Abstract:Combined chemodynamic/photothermal therapy has great potential in tumor treatment. However, the presence of excessive glutathione (GSH) in the tumor microenvironment (TME) can attenuate its therapeutic effect, and other components in the TME have not been fully utilized as well. In this article, we designed a noble metal nanozyme called PdCu@BSA, which can be used for the combined chemodynamic therapy (CDT) and photothermal therapy (PTT) of tumor. In detail, PdCu@BSA has three different types of enzyme-like activities. Its catalase (CAT)-like activity can degrade extra H2O2 in the TME to create O2 and relieve the hypoxic situation. The glutathione oxidase (GSHox)-like activity can consume high level of GSH in the TME to reduce the consumption of reactive oxygen species (ROS). Peroxidase (POD)-like activity catalyzes H2O2 to form strong oxidized ·OH. The above enzyme-like activities enhance the effectiveness of CDT. Besides, PdCu@BSA has good photothermal effect and can be used for PTT when exposed to 1064 nm laser. Therefore, based on multiple enzyme-like activities and photothermal effects, PdCu@BSA can be employed for synergistic tumor therapy, resulting in good therapeutic outcome.

Abstract:In vivo chemical reactions activated by the tumor microenvironment (TME) are particularly promising for antitumor treatments. The amorphous form of CuSe and the catalytic effect of Au benefit the OH generation, and the photo-induced electron‒hole separation of the Janus NPs produces additional OH. The plasmonic electrons of Au facilitate the conversion from Cu to Cu. Both CuSe and Au contributes to the efficient photothermal conversion, further promoting the reactions. As a result, the HO utilization rate is largely increased, and remarkable generation of reactive oxygen species is achieved by cell endogenous HOin vitro/vivo. A competent tumor inhibition effect is afforded, with high-contrast multimodal imaging. This work opens up the route synergistically integrating photothermal therapy with chemodynamic therapy and photocatalytic therapy into tri-combination antitumor therapy, simply by heterojunction of semiconductor and noble metal.

Abstract:Developing nanomedicines with high-performance is a promising therapeutic strategy for cancer treatment. Herein, a multifunctional nanoplatform (CuS@COFs-BSA-FA/DOX) based on copper sulfide nanoparticles engineered covalent organic frameworks (CuS@COFs) has been established for synergistic photothermal therapy (PTT), chemotherapy and chemodynamic therapy (CDT). The high photothermal effect and excellent Fenton-like catalytic activity enabled CuS@COFs to generate a new minimally invasive PTT/CDT synergistic therapy. Moreover, with the pH-dependent fluorescence behavior and the inherited mesoporous structure, the nanocomposites can also be utilized as fluorescent probes for cancer cell imaging and drug-delivery carriers to encapsulate the chemotherapeutic agent, DOX. The densely coated PEI and BSA-FA layers on the surface of CuS@COFs not only increased the cancer-targeting efficiency but also prevented the nonspecific release of DOX from CuS@COFs. The acid tumor microenvironment and near-infrared light triggered the release of DOX for chemotherapy, which simultaneously augmented the CDT efficiency of CuS@COFs. Notably, the localized hyperthermia induced by PTT can further improve the CDT efficiency of the nanoplatform, leading to a synergistic PTT/chemotherapy/CDT effect. The nanoplatform possessed the capability of cancer cell-targeted imaging and achieved better therapeutic efficacy with negligible systemic toxicity both in vitro and in vivo. Our work opens up a powerful avenue for developing multifunctional COFs-based theranostic platform and shows strong potential in practical applications.

Abstract:The clinical applications of the photothermal therapy (PTT) and photodynamic therapy (PDT) are restricted by the limited tissue penetration depth of excitation light, while the enzyme catalysis therapy is hindered on account of the low treatment efficiency. Herein, we modify artificial nanozyme Pt nanoparticles on the surface of CuCo 2 S 4 nanoparticles to prepare ultrasmall copper-based ternary bimetallic chalcogenides CuCo 2 S 4 -Pt-PEG nanocomposites for PTT/PDT/enzyme catalysis synergistic therapy. The as-fabricated CuCo 2 S 4 -Pt-PEG nanocomposites display peroxidase (POD)-like and catalase (CAT)-like activities, which can catalyze hydrogen peroxide to emerge highly toxic hydroxyl radicals and oxygen, respectively. The integration of Pt nanozyme significantly improves the POD-like and CAT-like activity of CuCo 2 S 4 −PEG that can produce more hydroxyl radicals and oxygen to improve the effects of PDT. Furthermore, the nanocomposites exhibit outstanding photothermal properties derived from the enhanced photothermal conversion efficiency ( η = 78.46 %) exposed to 1064 nm NIR-II light irradiation with a low power density (0.8 W cm −2 ). In particular, the photothermal performance of CuCo 2 S 4 -Pt-PEG significantly enhances the enzyme-like activity. Additionally, the T 1 -weighted magnetic resonance and photoacoustic imaging endow CuCo 2 S 4 -Pt-PEG with the advantages of guiding the treatment process. The synergistic treatment demonstrates significant anticancer treatment efficiency while also elicits immune response that suppressed metastasis. This study could further promote the application of ternary bimetallic chalcogenides in cancer diagnosis and treatment.

Abstract:Abstract The efficient way to increase the therapeutic efficacy of nanomedicines is by encouraging the penetration and enhancing the retention of nanoparticles at the tumor site. However, it is a serious dilemma that small nanoparticles can penetrate deep into the tumor tissue but easily be cleared into the surrounding tissues. In order to solve this dilemma, a smart nanosystem is created to address this problem, ensuring both the effective penetration of tiny nanoparticles (NPs) and their appropriate retention at the tumor site. CuS NPs that is modified with peptides are prepared facilely, and can aggregate in situ through the intermolecular crosslinking reaction catalyzed by the transglutaminase (TGase) abundantly expressed at the tumor site, resulting in an outstanding photothermal effect for tumor therapy. Upon NIR irradiation, the photothermal effect of CuS‐K and CuS‐Q induced the disintegration of liposomes and prompted the release of CuS‐K, CuS‐Q, and indocyanine green (ICG). Simultaneously, CuS‐K and CuS‐Q aggregated under the catalysis of TGase after being internalized by tumor cells to enhance photothermal therapy. The current study provides valuable inspiration to design nanomedicines with prolonged circulation time in the blood system, better penetration, and retention at the tumor site, and multimodal tumor therapy to achieve the desired therapeutic efficacy.

Abstract:Multifunctional nanotheranostic systems with both therapeutic and imaging functions are highly desired for the development of more effective and less toxic anti-tumor drugs. Herein, a simple but effective method is reported to fabricate a novel PCN-CuS-FA-ICG-based nanoplatform for dual-modal imaging-guided synergistic photothermal/photodynamic therapy. Porphyrinic metal-organic frameworks with CuS NPs are obtained in aqueous solution via a simple post-synthesis strategy. Furthermore, to obtain a more effective therapy, indocyanine green (ICG) was incorporated into the multifunctional theranostic platform to promote the photothermal therapeutic effect. The as-prepared PCN-CuS-FA-ICG not only exhibits an excellent O-1(2) generation efficiency under 650 nm irradiation to achieve remarkable photodynamic cell killing, but also presents outstanding photothermal conversion under 808 nm irradiation to destroy tumor tissues by hyperthermia. In particular, the nanotherapeutic agent realized fluorescence and thermal imaging dual-modal imaging-guided cancer treatment. Meanwhile, in vivo experiments confirmed the evident accumulation of nanoparticles (NPs) at local tumors, and tumor growth was inhibited obviously via synergistic photothermal/photodynamic therapy with negligible side effects.

Abstract:Recent advancements in cancer treatment have underscored the inadequacy of conventional monotherapies in addressing complex malignant tumors. Consequently, there is a growing interest in synergistic therapies capable of overcoming the limitations of monotherapies, leading to more personalized and effective approaches. Among these, the combination of photothermal therapy (PTT) and chemotherapy has emerged as a promising avenue for tumor management. In this study, we present a novel approach utilizing thermoresponsive mesoporous silica nanoparticles (MSN) as a delivery system for the chemotherapeutic drug doxorubicin. By incorporating photothermal agent copper sulfide (CuS) nanoparticles into the MSN, the resulting composite material exhibits potent photothermal properties. Furthermore, the integration of an upper critical solution temperature (UCST) polymer within the silica outer layer serves as a "gatekeeper", enabling precise control over drug release kinetics. This innovative nanomaterial effectively merges thermoresponsive behavior with PTT, thereby minimizing the collateral damage associated with traditional chemotherapy on healthy tissues. Moreover, in both in vitro studies using mouse breast carcinoma cells (4 T1) and in vivo experiments utilizing a 4 T1 tumor-bearing mouse model, our nanomaterials demonstrated synergistic effects, enhancing the anti-tumor efficacy of combined PTT and chemotherapy. With its remarkable photothermal conversion efficiency, robust stability, and biocompatibility, the UCST-responsive nanoplatform holds immense potential for clinical applications.

Abstract:The novel nanocomposites were constructed through encapsulate Au nanoparticles and Ru nanoparticles into dendritic mesoporous silica (DMSN-Au-Ru NPs), which exhibit improved therapy effect due to cascade catalytic ability for synergistic therapy of cancer. Au nanoparticles with glucose oxidase-like property could catalyze the oxidation of glucose to produce H2O2, while Ru nanoparticles could decompose H2O2 and produce toxic 1O2 for improved photodynamic therapy (PDT). In addition, the nanocomposites have good photothermal performances under the irradiation of near infrared (NIR) light. Both in vitro and in vivo experiments show that the nanocomposites have good therapeutic effects due to the cascade catalytic effect and synergistic effect. These findings provides an effective way to design new generation of nanodrugs for highly efficient cancer treatment.

Abstract:Multifunctional nanoparticles are of significant importance for synergistic multimodal antitumor activity. Herein, zinc oxide (ZnO) was used as pH-sensitive nanoparticles for loading the chemotherapy agent doxorubicin (DOX) and the photosensitizer agent indocyanine green (ICG), and biocompatible low-molecular-weight heparin (LMHP) was used as the gatekeepers for synergistic photothermal therapy/photodynamic therapy/chemotherapy/immunotherapy. ZnO was decomposed into cytotoxic Zn2+ ions, leading to a tumor-specific release of ICG and DOX. ZnO simultaneously produced oxygen (O2) and reactive oxygen species (ROS) for photodynamic therapy (PDT). The released ICG under laser irradiation produced ROS for PDT and raised the tumor temperature for photothermal therapy (PTT). The released DOX directly caused tumor cell death for chemotherapy. Both DOX and ICG also induced immunogenic cell death (ICD) for immunotherapy. The in vivo and in vitro results presented a superior inhibition of tumor progression, metastasis and recurrence. Therefore, this study could provide an efficient approach for designing multifunctional nanoparticles for synergistic multimodal antitumor therapy.
Novel theranostic nanoagent based on CuMo2S3-PEG-Gd for MRI-guided photothermal/photodynamic/chemodynamic therapy

Abstract:Cancer is a severe disease, which have troubled human being for a long time. The development of nanotechnology has provided a new way for cancer treatment. It is a promising strategy to integrate imaging and therapeutic functions into one single nanoplatform to achieve efficient combination of diagnosis and treatment. Herein, we exploited novel CuMo2S3-PEG-Gd nanocomposites (NCs) for magnetic resonance imaging (MRI), guiding the photothermal therapy (PTT)/photodynamic therapy (PDT)/chemodynamic therapy (CDT). The experimental results showed that CuMo2S3-PEG-Gd NCs have a high photothermal conversion efficiency (40.6%), excellent biocompatibility and good biosecurity. The CuMo2S3-PEG-Gd NCs exhibited a clear MRI performance for tumor due to connecting Gd, which can guide in vivo therapy to improve the therapeutic effect. Moreover, both in vitro and in vivo therapeutic results of CuMo2S3-PEG-Gd NCs exhibited that the PTT/PDT/CDT achieved a remarkably synergistic effect, which could efficiently inhibit the tumor growth. Thus, CuMo2S3-PEG-Gd NCs, which integrated imaging with multiple therapies, have a good potential as theranostic agent for tumor.Graphical abstract

Abstract:Designing a multifunctional nanoplatform that combines multiple treatments has emerged as an innovative cancer treatment strategy. A simple and clear route is put forward to develop Cu2+-doped zinc phosphate coated prussian blue nanoparticles (designated as PB@Cu2+/ZnP NPs) integrating tri-modal therapy (chemo, chemodynamic and photothermal therapy) for maximizing anti-tumor efficacy. The obtained PB@Cu2+/ZnP NPs possess drug loading capacity due to the mesoporous structure present in the Cu2+-doped ZnP shell. In addition, the Cu2+-doped ZnP shell can gradually degrade in response to the mildly acidic tumor microenvironment to release DOX and Cu2+, where the released drug plays the role of chemotherapy agent and the Cu2+ can react with intracellular glutathione to achieve a Cu-mediated Fenton-like reaction for chemodynamic therapy. Moreover, under laser irradiation, the heat garnered by the photothermal conversion of PB can be applied for photothermal therapy and enhance the generation of toxic ˙OH as well as the amount of DOX released, further boosting chemo- and chemodynamic therapy to realize a combined therapy. Importantly, the PB@Cu2+/ZnP NPs effectively limit the growth of tumors via the coordinated action of chemo/chemodynamic/photothermal therapy and no noticeable systematic toxicity can be found in mice. Taken together, the PB@Cu2+/ZnP NPs can act as a prospective therapeutic nanoplatform for multi-modal therapy of tumors.

A Three-in-one ZIFs-Derived CuCo(O)/GOx@PCNs Hybrid Cascade Nanozyme for Immunotherapy/Enhanced Starvation/Photothermal Therapy

Abstract:Glucose oxidase (GOx) is regarded as an ideal endogenous natural enzyme for tumor starvation therapy and photothermal therapy (PTT) is a promising strategy for the ablation of primary tumor. In this work, Cu-doped cobalt oxide and porous carbon nanocomposites (CuCo(O)@PCNs) were synthesized from double-layered and GOx was loaded in the porous carbon to form a CuCo(O)/ hybrid nanozyme. CuCo(O) was characterized as the Cu0.3Co2.7O4 phase through X-ray diffraction analysis and it can react with H2O2 to generate O2 and alleviate tumor hypoxia, resulting in the recovered enzymatic activity of GOx and the enhanced starvation therapy. The porous nanocarbon can ablate the primary tumor because of its high photothermal conversion efficiency of 40.04%. The three-in-one functions of oxygen supply, glucose consumption, and photothermal conversion were realized in the ZIFs-derived CuCo(O)/ nanozyme and the starvation therapy effect was improved by PTT and oxygen supplement. Furthermore, the inhibition effect of CuCo(O)/ on metastatic tumor is similar to combined therapy of the nanozyme and the immune checkpoint-blocking antibody, α-PD-1. The related antitumor immune mechanism was studied through the analysis of immune-related proinflammatory cytokines and the activated T cells. Abstract:Nanozymes are nanomaterials with enzyme-like properties that have attracted significant interest owing to their high stability, easy preparation, and tunable catalytic properties, especially in the field of cancer therapy. However, the unfavorable catalytic effects of nanozymes in the acidic tumor microenvironment have limited their applications. Herein, we developed a biomimetic erythrocyte membrane-camouflaged ultrasmall black phosphorus quantum dots (BPQDs) nanozymes that simultaneously exhibited an exceptional near-infrared (NIR) photothermal property and dramatically photothermal-enhanced glucose oxidase (GOx)-like activity in the acidic tumor microenvironment. We demonstrated the engineered BPQDs gave a photothermal conversion efficiency of 28.9% that could rapidly heat the tumor up to 50 °C while effectively localized into tumors via homing peptide iRGD leading after intravenously injection. Meanwhile, the significantly enhanced GOx-like activity of BPQDs under NIR irradiation was capable of catalytical generating massive toxic reactive oxygen species via using cellular glucose. By combining the intrinsic photothermal property and the unique photothermal-enhanced GOx-like catalytic activity, the developed BPQDs were demonstrated to be an effective therapeutic strategy for inhibiting tumor growth in vivo. We believe that this work will provide a novel perspective for the development of nanozymes in tumor catalytic therapy.

Abstract:Nanozyme-based catalytic therapy, an emerging therapeutic pattern, has significantly incorporated in the advancement of tumor therapy by generating lethal reactive oxygen species. Nevertheless, most of the nanozymes have mono catalytic performances with H2O2 in the tumor microenvironment (TME), which lowers their therapeutic efficiency. Herein, we design a newly-developed single-atom Fe dispersed N-doped mesoporous carbon nanospheres (SAFe-NMCNs) nanozyme with high H2O2 affinity for photothermal-augmented nanocatalytic therapy. The SAFe-NMCNs nanozyme possesses dual enzyme-mimic catalytic activity which not only acts as a catalase-mimic role to achieve ultrasonic imaging in tumor site by O2 generation, but also exhibits the superior peroxidase-mimic catalytic performance to generate •OH for nanocatalytic therapy. Besides, the SAFe-NMCNs nanozyme with strong optical absorption in the second near-infrared (NIR-II) region shows excellent photothermal conversion performance. The peroxidase-mimic catalytic process of SAFe-NMCNs nanozyme is realized using density functional theory (DFT). Both in vitro and in vivo results indicate that the SAFe-NMCNs nanozyme can efficiently suppress tumor cells growth by a synergistic therapy effect with photothermal-augmented nanocatalytic therapy. The work developed a single-atom-coordinated nanozyme with dual-enzyme catalytic performance and achieve hyperthermia-augmented nanocatalytic therapy effect, can open a window for potential biological applications.

Abstract:The main challenges of nanozyme-based tumor catalytic therapy (NCT) lie in the unsatisfactory catalytic activity accompanied by a complex tumor microenvironment (TME). A few nanozymes have been designed to possess both enzyme-like catalytic activities and photothermal properties; however, the previously reported nanozymes mainly utilize the inefficient and unsafe NIR-I laser, which has a low maximum permissible exposure limit and a limited penetration depth. Herein, we report for the first time an all-in-one strategy to realize mild NIR-II photothermally amplified NCT by synthesizing amorphous CoSnO 3 nanocubes with efficient triple enzyme-like catalytic activities and photothermal conversion properties. The presence of Co 2+ and Sn 4+ endows CoSnO 3 nanocubes with the triple enzyme-like catalytic activities, not only achieving enhanced reactive oxygen species (ROS) generation through the Co 2+ -mediated peroxidase-like catalytic reaction to generate ̇OH and Sn 4+ -mediated depletion of overexpressed GSH, but also realizing the catalytic decomposition of endogenous H 2 O 2 for relieving tumor hypoxia. More importantly, the obtained CoSnO 3 nanocubes with a high photothermal conversion efficiency of 82.1% at 1064 nm could achieve mild hyperthermia (43 °C), which further improves the triple enzyme-like catalytic activities of the CoSnO 3 nanozyme. The synergetic therapeutic efficacy of the NIR-II-responsive CoSnO 3 nanozyme through mild NIR-II PTT-enhanced NCT could realize all-in-one multimodal tumor therapy to completely eliminate tumors without recurrence. This study will open a new avenue to explore NIR-II-photoresponsive nanozymes for efficient tumor therapy.

Abstract:Recently, nanozymes show increasing biological applications and promising possibilities for therapeutic intervention, while their mediated therapeutic outcomes are severely compromised due to their insufficient catalytic activity and specificity. Herein, ternary FeCoMn single atoms were incorporated into N-doped hollow mesoporous carbon nanospheres by in situ confinement pyrolysis at 800 °C as high-efficiency nanozyme. The confinement strategy endows the as-prepared nanozyme with the enhanced catalase- and oxidase-like activities. Specifically, the FeCoMn TSAs/N-HCSs nanozyme can decompose intracellular H2O2 to generate O2 and subsequently convert O2 to cytotoxic superoxide radicals (O2∙-), which can initiate cascade enzymatic reactions in tumor microenvironment (TME) for chemodynamic therapy (CDT). Moreover, the cancer therapy was largely enhanced by loading with doxorubicin (DOX). Impressively, the FeCoMn TSAs/N-HCSs nanozyme-mediated CDT and the DOX-induced chemotherapy endow the DOX@FeCoMn TSAs/N-HCSs with effective tumor inhibition, showing the superior therapeutic efficacy.
Photothermal amplified multizyme activity for synergistic photothermal-catalytic tumor therapy

Abstract:Nano-enzymatic catalytic therapy has been widely explored as a promising tumor therapeutic method with specific responsiveness to the tumor microenvironment (TME). However, the inherent lower and simplex catalytic efficiency impairs their anti-tumor efficacy. Therefore, developing novel nanozymes with relatively high and multiple catalytic characteristics, simultaneously enhancing the enzyme-like activity of nanozymes using the proper method, photothermal promoted catalytic property, is a reliable way. In this paper, we report a manganese oxide/nitrogen-doped carbon composite nanoparticles (MnO-N/C NPs) with multi-enzyme mimetic activity and photothermal conversional effect. The peroxidase (POD)-like/oxidase (OXD)-like/catalase (CAT)-like activity of MnO-N/C nanozymes was accelerated upon exposure to an 808 nm NIR laser. In vitro and in vivo results proved that the MnO-N/C NPs shown excellent magnetic resonance imaging (MRI) guided synergistic photothermal-enhanced catalytic treatment and photothermal therapy of liver cancer. The photothermal enhanced multi-enzyme activity maximizes the efficacy of catalytic and photothermal therapy while reducing harm to healthy cells, thereby offering valuable insights for the development of next-generation photothermal nanozymes to enhance tumor therapy.

Abstract:Transition-metal chalcogenide compounds with facile preparation and multifunctional elements act as ideal photothermal agents for cancer theranostics. This work synthesizes Cu 7.2 S 4 /5MoS 2 composite nanoflowers and investigates the crystal growth mechanism to optimize the synthesis strategy and obtain excellent photothermal therapy agents. Cu 7.2 S 4 /5MoS 2 exhibits a high photothermal conversion efficiency of 58.7% and acts as a theranostic nanoplatform and demonstrated an effective photothermal–chemodynamic–photodynamic synergetic therapeutic effect in both in vitro and in vivo tests. Moreover, Cu 7.2 S 4 /5MoS 2 shows strong photoacoustic signal amplitudes and computed tomographic contrast enhancement in vivo . These results suggest a potential application of Cu 7.2 S 4 /5MoS 2 composite nanoflowers as photo/H 2 O 2 -responsive therapeutic agents against tumors.
