Abstract:Real-time quantification of glutathione (GSH) concentrations in living cells is essential for understanding GSH-related pathological events. However, most existing fluorescent probes are not suitable for this purpose due to their irreversible response mechanisms. Only a few probes have been successfully developed to realize this goal to date. Herein, we presented a fluorescent probe (QG-S) for quantifying cellular GSH and monitoring GSH dynamic variations. The probe QG-S was successfully employed in real-time monitoring of the dynamic variations of GSH in living cells upon reversible treatment with a GSH inhibitor (N-methylmaleimide) by using confocal microscopy. Moreover, the probe QG-S showed extremely low cytotoxicity and was facilely used to quantitatively determine the GSH concentration (4.25 mM) in living HeLa cells.

Abstract:A novel photoconvertible fluorescent probe, which can be activated by intracellular thiols, has been synthesized. Such a molecular probe comprises three parts: a 7-aminocoumarin phototrigger, a thiol-removable energy acceptor, and a caged fluorescein scaffold with intracellular thiols reactivity as the fluorescent reporter. Extracellularly, the energy acceptor blocks the emission of the coumarin that regulates the photocleavage and photoactivation of the fluorescein. Intracelluarly, the high concentration of thiols releases the energy acceptor, thus activating the S1 state of the phototrigger, which emits coumarin blue fluorescence for pre-visualization and liberates the caged green-fluorescent fluorescein to highlight the specific cell upon illumination. Compared to traditional photoactivated organic dyes, the intracellular thiols activated probe requires double activations: one by intracellular thiols and the other by light activation. The dual activations restrict fluorescence precisely inside live cells and at the particular spatial region of light activation, thus a probe with precise spatial accuracy in live cells.

Abstract:In the last few decades, growing numbers of fluorescent probes have been developed to detect intracellular GSH. However, the majority of probes for GSH were irreversible without monitoring the changes of intracellular GSH concentration. Therefore, recently, fluorescent probes for monitoring concentrations of GSH in real-time in living cells have come into being to address this challenge. This Perspective aimed at the development of reversible probes for GSH was organized by structural features, chemical reactions, and physicochemical properties. The reversible probes designed by a coumarin skeleton as a read-out fluorophore and the Michael addition reaction as a response mechanism accounted for most of the reported reversible probes. The performances of reversible fluorescent probes based on Michael addition could be roughly predicted by fundamental laws of kinetics and thermodynamics in physical chemistry. Essentially, the design principles included a highly reactive site for GSH, a small thermodynamic driving force, a desirable Kd of 1–10 mM, and excellent cell membrane permeability. Prospectively, the development of various mechanisms and fluorophores will be effective measures to enrich the types of reversible probes for GSH.This article has not yet been cited by other publications.

Abstract:Cysteine (Cys) plays important roles in many physiological processes of eukaryotic cells and its detection in cells is of fundamental significance. However, glutathione (GSH), homocysteine, N-acetyl-L-cysteine and other thiols greatly hamper the detection of Cys. In particular, GSH strongly interferes with the detection of cellular Cys (30-200 mu M) due to its high intracellular concentration (1-10 mM). In this work, an off on fluorescent probe (HOTA) for the detection of Cys is presented. This probe possesses both excellent sensitivity and satisfactory selectivity for cellular Cys detection: with the addition of 200 mu M Cys, the fluorescence intensity of the probe (10 mu M) enhanced 117-fold and the detection limit was calculated to be 13.47 mu M, which is lower than the cellular Cys concentration; the probe also selectively detected 30-200 mu M cysteine over 1-10 mM glutathione. Consequently, cell imaging experiments were performed with probe HOTA. Furthermore, the results of the thiol-blocking and GSH synthesis inhibiting experiments confirmed that the intracellular emission mainly originates from the interaction between Cys and HOTA.

Abstract:Biological thiols, such as glutathione, cysteine, and homocysteine, are closely associated with human health and disease. In this work, we demonstrated a general strategy for the design of fluorescent probes for the detection of various biological related thiols. The preferred strategy we employed was to introduce a 3,5-dinitropyridyl moiety to a selected fluorophore via ether linkage to afford fluorescent probe. Once the ether bond was thiolyzed by biothiol, the hydroxy-functionalized fluorescent dye would be released and fluorescence emission restored. Herein, some novel fluorescent probes based on this strategy have been developed for sensing biothiols in living cells. All these probes presented here showed desirable properties, such as high sensitivity, high selectivity and preferred reaction kinetics. Moreover, the probes have been used for imaging of intracellular biothiols with emission colors ranging from green to near-infrared (NIR).

Abstract:This study aimed to develop a novel and practical fluorescent method for GSH detection in complex biological samples. To this end, a series of coumarin-based fluorescent probes was designed and synthesized using various aliphatic halogens as the sensing group. By using a new evaluation method of GSH/Cys/Hcy coexisting conditions, the probe with chloropropionate (CBF3) showed a high selectivity, excellent sensitivity, good stability for GSH detection. The reaction mechanism is proposed as nucleophilic substitution/cyclization and intramolecular charge transfer (ICT), which was confirmed by LC-MS and NMR analysis, as well as density functional theory calculations. In addition, CBF3 was demonstrated to be competent not only for the quantitative detection of GSH in real serum samples, but also for sensing GSH changes in different oxidative stress models in living cells and nematodes. This study showed a practical strategy for constructing GSH-specific fluorescent probes, and provided a sensitive tool for real-time sensing of GSH in real biological samples. The findings would greatly facilitate further investigations on GSH-associated clinical diagnosis and biomedical studies.

Abstract:Glutathione (GSH) plays an important role in the body's biochemical defense system, and the detection of GSH in a physiological system is an important tool for understanding redox homeostasis. Protection-deprotection strategies have proven to be the most reliable, among existing detection methods. However, the understanding of how various electronic and steric effects influence a probe's ability to recognize a substrate is still lacking. In this study, we have analyzed various substituent effects on a GSH probe template via theoretical calculations and constructed the performance regulation and control strategy for this kind of probe. We then developed a series of guided probes using eighteen different acrylic ester derivatives to mask the fluorescence of fluorescein. The optical performance differences between the guided probes strongly supported the applicability of our proposed guiding strategy. Moreover, the positively guided probes are excellent for imaging GSH distribution in living cells and mice.

Abstract:Two probes, structurally symmetric CBFB and asymmetric CBFM, constructed by a D–A–D (donor–acceptor–donor) type curcuminoid as the fluorophore and the DNBS (2,4-dinitrobenzenesulfonyl) group as the biothiol recognition site were designed and synthesized here. The DNBS group can quench the emission of the fluorophore by the PET (photoinduced electron transfer) process, and in the presence of biothiols, the emission of the probe was switched on as a result of the cleavage of the quencher by a nucleophilic aromatic substitution reaction. Experimental analyses and theoretical calculations revealed that two recognition moieties in the molecule can quench the fluorescence more efficiently, therefore, CBFB showed a much higher SNR (signal to noise ratio) than CBFM in biothiol detection with an emission maximum at 610 nm. This “low background” and “turn-on” fluorescent probe, CBFB, was successfully utilized to map endogenous biothiols in living cells.

Abstract:Intracellular biothiols, including cysteine (Cys), glutathione (GSH), and homocysteine (Hcy), play a critical role in many physiological and pathological processes. Among them, GSH is the most abundant non-protein mercaptan (1–10 mM) in cells, and the change in GSH concentration level is closely related to the occurrence of many diseases, such as Parkinson's disease, Alzheimer's disease, and neurological diseases. Fluorescent probes have attracted much attention due to their advantages of high specificity, high sensitivity, high selectivity, low cost, and high quantum yield. Methods that use optical probes for selective detection of GSH in vitro and in vivo are in high demand. In this paper, we reviewed the most recent five years of research on fluorescence probes for the detection of GSH, including the specific detection of GSH, dual-channel identification of GSH and other substances, and the detection of GSH and other biothiols. According to the type of fluorophore, we classified GSH fluorescent probes into eight classes, including BODIPY, 1,8-Naphthalimide, coumarin, xanthene, rhodamine, cyanine, benzothiazoles, and others. In addition, we roundly discuss the synthesis, detection mechanism, photophysical properties, and biological applications of fluorescent probes. We hope that this review will inspire the exploration of new fluorescent probes for GSH and other related analyses.

Abstract:A new fluorescent probe based on fluorescein fluorophore displays an excellent selectivity and sensitivity for GSH in buffer solution containing CTAB. The fluorescence intensity of sensor is linearly proportional to GSH concentrations ranging from 0 to 10 μM, and as low as 22 nM concentration of GSH was evaluated with 3.3 σ/s. The optical changes are attributed to the release of free fluorescein after the electron withdrawing dinitrophenyl ether reacted efficiently with GSH in CTAB micelles. The enhanced selectivity toward GSH over Cys and Hcy considered to be related to the carboxyl groups in thiols. The confocal microscopy experiments implied that this probe is a promising candidate for imaging of GSH in living cells.

Abstract:Glutathione (GSH), as one of the most important biological mercaptans, is involved in a variety of biological processes and is considered an important biomarker in early diagnosis and treatment as well as disease stage monitoring. Rapid and accurate detection of GSH in complex biological systems is of great significance for the analysis of pathology. Fluorescence imaging technology is widely used because of its advantages of high sensitivity, high resolution and non-destructive. In this paper, the latest research progress of GSH-responsive organic small molecule fluorescence probes in recent five years is summed up, and their response mechanisms are classified and discussed. In addition, the probe design strategy, sensing mechanism and biological application are also discussed in this review. Abstract:Biothiols, including homocysteine (Hcy), cysteine (Cys) and reduced glutathione (GSH), play various roles in physiological and pathological processes. Because these biothiols possess similar in structures, it is difficult to discriminate Hcy, Cys and GSH from one another. In this work, a novel fluorescent probe, 4-BrCP, based on coumarin as a fluorophore to discriminate GSH from Hcy and Cys was rationally designed and synthesized in three steps with good total yield. The benzothiazol ring played the role of a fluorescent emission wavelength adjuster, while the ester part served as a fluorescent quencher and reactive sites with Hcy, Cys and GSH. The results of responsive experiment indicated that the probe could discriminate GSH from Hcy and Cys distinctly and exhibited a relatively high fluorescence quantum yield (0.84) and low detection limit (9.8 nM). In addition, the probe 4-BrCP also showed good stability and low toxicity. The reaction mechanism of 4-BrCP with GSH was speculated on according to the LC-MS data. Most importantly, because of the length of the skeletons of Hcy, Cys and GSH, different dynamic fluorescent phenomena were observed. Furthermore, imaging experiments suggested that the probe could be used to monitor GSH in living cells and organisms.

Abstract:Glutathione (GSH) is the most abundant biological thiol in cells. It can participate in metabolic processes, regulate the body's oxidation level and is essential for various cell functions. An abnormal concentration of glutathione in the cell is directly related to some diseases. Therefore, it is very important to develop fast, sensitive and reversible detection methods for GSH. Herein, we designed a reversible fluorescent probe (named GeP) for sensing GSH based on the nucleophilic addition and dissociation of intracellular GSH to GeP. The probe GeP showed a fast response time and a 20-fold fluorescence change toward GSH. It exhibited excellent mitochondrial-targeted performance and could be used to monitor GSH in mitochondria. Importantly, GeP could also enable superresolution fluorescence imaging of mitochondria through stochastic optical reconstruction microscopy.

Abstract:Glutathione is a major endogenous antioxidant in biological systems. As a result, highly selective fluorescent probes are significant components of studies aimed at gaining a better understanding of the role(s) glutathione plays in biological functions. In this study, we developed a dual-channel turn-on fluorescent probe for glutathione, comprising naphthalimide and cyanine, bridged by a thiol-reactive sulfonamide moiety. The probe selectively responds to glutathione and is not affected by cysteine and homocysteine. Important information has been gained about the chemical mechanism for operation of the probe, which responds to glutathione in visible and near-infrared channels synchronously and independently. Moreover, this probe exhibits the remarkable capability of targeting mitochondria and dual-channel tracking of intracellular glutathione in a spatiotemporal and synchronous manner. We believe that the strategy used to design the new dual-channel fluorescence-on probe will be applicable to the design of fluorescent probes that display specific responses in multiple channels.

Abstract:We synthesized six 1-oxo-1H-phenalene-2,3-dicarbonitrile (OPD)-based probes with various leaving groups using an arylthioether linker and for the first time identified the probe O-NH2 capable of highly selective detection of glutathione over cysteine/homocysteine in vitro and in vivo based on an aromatic nuclear substitution reaction (SNAr) mechanism. The fluorescence of the probe O-NH2 was quenched because of the photoinduced electron transfer (PET) process, but switched on by a glutathione-triggered specific recognition reaction between the probe O-NH2 and glutathione. The recognition mechanism for glutathione was explored and verified by theoretical calculations and ESI-MS analysis. Using O-NH2 as the probe, the GSH fluorescence images were demonstrated in HeLa cells and the intracellular GSH levels in different imatinib-resistant K562 tumor cells were firstly determined. Further, O-NH2 was utilized to detect glutathione in D. magna and zebrafish embryos. The combined results indicate that O-NH2 can be applied as an effective tool for detecting glutathione in biological investigations.

Abstract:A fluorescent probe CTP2-IMC was designed for bioimaging of glutathione (GSH) in cancer cells with indomethacin (IMC), coumarin and bromide as the targeting group, fluorophore and receptor, respectively. Due to the π-π interaction between coumarin and IMC, CTP2-IMC mainly exists in the form of folded state in aqueous solution. The non-radiative transitions caused by the photo-induced electron transfer (PET) process from IMC to the fluorophore as well as the heavy-atom effect led to non-fluorescent of CTP2-IMC . The substitution of Br by GSH and unfolded conformation induced by IMC acceptor on cancer cells resulted in significant fluorescence enhancement, which enabled CTP2-IMC to bioimage GSH in cancer cells rather than in normal one.

Abstract:Biological thiols importantly regulate the intracellular redox activity and metabolic level, but many of the developed probes for biothiols are facing difficulty in effectively distinguishing GSH from Cys/Hcy due to the similarity in mechanism. In this work, despite the previous pattern of "Logic Gate", we reported the concept of "Fluorescence Fusion" for the first time to achieve only one excitation-emission process. The exploited the probe, MZ-NBD, could quickly measure GSH in 10 min with a large Stokes shift (130 nm). Though the reacting mechanism was similar, only GSH could cause the "Fluorescence Fusion" with only one strong fluorescence response while Cys/Hcy caused two peaks. Adjusting the excitation wavelength could hardly split the fused peak into two. Though image recognition by artificial intelligence could easily distinguish the patterns of peaks, here we used the signal-treating method to realize the high selectivity towards GSH. Moreover, MZ-NBD could be utilized for rapid detection of GSH in living MCF-7 cells, which was more suitable for GSH than using the "Logic Gate" strategy. More than introducing a novel probe with the new concept, this work was meaningful as the linker of traditional reaction-based fluorescent probes and potential image recognition by artificial intelligence, thus led to various future researches in inter-disciplines.
Thiolation for Enhancing Photostability of Fluorophores at the Single-Molecule Level

Abstract:Fluorescent probes are essential for single-molecule imaging. However, their application in biological systems is often limited by the short photobleaching lifetime. To overcome this, we developed a novel thiolation strategy for squaraine dyes. By introducing thiolation of the central cyclobutene of squaraine (thio-squaraine), we observed a ≈5-fold increase in photobleaching lifetime. Our single-molecule data analysis attributes this improvement to improved photostability resulting from thiolation. Interestingly, bulk measurements show rapid oxidation of thio-squaraine to its oxo-analogue under irradiation, giving the perception of inferior photostability. This discrepancy between bulk and single-molecule environments can be ascribed to the factors in the latter, including larger intermolecular distances and restricted mobility, which reduce the interactions between a fluorophore and reactive oxygen species produced by other fluorophores, ultimately impacting photobleaching and photoconversion rate. We demonstrate the remarkable performance of thio-squaraine probes in various imaging buffers, such as glucose oxidase with catalase (GLOX) and GLOX+trolox. We successfully employed these photostable probes for single-molecule tracking of CD56 membrane protein and monitoring mitochondria movements in live neurons. CD56 tracking revealed distinct motion states and the corresponding protein fractions.
