Abstract:The rising incidence of cutaneous malignancies presents ongoing therapeutic challenges. While surgery is the primary treatment modality, its limitations, including complications such as non-healing wounds and scarring, necessitate adjunctive therapies for optimal outcomes. But conventional methods like chemotherapy and phototherapy often fail to meet these needs. In response, microneedle patches (MNPs) have emerged, offering simultaneous delivery of multiple therapies directly to tumor sites. This review delves into three critical aspects of MNPs in treating skin malignancies: 1) what are the therapeutic combinations for different tumors? 2) what potential does MNP offer in the management of tumor-related symptoms? and 3) how to select suitable materials and design rational structures for enhanced therapeutic efficacy? Focusing on skin melanoma, basal cell carcinoma, and squamous cell carcinoma, we examine MNPs’ capacity for integrated multi-therapy, as well as their role in managing hemorrhage, pain, postoperative wounds, scars, and tumor indicators. Additionally, considerations for MNP storage, transportation, and side effects are discussed, emphasizing material selection and structural design. The review also discussed the ongoing dilemmas and future perspectives of MNPs in the clinical translation and application for cutaneous malignancies.
A novel multifunctional microneedle patch for synergistic photothermal- gas therapy against maxillofacial malignant melanoma and associated skin defects

Abstract:Considering the high recrudescence and the long-lasting unhealed large-sized wound that affect the aesthetics and cause dysfunction after resection of maxillofacial malignant skin tumors, a groundbreaking strategy is urgently needed. Photothermal therapy (PTT), which has become a complementary treatment of tumors, however, is powerless in tissue defect regeneration. Therefore, a novel multifunctional sodium nitroprusside and Fe 2+ ions loaded microneedles (SNP-Fe@MNs) platform was fabricated by accomplishing desirable NIR-responsive photothermal effect while burst releasing nitric oxide (NO) after the ultraviolet radiation for the ablation of melanoma. Moreover, the steady releasing of NO in the long term by the platform can exert its angiogenic effects via upregulating multiple related pathways to promote tissue regeneration. Thus, the therapeutic dilemma caused by postoperative maxillofacial skin malignancies could be conquered through promoting tumor cell apoptosis via synergistic PTT—gas therapy and subsequent regeneration process in one step. The bio-application of SNP-Fe@MNs could be further popularized based on its ideal bioactivity and appealing features as a strategy for synergistic therapy of other tumors occurred in skin.

Accelerated Cascade Melanoma Therapy using Enzyme-Nanozyme-Integrated Dissolvable Polymeric Microneedles

Abstract:Dissolvable polymeric microneedles (DPMNs) have emerged as a powerful technology for the localized treatment of diseases, such as melanoma. Herein, we fabricated a DPMN patch containing a potent enzyme-nanozyme composite that transforms the upregulated glucose consumption of cancerous cells into lethal reactive oxygen species via a cascade reaction accelerated by endogenous chloride ions and external near-infrared (NIR) irradiation. This was accomplished by combining glucose oxidase (Gox) with a NIR-responsive chloroperoxidase-like copper sulfide (CuS) nanozyme. In contrast with subcutaneous injection, the microneedle system highly localizes the treatment, enhancing nanomedicine uptake by the tumor and reducing its systemic exposure to the kidneys and spleen. NIR irradiation further controls the potency and toxicity of the formulation by thermally disabling Gox. In a mouse melanoma model, this unique combination of photothermal, starvation, and chemodynamic therapies resulted in complete tumor eradication (99.2 ± 0.8% reduction in tumor volume within 10 d) without producing signs of systemic toxicity. By comparison, other treatment combinations only resulted in a 42–76.5% reduction in tumor growth. The microneedle patch design is therefore not only highly potent but also with regulated toxicity and improved safety.

Abstract:Summary form only given. In recent years, atmospheric pressure nonequilibrium plasmas (APNPs) have been used in many biomedical applications such as bacteria, yeasts, fungi and algae inactivation, surface treatment for cell sheet engineering to controlled cell cultivation, promote blood coagulation and root canal of teeth sterilization. The APNPs can operate at room temperature and APNPs can form numerous short-lived but highly active chemical particles, such as reactive oxygen species, hydroxyl radical (OH), and other excited species. These active chemical particles are essential for various biological processes in cells and human body, reactive oxygen species are involved in initiation, progression and metastasis of cancers and nitric oxide (NO) can induce apoptosis, necrosis or protect cells from death, depending on the cell type and the amount, duration, and site of NO production. Therefore, it is important to study the impact of plasma on cancer cells. In this paper, human hepatocellular carcinoma cell (HepG2) is treated by a single electrode plasma jet device. Further investigation by using the flow cytometric analysis demonstrates that the plasma treatment increases the percentage of apoptotic cells being associated with cell cycle arrest at G2/M phase. Besides, the Reverse Transcriptase PCR assay shows that the Cyclin B1 and Cdc2 are decreased in transcription level after plasma treatment, while the expression of the p21 Cdk inhibitor, as well as tumor suppressor p53, is enhanced. On the other hand, the levels of certain pro-apoptotic or anti-apoptotic genes are checked, the experimental results suggests that the plasma induces apoptosis through shifting the Bax/Bcl-2 ratio to trigger HepG2 cell apoptosis. Results of the present investigation indicate that the plasma jet device may have potential therapeutic activity, such as sterilization of living tissue and tumor therapy of the postoperative treatment process.
A responsive microneedle system for efficient anti-melanoma by combining self-enhanced chemodynamic therapy with photothermal therapy

Abstract:Traditional therapies for melanoma suffer from many restrictions, that calls for the development of new treatments with high effectiveness and safety. Recently, employing transdermal microneedle (MN) systems for delivering therapeutic agents has obtained increasing interest as the efficient, minimally invasive, painless, and safe natures. Designing an “all-in-one” multifunctional MN system without complicated integration that can realize enhanced tumor therapy has presented huge promising for clinical application but is still a challenging strategy. Herein, a simple responsive MN system is developed for efficient anti-melanoma by combining self-enhanced chemodynamic therapy (CDT) and photothermal therapy (PTT). This MN system, constructed by biocompatible poly(vinylpyrrolidone) (PVP) as substrate and PVP-stabilized CuO2 nanoparticles (CuO2 NPs) as therapeutic agent, shows good skin-insertion capability and dissolving property that benefit in penetrating skin efficiently to deliver the therapeutic CuO2 NPs into melanoma site. The released CuO2 NPs from MN system can not only produce copper ion (Cu2+) and hydrogen peroxide (H2O2) in response to the acidic environment for catalytic hydroxyl radical (OH) generation and thus achieve the H2O2 self-supplying CDT, but also can be utilized as a glutathione (GSH)-scavenging agent for further enhancing CDT. Moreover, owing to the loading of CuO2 NPs, such MN system exhibits excellent near-infrared (NIR) photothermal property that can induce local hyperthermia at the melanoma area for PTT. In result, the designed MN system realizes the efficient tumor inhibition with minimal side effects in vivo by the combination therapy, offering a safe and effective melanoma treatment option for patients.

Abstract:Melanoma, the most aggressive form of skin cancer, is characterized by an increasing incidence rate. However, conventional treatment methods such as surgery, chemotherapy, radiotherapy, and immunotherapy have limitations that hinder their widespread application. In this study, we aim to develop a platinum nanozyme (PtNP) co-loaded reactive oxygen species (ROS)-responsive prodrug integrated with dissolvable microneedle for chemotherapy and photodynamic therapy of melanoma. The utilization of microneedle can significantly enhance the efficiency of transdermal drug delivery while improving treatment efficacy and minimizing toxic side effects. The nanodrug system incorporates a prodrug composed of chemotherapeutic agent, photosensitizer, and ROS-responsive chemical bond. Upon laser irradiation, it generates ROS for effective photodynamic therapy while precisely controlling the release behavior of camptothecin (CPT) within the prodrug formulation. Furthermore, PtNP in the nanodrug exhibits nanozyme-like activity by catalyzing the decomposition of hydrogen peroxide into oxygen to overcome hypoxia-related challenges and enhance the effectiveness of photodynamic therapy. The integration of the nanodrug complex with dissolvable microneedle presents a synergistic approach for the combined delivery of chemotherapy and photodynamic therapy to melanoma patients, offering novel strategies and avenues for clinical treatment.

Multiple RONS-Loaded Plasma-Activated Ice Microneedle Patches for Transdermal Treatment of Psoriasis

Abstract:Cold atmospheric plasma (CAP) is a fledgling therapeutic technique for psoriasis treatment with noninvasiveness, but clinical adoption has been stifled by the insufficient production and delivery of plasma-generated reactive oxygen and nitrogen species (RONS). Herein, patches of air-discharge plasma-activated ice microneedles (PA-IMNs) loaded with multiple RONS are designed for local transdermal delivery to treat psoriasis as an alternative to direct CAP irradiation treatment. By mixing two RONS generated by the air-discharge plasma in the NOx mode and O3 mode, abundant high-valence RONS are produced and incorporated into PA-IMNs via complex gas-gas and gas-liquid reactions. The PA-IMNs abrogate keratinocyte overproliferation by inducing reactive oxygen species (ROS)-mediated loss of the mitochondrial membrane potential and apoptosis of keratinocytes. The in vivo transdermal treatment confirms that PA-IMNs produce significant anti-inflammatory and therapeutic actions for imiquimod (IMQ)-induced psoriasis-like dermatitis in mice by inhibiting the release of associated inflammatory factors while showing no evident systemic toxicity. Therefore, PA-IMNs have a large potential in transdermal delivery platforms as they overcome the limitations of using CAP directly in the clinical treatment of psoriasis.
Living photosynthetic microneedle patches for in situ oxygenation and postsurgical melanoma therapy

Abstract:Surgical excision remains the principal treatment for melanoma, while tumor recurrence and delayed wound healing often occur due to the residual tumor cells and hypoxic microenvironment in the postoperative skin wounds. Herein, we present a living photosynthetic microneedle (MN) patch (namely MA/CM@MN) loaded with microalgae (MA) and cuttlefish melanin (CM) for postsurgical melanoma therapy and skin wound healing. Benefiting from the oxygenic photosynthesis of the alive MA in the MN base, the MA/CM@MN can generate oxygen under light exposure, thus facilitating skin cell proliferation and protecting cells against hypoxia-induced cell death. In addition, with CM nanoparticles embedded in the MN tips, the MA/CM@MN can be effectively heated up under near-infrared (NIR) irradiation, contributing to a strong tumor killing efficacy on melanoma cells in vitro. Further experiments demonstrate that the NIR-irradiated MA/CM@MN effectively prevents local tumor recurrence and simultaneously promotes the healing of tumor-induced wounds after incomplete tumor resection in melanoma-bearing mice, probably because the MA/CM@MN can inhibit tumor cell proliferation, stimulate tumor cell apoptosis, and mitigate tissue hypoxia in light. These results indicate that the living photosynthetic MN patch offers an effective therapeutic strategy for postoperative cancer therapy and wound healing applications.

Abstract:High aggressiveness and recurrence of melanoma tumors require multiple systemic drug administrations, causing discomfort and severe side effects to the patients. Topical treatment strategies that provide repetitively controllable and precise drug administrations will greatly improve treatment effects. Methods: In this study, a spatiotemporally controlled pulsatile release system, which combined dissolving microneedles (DMNs) and thermal-sensitive solid lipid nanoparticles (SLNs), was constructed to realize multiple doses of dual-modal chemo-photothermal therapy in a single administration. Paclitaxel (PTX) and photothermal agent IR-780 were encapsulated into SLNs and were concentrated in the tips of DMNs (PTX/IR-780 SLNs @DMNs). Equipped with several needles, the DMN patch could be directly inserted into the tumor site and provide a stable "Zone accumulation" to constrain the PTX/IR-780 SLNs at the tumor site with uniform distribution. Results: In vitro experiments showed that after irradiation with near-infrared light, the PTX/IR-780 SLNs gradually underwent phase transition, thereby accelerating the release of PTX. When irradiation was switched off, the PTX/IR-780 SLNs cooled to re-solidify with limited drug release. Compared with intravenous and intratumoral injections, very few SLNs from PTX/IR-780 SLNs @DMNs were distributed into other organs, resulting in enhanced bioavailability at the tumor site and good safety. In vivo analysis revealed that PTX/IR-780 SLNs @DMNs exhibited significant anti-tumor efficacy. In particular, the primary tumor was completely eradicated with a curable rate of 100% in 30 days and the highest survival rate of 66.67% after 100 days of treatment. Conclusion: Herein, we developed a DMN system with a unique spatiotemporally controlled pulsatile release feature that provides a user-friendly and low-toxicity treatment route for patients who need long-term and repeat treatments.

Abstract:Melanoma is the most lethal malignancy in skin cancer and may occur at any site and express melanocytes. Due to malignant melanoma's invasion and migration nature, conventional therapies make it challenging to remove the whole tumor tissue while undertaking the high risks of tumor recurrence. Regarding the emerging targeted therapies and immunotherapy, drug resistance and low immunotherapeutic activity remain significant challenges. It is thus becoming urgently important to develop alternative strategies for melanoma therapy. Herein, we develop a novel bifunctional protein-based photo-thermal bioplaster (PPTB) for non-invasive tumor therapy and skin tissue regeneration. The complexation of adhesive protein and gold nanorods (GNRs) endow the obtained PPTB with good biocompatibility, controllable near-infrared (NIR) light-mediated adhesion performance, and high photo-thermal performance. Therefore, the PPTB bioagent facilitates skin adhesion and effectively transfers heat from skin to tumor. This behavior endows PPTB capability to eradicate skin tumors conveniently. Abstract:A dually‐layered "microneedle rocket" is designed for melanoma treatment. The upper layer consists of mesoporous silica nanoparticles covalently bonded with a photosensitizer (PcNP) alongside a targeted therapy drugs (TRA). The lower layer comprises a hydrogel matrix with controllable cross‐linking density encapsulating collagenase (CLG) for remodeling the extracellular matrix (ECM). The rocket‐like microneedles achieve deep penetration of nanoparticles (PcNP/TRA) for efficacious photodynamic therapy and targeted therapy. Photodynamic therapy (PDT) proves highly effective in addressing melanoma, and its synergy with targeted therapy offers a promising avenue in tumor treatment. However, the therapeutic outcome is largely impeded by the challenge of current photosensitizers and targeted drugs in reaching deep tumor tissues. Herein, a dually‐layered "microneedle rocket" termed PcNP/TRA‐HA‐Tyr/CLG‐MN is designed. The upper layer of the microneedle (MN) is composed of photodynamically active mesoporous silica nanoparticles, featuring photosensitizers covalently bonded to them. Within the mesopores of these nanoparticles lies trametinib (TRA), a compound that specifically targets the hyperactive MEK pathway present in melanoma cells. The lower layer is composed of an enzyme‐mediated hyaluronic acid‐tyramine hydrogel (HA‐Tyr/CLG) with collagenase (CLG), serving as rocket thrusters for remodeling the extracellular matrix (ECM) in tumor microenvironment. Among the three types of MNs prepared, the PcNP/TRA‐HA‐Tyr(II)/CLG‐MN exhibits the deepest penetration in tumor tissues and the longest retention time in vivo. Notably, the administrations of PcNP/TRA‐HA‐Tyr(II)/CLG‐MN alongside light irradiation significantly suppress the growth of A375‐xenografted tumors in mice. Together, the strategy of combining mesoporous silica nanoparticles, enzymatically cross‐linked hydrogels and CLG‐mediated ECM remodeling enables deep drug penetration and efficacious combination treatment against melanoma, showcasing significant potential in the realm of cancer nanomedicine.

Abstract:Melanoma is one of the most significant and dangerous superficial skin tumors with a high fatality rate, thanks to its high invasion rate, drug resistance and frequent metastasis properties. Unfortunately, researchers for decades have demonstrated that the outcome of using conventional therapies like chemotherapy and immunotherapy with normal drug delivery routes, such as an oral route to treat melanoma was not satisfactory. The severe adverse effects, slow drug delivery efficiency and low drug accumulation at targeted malignancy sites all lead to poor anti-cancer efficacy and terrible treatment experience. As a novel transdermal drug delivery system, microneedles (MNs) have emerged as an effective solution to help improve the low cure rate of melanoma. The excellent characteristics of MNs make it easy to penetrate the stratum corneum (SC) and then locally deliver the drug towards the lesion without drug leakage to mitigate the occurrence of side effects and increase the drug accumulation. Therefore, loading chemotherapeutic drugs or immunotherapy drugs in MNs can address the problems mentioned above, and MNs play a crucial role in improving the curative effect of conventional treatment methods. Notably, novel tumor therapies like photothermal therapy (PTT), photodynamic therapy (PDT) and chemodynamic therapy (CDT) have shown good application prospects in the treatment of melanoma, and MNs provide a valid platform for the combination of conventional therapies and novel therapies by encompassing different therapeutic materials in the matrix of MNs. The synergistic effect of multiple therapies can enhance the therapeutic efficacy compared to single therapies, showing great potential in melanoma treatment. Dissolving MNs have been the most commonly used microneedles in the treatment of melanoma in recent years, mainly because of their simple fabrication procedure and enough drug loading. So, considering the increasing use of dissolving MNs, this review collects research studies published in the last four years (2020-2024) that have rarely been included in other reviews to update the progress of applications of dissolving MNs in anti-melanoma treatment, especially in synergistic therapies. This review also presents current design and fabrication methods of dissolving MNs; the limitations of microneedle technology in the treatment of melanoma are comprehensively discussed. This review can provide valuable guidance for their future development.

Abstract:Melanoma is a malignancy of the skin that is resistant to conventional treatment, necessitating the development of effective and safe new therapies. The percutaneous microneedle (MN) system has garnered increasing interest as a viable treatment option due to its high efficacy, minimal invasiveness, painlessness, and secure benefits. In this investigation, a sensitive MN system with multiple functions was created to combat melanoma effectively. This MN system utilized polyvinylpyrrolidone (PVP) as microneedle substrates and biocompatibility panax notoginseng polysaccharide (PNPS) as microneedle tips, which encapsulated PVP-stabilized CuO2 nanoparticles as a therapeutic agent and disulfiram-containing F127 micelles to enhance the tumor treatment effect. The MN system had sufficient mechanical properties to pierce the skin, and the excellent water solubility of PNPS brought high-speed dissolution properties under the bio conditions, allowing the MNs to effectively penetrate the skin and deliver the CuO2 nanoparticles as well as the drug-loaded micelles to the melanoma site. CuO2 nanoparticles released by the MN system generated Cu2+ and H2O2 in the tumor acidic environment to achieve self-supply of hydrogen peroxide to chemodynamic therapy (CDT). In addition, Cu2+ was chelated with disulfiram to produce CuET, which killed tumor cells. And the MN system had excellent near-infrared (NIR) photothermal properties due to the loading of CuO2 nanoparticles and induced localized thermotherapy in the melanoma region to further inhibit tumor growth. Thus, the designed MN system accomplished effective tumor suppression and minimal side effects in vivo via combined therapy, offering patients a safe and effective option for melanoma treatment.
Comparative Anthelmintic Efficacy of Caesalpinia Crista, Nigella Sativa and Oxfendazole in Broilers with Experimentally Induced Ascaridia Galli Infection -

Abstract:The study was conducted to explore the effect of two herbal drugs, Caesalpinia crista and Nigella sativa in comparison with Oxfendazole in broilers experimentally induced with Ascaridia galli infection .A total of 130 day old broiler chicks were divided into five groups i.e. A, B, C, D and E each group having 26 birds. Group E was kept as negative control, while all other groups were induced with experimental Ascardia galli infection on 15 day. Group A was kept as positive control and groups B and C were treated with Caesalpinia crista and Nigella sativa as methanolic extract orally once @50mg/kg body weight and group D was medicated with Oxfendazole @10mg/kg body weight orally once on 25 day post infection. On 25th day post infection before treatment 4 birds from each group were slaughtered, and worm count was done. The efficacy was confirmed by counting the worm, slaughtering all the bird on 30 day post infection. The mean worm count was significantly different (p<0.05) between the treated and non treated groups. The efficacy of the plant extracts were less as compare to Oxfendazole. The efficacy of Caesalpinia crista and Nigella sativa was 70.83% and 57.5% respectively while the efficacy of Oxfendazole was 90.83%.In vitro treatment at 37±1°C revealed that a serial dose of the plant extract caused dose-dependent vermicidal effect on the worm (significant efficacy at P<0.05), except at the lowest dose (i.e. 5 mg/mL). The efficacy of drugs was calculated on the basis of reduction in total worm count after treatment.

Abstract:Malignant melanoma is a challenging problem worldwide, because the remaining tumor cells and extensive skin defects following surgical resection are difficult to treat. Biomaterial-mediated immunotherapy has emerged as a superior strategy for anti-tumor applications in recent years. Herein, a unique double-layer MNP was developed to address the problem of malignant melanoma. Hydroxyapatite (HAP) and short-chain peptides from tumor cells were self-assembled to prepare the bioinspired nano-vaccine, and then they were loaded onto the microneedle tips of methacrylated gelatin (GelMA)-based MNP. The products (dubbed HVMN) demonstrated relatively good biocompatibility and immune activity, inhibiting the proliferation and inducing apoptosis of malignant melanoma in a B16 cell-bearing model of C57BL/6 mice, and promoting skin tissue regeneration in a full thickness skin defect model of SD rats in 15 days. The putative molecular pathways were examined preliminarily. In conclusion, this research will develop a competitive microneedle patch with dual anti-tumor and pro-regenerative properties for the postoperative treatment of malignant melanoma.

Abstract:Photodynamic therapy (PDT) is a potential treatment strategy for melanoma. As a second-generation photosensitizer, Zinc phthalocyanine (ZnPc) has many advantages for anti-tumor PDTs, such as strong absorption in the red and near infrared regions, high photo and chemical stability, etc. However, ZnPc has a poor water solubility and is apt to aggregate due to the π-π interaction between molecules, which limits its applications. In this study, various solvents and surfactants were screened for dissolving ZnPc and preparing ZnPc@SDC-TPGS micelle and thermosensitive in situ gel. After the cytotoxic effects of thermosensitive gels on PDT were tested, the antitumor effects on PDT of them in mice by intratumoral injection were evaluated, including body weight, and tumor weight, volume and morphology. The cell death pathway and the relationship of reactive oxygen species yield with apoptotic rate of tumor cells induced by ZnPc in situ gel were investigated. The results were that N-methyl-pyrrolidone (NMP) mixed with 2% SDC and aqueous solution containing 2% TPGS and 2% SDC were used to synthesize ZnPc@SDC-TPGS micelle and the thermosensitive in situ gel. The cytotoxic effects of thermosensitive gels showed good tumor suppression of ZnPc@SDC-TPGS in situ gel and no toxicity of the blank gel. Intratumoral injection in situ gel containing 3μg ZnPc under irradiation demonstrated good tumor inhibition in mice with melanoma. Apoptosis has been established as the primary pathway of cell death, and the production of reactive oxygen species (ROS) plays a crucial role in cellular apoptosis induced by ZnPc@SDC-TPGS in situ gel. In conclusion, the intra-tumoral injection of ZnPc@SDC-TPGS thermosensitive in situ gel provides a promising local treatment option for melanoma.

Abstract:Melanoma has emerged as a significant threat to human life and health. Microneedle (MN)‐mediated transdermal drug delivery (TDD) has garnered attention in melanoma treatment for bypassing the first‐pass effect. However, the propensity of melanoma to metastasize presents substantial challenges for MN mediated local treatment. Developing systemic therapies, such as immunotherapy in combination with TDD, is crucial for achieving effective melanoma treatment. Herein, a polyvinyl alcohol (PVA) MN‐mediated multifunctional TDD system, designated MN@PDA@1‐MT/CUR/DOX@HA (MN@PMCDH), was developed for synergetic chemotherapy/photothermal/immunotherapy of melanoma. PMCDH nanomedicines penetrate deep skin layers through MNs, accumulate at tumor sites guided by hyaluronic acid (HA), and selectively release drugs in response to the acidic tumor microenvironment and near‐infrared (NIR) stimulation. Released curcumin (CUR) significantly enhances the efficacy of photothermal therapy (PTT) and chemotherapy, as well as improves the induction of immunogenic cell death (ICD) by increasing melanoma sensitivity to polydopamine (PDA)‐mediated photothermal effects and doxorubicin (DOX). Moreover, the incorporation of 1‐methyltryptophan (1‐MT) to reverse the tumor immunosuppressive microenvironment can further enhance the effects of immunotherapy. In vitro studies revealed that the MN@PMCDH system can effectively induce ICD and inhibit tumor cell growth. Additionally, remarkable deep tumor cell inhibition effects are also achieved in 3D tumor models.

Abstract:The multifunctional porous silicon‐based nanozyme complex (CuGQD/PdNPs@PSi) possesses synergistic and hyperthermia‐enhanced nanocatalytic activities. The unique confinement effect of PSi host endows CuGQD/PdNPs@PSi with superior mimic‐peroxidase and glutathione oxidase activities. Moreover, photothermal stimulation can further improve the mimic enzymatic performance of CuGQD/PdNPs@PSi, which can induce efficient ferroptosis pathway. Therefore, the microneedles integrated with CuGQD/PdNPs@PSi can achieve satisfactory ferroptosis‐based therapy for melanoma. Superficial melanoma is the deadliest form of skin cancer without desirable clinically therapeutic options. Nanozymes, artificial nanomaterials with physicochemical performance and enzyme catalytic properties, have attracted considerable attention for antitumor therapy. However, the therapeutic efficiency of nanozymes is vulnerable to the tumor microenvironment (TME) and delivery process. Herein, a microneedle (MN) patch that integrates porous silicon (PSi) loaded with dual nanozymes is devised to bidirectionally regulate TME and accurately deliver nanocomplex to initiate ferroptosis for melanoma treatment. Benefitting from the channel confinement effect of PSi, the copper‐doped graphene quantum dots and palladium nanoparticles coloaded PSi (CuGQD/PdNPs@PSi) exhibit synergistic effect with enhanced mimicking peroxidase and glutathione oxidase activities, which are ≈2–3‐fold higher than those of monoconfined nanozyme or nonconfined nanozyme complexes. Additionally, the synergistic catalytic performance of CuGQD/PdNPs@PSi can be improved via photostimuli hyperthermia. The CuGQD/PdNPs@PSi can induce ferroptosis manifested by upregulation of lipid peroxides and inactivation of glutathione peroxidase 4. Furthermore, loading of nanocomplexes into MNs for administration resulted in a satisfactory melanoma growth inhibition of 98.8% within 14 days. Therefore, MNs encapsulated with CuGQD/PdNPs@PSi can provide a potentially nanocatalytic strategy for ferroptosis‐inducing tumor treatment while also meeting the medical needs of eradicating superficial tumors.

Abstract:Melanoma is a common malignant skin tumor with highly invasive features and a high metastasis rate that can be difficult to treat clinically. Large‐scale resection of primary cutaneous melanoma is often used to avoid postoperative recurrence. For advanced patients, radiotherapy, targeted therapy and immunotherapy are often needed. Low‐temperature plasma has been proved to have significant antitumor effects on a variety of cancer cell lines cultured in vitro . The main limitation of direct low‐temperature plasma treatment is that it has weak penetration ability and can only treat superficial lesions. In recent years, research on low‐temperature plasma‐activated solution has revealed that it also have good antitumor effects and low‐temperature plasma penetration depth problems can be solved by local injection. The present study revealed that low‐temperature plasma‐activated phosphate buffer solution exhibited good antitumor effects and biosafety against melanoma in vitro and in vivo . It demonstrated that low‐temperature plasma‐activated solution has antitumor effects due to its regulation of intracellular redox, destruction of mitochondrial function and DNA damage. In vivo experiments demonstrated that treatment with low‐temperature plasma‐activated solution not only exhibited antitumor effects but also caused no significant damage to hematopoietic function or liver and kidney functions in mice. All these results demonstrated that low‐temperature plasma‐activated solution represent a promising antitumor treatment strategy.
