Enhanced conversion of corncob into furfurylamine via chemoenzymatic cascade catalysis in a toluene–water medium

Abstract:Furfurylamine is recognized as an attractive platform molecule for production of fibers, perfumes, additives, medicines and agrochemicals. It can be synthesized from biomacromolecule-derived furfural by ω-transaminase ( At AT) from Aspergillus terreus . To efficiently transform biomacromolecule (xylan in lignocellulosic biomass) into furfurylamine, a hybrid strategy by bridging biomass-based heterogeneous catalyst and robust mutant At AT biocatalyst was attempted to transform biomass in a toluene–water medium. Using rice stalk (RS) as carrier, tin-based solid acid catalyst Sn-RS was prepared to catalyze corncob (75 g/L) into furfural (121 mM FAL, 67.8% yield) at 170 °C in a toluene–water (2:8, v/v) medium after 20 min. Moreover, recombinant E. coli NDTS was constructed by mutating Asparagine to Aspartic acid at position 245 in At AT using site-directed mutation strategy. 200 mM furfural could be biologically aminated to furfurylamine (92.4% yield) with robust ω- transaminases biocatalyst (NDTS cell). Furfural derived from corncob could be fully aminated to furfurylamine with NDTS cell at 35 °C in toluene–water, achieving the productivity of 0.54 g furfurylamine/(g xylan). This work provides an efficient, green and sustainable approach for manufacturing value-added furan-based chemicals.
