Abstract:Many current reports in the scientific literature describe novel fluorescent probes intended to provide information on various structures or properties of live cells by using microscopic imaging. Unfortunately, many such reports fail to provide key information regarding the staining process. It is often the case that neither the necessary minimum technical detail (probe concentration, solvent and cosolute, temperature and time of staining, and details of post-staining washes) nor a discussion of the proposed staining mechanism are provided. Such omissions make it unnecessarily difficult for biomedical end-users to try out reported novel probes in their own laboratories. The validity of these criticisms is explored and demonstrated by a detailed analysis of 75 non-cherry-picked articles describing novel fluorescent probes for the detection of lipid droplets in live cells. This dataset also suggests that papers from journals with high journal impact factors or from better-known research groups are no more likely to provide better protocol information or discussion of the mechanism than papers from less prestigious sources. Comments on possible reasons for this suboptimal reporting are offered. The use of a suitable information/feature checklist, following best practice in many leading chemical and biological journals, is suggested as a mechanism for ameliorating this situation, with a draft checklist being provided.

Abstract:In the past few decades there has been a revolution in the field of optical microscopy with emerging capabilities such as super-resolution and single-molecule fluorescence techniques. Combined with the classical advantages of fluorescence imaging, such as chemical labeling specificity, and noninvasive sample preparation and imaging, these methods have enabled significant advances in our polymer community. This Viewpoint discusses several of these capabilities and how they can uniquely offer information where other characterization techniques are limited. Several examples are highlighted that demonstrate the ability of fluorescence microscopy to understand key questions in polymer science such as single-molecule diffusion and orientation, 3D nanostructural morphology, and interfacial and multicomponent dynamics. Finally, we briefly discuss opportunities for further advances in techniques that may allow them to make an even greater contribution in polymer science.

Abstract:This Editorial celebrates the 70 th anniversary of the European Journal of Histochemistry since its foundation as Rivista di Istochimica Normale e Patologica , and introduces a Special Collection of selected articles on the application of the histochemical approach for investigating cell biological features and processes in animals and plants, and under diseased conditions. The year 2024 is a special one for histochemists, as 100 years ago J.W. Robert Feulgen and H. Rossenbeck introduced the histochemical procedure for the specific stoichiometric staining of DNA in histological samples: to commemorate this influential publication, three papers in the present issue are devoted to the application of the Feulgen reaction at light and electron microscopy, and in cytometry.

Abstract:Fluorescence, a form of photoluminescence, is the emission of light at a longer wavelength by a substance when exposed to shorter-wavelength energy. Biofluorescence, which can be observed in living organisms, involves the absorption of light at one wavelength and re-emission at a longer wavelength due to fluorophores in specialized cells or structures. While initially studied in marine organisms, attention has shifted to exploring biofluorescence in terrestrial organisms, revealing roles in reproduction, camouflage, communication, and prey attraction across phyla. Community science databases engage the public in data collection, fostering scientific discovery and strengthening the science-society connection. Such databases have become valuable tools and have aided scientists in understanding the natural history of many different traits in organisms. This paper introduces Finding Fluorescence, the first biofluorescence-based community science website established in 2020 to gather public observations of biofluorescent organisms. The study presents at least 15 novel biofluorescence accounts spanning five phyla, 15 families, and 15 species. The observations collected from Finding Fluorescence contribute to our understanding of fluorescence in organisms and provide insight into possible ecological functions. We emphasize the importance of community engagement in scientific exploration and encourage future studies to incorporate such aspects into their research.

Abstract:We review some of the progress of single-molecule optical experiments in the past 20 years and propose some perspectives for the coming years. We particularly focus on methodological advances in fluorescence, super-resolution, photothermal contrast, and interferometric scattering and briefly discuss a few of the applications. These advances have enabled the exploration of new emitters and quantum optics; the chemistry and biology of complex heterogeneous systems, nanoparticles, and plasmonics; and the detection and study of non-fluorescing and non-absorbing nano-objects. We conclude by proposing some ideas for future experiments. The field will move toward more and better signals of a broader variety of objects and toward a sharper view of the surprising complexity of the nanoscale world of single (bio-)molecules, nanoparticles, and their nano-environments.

Abstract:Small-molecule fluorophores, such as fluorescein and rhodamine derivatives, are critical tools in modern biochemical and biological research. The field of chemical dyes is old; colored molecules were first discovered in the 1800s, and the fluorescein and rhodamine scaffolds have been known for over a century. Nevertheless, there has been a renaissance in using these dyes to create tools for biochemistry and biology. The application of modern chemistry, biochemistry, molecular genetics, and optical physics to these old structures enables and drives the development of novel, sophisticated fluorescent dyes. This critical review focuses on an important example of chemical biology-the melding of old and new chemical knowledge-leading to useful molecules for advanced biochemical and biological experiments.

Abstract:This manuscript commemorates the 100th anniversary of APMIS, highlighting its evolution from a regional journal, founded in 1924 as Acta Pathologica et Microbiologica Scandinavica, to an international platform fostering global collaboration in pathology, microbiology, and immunology. The journal's inception was driven by Ulrik Quensel's vision in 1919, leading to the establishment of the Northern Pathological Society and the launch of the journal in 1924. APMIS has consistently published landmark research, including significant contributions from prominent. These studies have advanced understanding in fields such as pathology, microbiology, and immunology. The journal expanded its scope in the 1970s to include immunology, rebranding as APMIS in the mid‐1980s. Recent decades have seen a continued commitment to cutting‐edge research and an increasing impact factor. As APMIS transitions to an Open Access model under Wiley, it will be renamed the PMI Journal (Pathology, Microbiology, and Immunology) to reflect its global reach and dedication to scientific excellence. This centennial celebration acknowledges the contributions of editors, authors, and readers, looking forward to future advancements in biomedical research.

Abstract:Fluorescent and colorimetric probes are gaining interest in analyte detection, imaging, and real-time applications. However, it faces several challenges in achieving high sensitivity and selectivity, maintaining a strong signal-to-noise ratio, detecting analytes at extremely low concentrations, ensuring stability under different conditions and addressing issues related to cost, accessibility and instrumentation. Additionally, ensuring biological compatibility and enabling real-time monitoring are also challenges that need to overcome. We have investigated the sensing application of probe molecules towards various analytes like metal ions, anions, biomolecules, reactive oxygen species (ROS), reactive sulphur species (RSS), toxic agents and nanoparticles performed by our group together with critically comparing with the existing reports. This featured article provides the impact of fluorescent and colorimetric probes in advancing analytical techniques, imaging and real-time applications that transcend traditional boundaries.
