Abstract:To sensitively detect multiple and cross-species disease-related targets from a single biological sample in a quick and reliable manner is of high importance in accurately diagnosing and monitoring diseases. Herein, a surface-enhanced Raman scattering (SERS) sensor based on a functionalized multiple-armed tetrahedral DNA nanostructure (FMTDN) immobilized silver nanorod (AgNR) array substrate and Au nanoparticle (AuNP) SERS tags is constructed to achieve both multiplex detection and enhanced sensitivity using a sandwich strategy. The sensor can achieve single, dual, and triple biomarker detections of three lung cancer-related nucleic acid and protein biomarkers, i.e., miRNA-21, miRNA-486 and carcinoembryonic antigen (CEA) in human serum. The enhanced SERS signals in multiplex detections are due to the DNA self-assembled AuNP clusters on the silver nanorod array during the assay, and the experimentally obtained relative enhancement factor ratios, 150 for AuNP dimers and 840 for AuNP trimers, qualitatively agree with the numerically calculated local electric field enhancements. The proposed FMTDN-functionalized AgNR SERS sensor is capable of multiplex and cross-species detection of nucleic acid and protein biomarkers with improved sensitivity, which has great potential for the screening and clinical diagnosis of cancer in the early stage.

Abstract:Surface-enhanced Raman scattering (SERS) enables rapid detection of single molecules with high specificity. However, quantitative and sensitive SERS analysis has been a challenge due to the lack of reliable SERS-active materials. In this study, we developed a quantitative SERS-based immunoassay using enzyme-guided Ag growth on Raman labeling compound (RLC)-immobilized gold nanoparticle (Au NP)-assembled silica NPs (SiO2@Au-RLC@Ag). The enzyme amplified Ag+ reduction as well as Ag growth on the RLC-immobilized Au NP-assembled silica NPs (SiO2@Au-RLC), which resulted in a significant increase in SERS signal. In the presence of target antigens such as immunoglobulinG (IgG) or prostate-specific antigen (PSA), Ab1-Antigen-Ab2 immune complex with alkaline phosphatase triggered an enzyme- catalyzed reaction to convert 2-phospho-L-ascorbic acid (2-phospho-L-AA) to ascorbic acid (AA). As produced AA reduced Ag+ to Ag, forming an Ag hot spot on the surface of SiO2@Au-RLC, which enhanced the SERS signal of SiO2@Au-RLC@Ag in a solution with a target antigen concentration. The plasmonic immunoassay for IgG detection showed a high linearity of SERS intensity in the range of 0.6 to 9.0 ng/mL with a detection limit (LOD) of 0.09 ng/mL, while an LOD of 0.006 ng/mL was obtained for PSA. The results indicate that the sensitivity of our novel SERS-based immunoassay is higher than that of conventional enzyme-based colorimetric immunoassays.

Abstract:Among various biomimetic polymer materials, polydimethylsiloxane (PDMS) stands out as an ideal matrix for surface-enhanced Raman scattering (SERS) due to its unique intrinsic Raman signal and tenacity. In order to realize the precise detection of prostate-specific antigen (PSA), we proposed a sandwich-type SERS-active immunostructure composed of PDMS@silver nanoparticles (Ag NPs)@ZIF-67 biomimetic film as the immunosubstrate and gold nanorods (Au NRs) as immunoprobes. Due to the synergistic effect of electromagnetic enhancement facilitated by biomimetic surfaces and chemical enhancement achieved by ZIF-67, this structure enabled an ultrasensitive and selective detection of PSA across a broad range from 10 −3 to 10 −9 mg/mL. The achieved limit of detection was as low as 3.0 × 10 −10 mg/mL. Particularly, the intrinsic Raman signal of PDMS matrix at 2905 cm −1 was employed as a potential internal standard (IS) in the detection, achieving a high coefficient of determination (R 2 ) value of 0.996. This multifunctional SERS substrate-mediated immunoassay holds vast potential for early diagnosis of prostate cancer, offering promising prospects for clinical applications.

Abstract:Biomarker testing plays a crucial role in the early detection of liver cancer. Herein, we developed a dual-signal amplification approach utilizing magnetic aggregation and a recognition competition strategy for the simultaneous detection of alpha-fetoprotein (AFP) and manganese superoxide dismutase (MnSOD) in serum. 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 were synthesized by functionalizing Raman signaling molecules and aptamer complementary chains onto the surface of gold nanoparticles (AuNPs). The detection complex Raman signal molecule@AuNPs@H-Fe3O4@cDNA was assembled by conjugating 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 with two nucleic acid aptamers (cDNA1 and cDNA2) modified with Fe3O4 through partial base complementary pairing. The target protein exhibited specific binding with the aptamer, leading to the competitive displacement of 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 from the Fe3O4 array surface, consequently resulting in a reduction of the Surface-Enhanced Raman Spectroscopy (SERS) signal. Through this approach, the limit of detection (LOD) for AFP and MnSOD in serum was achieved at remarkably low levels of 5.89 pg/mL and 6.23 pg/mL, respectively, with a reaction incubation period of only 5 min. Finally, the platform was utilized for the quantification of AFP and MnSOD in the serum of a nude mouse hepatocellular carcinoma model. The results obtained through SERS were consistent with those from enzyme-linked immunosorbent assay (ELISA), validating its accuracy. This methodology presents a novel approach for the swift and concurrent detection of proteins, holding significant clinical promise for the early diagnosis of hepatocellular carcinoma.
Ag@Au Core–Shell Porous Nanocages with Outstanding SERS Activity for Highly Sensitive SERS Immunoassay

Abstract:A highly sensitive immunoassay of biomarkers has been achieved using 4-mercaptobenzoic acid-labeled Ag@Au core–shell porous nanocage tags and α-fetoprotein immuno-sensing chips. The Ag@Au porous nanocages were uniquely synthesized by using an Ag core as a self-sacrificial template and reducing agent, where the slow reaction process led to the formation of a porous Au layer. The size of the remaining Ag core and surface roughness of the Au shell were controlled by adjusting the chloroauric acid concentration. The porous cage exhibited excellent surface-enhanced Raman spectroscopy (SERS) activity, presumably due to a synergetic interaction between newly generated hot spots in the rough Au shell and the retained SERS activity of the Ag core. Using α-fetoprotein as a model analyte for immunoassay, the SERS signal had a wide linear range of 0.20 ng mL−1 to 500.0 ng mL−1 with a detection limit of 0.12 ng mL−1. Without the need of further signal amplification, the as-prepared Ag@Au bimetallic nanocages can be directly used for highly sensitive SERS assays of other biomarkers in biomedical research, diagnostics, etc.

Abstract:Sensitive surface-enhanced Raman scattering (SERS) assays of glycoproteins have been proposed using p-aminothiophenol (PATP)-embedded Ag core-Au satellite nanostructures modified with p-mercaptophenylboronic acid (PMBA) and the self-assembled monolayer of PMBA on a smooth gold-coated wafer. The apparent Raman probe PATP on the surfaces of the Ag cores underwent a photodimerization to generate 4,4'-dimercaptoazobenzene (DMAB) in situ upon excitation of laser, and the in situ-generated DMAB acted as the actual Raman probe with considerably strong SERS signals, which was further enhanced by the plasmonic coupling of the Ag core-Au satellite nanostructures due to the synergistic effect. The sandwich assays of glycoproteins showed high sensitivity and excellent selectivity against non-glycoproteins. The Ag core-Au satellite SERS nanostructures can be used for high sensitive SERS assays of other analytes.
Hospital type and patient outcomes: an empirical examination using AMI readmission and mortality records.

Abstract:This paper investigates whether there are differences in patient outcomes across different types of hospitals using patient-level data on readmission and mortality associated with acute myocardial infarction (AMI). Hospitals are grouped according to their ownership type (private, public teaching, public non-teaching) and their location (metropolitan, country and remote country). Using data collected from 130 Victorian hospitals on 19,000 patients admitted to a hospital with their first AMI between January 2001 and December 2003, we consider how the likelihood of unplanned re-admission and mortality varies across hospital type. We find that there are significant differences across hospital types in the observed patient outcomes - private hospitals persistently outperform public hospitals.

Abstract:A multiplexed electrochemical immunoassay method was developed for simultaneous ultrasensitive measurement of tumor markers based on electrochemical stripping analysis of silver nanoparticles (Ag NPs). The Ag NPs were deposited on a disposable immunosensor array with a reduction reaction catalyzed by nanogold labels. The immunosensor array was prepared by covalently immobilizing capture antibodies on chitosan modified screen-printed carbon electrodes. Through a sandwich-type immunoreaction, antibody-functionalized Au NPs were captured onto immunosensor surface to induce the silver deposition from a silver enhancer solution. The deposited Ag NPs could be directly measured by anodic stripping analysis in KCl solution. The catalytic deposition enhanced the analytical sensitivity for detection of protein markers. The interference of dissolved oxygen could be avoided as the detection was performed with positive stripping potential range. Using carcinoembryonic antigen and α-fetoprotein as model analytes, the proposed multiplexed immunoassay method showed wide linear ranges of three orders of magnitude with the detection limits down to 3.5 and 3.9pgmL−1, respectively. The localized silver deposition, as well as the stripping detection process, eliminated completely the electrochemical cross talk between adjacent immunosensors. The immunosensor array exhibited acceptable reproducibility, stability and accuracy, showing a promising potential in multianalyte determination for clinical application.

Abstract:The selective discrimination of multiple microRNAs (miRNAs) in biological samples holds great potential for the early diagnosis of various cancers. In the present study, we present an extremely sensitive surface-enhanced Raman scattering (SERS) assay for multiplex detection of cancer-related miRNAs-miR-141, miR-429 and miR-200b-derived from the miR-200 family, using three pairs of silica-coated, analyte-embedded Au nanoparticles (SA@GNPs) and Au-coated paramagnetic nanoparticles (Au@MNPs) conjugated with DNA probes specific for each miRNA target. The SA@GNPs with an optimal silica shell (similar to 3 nm) were prepared to provide an effective barrier to prevent the protein-induced disassociation of Raman analytes from the GNPs, therefore allowing more reproducible SERS signaling than uncoated GNPs in serum solution. Moreover, this thin silica shell minimized the interparticle distance between plasmonic nanostructures in dense magnetic concentrates, significantly impacting the formation of hot spots for SERS enhancement. The target miRNA detection was achieved by SERS excitation of magnetically concentrated SA@GNPs/target miRNA/Au@MNP complexes, which are formed by target-specific hybridization reactions. The limit of detection (LOD) of miRNAs in human serum was 120 fM. The discrimination of miR-141, miR-429, and miR-200b in multiplex detection was realized by identifying diagnostic peaks of analytes in mixed Raman spectra. Due to the high-level detection sensitivity and assay reproducibility, the proposed SERS assay offers opportunities for multiplex detection of serum miRNAs and cancer diagnosis in a clinical setting.

Abstract:It is tremendously desirable for the timely and effective detection of cancer to facilitate the ultra-highly sensitive monitoring of tumor marker in clinical serum sample. In this study, an electromagnetic and chemical synergistically enabled recyclable immunoassay based on surface-enhanced Raman scattering (SERS) was proposed by realizing the anisotropic growth of sea-urchin-like gold nanoflowers (Au NFs) on two-dimensional red phosphorus (RP) nanoplates. Besides the achieved enhancement factor as high as 2.24 × 106, it was found that the photocatalytic and SERS activities were kept at a high level for the hybrid substrate of RP/Au NFs throughout 7 cycles of immunoassay. In combination with a non-metallic immunoprobe, the limit of detection was drove to 7.41 × 10−5 IU·mL−1 for cancer antigen 19–9. The comparative experiments of nonspecific monitoring verified the promising selectivity of this strategy. Considering the intriguing features of high sensitivity, recyclability, and specificity, the proposed multifunctional RP/Au NFs exhibited its superior role in the early detection of cancer and can be adapted for point-of-care diagnosis.

Abstract:A sensitive dual immunoassay was proposed for the determination of carcinoembryonic antigen (CEA) and α-fetoprotein (AFP) based on signal amplification. Monoclonal antibodies immobilized on magnetic mesoporous silica particles (Fe_3O_4/SiO_2) were prepared as the primary probe. Horseradish peroxidase (HRP) labeled antibodies co-coated with HRP on gold nanoparticles (AuNPs) were used as the secondary probe to achieve signal amplification. HRP tags were retained in the flow cells after a sandwich immunoassay. By controlling two switches on the two channels, chemiluminescent substrates were injected orderly man way, and then signals for CEA and AFP were sequentially detected by HRP-luminol-H_2O_2. Due to the increased amount of HRP on AuNPs and the increased amount of monoclonal antibodies on Fe_3O_4/SiO_2, the signals were largely amplified. Under the optimal conditions, CEA and AFP could be detected in the linear ranges of 1.0 – 80 and 1.0 – 75 ng mL^−1 with detection limits of 0.25 and 0.5 ng mL^−1, respectively.

Does Packing the Eustachian Tube Impact Cerebrospin al Fluid Rhinorrhea Rates in Translabyrinthine Vestibular Schwannoma Resections?

Abstract:Objective: To calculate cerebrospinal fluid (CSF) leak rates for translabyrinthine (TL), middle cranial fossa (MCF), and retrosigmoid/suboccipital (SO) craniotomies performed for removal of vestibular schwannoma (VS) and analyze whether packing the eustachian tube (ET) in TL VS resections impacts CSF rhinorrhea rates. Study Design: Retrospective. Setting: Tertiary care center. Methods: Chart review. Results: Three hundred fifty-nine VS resections were reviewed in 356 patients ranging from 10 to 86 years of age. Two hundred thirty-one TL, 70 MCF, 53 SO, and 5 combined TL/SO procedures were analyzed. Total CSF leak rates (incisional, otorrhea, and rhinorrhea) were 14.2% for TL, 11.4% for MCF, and 13.2% for SO approaches. Differences in overall CSF leak rates were not statistically significant. For those who underwent TL craniotomies, 2 groups of patients were identified based on whether their ETs were packed during surgery. In 1 group, the incus was removed, the aditus enlarged, the ET packed, and the middle ear filled with muscle. In the second group, the aditus, epitympanum and middle ear were packed without removing the incus, and the ET was not packed. Of 148 patients who had their ET packed, 12 developed CSF rhinorrhea (8.1%). The CSF rhinorrhea rate for patients who did not have ET packing was 5.9% (3 of 51 patients). This difference was not statistically significant (p = 0.80). When Proplast was used to pack the ET (121 patients), the CSF rhinorrhea rate was 5.8%. Unfortunately, this material extruded in 4 of 121 patients (3.3%) and presented clinically as delayed purulent otorrhea. Conclusion: Cerebrospinal fluid leak rates were similar in patients undergoing TL, SO, and MCF approaches, and CSF rhinorrhea was not decreased by ET packing. Patients whose ETs are packed with Proplast are at risk for extrusion and otorrhea years after their initial VS resection.

Abstract:An ultrasensitive enzyme-free electrochemical immunoassay was developed for detection of the fg/mL level carcinoembryonic antigen (CEA) by using a double strand DNA@Au nanoparticle (dsDNA@AuNP) tag and hexaammineruthenium(III) chloride (RuHex) as the electroactive indicator. The dsDNA@AuNP was synthesized by one-pot hybrid polymerization of dsDNA on initiator DNA modified AuNPs via hybridization chain reaction. The immunosensor was prepared by covalently cross-linking capture antibody on chitosan/AuNP nanocomposite modified glass carbon electrode. The AuNPs accelerated the electron transfer and led to high detection sensitivity. With a sandwich-type immunoreaction and a biotin-streptavidin affinity reaction, the dsDNA@AuNP tag was conjugated on the immunocomplex to bring a high amount of RuHex to the electrode surface via electrostatic interaction, resulting in an amplified electrochemical signal. Under optimal conditions, the proposed sensing platform showed a wide linear detection range from 10 fg/mL to 10 ng/mL along with a detection limit of 3.2 fg/mL for CEA. The immunosensor exhibited high sensitivity and good stability, showing a promising application in early cancer diagnosis and could be extended to sensitive electrochemical biosensing of other analytes.

Abstract:Quantitative molecular detection based on surface-enhanced Raman spectroscopy (SERS) is still a great challenge because of the highly nonuniform distribution of the SERS hot spots and the nondeterministic spatial and spectral overlap of the analyte with the hot spot. Here, we report a nanoparticle-on-mirror plasmonic sensor excited by surface plasmon polaritons for quantitative SERS detection of alpha fetoprotein in serum with ultrahigh sensitivity. The uniform gaps between the nanoparticles and gold film and the alignment of the gap modes relative to the excitation electric field endow this substrate with a uniform and strong SERS enhancement. The limit of detection reaches 1.45 fM, 697 times higher than that under normal excitation and 7800 times higher than a commercial enzyme-linked immunosorbent assay kit. This approach offers a potential solution to overcome the bottleneck in the field of SERS-based biosensing.

Abstract:The design of a rapid, simple, and ultrasensitive detection platform of protein biomarkers has become an urgent need in early diagnosis and treatment. Surface-enhanced Raman scattering (SERS) technology, with the unique advantages of ultra-sensitivity and fingerprint recognition, has attracted much attention in biomarker detection. An innovative aptamer SERS biosensor for highly sensitive protein biomarker detection was presented. This biosensor contains a SERS tag (reporter-labeled Au nano-bridged nanogaps particles, Au NNPs) and novel magnetic capture substrate (Ag-coated Fe3O4-Au NPs, Ag MNPs). C-reactive protein (CRP), a representative biomarker for diagnosing, was used to assess the SERS performance of this biosensor. Aptamers against CRP were modified on both SERS tag and magnetic capture substrate for specific recognition by Au NNPs-CRP-Ag MNPs sandwich assay. This strategy achieved sensitive detection of CRP with a limit of detection (LOD) of 10 fM (1.14 pg/mL), and is much lower than reported analytical methods. Besides, this method exhibits excellent selectivity and specificity for CRP under the interference of other proteins and displays high accuracy in the detection of actual human serum samples. This proposed detection strategy can be extended to the sensitive detection of other biomarker proteins and hold great promise in the clinical diagnosis of diseases.
A fluorescent platform integrated with a "one-pot" nicking endonuclease signal amplification and magnetic separation for simultaneous detection of tumor markers

Abstract:Although Enzyme-linked immunosorbent assay (ELISA) has been widely used for biomedical research, simultaneous sensitive and cost-effective detection of multiple biomarkers is challenging. Herein, we proposed a "one-pot" nicking endonuclease signal amplification (NESA)-based fluorescent aptasensor for simultaneous detection of carcinoembryonic antigen (CEA) and alpha fetoprotein (AFP). Firstly, two aptamers were synchronously immobilized on the surface of magnetic nanoparticles (MNPs) by coupling with two complementary DNA (cDNA). CEA and AFP specifically recognized the aptamers and then the released cDNA (ssDNA) from the double-strands (dsDNA) triggers NESA, further breaking two detection probes which were labeled with the fluorescent dye (FAM and ROX) and its quencher (BHQ1 and BHQ2) at the same time. Then, the fluorescence signal of FAM and ROX were restored separately. The results indicated that the fluorescence intensity at the emission wavelength of 518 nm and 610 nm had a positive correlation with CEA and AFP concentrations, respectively. Under the optimum conditions, wider liner range of 1-500 ng mL-1 to CEA and 5-800 ng mL-1 to AFP of this fluorescent aptasensor were successfully obtained, achieving a detection limit of CEA and AFP were 0.7 ng mL-1 and 2 ng mL-1, respectively. Hence, it turned out that the aptasensor strategy can be a promising candidate for developing a newly fluorescence assay for the simultaneous quantitative detection of multiple tumor markers in matrix samples by changing the corresponding sequences of aptamer and fluorescent signal probe, which has great potential for the screening of early cancer.

Abstract:A surface-enhanced Raman scattering (SERS) technique shows extraordinary features for a range of biological and biomedical applications. Herein, a series of novel bioorthogonal SERS nanoprobes were constructed with Gold nanoflower (AuNF) and Raman reporters, the signals of which were located in a Raman-silent region of biological samples. AS1411 aptamer was also co-conjugated with AuNF through a self-assembled monolayer coverage strategy. Multiplex SERS imaging using these nanoprobes with three different bioorthogonal small-molecule Raman reporters is successfully achieved with high multiplexing capacity in a biologically Raman-silent region. These Raman nanoprobes co-conjugated with AS1411 showed high affinity for tumor cells with overexpressed nucleolin and can be used for selective tumor cell screening and tissue imaging.
Gold nanoring core-shell satellites with abundant built-in hotspots and great analyte penetration: An immunoassay platform for the SERS/fluorescence-based detection of carcinoembryonic antigen

Abstract:We designed an ingenious plasmonic core-shell-satellite nanostructure (2-AuNR) including a hollow porous gold nanoshell (HPGNP) core, a protective silica shell with mesoporous (mSiO2) interlayer, and a high level of gold nanoring (AuNR) satellite coverage by in situ etching. The HPGNP core modified with 4-mercaptobenzoic acid (4-MBA) possessed relatively strong local-field enhancement. The mSiO2 coating acted as a continuous interior nanogap and protective shell, and the variable external satellites in the morphology had tunable optical responses and designed "hotspot" regions. Specifically, AuNR satellites exhibited superior plasmonic properties, intense core-satellite plasmon coupling with the HPGNP core, and favorable biocompatibility. Meanwhile, the nanoassemblies had larger surface-to-volume ratios for molecular modification and were penetrable for analyte. With their abundant built-in hotspots and less hindrance for analyte penetration into the hollow interior, the labeled nanoassemblies were successfully used in SERS/fluorescence sensing system for carcinoembryonic antigen (CEA) detection. These immunoprobes with high SERS activity could be used in SERS-based immunoassay for CEA detection with a linear response ranging from 1 pg/mL to 1000 pg/mL. CEA detection in the fluorescence sensing system showed that the fluorescence quenching efficiency gradually decreased as the CEA level increased, with a linear response of 0.5–400 pg/mL. This SERS/fluorescence-based immunoassay had the same CEA detection limit of 0.1 pg/mL. The fine-engineered penetrable nanoassemblies with Au ring satellites can provide an insight into the rational and optimal design of multifunctional nanostructures. They can also broaden the field of vision for the design of multifunctional sensing probes in biochemical detection.

Abstract:A novel electrochemical magnetoimmunosensor for the rapid and sensitive detection of carcinoembryonic antigen (CEA) was fabricated based on a combination of high-efficiency immunomagnetic separation, bifunctional Au-nanoparticle (bi-AuNP) probes, and enzyme catalytic amplification. The reaction carrier magnetic beads (MBs) effectively reduced the toxicity of the complex sample to the working electrode, and the signal carrier bi-AuNP probes loaded a large amount of signal molecules, both of which enhanced the signal-to-noise ratio and further improved the detection sensitivity. A detection limit as low as 0.11 pg/mL was achieved for CEA detection based on the immunomagnetic separation and bi-AuNP probe-based multiamplification strategy, and the strategy was further successfully applied in human serum samples. The transducer was regenerated via a simple washing procedure, which enabled the detection of all samples on a single electrode with high reproducibility. The proposed strategy, which has the merits of high sensitivity, selectivity, and reproducibility exhibits great potential for detection in complex samples.
