Abstract:Herein, we disclose the metal-free synthesis of arylamines via the direct amination of phenols using aminating reagents. This reaction procedure uses easy accessible aminating reagents and provides a versatile synthetic route to a broad range of arylamines with various functionalities in good to excellent yield. By using a two-step route of amination and oxidative coupling reaction, we synthesized three naturally occurring carbazole alkaloids: murrayafoline A, mukonine, and clausenine from two commercially available phenols.

Abstract:Some acyclic acidic ionic liquids were prepared and used as the catalysts for the one-pot, three-component synthesis of -aminophosphonates from aldehydes, amines, and trimethylphosphite at room temperature in water. The products could simply be separated from the system of catalyst/water, and the catalysts could be reused at least six times without noticeably decreasing the catalytic activity.

Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:An efficient and environmentally friendly method for the synthesis of α‐amino phosphonates has been reported using the one‐pot, three‐component reaction of various substituted anilines and benzaldehydes with diethyl phosphite in acetonitrile at 100°C. The advantages of this method are catalyst‐free and works well with a broad substrate scope of various aromatic anilines and aldehydes containing both electron‐donating and withdrawing groups.

Abstract:Herein, we report a highly efficient ZnI2-triggered oxidative cross-coupling reaction of P(O)-H compounds and amines. This operationally simple protocol provides unprecedented generic access to phosphinic amides/phosphoramidate derivatives in good yields and short reaction time. Besides, the reaction proceeds under mild conditions, which avoids the use of hazardous reagents, and is applicable to scale-up syntheses as well as late-stage functionalization of drug molecules. The stereospecific coupling is also achieved from readily available optically enriched P(O)-H compounds.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:Inspired by the diverse bioactivities of α-amino phosphine oxides, an efficient strategy for the synthesis of less researched α-(hydroxyamino)diarylphosphine oxides has been developed and their antitumor activities are explored. Under water as a solvent and catalyst-free conditions, the addition of nitrones and diphenylphosphine oxide occurs smoothly to afford α-(hydroxyamino) diarylphosphine oxides in high yields. This reaction features a wide substrate scope, facile starting materials, atom economy, and easy purification. Moreover, the biological evaluation revealed that two synthesized derivatives 5e and 5f could serve as interesting anti-cancer agents for further development.

Abstract:ynthesis of valuable compounds directly from small and simple molecules is an area of great interest in chemical research. However, the preparation of most organophosphorus compounds from white phosphorus currently relies on inefficient, stepwise procedures that result in significant environmental pollution and poor atom economy. We herein describe a novel and high-yielding route to generate various phosphorothioates with diverse substituents from white phosphorus, disulfides, and alcohols in a single reaction step. This innovative three-component coupling reaction utilizes basic industrial raw materials and offers several advantages, including operational simplicity, mild reaction conditions, a broad substrate scope, and high product selectivity.

Abstract:An efficient enantioselective synthesis of cyclic α-aminophosphonates via multicomponent reactions of 2-alkynylbenzaldehydes, amines, and dimethylphosphonate has been developed with the use of a chiral silver spirocyclic phosphate as the catalyst. This protocol provides straightforward access to a series of chiral C1-phosphonylated 1,2-dihydroisoquinoline derivatives with high yields (up to 99%) and high enantioselectivities (up to 94% ee) for a broad substrate scope. Abstract:The practical synthesis of C-stereogenic α-aminophosphine oxides, which exhibit a wide range of applications in medicinal chemistry, biochemistry, material science, and asymmetric catalysis, represents an unmet need. Herein, a Ni/(S,S)-BenzP* catalyst system is developed for asymmetric synthesis of branched α-aminophosphine oxides through an enantioselective Markovnikov addition of H-phosphine oxides to 2-azadienes. A variety of readily available 2-azadienes and H-phosphine oxides undergo hydrophosphinylation with high enantioselectivities (up to 99%) and good yields (up to 96%). These products can be readily hydrolyzed to afford synthetically valuable chiral α-aminophosphine oxides, which are key building blocks for the synthesis of bioactive compounds (such as phospholeucine synthesized herein), organocatalysts, and ligands. Mechanistic studies suggest a hydrofunctionalization process.

Abstract:A protocol for the synthesis of α-amino-vinylphosphine oxides by phosphinoenamination reaction between alkyl nitriles and phosphine oxides was developed. The combination of Mn(OAc)2 as a Lewis acid and guanidine as a Lewis base was found to be an efficient catalytic system for this reaction. A series of alkyl nitriles and phosphine oxides are compatible with this conversion, furnishing the desired products in up to 95% yield under mild conditions. Furthermore, this method demonstrates the capability of gram-scale synthesis.

Abstract:A one-pot synthetic strategy for diarylamines using only aromatic amines as starting materials has been developed. This method involved a PhI(OAc)2-induced oxidative dearomatization of N-sulfonyl protected para-substituted anilines, a Bi(OTf)3-catalyzed imino exchange reaction between N-sulfonyl cyclohexadienimines and aromatic amines, and a CF3COOH/Zn mediated reductive aromatization of the resulting N-aryl cyclohexadienimines.

Abstract:An efficient, LiN(SiMe3)2‐mediated phosphorylation of terminal alkynes with chlorodiphenylphosphine oxide has been developed to synthesize alkynylphosphine oxides. This transition‐metal‐free protocol was featured with simple operation, mild reaction conditions, and broad substrate scope. The tolerance of halogen and ester substituents displayed the advantages of this new method over the traditional ones. In addition, the application potential was demonstrated by the gram‐scale reaction. When using diisopropyl chlorophosphate and chlorodiphenylphosphine, the corresponding alkynylphosphate and alkynylphosphine could also been produced.

Abstract:A metal‐free three‐component reaction for the direct synthesis of α‐sulfonamide phosphonates involving sulfonamide derivatives, benzaldehyde derivatives, and diethyl phosphite, has been achieved with high yields using ethanol as a green solvent. This efficient method reduces reaction time and minimizes the use of excess reagents. Hitherto, no reports have been found in the literature for the direct synthesis of α‐sulfonamide phosphonates from the readily available aldehydes, diethyl phosphite, and sulfonamides, emphasizing the novelty and significance of this work.
