cells. In this paper, human hepatocellular carcinoma cell (HepG2) is treated by a single electrode plasma jet device. Further investigation by using the flow cytometric analysis demonstrates that the plasma treatment increases the percentage of apoptotic cells being associated with cell cycle arrest at G2/M phase. Besides, the Reverse Transcriptase PCR assay shows that the Cyclin B1 and Cdc2 are decreased in transcription level after plasma treatment, while the expression of the p21 Cdk inhibitor, as well as tumor suppressor p53, is enhanced. On the other hand, the levels of certain pro-apoptotic or anti-apoptotic genes are checked, the experimental results suggests that the plasma induces apoptosis through shifting the Bax/Bcl-2 ratio to trigger HepG2 cell apoptosis. Results of the present investigation indicate that the plasma jet device may have potential therapeutic activity, such as sterilization of living tissue and tumor therapy of the postoperative treatment process.
Plasma‐Induced Death of HepG2 Cancer Cells: Intracellular Effects of Reactive Species

Abstract:Reports show that cold atmospheric-pressure plasmas can induce death of cancer cells in several minutes. However, very little is presently known about the mechanism of the plasma-induced death of cancer cells. In this paper, an atmospheric-pressure plasma plume is used to treat HepG2 cells. The experimental results show that the plasma can effectively control the intracellular concentrations of ROS, NO and lipid peroxide. It is shown that these concentrations are directly related to the mechanism of the HepG2 death, which involves several stages. First, the plasma generates NO species, which increases the NO concentration in the extracellular medium. Second, the intracellular NO concentration is increased due to the NO diffusion from the medium. Third, an increase in the intracellular NO concentration leads to the increase of the intracellular ROS concentration. Fourth, the increased oxidative stress results in more effective lipid peroxidation and consequently, cell injury. The combined action of NO, ROS and lipid peroxide species eventually results in the HepG2 cell death. The mechanism of death of human hepatocellular carcinoma cells (HepG2) induced by atmospheric-pressure room-temperature plasma, related to the plasma-controlled intracellular concentrations of reactive oxygen species (ROS), nitric oxide (NO) and lipid peroxide is revealed. Only 34.75 s are required to reduce the number of the viable HepG2 cells by 50%.

Abstract:The potential antitumor application of low-temperature plasma (LTP) has attracted wide attention from researchers. The mechanism of plasma antitumor action may be related to LTP-induced reactive oxygen and nitrogen species. The aim of this study was to identify the apoptotic effects of plasma on melanoma cells (B16) in vivo and in vitro. For this purpose, a helium atmospheric-pressure plasma jet (APPJ) was used to generate the plasma-activated saline and medium (PAS and PAM). The PAS was injected subcutaneously to treat B16-tumor bearing mice in vivo and the PAM was used to treat B16 cells in vitro. Catalase (CAT) was added into the PAM to degrade H2O2 as the intervention group. The tumor nodules' weight, cryosections, B16 cell viability, apoptosis, DNA damage, p53 pathway-related protein and the concentration of H2O2 in PAM were detected. The in vivo results showed that PAS could inhibit tumor growth and induce apoptosis. The in vitro results revealed the concentration of H2O2 in the PAM increased as LTP treatment time was longer, while the cell viability decreased as APPJ irradiation time was increased. Flow cytometry results showed that, compared with the control group, all PAM treatment groups showed a higher apoptotic rate. A single cell gel assay and an 8-OHdG assay showed that the PAM could induce DNA damage. A western blotting assay indicated that the PAM could increase the expressions of p53, p53-induced gene 8 protein, PIG3, cytoplasmic cytochrome c, cleaved-caspase-9 and -3. However, CAT could alleviate all these changes effectively. These findings suggested that plasma exerted an apoptotic effect on B16 cells mainly by activating the p53/PIGs/caspase pathway.
Systemic Study on the Safety of Immuno-Deficient Nude Mice Treated by Atmospheric Plasma-Activated Water

Abstract:Cold atmospheric-pressure plasma is a new technology, widely used in many fields of biomedicine, especially in cancer treatment. Cold plasma can selectively kill a variety of tumor cells, and its biological safety in clinical trials is also very important. In many cases, the patient's immune level is relatively low, so we first studied the safety assessment of plasma treatment in an immuno-compromised animal model. In this study, we examined the safety of immuno-deficient nude mice by oral lavage treatment of plasma-activated water, and studied the growth status, main organs and blood biochemical indexes. Acute toxicity test results showed that the maximum dose of plasma treatment for 15 min had no lethal effect and other acute toxicity. There were no significant changes in body weight and survival status of mice after 2 min and 4 min of plasma-activated water (PAW) treatment for 2 weeks. After treatment, the major organs, including heart, liver, spleen, lung and kidney, were not significantly changed in organ coefficient and tissue structure. Blood biochemical markers showed that blood neutrophils and mononuclear cells were slightly increased, and the others remained unchanged. Liver function, renal function, electrolytes, glucose metabolism and lipid metabolism were not affected by different doses of PAW treatment. The above results indicate that PAW treatment can be used to treat immuno-deficient nude mice without significant safety problems.
Cold Atmospheric Plasma As a Potential Tool for Multiple Myeloma Treatment

Abstract:Multiple myeloma (MM) is a fatal and incurable hematological malignancy thus new therapy need to be developed. Cold atmospheric plasma, a new technology that could generate various active species, could efficiently induce various tumor cells apoptosis. More details about the interaction of plasma and tumor cells need to be addressed before the application of gas plasma in clinical cancer treatment. In this study, we demonstrate that He+O2 plasma could efficiently induce myeloma cell apoptosis through the activation of CD95 and downstream caspase cascades. Extracellular and intracellular reactive oxygen species (ROS) accumulation is essential for CD95-mediated cell apoptosis in response to plasma treatment. Furthermore, p53 is shown to be a key transcription factor in activating CD95 and caspase cascades. More importantly, we demonstrate that CD95 expression is higher in tumor cells than in normal cells in both MM cell lines and MM clinical samples, which suggests that CD95 could be a favorable target for plasma treatment as it could selectively inactivate myeloma tumor cells. Our results illustrate the molecular details of plasma induced myeloma cell apoptosis and it shows that gas plasma could be a potential tool for myeloma therapy in the future.
Effects of Atmospheric Pressure Plasma Jet with Floating Electrode on Murine Melanoma and Fibroblast Cells

Abstract:Atmospheric pressure cold plasma jets have been recently shown as a highly promising tool in certain cancer therapies. In this paper, an atmospheric pressure plasma jet (APPJ) with a one inner floating and two outer electrode configuration using helium gas for medical applications is developed. Subjected to a range of applied voltages with a frequency of 19.8 kHz at a fixed rate of gas flow (i.e., 3 l/min), electrical and optical characteristics of the APPJ are investigated. Compared with the device only with two outer electrodes, higher discharge current, longer jet, and more active species in the plasma plume at the same applied voltage together with the lower gas breakdown voltage can be achieved through embedding a floating inner electrode. Employing the APPJ with a floating electrode, the effects of identical plasma treatment time durations on murine melanoma cancer and normal fibroblast cells cultured in vitro are evaluated. The results of cell viability, cell apoptosis, and DNA damage detection show that the plasma can inactivate melanoma cells in a time-dependent manner from 10 s to 60 s compared with the control group (p < 0.05). However, for fibroblast cells compared with their control group, the plasma with treatment time from 30 s to 60 s can induce significant changes (p < 0.05), showing a less cytotoxic effect than that on melanoma cells at the same treatment time. The different basal reactive oxygen species level and antioxidant superoxide dismutase level of two kinds of cells may account for their different responses towards the identical plasma exposure.

Abstract:Small RNAs (sRNAs), particularly microRNAs (miRNAs), are functional molecules that modulate mRNA transcripts and have been implicated in the etiology of various types of cancer. Cold atmospheric plasma (CAP) is a physical technology widely used in the field of cancer treatment after exhibiting extensive lethality on cancer cells. However, few studies have reported the exact role of miRNAs in CAP-induced anti-cancer effects. The aim of the present study was to determine whether miRNAs are involved in CAP-induced cytotoxicity by using high-throughput sequencing. Our research demonstrated that 28 miRNAs were significantly changed (17 upregulated and 11downregulated) following 24 h of treatment with a room-temperature argon plasma jet for 90 s compared with that of the untreated group in human chronic myeloid leukemia K562 cells. GO enrichment analysis revealed that these target genes were related to cell organelles, protein binding, and single-organism processes. Furthermore, KEGG pathway analysis demonstrated that the target genes of differentially expressed miRNAs were primarily involved in the cAMP signaling pathway, AMPK signaling pathway, and phosphatidylinositol signaling system. Taken together, our study demonstrated that CAP treatment could significantly alter the small RNA expression profile of chronic myeloid leukemia cells and provide a novel theoretical insight for elucidating the molecular mechanisms in CAP biomedicine application.
