Abstract:Nanocatalytic therapy, based on the nanozyme-triggered production of reactive oxygen species (ROS) in tumor microenvironment (TME), has demonstrated potential in tumor therapy, but nanozymes still face challenges of activity and specificity that compromise the therapeutic efficacy. Herein, we develop an innovative strategy using a single-atom nanozyme to initiate cascade enzymatic reactions in TME for tumor-specific treatment. The cobalt (Co)-single-atom nanozyme, with Co- N coordination on N -doped porous carbon ( Co-SAs@NC ), displays catalase-like activity that decompose cellular endogenous H 2 O 2 to produce O 2 , and subsequent oxidase-like activity that converts O 2 into cytotoxic superoxide radicals to efficiently kill tumor cells. By incorporation with doxorubicin, the therapy achieves a significantly enhanced antitumor effect in vivo . Our findings show that cascade TME-specific catalytic therapy combining with chemotherapy is a promising strategy for efficient tumor therapy.

Development of an Intelligent Heterojunction Fenton Catalyst for Chemodynamic/starvation Synergistic Cancer Therapy

Abstract:Chemodynamic therapy (CDT) as an emerging modality in cancer treatment, its implementation remains a daunting challenge by the lack of smart Fenton catalyst under acidic tumor microenvironments. Herein, we have successfully constructed a Fe3O4 @MIL-10 0 heterojunction by growing Fe-based metal-organic framework (MIL-100) onto the surface of Fe3O4 nanoparticles. The as-made heterojunction after encapsu-lating glucose oxidase (termed FMG) is demonstrated as a pH-responsive intelligent Fenton nanosystem with the synergistic effect of starvation therapy (ST). Density functional theory (DFT) calculations reveal that such heterojunction could greatly reduce the energy barrier of the Fenton reaction, which better ex-plains the mechanism of Fenton performance improvement. Moreover, the encapsulated glucose oxidase has successfully activated the ST process, in which its generated H2O2 and gluconic acid further improve the CDT efficiency. More O 2 from the enhanced CDT in turn promotes the enzymatic reaction of glucose oxidase. The Fenton/cascade enzymatic reaction operates in a self-feedback manner as proposed. In vitro and in vivo experiments demonstrate that such intelligent Fenton nanoreactors provide a powerful anti-cancer mechanism for effective tumor ablation with enough safety. This work provides insights into the developments of MOF-based heterojunctions as powerful anticancer treatment nanoreactors. Covalent organic framework-based nanozyme for cascade-amplified synergistic cancer therapy

Abstract:Due to the limited concentration of hydrogen peroxide (H 2 O 2 ) in the tumor microenvironment, peroxidase (POD)-mimicking nanozyme-mediated catalytic therapy usually cannot completely eliminate tumor issues. In this study, we report an H 2 O 2 -responsive nanozyme constructed by loading 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) into POD-like Fe-porphyrin covalent organic frameworks (COFs) (Fe-DhaTph). The resulting ABTS@Fe-DhaTph can function as a multifunctional therapeutic nanoagent for combination therapy of catalytic therapy, photothermal therapy (PTT), and photodynamic therapy (PDT). In the tumor microenvironment, ABTS@Fe-DhaTph can catalyze the decomposition of endogenous H 2 O 2 to produce highly cytotoxic hydroxyl radicals (·OH) for in situ tumor catalytic therapy. Meanwhile, the loaded ABTS is oxidized by H 2 O 2 with the assistance of ABTS@Fe-DhaTph, and the resulting oxABTS, with strong near-infrared absorbance, can enable the H 2 O 2 -dependent PTT. Besides Fe-DhaTph-induced PDT, the generated oxABTS with glutathione (GSH)-depletion ability can synergistically enhance the generation of reactive oxygen species, thereby improving catalytic therapy and PDT. To the best of our knowledge, ABTS@Fe-DhaTph is the first COF-based nanozyme to serve as a cascade-amplified synergistic therapeutic nanoagent for cancer treatment.

Atomically dispersed bimetallic active sites as H2O2 self-supplied nanozyme for effective chemodynamic therapy, chemotherapy and starvation therapy

Abstract:Tumor microenvironment (TME)-responsive chemodynamic therapy (CDT) is severely hindered by insufficient intracellular H2O2 level that seriously deteriorates antitumor efficacy, albeit with its extensively experimental and theoretical research. Herein, we designed atomically dispersed FeCo dual active sites anchored in porous carbon polyhedra (termed FeCo/PCP), followed by loading with glucose oxidase (GOx) and anticancer doxorubicin (DOX), named FeCo/PCP-GOx-DOX, which converted glucose into toxic hydroxyl radicals. The loaded GOx can either decompose glucose to self-supply H2O2 or provide fewer nutrients to feed the tumor cells. The as-prepared nanozyme exhibited the enhanced in vitro cytotoxicity at high glucose by contrast with those at less or even free of glucose, suggesting sufficient accumulation of H2O2 and continual transformation to OH for CDT. Besides, the FeCo/PCP-GOx-DOX can subtly integrate starvation therapy, the FeCo/PCP-initiated CDT, and DOX-inducible chemotherapy (CT), greatly enhancing the therapeutic efficacy than each monotherapy.
Ultrathin 2D Pd/Cu Single-Atom MOF Nanozyme to Synergistically Overcome Chemoresistance for Multienzyme Catalytic Cancer Therapy

Abstract:Single-atom nanozymes (SAzymes) have obtained increasing interest to mimic natural enzymes for efficient cancer therapy, while challenged by chemoresistance from cellular redox homeostasis and the interface of reductive species in tumor microenvironment (TME). Herein, a dual single-atomic ultrathin 2D metal organic framework (MOF) nanosheet of multienzyme (Pd/Cu SAzyme@Dzy) is prepared to synergistically overcome chemoresistance for multienzyme enhanced cancer catalytic therapy. The Pd SAzyme exhibits peroxidase (POD)-like catalytic activity for overcoming chemoresistance via disturbing cellular redox balance. This is further enhanced by cascade generation of more ∙OH via Cu+ -catalyzed POD-like reactions, initiated by in situ-reduction of Cu2+ into Cu+ upon GSH depletion. This process can also avoid the consumption of ∙OH by endogenous reductive GSH in TME, ensuring the adequate amount of ∙OH for highly efficient therapy. Besides, the DNAzyme is also delivered for gene therapy of silencing cancer-cell-targeting VEGFR2 protein to further enhance the therapy. Based on both experiments and theoretical calculations, the synergetic multienzyme-based cancer therapy is examined and the enhancement by the cascade tumor antichemoresistance is revealed.
Dual isolated bimetal single-atom catalysts for tumor ROS cycle and parallel catalytic therapy

Abstract:The application of emerging nanocatalytic drugs in tumor therapy mainly depends on reactive oxygen species (ROS) production in tumors. However, current nanocatalysts have drawbacks such as catalytic inefficiency and high toxicity. Herein, we designed a paradigm of synergy and "division of labor" bimetallic dual active sites single-atom catalyst (SAC) combined with ROS circulation and parallel catalytic therapy for efficient tumor therapy. Both Fe and Co atoms were isolated in the N-doped carbon material with a sharp-angled dodecahedron in a monodisperse state to form an independent bimetallic non-alloy structured atom pair (FeCo-DIA/NC). FeCo-DIA/NC synergistically initiates both Fenton and Fenton-like reactions, directly and simultaneously catalyzing H2O2 and O2 into ROS, including hydroxyl radicals (•OH), superoxide ions (O2•−), and singlet oxygen (1O2). The above ROS are further converted into H2O2 in the acid tumor microenvironment (TME), forming a "ROS Cycle" for highly efficient tumor inhibition. FeCo-DIA/NC exhibits higher activity for Fenton and Fenton-like reactions than single-atom Fe dispersed on N-doped carbon material (Fe-SIA/NC) and single-atom Co dispersed on N-doped carbon material (Co-SIA/NC). The in vitro and in vivo results show that the FeCo-DIA/NC catalyst significantly induces cell apoptosis and inhibits tumor growth. Furthermore, extremely low metal concentrations and high therapeutic effects were achieved simultaneously in FeCo-DIA/NC, highlighting the promising clinical potential. This bimetallic dual-active site monodisperse catalyst provides an important example for applying SACs in the biomedical field, moreover, the highly effective and parallel catalytic "ROS Cycle" provides an opportunity to design nanocatalysts for oncology.

Asymmetric Coordination of Iron Single‐Atom Nanozymes with Efficient Self‐Cascade Catalysis for Ferroptosis Therapy

Abstract:Asymmetrically coordinated Fe‐N3S1 single‐atom nanozymes (Fe‐S/N‐C SAzymes) exhibit significantly stronger peroxidase (POD)‐ and glutathione oxidase (GSHOx)‐like activities than those of their symmetric Fe‐N4 counterparts (Fe‐N‐C) SAzymes and the classical Fe3O4 nanozyme. By camouflaging with macrophage membranes, the tumor‐targeting nanocatalytic agents (M@Fe‐S/N‐C) trigger enhanced self‐cascade catalysis in the tumor microenvironment for ferroptosis‐based tumor‐specific therapy. Single‐atom nanozymes (SAzymes) hold great promise in tumor therapy due to their maximized atomic utilization and well‐defined electronic structures. However, they still face challenges of activity, specificity, and targeting that impede therapeutic efficacy. Herein, a practical strategy is reported to construct asymmetric N, S‐coordinated Fe SAzymes (Fe‐S/N‐C). Benefiting from the regulatory influence of S atoms on the disruption of local charge symmetry of center Fe atoms, the Fe‐S/N‐C SAzymes exhibit significantly enhanced peroxidase (POD)‐ and glutathione oxidase (GSHOx)‐like activities, with catalytic efficiencies being 6.33 and 47.88 times higher than their symmetric Fe‐N4 counterparts, respectively. Theoretical calculations demonstrate that the asymmetric atomic interface configuration increases electron localization around center Fe sites, thus facilitating the adsorption and activation of H2O2 and O2. By camouflaging with macrophage membranes, the tumor‐targeting nanocatalytic agents (M@Fe‐S/N‐C) trigger enhanced self‐cascade catalysis in the tumor microenvironment for ferroptosis‐based tumor‐specific therapy. These results open up a promising avenue for addressing the limitations associated with current SAzymes‐based tumor therapies.

Abstract:Chemodynamic therapy (CDT) based on multifunctional nanoparticles (NPs) has occurred as an attractive cancer treatment that covers the shortage of nonnegligible limitations from single therapy. Herein, we synthesize a safe and universal CDT nanoparticle—iron single-atom nanoparticle (Fe SANP), in which Fe single-atom as the active catalytic site is anchored in a carbon framework with encapsulated doxorubicin (DOX). Fe SANPs work as catalase-like nanozyme for hydroxyl radicals (•OH) production. The prepared NPs (denoted as DOX@Fe SANPs) can efficiently mediate peroxidase-like activity with the existence of H2O2 that enhances cancer cell elimination by generating abundant •OH. DOX@Fe SANPs exhibit a pH-induced degradable character that contributes to specific drug release in the tumor microenvironment (TME). DOX@Fe SANPs produce ignorable systematic toxicity after biodegradation and drug delivery processes. Collectively, this study highlights the efficient anti-tumor performances of the iron single-atom nanocatalysts containing an anti-cancer drug DOX.
Single-atom and clustered iron-embedded graphitic nitrogen-doped carbon nanospheres for cancer imaging and ferroptosis

Abstract:Despite development in nanozymes for cancer treatment, challenges in their synthesis and structural optimization for peak catalytic activity persist. A multifunctional enzyme-like nanoparticle using a controllable synthesis that offers a clear structure–activity relationship was developed. The nitrogen-doped mesoporous carbon nanospheres (MCNs) with iron coordination were produced via an in-situ iron-catalyzed pyrolysis, which allowed for precise adjustment of iron content. Not only do Fe/MCN exhibit high graphitization for enhanced photothermal conversion but also feature co-doping with both single atoms and atom clusters, enhancing their enzyme-like activities. These activities included oxidase, peroxidase, catalase, and glutathione oxidase, leading to synergistic effects in chemodynamic, photodynamic therapies, and hypoxia alleviation. Additionally, Fe/N-MCNs induced potent immunogenic cell death, aided by ROS, ferroptosis, and ferroptosis-sensitized photothermal therapy. Fe/N-MCN also provided excellent photoacoustic and magnetic resonance imaging capabilities, establishing a multifaceted platform for the treatment of breast cancer and the inhibition of postoperative recurrence and metastasis.

Abstract:Rationale: Cancer continues to be a significant public health issue. Traditional treatments such as surgery, radiotherapy, and chemotherapy often fall short because of intrinsic issues such as lack of specificity and poor drug delivery, leading to insufficient drug concentration at the tumor site and/or potential side effects. Consequently, improving the delivery of conventional chemotherapy drugs like doxorubicin (DOX) is crucial for their therapeutic efficacy. Successful cancer treatment is achieved when regulated cell death (RCD) of cancer cells, which includes apoptotic and non-apoptotic processes such as ferroptosis, is fundamental to successful cancer treatment. The developing field of nanozymes holds considerable promise for innovative cancer treatment approaches. Methods: A dual-metallic nanozyme system encapsulated with DOX was created, derived from metal-organic frameworks (MOFs), designed to combat tumors by depleting glutathione (GSH) and concurrently liberating DOX. The initial phase of the study examined the GSH oxidase-mimicking function of the dimetallic nanozyme (ZIF-8/SrSe) through enzyme kinetic assays and Density Functional Theory (DFT) simulations. Following this, we probed the ability of ZIF-8/SrSe@DOX to release DOX in response to the tumor microenvironment in vitro, alongside examining its anticancer capabilities and mechanisms prompting apoptosis or ferroptosis in cancer cells. Moreover, we established tumor-bearing animal models to corroborate the anti-tumor effectiveness of our nanozyme complex and to identify the involved apoptotic and ferroptotic pathways implicated. Results: Enzyme kinetic analyses demonstrated that the ZIF-8/SrSe nanozyme exhibits substantial GSH oxidase-like activity, effectively oxidizing reduced GSH to glutathione disulfide (GSSG), while also inhibiting glutathione peroxidase 4 (GPX4) and solute carrier family 7 member 11 (SLC7A11). This inhibition led to an imbalance in iron homeostasis, pronounced caspase activation, and subsequent induction of apoptosis and ferroptosis in tumor cells. Additionally, the ZIF-8/SrSe@DOX nanoparticles efficiently delivered DOX, causing DNA damage and further promoting apoptotic and ferroptotic pathways. Conclusions: This research outlines the design of a novel platform that combines chemotherapeutic agents with a Fenton reaction catalyst, offering a promising strategy for cancer therapy that leverages the synergistic effects of apoptosis and ferroptosis.

Modulating the Coordination Environment of Carbon‐Dot‐Supported Fe Single‐Atom Nanozymes for Enhanced Tumor Therapy

Abstract:Introducing phenanthroline significantly increases the content of pyrrolic N in carbon dot (CD)‐supported Fe single‐atom nanozymes (ph‐CDs‐Fe SAzyme) and enhances their peroxidase‐like activity. Combing the excellent tumor penetration ability, high peroxidase‐like activity, and excellent photothermal efficiency, ph‐CDs‐Fe SAzyme can locally trigger strong and synergistic chemodynamic and photothermal antitumor therapy. Herein, carbon dot (CD)‐supported Fe single‐atom nanozymes with high content of pyrrolic N and ultrasmall size (ph‐CDs‐Fe SAzyme) are fabricated by a phenanthroline‐mediated ligand‐assisted strategy. Compared with phenanthroline‐free nanozymes (CDs‐Fe SAzyme), ph‐CDs‐Fe SAzyme exhibit higher peroxidase (POD)‐like activity due to their structure similar to that of ferriporphyrin in natural POD. Aberration‐corrected high‐angle annular dark field scanning transmission electron microscopy (HAADF‐STEM) and X‐ray absorption fine structure spectroscopy (XAFS) analyses show that metal Fe is dispersed in ph‐CDs‐Fe SAzyme as single atoms. Steady‐state kinetic studies show that the maximum velocity (Vmax) and turnover number (kcat) of H2O2 homolytic cleavage catalyzed by ph‐CDs‐Fe SAzyme are 3.0 and 6.2 more than those of the reaction catalyzed by CDs‐Fe SAzyme. Density functional theory (DFT) calculations show that the energy barrier of the reaction catalyzed by ph‐CDs‐Fe SAzyme is lower than that catalyzed by CDs‐Fe SAzyme. Antitumor efficacy experiments show that ph‐CDs‐Fe SAzyme can efficiently inhibit the growth of tumor cells both in vitro and in vivo by synergistic chemodynamic and photothermal effects. Here a new paradigm is provided for the development of efficient antitumor therapeutic approaches based on SAzyme with POD‐like activity.

Abstract:Simple biomolecules-based supramolecular self-assembly hold great promise on the fabrication of nanozyme for mimicking natural enzyme, both structurally and functionally. However, it remains a formidable challenge to design of tumor-specific nanozyme with promoted therapeutic efficiencies starting from such small biological molecule combinations and their cooperative interactions. Inspired by the metalloenzyme in living systems, i.e., peroxidase, herein the construction of metallo-nanozyme through a facile multicomponent coordination self-assembly based on the combination of amino acid, chemotherapeutic motif and metal ions is reported. The resulting metallo-nanozyme possess uniform size distribution, well-defined spherical nanostructure and high chemical drugs contents. Most importantly, the metallo-nanozyme depletes specifically high-level glutathione (GSH) in tumor cell and converts Fe3+ to Fe2+ for subsequent transformation of overproduced hydrogen peroxide (H2O2) into highly cytotoxic hydroxyl radical (·OH) in peroxidase-like catalytic manner. Meanwhile, the metallo-nanozyme are also activated in situ to release the chemical drugs in tumor cell for enhanced chemotherapy. In vitro and in vivo evaluations demonstrate that the supramolecular metallo-nanozyme suppresses remarkably tumor growth via combined catalytic-chemotherapy and without any systemic toxicity. Therefore, this study demonstrates that the tumor-specific biomimetic nanozyme with advanced catalytic therapeutic efficacy could be achieved through cooperative coordination of small biomolecules or therapeutic drugs, opening up opportunities in the development of catalytic anticancer nanozyme for efficiently combat cancers.

Abstract:Clinical applications of nanozyme-initiated chemodynamic therapy (NCDT) have been severely limited by the poor catalytic efficiency of nanozymes, insufficient endogenous hydrogen peroxide (H2 O2 ) content, and its off-target consumption. Herein, the authors developed a hollow mesoporous Mn/Zr-co-doped CeO2 tandem nanozyme (PHMZCO-AT) with regulated multi-enzymatic activities, that is, the enhancement of superoxide dismutase (SOD)-like and peroxidase (POD)-like activities and inhibition of catalase (CAT)-like activity. PHMZCO-AT as a H2 O2 homeostasis disruptor promotes H2 O2 evolution and restrains off-target elimination of H2 O2 to achieve intensive NCDT. PHMZCO-AT with SOD-like activity catalyzes endogenous superoxide anion (O2 •- ) into H2 O2 in the tumor region. The suppression of CAT activity and depletion of glutathione by PHMZCO-AT largely weaken the off-target decomposition of H2 O2 to H2 O. Elevated H2 O2 is then catalyzed by the downstream POD-like activity of PHMZCO-AT to generate toxic hydroxyl radicals, further inducing tumor apoptosis and death. Abstract:The tumor microenvironment (TME) responsive multimodal theranostic nanoplatforms have attracted great attentions for precision medicine. To design well-defined cascade catalytic therapy and achieve the synergistic effect of different components in a nanoplatform, herein a spindle-shaped nanodrug termed as FeCu-GOx PNzyme-MTO was constructed by in-situ growth of bimetallic metal nanozyme (FeCu PNzyme), and simultaneous encapsulation of glucose oxidase (GOx) as well as fluorescent and chemotherapeutic mitoxanthrone (MTO). Interestingly, the obtained FeCu-GOx PNzyme is able to efficiently upregulate endogenous H2O2 level and down-regulate acidity via inducing the catalytic decomposition of intratumoral glucose to gluconic acid and H2O2 in TME, which is benefit to the cascade catalytic reactions for chemodynamic therapy (CDT). After efficient loading of MTO, such intelligent nanozyme triggers reactive oxygen species (ROS) generation in situ with tumor stimuli, self-augment ROS level upon intrinsic photothermal-conversion efficiency (53.05%) and glutathione (GSH) depletion, and specifically TME-responsive release of MTO to achieve high chemotherapeutic efficacy and biosafety. Both in vitro cellular assays and in vivo tumor-xenograft experiments proved that the tumor-specific cytotoxicity, biodegradability and potent antitumor efficiency of nanodrug, with co-contributions from highly cytotoxic ROS generated and precise drug delivery within tumors. This work thus presents the friendly construction of biocompatible FeCu-GOx PNzyme-MTO as effective nanodrug to provided a promising strategy for a new perspective on effective tumor therapy.

Abstract:Here, we report Fe-based single-atom nanozymes, which were fabricated by embedding Fe atoms into N-doped hollow carbon spheres. The nanozymes (FeSA-HNCSB) exhibit the efficient catalytic generation of ROS in the tumor microenvironment (TME). They possess multiple enzyme-mimicking activities, including catalase-like, oxidase-like, and peroxidase-like activities. Furthermore, the FeSA-HNCSB nanozymes also exhibit good photothermal conversion efficiency in the near-infrared (NIR) region, making them potential candidates for photothermal therapy (PTT) and photothermal-enhanced catalytic therapy. This work provides a new paradigm for integrating parallel catalytic therapy and PTT to improve the effectiveness of tumor treatment.

MOF-derived cobalt-iron containing nanocomposite with cascade-catalytic activities for multimodal synergistic tumor therapy

Abstract:Reactive oxygen species (ROS)-driven chemodynamic therapy has emerged as a promising anti-tumor strategy. However, the insufficient hydrogen peroxide (H2O2) supply in tumor microenvironment results in a low Fenton reaction rate and subsequently poor ROS production and therapeutic efficacy. Herein, we report on a new nanocomposite MIL-53@ZIF-67/S loaded with doxorubicin and glucose oxidase, which is decomposed under the acidic tumor microenvironment to release Fe3+, Co3+, glucose oxidase, and doxorubicin. The released content leads to synergistic anti-tumor effect through the following manners: 1) doxorubicin is directly used for chemotherapy; 2) Fe3+and Co3+ result in glutathione depletion and Fenton reaction activation through Fe2+ and Co2+ generation to achieve chemodynamic therapy; 3) glucose oxidase continuously catalyzes glucose consumption to induce starvation of the cancer cells, and 4) at the same time the produced gluconic acid and H2O2 significantly promote Fenton reaction and further boost chemodynamic therapy. This work not only demonstrates the high anti-tumor effect of the new nanocomposite, but also provides an innovative strategy for the development of a multi-in-one nanoplatform for cancer therapy.

Abstract:Nanozymes, which are inorganic nanomaterials mimicking natural enzyme activities, are bringing enormous opportunities to theranostics. Herein, a cytochrome c oxidase‐like nanozyme (copper‐silver alloy nanoparticle, Cu‐Ag NP) has been demonstrated for nanocatalytic cancer therapy. Loaded with bioreductive predrug (AQ4N), this Cu‐Ag nanozyme unprecedentedly enables simultaneous starvation, ferroptosis, and chemical therapy with high specificity, which is able to totally eliminate tumor and greatly prolong the survival rate for 4T1 tumor‐bearing mice. Abstract:Tumor therapy employing metal-based nanomaterials to convert the abundant H 2 O 2 in tumor microenvironment (TME) to oxygen (O 2 ) and hydroxy radical (·OH) has attracted substantial attention. However, the generally complex structure of metal nanosystems may have poor catalytic selectivity towards the target and hence cause undesired side reactions. Single-atom catalysts (SACs) with high atomic utilization, composition of identical active site and tunable reaction pathway can be harnessed to realize the well-controlled and highly-selective conversion of H 2 O 2 for cancer therapy. Herein, a series of dual single-atom catalysts (DSACs) containing two metal centers (Fe-Se 6 , Mn-Se 6 ) are proposed. As guided by theoretical calculations, DSACs with equal proportions of Fe and Mn (termed as Fe/Mn@PSe 3 ) exhibit the optimal reaction barriers towards the production of both ·OH and O 2 by catalyzing H 2 O 2 . In response to the elevated TME H 2 O 2 , Fe/Mn@PSe 3 can produce ·OH to trigger chemodynamic therapy (CDT), and boost O 2 generation to alleviate tumor hypoxia and its mediated immunosuppression. In addition, its mild hyperthermia feature enhances the anti-tumor effects of CDT and immune therapy, causing an efficient synergistic tumor suppression outcome. The study provides new insights into highly selective nanomaterial design that preferentially activating specific catalytic processes within tumor, functioning as promising candidates for cancer therapy.

Abstract:Tumor microenvironment (TME)‐responsive nanozyme‐catalyzed cancer therapy shows great potential due to its specificity and efficiency. However, breaking the self‐adaption of tumors and improving the sustainable remodeling TME ability remains a major challenge for developing novel nanozymes. In this study, a rapid method is firstly developed to synthesize unprecedented trimetalic nanozyme (AuMnCu, AMC) with a targeting peptide (AMCc), which exhibits excellent peroxidase‐like, catalase‐like, and glucose oxidase‐like activities. AMCc consumes endogenous H2O2 and produce O2, while the released Cu and Mn ions in TME catalyze glucose oxidation reaction to generate H2O2 and gluconic acid, which achieves the starvation therapy by depleting the energy and enhances the chemodynamic therapy effect by lowering the pH of the TME and producing extra H2O2. Meanwhile, the reactive oxygen species damage was amplified, as AMCc can constantly oxidize intracellular reductive glutathione through the cyclic valence alternation of Cu and Mn ions, and the generated Cu+ elevate the production of ·OH from H2O2. Further studies depict that the well‐designed AMCc exhibit the excellent photothermal performance, and achieve TME‐responsive sustainable starvation/photothermal‐enhanced chemodynamic synergistic effects in vitro and in vivo. Precise Tuning of the D-Band Center of Dual-Atomic Enzymes for Catalytic Therapy

Abstract:Single-atom nanozyme-based catalytic therapy is of great interest in the field of tumor catalytic therapy; however, their development suffers from the low affinity of nanozymes to the substrates (H2O2 or O2), leading to deficient catalytic activity in the tumor microenvironment. Herein, we report a new strategy for precisely tuning the d-band center of dual-atomic sites to enhance the affinity of metal atomic sites and substrates on a class of edge-rich N-doped porous carbon dual-atomic sites Fe-Mn (Fe1Mn1-NCe) for greatly boosting multiple-enzyme-like catalytic activities. The as-made Fe1Mn1-NCe achieved a much higher catalytic efficiency (Kcat/Km = 4.01 × 105 S-1·M-1) than Fe1-NCe (Kcat/Km = 2.41 × 104 S-1·M-1) with an outstanding stability of over 90% activity retention after 1 year, which is the best among the reported dual-atom nanozymes. Theoretical calculations reveal that the synergetic effect of Mn upshifts the d-band center of Fe from -1.113 to -0.564 eV and enhances the adsorption capacity for the substrate, thus accelerating the dissociation of H2O2 and weakening the O-O bond on O2. We further demonstrated that the superior enzyme-like catalytic activity of Fe1Mn1-NCe combined with photothermal therapy could effectively inhibit tumor growth in vivo, with an inhibition rate of up to 95.74%, which is the highest value among the dual-atom artificial enzyme therapies reported so far.
