Abstract:<p>The unique Raman spectra of various polymers render polymeric microbeads to be attractive in Raman‐based multiplex analysis and simultaneous biomolecular recognition as self‐encoded supports. The combination of polymer Raman signals with highly sensitive surface‐enhanced Raman scattering (SERS) codes will be able to significantly enhance the combinatory library in bioanalysis. However, there is lack of study on the preparation and spectroscopic analysis of self‐encoded microbeads with both Raman and SERS signatures. This study establishes a robust synthesis and spectral analysis protocol of the microbeads with bi‐codes, namely Raman and SERS. As modal examples, surface carboxylic acid functionalized monodisperse microbeads of polystyrene and poly(4‐tert‐butylstyrene) were conjugated with the amine group of 4‐aminothiophenol, followed by the adsorption of gold nanoparticles (AuNP) on microbead surfaces. The thiol group of 4‐aminothiophenol forms self‐assembled monolayers with AuNPs. The resulting microbeads are Raman and SERS dual self‐encoded as evident from their Raman spectroscopic and imaging analysis. The synthesis and spectroscopic analysis of the Raman and SERS dual‐encoded systems suggest the potential expansion of multiplex library in suspension bioassays.</p>

Abstract:Suspension microsphere immunoassays are rapidly gaining attention in multiplex bioassays. Accurate detection of multiple analytes from a single measurement is critical in modern bioanalysis, which always requires complex encoding systems. In this study, a novel bioassay with Raman-coded antibody supports (polymer microbeads with different Raman signatures) and surface-enhanced Raman scattering (SERS)-coded nanotags (organic thiols on a gold nanoparticle surface with different SERS signatures) was developed as a model fluorescent, label-free, bead-based multiplex immunoassay system. The developed homogeneous immunoassays included two surface-functionalized monodisperse Raman-coded microbeads of polystyrene and poly(4-tert-butylstyrene) as the immune solid supports, and two epitope modified nanotags (self-assembled 4-mercaptobenzoic acid or 3-mercaptopropionic acid on gold nanoparticles) as the SERS-coded reporters. Such multiplex Raman/SERS-based microsphere immunoassays could selectively identify specific paratope-epitope interactions from one mixture sample solution under a single laser illumination, and thus hold great promise in future suspension multiplex analysis for diverse biomedical applications.

Abstract:We analyze microstructured multilayer films of poly(ethyleneimine) (PEI) and DNA by employing Raman and Surface enhanced Raman spectroscopy (SERS). The microstructuring of the samples allows a simultaneous measurement of signal and reference in a single analytic process. Silver nanoparticles are implemented in the microstructured multilayers for SERS measurements. The recorded SERS spectra of PEI/DNA are dominated by the Raman bands of the DNA bases which show a larger mean enhancement than bands belonging to DNA backbone vibrations. Our results show that the combination of SERS and microstructured multilayer films provides an adapted way to characterize the polyelectrolytes as well as to measure the enhancement factor and the distance dependence for the SERS active silver nanoparticles. Furthermore, microstructured polyelectrolyte films containing SERS active nanoparticles are used for sensing molecules.

Abstract:A new type of optical probes, featuring surface enhanced Raman scattering (SERS) and fluorescence spectra dual-mode encoding, has been reported in this article. Based on the uniform micrometer-sized melamine resin/Ag nanoparticles (MRM/Ag-NPs) composite microspheres, the SERS reporters and fluorescent probes were successfully fixed onto the different layers of the MEM/Ag-NPs microspheres, which supported the sensitive DNA detecton. The two spectroscopic methods commonly considered to be contradictive to each other, yet the optical signals were separable in the experiments. The dual-encoding strategy and single microsphere detecton method put the number of available independent codes to be rough the multiple of those available in the two optical detection channels, which increases far more rapidly than the summation of the two channels. As a proof of cencept, the utility of this dual spectrum mode SERS-fluoresecence encoded microsphere (SFEM) was demonstrated in a specific DNA detection using complimentary ssDNA functionalized magnetic beads as the DNA capturing and separation agents. Excellent encoding results were demonstrated from the decoding of the SERS and fluorescence signals of the SFEM. The method appears to be general in scope and we expect that the SERS-fluoresecence encoded microspheres system is applicable to multiplex bioassays of a variety of biomolecules.

Abstract:In 1997, the discovery of single molecule-surface enhanced Raman spectroscopy (SM-SERS) rekindled broad interests owing to its ultrahigh enhancement factor up to the 1014-1015 level. However, regretfully, the advantage of SM-SERS with an ultralow detection limit has not yet been fully utilized in commercialized applications. Here, we report a strategy, which we name confined-enhanced Raman spectroscopy, in which the overall Raman properties can be remarkably improved with in situ-formed active nanoshell on the surface of silver or gold nanoparticles. The nanoshell can confine and anchor molecules onto the surface of plasmonic nanoparticles and avoid desorption from hot spots so that the "on and off"blinking effect can be eliminated. It is the first time the single-molecule detection of analytes with super sensitivity, high stability, and reproducibility based on gold nanoparticles has been realized. In addition, this strategy is suitable for SERS detection in diverse molecule systems, including biomedical diagnosis, catalytic reaction, etc.
Multicolor Raman Beads for Multiplexed Tumor Cell and Tissue Imaging and in Vivo Tumor Spectral Detection

Abstract:Developing new nanomaterials with strong and distinctive Raman vibrations in the biological Raman-silent region (1800-2800 cm<sup>-1</sup>) were highly desirable for Raman hyperspectral detection and imaging in living cells and animals. Herein, polymeric nanoparticles with monomers containing alkyne, cyanide, azide, and carbon-deuterate were prepared as Raman-active nanomaterials (Raman beads) for bioimaging applications. Intense Raman signals were obtained due to the high density of alkyne, cyanide, azide, and carbon-deuterate in single nanoparticles, in absence of metal (such as Au or Ag) as Raman enhancers. We have developed a library of Raman beads for frequency multiplexing through the end-capping substitutions of monomers and demonstrated five-color SRS imaging of mixed nanoparticles with distinct Raman frequencies. In addition, with further surface functionalization of targeting moieties (such as nucleic acid aptamers and targeting peptides), targetable Raman beads were successfully used as probes for tumor targeting and Raman spectroscopic detection, including multicolor SRS imaging in living tumor cells and tissues with high specificity. Further in vivo studies indicated that Raman beads anchored with targeting moieties were successfully employed to target tumors in living mice after tail intravenous injection, and Raman spectral detection of tumor in live mice was achieved only through spontaneous Raman signal at the biological Raman-silent region without any signal enhancement due to a high density of Raman reporters in Raman beads. With further copolymerization of these monomers, Raman beads with supermultiplex barcoding could be readily achieved.

Hydrogel-based Microbeads for Raman-encoded Suspension Array Using the Reversed-Phase Suspension Polymerization Method and Ultraviolet Light Curing

Abstract:A one-step synthesis using the reversed-phase suspension polymerization method and ultraviolet light curing is proposed for preparing the Raman-encoded suspension array (SA). The encoded microcarriers are prepared by doping the Raman reporter molecules into an aqueous phase, and then dispersing the aqueous phase in an oil phase and curing by ultraviolet light irradiation. The multiplexed biomolecule detection and various concentration experiments confirm the qualitative and quantitative analysis capabilities of the Raman-encoded SA with a limit of detection of 52.68 pM. The narrow bandwidth of the Raman spectrum can achieve a large number of codes in the available spectral range and the independence between the encoding channel and the fluorescent label channel provides the encoding method with high accuracy. This preparation method is simple and easy to operate, low in cost, and high in efficiency. A large number of hydrogel-based encoding microbeads could be quickly obtained with good biocompatibility. Most importantly, concentrating plenty of Raman reporter molecules inside the microbeads increases the signal intensity and means the molecular assembly is not limited by the functional groups; thus, the types of materials available for Raman encoding method are expanded. Furthermore, the signal intensity–related encoding method is verified by doping different proportions of Raman reporter molecules with our proposed synthesis method, which further increases the detection throughput of Raman-encoded SA.

Abstract:Bead-based assays have drawn more and more attention in biomedical fields. Herein, we proposed a novel approach to achieve multiplex analysis on a single porous hydrogel bead (PHB) with Raman dyes (RDs) encoded core-shell surface-enhanced Raman scattering (SERS) nanotags. Because of the amplified signal of RDs by core-shell metal structure of the nanotag and the high surface area to volume ratio (SVR) of the PHB, the sensitivity and linear dynamic range (LDR) of the as-proposed multiplex analysis method are significantly improved. We anticipate this approach to be widely used in the multiplex assays and biosensors.

Abstract:The rapid growth of demand for high-throughput multiplexed biochips from modern biotechnology has led to growing interest in suspension array based on multi-channel encoded microbeads. We prepare dual-spectra encoded PEGDA microbeads (DSEPM) by reversed-phase microemulsion UV curing method and layer-by-layer electrostatic self-assembly method. Excitation of the synthesized DSEPM results in two spectra, including fluorescence spectra from quantum dots and laser induced breakdown spectra from nanoparticles with specific elements. With further surface modification and bio-probes grafting, we use DSEPM to carry a series of detection experiments of biomolecules. The adsorption experiment to two types of anti-IgG in mixture sample has demonstrated the availability of DSEPM in multiplexing. Then, the contrast experiment has verified the specificity of DSEPM in detection. Finally, we carry out the concentration gradient experiment and obtain the response curve to show the performance of DSEPM in quantitative analysis. The results indicate our method provide an effective way to improve multiplexed biochips with more coding capacity, accuracy and stability.

Abstract:A new type of polymer core and silver nanoparticle/silica shell multifunctional composite microsphere is reported. The monodisperse polymer core of poly(styrene-co-acrylic acid), prepared by surfactant-free emulsion polymerization, serves as the supporting template to facilitate the synthesis and handling of the highly functionalized substrates to achieve consistent mechanical and surface chemical properties. The shell of silver nanoparticles, formed in situ by controlled interfacial reduction of AgNO3 with polyvinylpyrrolidone (PVP), serves as the optimal metal enhancer for surface enhanced Raman spectroscopy (SERS). The Ag nanoparticle shell is stabilized with a silica layer using a sol-gel process, which is also functionalizable with reactive epoxides for further bio-conjugation using epoxy ring-opening methods. Preliminary results indicated that the microspheres are easily prepared with exceptional chemical and physical uniformity. SERS experiments with 4-aminobenzenethiol (4-ABT) as the indicator showed that the resulting multifunctional microspheres allowed the production of highly consistent enhancement of the Raman signals down to nM concentrations of 4-ABT, making them highly desirable candidates as the enhancers for high performance SERS analysis and as SERS optical labels in biomedical imaging. The microspheres are robust and the high density of the SERS active Ag nanoparticles on the microspheres provided unique signal averaging for the method to be potentially quantitative which is an urgent goal for SERS substrate design and SERS analysis.

Abstract:The surface enhanced Raman scattering (SERS) reporters are rather limited, and the Raman peaks still overlap in varying degrees, making SERS multiplex coding a critical bottleneck in the exploration of SERS nanotechnology. Herein, we design a general strategy to expand the SERS probe scope to 26 probes of six types, which can be further expanded within a limited range, with stable performance and structure. By constructing (Au-aggregate)@ and (Au-aggregate)@silica nanocomposites, we develop optimal enhancement strategies for each Raman molecules. Mixed signal-ligand SERS probes improve the complexity of Raman spectra and expand the coding capacity. By integrating the strategies, SERS inks are produced and applied in anti-counterfeiting. With these improvements, this work breaks the constrains of probe selection, bringing SERS one step closer to the sensor or anti-counterfeiting application.

Abstract:In this study, a class of surface enhanced Raman spectroscopy (SERS) encoded core-shell nanospheres was synthesized as nano-SERS-tags for detecting specific DNA targets based on the sandwich hybridization assays. These core-shell nanospheres were synthesized by first depositing a layer of Ag-NPs (nanoparticles) onto the poly(styrene-co-acrylic acid) core and then the formation of a layer of uniform silica as the outer shell. The Ag-NPs served as SERS substrates with Raman active molecular probes adsorbed onto the Ag-NPs as indicative SERS molecular barcodes, and the silica coating shell was used for protecting the Ag-NPs and the Raman molecules from the exterior chemical and biological interference. The silica surfaces of nano-SERS-tags were further conjugated with probe DNA (pDNA) (nano-SERS-probes). The detection of single-stranded oligonucleotide (ssDNA) targets was successfully accomplished using the nano-SERS-probes in a chip-based sandwich hybridization assay in a mixed ssDNA target solution. The as-prepared nano-SERS-probes exhibited high chemical stability during the laser SERS experiments and the results were reproducible after a long-term storage. At least four different tags (a four "color" system) were quantitatively differentiated when simultaneously applied in the assays, indicating an excellent multiplexing potential of the method. Therefore, the as-prepared nano-SERS-probes are suitable for high specific detection of biomolecules with high sensitivity and remarkable multiplexing capability associated with the SERS method.

Abstract:Abstract The utilization of nanoparticle-polymer bead hybrid nanostructures as a SERS substrate depends on the control of the deposition, density, and distribution of nanoparticles on the bead surface. Here we demonstrate the fabrication of a large area SERS substate via a two- step DNA mediated assembly of gold nanoprisms and polystyrene (PS) beads into a large ensemble of beads that are densely coated with nanoprisms. First, nanoprisms are loaded on PS beads through DNA hybridization. The close packed arrangement of anisotropic nanoprisms in different orientations on a bead surface results in a plasmonic substrate with a variable nanogap size ranging 1–20 nm. Nanoprisms-coated beads are then assembled into a large stack or aggregate of beads using a DNA-induced crystallization approach. Each aggregate consists of 20–50 nanoprisms-coated beads, leading to the formation a large area of three-dimensional SERS substrate with a high-density of hot spots for SERS enhancement. An excellent enhancement factor (EF) of 1.09 × 10 5 and a very high detection sensitivity (up to 10 –10 M) are observed for the analysis of a probe molecule (Methylene blue) using the SERS substrate.

Abstract:We have developed a method to fabricate higher order surface enhanced Raman scattering (SERS) active structure with gold-decorated polystyrene microparticles (Au/PS microparticles) to serve a wide range of applications in life science and environmental analysis using alternating-current dielectrophoresis (DEP). It is within several tens of seconds to form or gather the SERS—active ‘hot spots’ and analyte. The immobilization of Au/PS microparticles can be demonstrated and qualitatively explained by DLVO and DEP calculation. Our method enables us to detect 4, 4′-bipyridine (4bpy) at 100pM by an easy operation of in situ SERS platform using fiber-type simple Raman spectrometer. This platform can easily generate SERS-active ‘hot spots’ with SERS enhancement factor ∼ 10 8 in specific region on demand.

Abstract:A quasi-periodic array of 3D gold-nanoparticle-capped SiO2 microspheres (Au@SiO2) was designed and prepared with a facile approach to enhance the Raman signal intensity of adsorbed biomolecules. Through adjusting the thickness and annealing of Au thin films initially deposited on arrays of self-assembled SiO2 microspheres, we were able to control the diameter of Au nanoparticles and their interparticle spacing to produce two types of plasmonic near-field hot spots, locating at the gaps of such densely arranged Au nanoparticles on individual SiO2 microspheres and in the gap regions of neighboring SiO2 microspheres, respectively. Such double near-field enhancement mechanism leads to a surface-enhanced Raman scattering (SERS) enhancement factor up to 3×106 for Rhodamine 6G molecules. The SERS signal intensity was highly uniform with a relative standard deviation of 4.5%. This 3D SERS substrate has significant potential for various applications in the field of SERS detection of analytes and wearable biosensing.

Abstract:Surface-enhanced Raman spectroscopy (SERS) can boost the pristine Raman signal significantly which could be exploited for producing innovative sensing devices with advanced properties. However, the inherent complexity of SERS systems restricts their further applications in rapid detection, especially in situ detection in narrow areas. Here, we construct an efficient and flexible SERS-based Lab-on-Fiber (LOF) sensor by integrating Ag/Au nanocap arrays obtained by Ag/Au coating polystyrene nanospheres on the optical fiber face. We obtain rich "hot spots" at the nanogaps between neighboring nanocaps, and further achieve SERS performance with the assistance of laser-induced thermophoresis on the metal film that can achieve efficiency aggregation of detected molecules. We achieve a high Raman enhancement with a low detection limitation of 10−7 mol/L for the most efficient samples based on the above sensor. This sensor also exhibits good repeatability and stability under multiple detections, revealing the potential application for in situ detection based on the reflexivity of the optical fiber.

Abstract:A novel nanoparticle-based imaging strategy is introduced that couples biocompatible organic nanoparticles and stimulated Raman scattering (SRS) microscopy. Polymer nanoparticles with vibrational labels incorporated were readily prepared for multi-color SRS imaging with excellent photo-stability. The Raman-active polymer dots are nontoxic, rapidly enter various cell types, and are applied in multiplexed cell-type sorting.
Ultrasensitive multiplex SERS immunoassay based on porous Au-Ag alloy nanoparticle-amplified Raman signal probe and encoded photonic crystal beads

Abstract:An ultrasensitive multiplex surface-enhanced Raman scattering (SERS) immunoassay was developed using porous Au-Ag alloy nanoparticles (p-AuAg NPs) as Raman signal amplification probe coupling with encoded photonic crystal microsphere. p-AuAg NPs were synthesized and modified with the second antibody (Ab2) and Raman tag (mercaptobenzoic acid, MBA) to prepare a Raman signal-amplified probe. The high porosity of the p-AuAg NPs enables significant coupling of the localized surface plasmon resonance and thus abundant inherent hotspots for Raman signal enhancement. 3D-ordered silver nanoparticles-coated silica photonic crystal beads (Ag/SPCBs) were prepared as encoded SERS substrate for multiplex detection using their reflection peaks. The signal-amplified probe was used for multiplex detection of tumor markers carcinoembryonic antigen (CEA) and alpha fetoprotein (AFP). The wide linear ranges of 10-7-103 ng/mL for CEA and 10-4-103 ng/mL for AFP with detection limits of 1.22 × 10-8 ng/mL and 2.47 × 10-5 ng/mL for CEA and AFP at a signal-to-noise ratio of 3 were obtained. The proposed multiplex SERS immunoassay method displays ultrahigh sensitivity, wide linear range, and excellent specificity, which can be successfully applied to measure clinical serum samples with satisfactory results. The research provides a novel SERS signal enhancement strategy for the multiplex bioassay.
