Abstract:Enzymatic biosensor (EBS) is the most promising substitute for traditional analytical methods on determination of organophosphorus pesticides (OPs). The merits of simple, inexpensive and miniature make it potential for environmental monitoring. Enzymatic determination of OPs is often based on cholinesterase (acetylcholineaterase and butyrylcholinesterase) and organophosphorus hydrolase. Various methods have been applied for enzyme immobilization in the construction of EBS for determination of OPs. Since the chemical and physical properties of the polymers may be tailored for particular needs, they gained importance as the substrate for enzyme immobilization. Application of single-use screen-printed enzymatic electrodes are most intensively developed procedures.

Abstract:Organophosphorus compounds are highly toxic chemicals and widely used as insecticides,plasticizers and flame retardants.Due to the difficulty of degradation these toxic chemicals have gradually accumulated in agricultural products,waters and soils,raising serious public concerns regarding health,environment and food safety.The enzymatic degradation of organophosphorus compounds is highly efficient and environmental friendly,which is the current research hotspot.This paper reviewed recent advances in discovery,engineering and applications of organophosphorus hydrolases,and highlighted the current challenges and the future research directions with the aim to provide an insight into the biodegradation of organophosphorus compounds.

Abstract:The inhibition of acetylcholinesterase (AChE) activity based methods have been widely used for rapid detection of organophosphate (OP) pesticide residues. However, the sensitivity of the enzyme toward pesticides is not desirable and complex purification steps increase production cost. In this study, we optimized and synthesized the cDNA of carp AChE based on the preferred codon using Saccharomyces cerevisiae, which was ligated into expression vector pYD1. The AChE was successfully presented on yeast surface through a-agglutinin mediated display system. In shake-flask after induction with galactose in YPD medium for 24 h, this strain expressed AChE activity at a high level of 0.15 U/OD600/ml. The correct location of AChE was confirmed by immunofluorescence analysis. The whole cell catalyst was applied to visible spectrophotometry detection for OP pesticides based on inhibition rate. The inhibition rate was linear with paraoxon concentration in the range of 0.5 ng/ml-10 mu g/ml and parathion concentration in the range of 5 ng/ml-10 mu g/ml. The low detection limit was 0.136 ng/ml paraoxon and 3.72 ng/ml parathion (S/N = 3). The proposed method is applicable to analyzing OP pesticides in real samples. Therefore, the recombinant strain developed could be an efficient and affordable biomaterial for rapid and accurate detection of OP pesticide residues.

Abstract:<span>The organophosphorus-based OPs) nerve agents and pesticides have been applied in the agriculture industry for a long time. However, they were found to have a persistent effect on the environment and threaten human health. Traditional methods, including incineration and landfilling, could not thoroughly remove these organophosphorus compounds (OPs). Meanwhile, chemical hydrolysis for decontamination was also inhibited due to the presence of corrosive materials and high costs. Biological remediation for OPs employing microorganisms and organophosphorus-degrading enzymes is promising due to a mild and controllable procedure, environmental-friendly reactions, and high efficacy. A wide variety of enzymes have shown latent ability in degrading OPs hazards like organophosphorus hydrolase (OPH), organophosphorus acid anhydrolase (OPAA), the diisopropylfluorophosphatase (DFPase), and mammalian paraoxonase 1 (PON 1). To this end, increasing efforts have been made on these intriguing enzymes to increase their expression level, enhance the catalytic activity, modify the optimal substrate, and expand the practical application. In this review, the enzyme resource, crystal structure, molecular modification, and industry application were compared and discussed in detail. Moreover, the proposed ideas and positive results could be useful for the other relevant OPs-degrading enzymes.</span>

The advances of the organophosphate biodegradation, including the species of the organophosphorous pesticides-degrading microorganisms, organophosphorus hydrolase, the degradation related genes and the application of organophosphorus hydrolase are reviewed in this paper, it can provide a support for the solution of the pesticides pollution in the environment and pesticide residues in agricultural products.
Displaying of Acetylcholinesterase Mutants on Surface of Yeast for Ultra-Trace Fluorescence Detection of Organophosphate Pesticides with Gold Nanoclusters

Abstract:Acetylcholinesterase (AChE) has been widely applied on the enzyme inhibition-based detection of organophosphate pesticides (OPs). To improve the sensitivity of fluorometric OPs assay, great efforts were made to change the fluorometric probes or analytical strategies rather than improve the sensitivity of AChE towards OPs. In this work, AChE wild-type (WT) and mutants (E69Y and E69Y/F330L) from Drosophila were successfully displayed on the surface of yeast through a-agglutinin-mediated microbial surface display system. The location of AChE on yeast surface was confirmed by immunofluorescence analysis. Further, a fluorescence OPs detection method was developed by combining yeast surface-displayed AChE mutants and protein-directed electronegative fluorescent gold nanoclusters (Au NCs). Yeast surface-displayed AChE can catalyze the hydrolysis of acetylthiocholine to produce thiocholine. The electropositive thiocholine can not only bind with AuNCs by Au-S bond but also absorb Au NCs by the electrostatic interaction, leading to the aggregation of AuNCs and corresponding fluorescence quenching. When AChE was incubated with paraoxon, a typical model of OPs, the activity of AChE was inhibited and the thiocholine-induced aggregation of AuNCs was reduced. The fluorescence assay based on Au NCs and yest-AChE-E69Y/F330L exhibited the ultra-sensitivity for ultra-trace OPs and 2-6 orders of magnitude lower detection limit (3.3 x 10(-14) M) than those of AChE-WT-based method and other reported methods. In addition, the proposed method showed excellent reliability for the real samples assay. This work would provide an alternative strategy for the improvement of bio-analysis at its source.

Abstract:The biodegradation of organophosphorus compounds (OPs) by phosphotriesterase (PTE) is an efficient and ecofriendly way. However, the low catalytic activity and instability of PTE becomes the key limiting step for the practical application. In this study, a new multimetal-based organic-inorganic self-assembly hybrid system was proposed to ameliorate this situation. A series of characterization methods demonstrated that the synthesized multimetallic PTE hybrid complex (Co/MnHF@PTE) exhibited a hierarchical flower-like structure. The activity recovery and the effective encapsulation yield of the Co/MnHF@PTE was 408% and 84.5%, respectively. The kcat/Km of Co/MnHF@PTE was separately 2.9-, 1.3-, and 2.7-fold higher than MnHF@PTE, CoHF@PTE, and free PTE. The hybrid nanoflowers showed preferable durability, stability, and reusability. When Co/MnHF@PTE was applied in a pump-flow reactor, the degradation percent of methyl parathion could be maintained at more than 93% with the injection of 24 ml of reaction mixture. More importantly, the Co/MnHF@PTE can effectively break P-F or P-S bond of soman (GD) and nerve agent VX to produce non-toxic products through hydroxyl nucleophilic attack. This study provides a feasible method to improve the activity and stability of PTE, promoting the biodegradation of OPs in the environment.

Abstract:Background: Organophosphorus compounds, widely used in agriculture and industry, pose a serious threat to human health due to their acute neurotoxicity. Although traditional interventions for organophosphate poisoning are effective, they often come with significant side effects. Objective: This paper aims to evaluate the potential of enzymes within biological organisms as organophosphorus bioclearing agents. It analyses the technical challenges in current enzyme research, such as substrate specificity, stereoselectivity, and immunogenicity, while exploring recent advancements in the field. Methods: A comprehensive review of literature related to detoxifying enzymes or proteins was conducted. Existing studies on organophosphorus bioclearing agents were summarised, elucidating the biological detoxification mechanisms, with a particular focus on advancements in protein engineering and novel delivery methods. Results: Current bioclearing agents can be categorised into stoichiometric and catalytic bioclearing agents, both of which have shown some success in preventing organophosphate poisoning. Technological advancements have significantly improved various properties of bioclearing agents, yet challenges remain, particularly in substrate specificity, stereoselectivity, and immunogenicity. Future research will focus on expanding the substrate spectrum, enhancing catalytic efficiency, prolonging in vivo half-life, and developing convenient administration methods. Conclusion: With the progression of clinical trials, bioclearing agents are expected to become widely used as a new generation of therapeutic organophosphate detoxifiers.

Abstract:Extensive application of synthesized organophosphorus compounds (OPs) leads to pollutant accumulation and enhanced eco-toxicity. Hydrolysis of phosphotriester bonds catalyzed by evolved microbial enzymes is a key step for detoxification of OPs. Here, a new marine bacterial prolidase OPAA4301 exhibiting promiscuous phosphotriesterase activity was isolated and systematically characterized. The homo-tetrameric enzyme OPAA4301 can catalyze the hydrolysis of both amido bond and phosphotriester bond. Manganese ions were observed to be essential for its catalytic integrity, and in vitro substitution of manganese ions by different metal cofactors led to decreased activity. We also revealed cooperation pattern of metal ligands and substrate-binding residues on OP hydrolysis by mutational analysis. Metal-binding sites together with Arg418 in the large-binding pocket of the enzyme were found to be indispensable for catalytic ability. Substitution mutation of small- and large-binding pocket residues caused significant variation in phosphotriesterase activity, and leaving group sites appeared to be involved in the catalytic process as substrate affinity regulators. Our study gave an overall biochemical understanding on the organophosphorus hydrolysis pattern of the newly identified marine bacterial prolidase and provided ideas for protein engineering to expand its application in the bioremediation field.

Abstract:Organophosphorus compounds (OPs) are widely used as pesticides and chemical warfare agents (CWAs). These synthetic compounds reside in the environment and cause serious pollution problems. Organophosphorus compounds can be detoxified rapidly by hydrolysis on exposure to environment, which are liable to be influenced by abiotic and biotic factors. Microorganisms isolated from different niches are the predominant entities in the biosphere with an exceptional ability to metabolize various OPs for their growth. The focus of this work is organophosphorus hydrolase (OPH, E.C. 8.1.3.1), which catalyzes the hydrolysis of many organophosphorus compounds and greatly reduces the toxicity and even can completely mineralize them. Several OPH enzymes, including o-phenylenediamine dihydrochloride (OPD), methyl parathion hydrolase (MPH) mevalonate pyrophosphate decarboxylase (MPD) etc, have been identified which accomplish hydrolysis for specific classes of organophosphorus compounds. The functional gene encoding OPH protein have been cloned, expressed and purified in both prokaryotic and eukaryocyte expression vector. To increase the enzyme activity or enhance their broad-spectrum property, the wild activated OPH was used to modify activated sites by the chemical modification of specific amino acid residues with the use of appropriately designed coenzyme analogs. The applications of the functional strains and OPH enzymes in bioremediation of OPs pollutants included: (i) Bioremediation: the technologies can be generally classified as in situ or ex situ; (ii) immobilization: an immobilized enzyme is the OPH that is attached to an inert, insoluble material such as calcium alginate and agar; immobilized whole cell, the target cell is the functional strains that is capable of degrading special OPs; (iii) construction of genetically engineering bacteria. A versatile genetically engineering bacterium that gained more functional genes from different sources was constructed by gene engineering and enzyme engineering, showing a creative and promising application in OPs detoxification. Abstract:Organophosphorus compounds (OPs) have been widely used in agriculture for decades because of their high insecticidal efficiency, which maintains and increases crop yields worldwide. More importantly, OPs, as typical chemical warfare agents, are a serious concern and significant danger for military and civilian personnel. The widespread use of OPs, superfluous and unreasonable use, has caused great harm to the environment and food chain. Developing efficient and environmentally friendly solutions for the decontamination of OPs is a long-term challenge. Microbial enzymes show potential application as natural and green biocatalysts. Thus, utilizing OP-degrading enzymes for environmental decontamination presents significant advantages, as these enzymes can rapidly hydrolyze OPs; are environmentally friendly, nonflammable, and noncorrosive; and can be discarded safely and easily. Here, the properties, structure and catalytic mechanism of various typical OP-degrading enzymes are reviewed. The methods and effects utilized to improve the expression level, catalytic performance and stability of OP-degrading enzymes were systematically summarized. In addition, the immobilization of OP-degrading enzymes was explicated emphatically, and the latest progress of cascade reactions based on immobilized enzymes was discussed. Finally, the latest applications of OP-degrading enzymes were summarized, including biosensors, nanozyme mimics and medical detoxification. This review provides guidance for the future development of OP-degrading enzymes and promotes their application in the field of environmental bioremediation and medicine.

Abstract:An ultrasensitive amperometric acetylcholinesterase (AChE) biosensor was fabricated by controlled immobilization of AChE on gold nanoparticles/poly(dimethyldiallylammonium chloride) protected Prussian blue (Au–PDDA–PB) nanocomposite modified electrode surface for the detection of organophorous pesticide. The Au–PDDA–PB membrane served as an excellent matrix for the immobilization of enzyme, which not only enhanced electron transfer but also possessed a relatively large surface area. In addition, the surface hydrophilicity of the Au–PDDA–PB nanocomposite was finely controlled in the static water contact angle range of 25.6–78.1° by adjusting the ratio of gold nanoparticles to PDDA–PB. On an optimized hydrophobic surface, the AChE adopts an orientation with both good activity and stability, which has been proven by electrochemical methods. Benefit from the advantages of the Au–PDDA–PB nanocomposite and the good activity and stability of AChE, the biosensor shows significantly improved sensitivity to monocrotophos, a typical highly toxic organophorous pesticide, with wide linear range (1.0–1000pg/mL and 1.0–10ng/mL) and an ultra-low detection limit of 0.8pg/mL. The biosensor exhibits accuracy, good reproducibility and stability. This strategy may therefore provide useful information for the controlled immobilization of protein and the design of highly sensitive biosensors.

Abstract:In the past decades, the organophosphorus compounds had been widely used in the environment and food industries as pesticides. Owing to the life-threatening and long-lasting problems of organophosphorus insecticide (OPs), an effective detection and removal of OPs have garnered growing attention both in the scientific and practical fields in recent years. Bacterial organophosphorus hydrolases (OPHs) have been extensively studied due to their high specific activity against OPs. OPH could efficiently hydrolyze a broad range of substrates both including the OP pesticides and some nerve agents, suggesting a great potential for the remediation of OPs. Abstract:This paper reports the codisplay of organophosphorus hydrolase (OPH) and methyl parathion hydrolase (MPH)-green fluorescent protein (GFP) fusion on the cell surface of Escherichia coli using the truncated ice nucleation protein (INPNC) and Lpp-OmpA as the anchoring motifs. The surface localization of both OPH and MPH-GFP was demonstrated by cell fractionation, Western blot analysis, protease accessibility experiment, and immunofluorescence microscopy. Anchorage of the foreign proteins on the outer membrane neither inhibits cell growth nor affects cell viability. The recombinant strain can be used as a whole-cell biocatalyst and showed a broader substrate range than strains expressing either OPH or MPH. A mixture of six organophosphorus pesticides (OPs) (0.2 mM each) could be degraded completely within 5 h. The broader substrate specificity in combination with the rapid degradation rate makes the recombinant strain a promising candidate for detoxification of OPs. The fluorescence of surface-displayed GFP is very sensitive to environmental pH change. Because hydrolysis of OPs by OPH or MPH generates protons, the recombinant E. coli could be used as a whole-cell biosensor for the rapid detection of OPs by evaluating fluorescence changes as a function of OP concentrations.
Investigation of structure-activity relationships in organophosphates-cholinesterase interaction using docking analysis

Abstract:It is known than the most potent homologues in various series of O,O-dialkylphosphates are the dibutyl or diamyl derivatives toward mammalian cholinesterases (ChEs) (both Acetyl- and Butyryl-ChEs), and the dimethyl or diethyl ones toward insect AChEs. To investigate the ChE interaction with organophosphorus inhibitors (OPIs) in more detail, we have performed in silico docking of the series of O,O-dialkylfluorophosphates into active center of different ChEs - both from mammals (human and mouse AChEs and horse BChE), and from insects (spring grain aphid AChE belonging to AChE-1 type, and housefly AChE belonging to AChE-2 type). According to the modeling results, one radical is directed to the anionic site W84, another to the acyl pocket. In addition to well-known residues 288 and 290 (Torpedo AChE sequence numbering), we showed an essential influence of residue 400 - a short alkyl residue in mammalian ChEs and phenylalanine in insect ChEs. Phenylalanine in this position creates sterical hindrance for proper orientation of the OPI molecule, which increases the distance between the catalytic serine gamma-oxygen and phosphorus, and decreases the angle of nucleophylic attack. This suggestion was supported by docking of dibutylfluorophosphate into the active center of AChEs with in silico mutations. Thus, we suggest both the angle of nucleophylic attack and the distance between the catalytic serine and phosphorus atom as measures of productivity of OPI binding.
Advances in organophosphorus pesticides pollution: Current status and challenges in ecotoxicological, sustainable agriculture, and degradation strategies

Abstract:Organophosphorus pesticides (OPPs) are one of the most widely used types of pesticide that play an important role in the production process due to their effects on preventing pathogen infection and increasing yield. However, in the early development and application of OPPs, their toxicological effects and the issue of environmental pollution were not considered. With the long-term overuse of OPPs, their hazards to the ecological environment (including soil and water) and animal health have attracted increasing attention. Therefore, this review first clarified the classification, characteristics, applications of various OPPs, and the government's restriction requirements on various OPPs. Second, the toxicological effects and metabolic mechanisms of OPPs and their metabolites were introduced in organisms. Finally, the existing methods of degrading OPPs were summarized, and the challenges and further addressing strategy of OPPs in the sustainable development of agriculture, the environment, and ecology were prospected. However, methods to solve the environmental and ecological problems caused by OPPs from the three aspects of use source, use process, and degradation methods were proposed, which provided a theoretical basis for addressing the stability of the ecological environment and improving the structure of the pesticide industry in the future.

Abstract:There has been a rise in pesticide use as a result of the growing industrialization of agriculture. Organophosphorus pesticides have been widely applied as agricultural and domestic pest control agents for nearly five decades, and they remain as health and environmental hazards in water supplies, vegetables, fruits, and processed foods causing serious foodborne illness. Thus, the rapid and reliable detection of these harmful organophosphorus toxins with excellent sensitivity and selectivity is of utmost importance. Aptasensors are biosensors based on aptamers, which exhibit exceptional recognition capability for a variety of targets. Aptasensors offer numerous advantages over conventional approaches, including increased sensitivity, selectivity, design flexibility, and cost-effectiveness. As a result, interest in developing aptasensors continues to expand. This paper discusses the historical and modern advancements of aptasensors through the use of nanotechnology to enhance the signal, resulting in high sensitivity and detection accuracy. More importantly, this review summarizes the principles and strategies underlying different organophosphorus aptasensors, including electrochemical, electrochemiluminescent, fluorescent, and colorimetric ones.
