Abstract:Real-time quantification of glutathione (GSH) concentrations in living cells is essential for understanding GSH-related pathological events. However, most existing fluorescent probes are not suitable for this purpose due to their irreversible response mechanisms. Only a few probes have been successfully developed to realize this goal to date. Herein, we presented a fluorescent probe (QG-S) for quantifying cellular GSH and monitoring GSH dynamic variations. The probe QG-S was successfully employed in real-time monitoring of the dynamic variations of GSH in living cells upon reversible treatment with a GSH inhibitor (N-methylmaleimide) by using confocal microscopy. Moreover, the probe QG-S showed extremely low cytotoxicity and was facilely used to quantitatively determine the GSH concentration (4.25 mM) in living HeLa cells.

Abstract:We synthesized six 1-oxo-1H-phenalene-2,3-dicarbonitrile (OPD)-based probes with various leaving groups using an arylthioether linker and for the first time identified the probe O-NH2 capable of highly selective detection of glutathione over cysteine/homocysteine in vitro and in vivo based on an aromatic nuclear substitution reaction (SNAr) mechanism. The fluorescence of the probe O-NH2 was quenched because of the photoinduced electron transfer (PET) process, but switched on by a glutathione-triggered specific recognition reaction between the probe O-NH2 and glutathione. The recognition mechanism for glutathione was explored and verified by theoretical calculations and ESI-MS analysis. Using O-NH2 as the probe, the GSH fluorescence images were demonstrated in HeLa cells and the intracellular GSH levels in different imatinib-resistant K562 tumor cells were firstly determined. Further, O-NH2 was utilized to detect glutathione in D. magna and zebrafish embryos. The combined results indicate that O-NH2 can be applied as an effective tool for detecting glutathione in biological investigations.

Abstract:Schematic representation depicting the fluorescent sensing of GSH. The probe was developed by the addition of cystine (Cystine‐AuNCs) and further extended for the turn‐on sensing of GSH. The probe achieved sensitive detection of GSH with a low Limit of Detection (LoD) of 0.43 μM. Glutathione (GSH), an oxidative stress biomarker plays a major role in different pathological processes and lead to a vast number of diseases including cancer. Biologically, cystine uptake is a fundamental factor in maintaining GSH levels in cells; however, the interaction between cystine and GSH in biological systems is least explored in developing sensors for GSH. In the present work, we have explored this interaction as the fundamental factor in "turn‐on" sensing of GSH from body fluids using a fluorescent gold nanocluster platform. The experimental results revealed that the probe displayed appreciable performance for GSH over a linear range from 0.2 mM to 1.3 mM with a limit of detection (LoD) of 0.43 μM. The probe also revealed sensitive as well as selective response towards GSH even in the presence of possible co‐existing interferants and provided satisfactory recovery percentage of 94.17 % to 108.12 % from real serum samples. These findings of the present study can be extended in future applications of sensing GSH from clinical samples.

Abstract:The reported probe 1 links two rhodamine fluorophores via a hexamethylene linker offering flexibility and increased fluorescence quantum yield for GSH recognition. It shows selectivity to GSH over cysteine and homocysteine. This nontoxic, cell permeable probe has been successfully applied for imaging cellular GSH in MCF‐7 live cells with detection limit of 0.125 nM. A new colorimetric and fluorescence turn‐on chemodosimeter for selective detection of GSH over Cys and Hcy with 34‐fold enhancement in emission intensity is reported. Probe 1 exhibited ultra‐sensitivity toward GSH with 0.125 nM detection limit and successfully displayed GSH detection in MCF‐7 live cells. The mechanism of sensing is established by density functional theoretical calculations.

Abstract:A novel turn-on red fluorescent BODIPY-based probe (Probe 1) for the detection of glutathione was developed. Such a probe carries a para-dinitrophenoxy benzyl pyridinium moiety at the meso position of a BODIPY dye as self-immolative linker. Probe 1 responds selectively to glutathione with the detection limit of 109nM over other amino acids, common metal ions, reactive oxygen species, reactive nitrogen species, and reactive sulfur species. A novel electrostatic interaction to modulate the SNAr attack of glutathione was believed to play significant role for the observed selective response to glutathione. The cleavage of dinitrophenyl ether by glutathione leads to the production of para-hydroxybenzyl moiety which is able to self-immolate through an intramolecular 1,4-elimination reaction to release the fluorescent BODIPY dye. The low toxic probe has been successfully used to detect mitochondrial glutathione in living cells.

Abstract:A 2,7-naphthyridine derivative (AND-OH) was used as a fluorophore in the development of a novel fluorescent turn-on probe (AND-DNBS) by introducing a 2,4-dinitrobenzenesulfonate functionality. This fluorescent probe was successfully used for sensing glutathione (GSH) in aqueous media and showed favorable properties such as a rapid response time (45 s) and a significant Stokes shift (227 nm). The characteristics of the developed probe are better than those of another probe, namely ND-DNBS, which was also synthesized by our group and most other previously reported fluorescent probes for GSH. The AND-DNBS probe was able to detect GSH via a red emission in living A549 cells and zebrafishes. These results show that AND-DNBS has potential applications as an excellent probe for detecting GSH in various environments.

Abstract:Developing selective and sensitive fluorescent probes for detection of glutathione (GSH) concentration and intracellular distribution is of great significance for early diagnosis and treatment of diseases such as liver injury and cancers since GSH plays irreplaceable roles in regulating intracellular redox homeostasis. Herein, we present a new fluorescent probe which can be specifically activated by GSH through the conjugate addition and hydrolysis induced covalent-assembly approach in achieving of zero-background interference fluorescent off-on sensing. Besides, the probe exhibited prominent selectivity and sensitivity, low detection limit and cytotoxicity, thus successfully realized specific real-time monitoring and tracking of GSH levels in living cells. As a consequence, this work might provide a potentially promising candidate for validation the function of GSH in various physiological and pathological processes, which is beneficial for early diagnosis and therapeutics of related diseases.

Abstract:Biological thiols importantly regulate the intracellular redox activity and metabolic level, but many of the developed probes for biothiols are facing difficulty in effectively distinguishing GSH from Cys/Hcy due to the similarity in mechanism. In this work, despite the previous pattern of "Logic Gate", we reported the concept of "Fluorescence Fusion" for the first time to achieve only one excitation-emission process. The exploited the probe, MZ-NBD, could quickly measure GSH in 10 min with a large Stokes shift (130 nm). Though the reacting mechanism was similar, only GSH could cause the "Fluorescence Fusion" with only one strong fluorescence response while Cys/Hcy caused two peaks. Adjusting the excitation wavelength could hardly split the fused peak into two. Though image recognition by artificial intelligence could easily distinguish the patterns of peaks, here we used the signal-treating method to realize the high selectivity towards GSH. Moreover, MZ-NBD could be utilized for rapid detection of GSH in living MCF-7 cells, which was more suitable for GSH than using the "Logic Gate" strategy. More than introducing a novel probe with the new concept, this work was meaningful as the linker of traditional reaction-based fluorescent probes and potential image recognition by artificial intelligence, thus led to various future researches in inter-disciplines.

Abstract:This study aimed to develop a novel and practical fluorescent method for GSH detection in complex biological samples. To this end, a series of coumarin-based fluorescent probes was designed and synthesized using various aliphatic halogens as the sensing group. By using a new evaluation method of GSH/Cys/Hcy coexisting conditions, the probe with chloropropionate (CBF3) showed a high selectivity, excellent sensitivity, good stability for GSH detection. The reaction mechanism is proposed as nucleophilic substitution/cyclization and intramolecular charge transfer (ICT), which was confirmed by LC-MS and NMR analysis, as well as density functional theory calculations. In addition, CBF3 was demonstrated to be competent not only for the quantitative detection of GSH in real serum samples, but also for sensing GSH changes in different oxidative stress models in living cells and nematodes. This study showed a practical strategy for constructing GSH-specific fluorescent probes, and provided a sensitive tool for real-time sensing of GSH in real biological samples. The findings would greatly facilitate further investigations on GSH-associated clinical diagnosis and biomedical studies.

Abstract:The abnormally elevated expression level of glutathione (GSH) has been observed in various human cancer cells and tissue. Thus, effective methods for glutathione detection are of great importance in early diagnosis of cancer. However, many fluorescent probes for GSH detection suffer from the interference of the abundantly existent nucleophilic biomolecules in biological environment. In this work, we propose a sequential activation strategy to overcome this problem by designing and synthesizing a series of 1,3,5-triazinyl resorufin turn-on fluorescent probe (Probes 1–3 ). As two electrophilic sites are presented in probes, GSH sequentially reacts with the resorufin and the triazine moiety, resulting in significant fluorescence augmentation (up to 165.0-fold). Designed probes possess low limit of detection as low as 1.8 μM). Cellular fluorescent imaging has been successfully applied to selectively detect GSH in several living cells.

Abstract:A new fluorescent probe based on fluorescein fluorophore displays an excellent selectivity and sensitivity for GSH in buffer solution containing CTAB. The fluorescence intensity of sensor is linearly proportional to GSH concentrations ranging from 0 to 10 μM, and as low as 22 nM concentration of GSH was evaluated with 3.3 σ/s. The optical changes are attributed to the release of free fluorescein after the electron withdrawing dinitrophenyl ether reacted efficiently with GSH in CTAB micelles. The enhanced selectivity toward GSH over Cys and Hcy considered to be related to the carboxyl groups in thiols. The confocal microscopy experiments implied that this probe is a promising candidate for imaging of GSH in living cells.

Abstract:Cysteine (Cys) plays important roles in many physiological processes of eukaryotic cells and its detection in cells is of fundamental significance. However, glutathione (GSH), homocysteine, N-acetyl-L-cysteine and other thiols greatly hamper the detection of Cys. In particular, GSH strongly interferes with the detection of cellular Cys (30-200 mu M) due to its high intracellular concentration (1-10 mM). In this work, an off on fluorescent probe (HOTA) for the detection of Cys is presented. This probe possesses both excellent sensitivity and satisfactory selectivity for cellular Cys detection: with the addition of 200 mu M Cys, the fluorescence intensity of the probe (10 mu M) enhanced 117-fold and the detection limit was calculated to be 13.47 mu M, which is lower than the cellular Cys concentration; the probe also selectively detected 30-200 mu M cysteine over 1-10 mM glutathione. Consequently, cell imaging experiments were performed with probe HOTA. Furthermore, the results of the thiol-blocking and GSH synthesis inhibiting experiments confirmed that the intracellular emission mainly originates from the interaction between Cys and HOTA.

Abstract:Glutathione is a major endogenous antioxidant in biological systems. As a result, highly selective fluorescent probes are significant components of studies aimed at gaining a better understanding of the role(s) glutathione plays in biological functions. In this study, we developed a dual-channel turn-on fluorescent probe for glutathione, comprising naphthalimide and cyanine, bridged by a thiol-reactive sulfonamide moiety. The probe selectively responds to glutathione and is not affected by cysteine and homocysteine. Important information has been gained about the chemical mechanism for operation of the probe, which responds to glutathione in visible and near-infrared channels synchronously and independently. Moreover, this probe exhibits the remarkable capability of targeting mitochondria and dual-channel tracking of intracellular glutathione in a spatiotemporal and synchronous manner. We believe that the strategy used to design the new dual-channel fluorescence-on probe will be applicable to the design of fluorescent probes that display specific responses in multiple channels.

Abstract:In the last few decades, growing numbers of fluorescent probes have been developed to detect intracellular GSH. However, the majority of probes for GSH were irreversible without monitoring the changes of intracellular GSH concentration. Therefore, recently, fluorescent probes for monitoring concentrations of GSH in real-time in living cells have come into being to address this challenge. This Perspective aimed at the development of reversible probes for GSH was organized by structural features, chemical reactions, and physicochemical properties. The reversible probes designed by a coumarin skeleton as a read-out fluorophore and the Michael addition reaction as a response mechanism accounted for most of the reported reversible probes. The performances of reversible fluorescent probes based on Michael addition could be roughly predicted by fundamental laws of kinetics and thermodynamics in physical chemistry. Essentially, the design principles included a highly reactive site for GSH, a small thermodynamic driving force, a desirable <i>K</i><sub>d</sub> of 1–10 mM, and excellent cell membrane permeability. Prospectively, the development of various mechanisms and fluorophores will be effective measures to enrich the types of reversible probes for GSH.This article has not yet been cited by other publications.

Abstract:In this paper, we successfully synthesized a simple and versatile fluorescent probe. This probe was not only easily prepared with a high yield, but also showed rapid selective and sensitive responses for Cys/Hcy and GSH. The probe can be used as a naked-eye detector for Cys/Hcy and GSH from other analytes. As a fluorescent sensor, it can be used to simultaneously detect and discriminate Cys/Hcy from GSH with two fluorescent emission signals without spectral crosstalk.

Abstract:Glutathione (GSH), as a key regulator for the redox balance of biological processes, was detected with a metal-organic framework (MOF) (UiO-67-sbdc) fluorescence sensor. The fluorescence of UiO-67-sbdc could be effectively enhanced by GSH via an electronic effect between GSH and the ligand of MOF, thus achieving the recognition of GSH. It is the first attempt to design a MOF-based fluorescent sensor to distinguish GSH from cysteine (Cys). This sensor exhibited short response time (< 120 s), suitable linear range, and high selectivity towards GSH. Further research conducted in serum also showed a quantitative and selective detection of GSH. Thus, the reported UiO-67-sbdc in this work has its application prospect in clinically judging antioxidant capacity of the body.

Abstract:Glutathione (GSH) plays an important role in the body's biochemical defense system, and the detection of GSH in a physiological system is an important tool for understanding redox homeostasis. Protection-deprotection strategies have proven to be the most reliable, among existing detection methods. However, the understanding of how various electronic and steric effects influence a probe's ability to recognize a substrate is still lacking. In this study, we have analyzed various substituent effects on a GSH probe template via theoretical calculations and constructed the performance regulation and control strategy for this kind of probe. We then developed a series of guided probes using eighteen different acrylic ester derivatives to mask the fluorescence of fluorescein. The optical performance differences between the guided probes strongly supported the applicability of our proposed guiding strategy. Moreover, the positively guided probes are excellent for imaging GSH distribution in living cells and mice.

Abstract:Real-time and accurate detection of intracellular GSH content is crucial for clinical diagnosis, disease treatment, and pathogen detection. Creating fluorescent probes that possess aggregation induced emission (AIE) and precisely detect GSH remains a formidable challenge despite the widespread reporting of numerous fluorescent probes for GSH recognition. This may due to the absence of effective strategies. Herein, using a "turn off - turn on" strategy, a novel fluorescent probe DPND with AIE characteristic has been devised and synthesized. It can detect GSH selectively and uninhibitedly in the presence of various ions. The limit of detection of DPND-Cu2+ for GSH was calculated to be 1.17 μM, demonstrating the probe's ability to detect GSH quantitatively. More crucially, the probe exhibits low toxicity and solid biocompatibility while recognizing GSH in HeLa cells and mices, indicating that it may be a promising candidate for in vivo detection of GSH. Consequently, this work offers the potential to precisely tailor molecular recognition of GSH in addition to providing ongoing exploration of approaches for monitoring GSH.
