Abstract:Fused systems combining 1,2,4-triazole and 1,3,4-thiadiazole rings occupy a prominent place in modern bioorganic and medicinal chemistry due to the diverse range of their biological activities. Thus, compounds incorporating [1,2,4]triazolo[3,4- b ]- thiadiazole ring system are of essential importance and particular interest both in the pharmaceutical as well as agrochemical industries. Herein, an overview of certain synthetic methodologies for obtaining of heterocyclic compounds based on functionally substituted 1,2,4-triazolo[3,4- b ][1,3,4]thiadiazoles are presented.

The microreview focuses on nonfunctionalized in positions 4 and 5 derivatives rather than well-reported 4-CO 2 R and 4-COR ones. The examples reported herein are arranged in three principal categories: assembly of the pyrrolidine-2,3-dione core, its functionalization, and the construction of sophisticated 3D chiral carcasses with high medicinal prospects based on the core. The summary includes works published within a period from 2020 to 2024.

Abstract:Abstract: The pyranoquinoline frameworks have a wide distribution in natural products and have displayed a great amplitude of biological activities, attracting the attention of synthetic and medicinal chemists due to their usefulness in agrochemical and pharmaceutical industries. Among the twenty possible isomeric pyranoquinoline frameworks, research reports for only six of them that were found during the period of time covered in this review article (i.e. 2000 to 2023). According to the literature reports during this frame of time, the synthesis of pyranoquinoline derivatives was achieved by following three general synthetic approaches [i.e. (i) via multicomponent cyclizations (MCC), (ii) via bimolecular cyclizations (BMC) and via intramolecular cyclizations (IMC)], mediated by catalyst-free conditions or by diverse environmentally friendly catalysts, in which mechanistic proposal was discussed for several of such processes. Additionally, various obtained pyranoquinoline derivatives reported in this review were subjected to diverse biological evaluations, such as Parkinson ́s and Alzheimer ́s diseases, as antibacterials, antifungals, and anticancer drugs, among others, indicating the promising biological potential of this class of heterocyclic structure.

Abstract:The syntheses and bioevaluation of 3‐acyl‐2‐pyridones have garnered significant interest in recent years as a novel class of compounds with diverse biological activities. The bioactivities cover a wide range of therapeutic areas including antimicrobial, antitumor, anti‐inflammatory, antifungal, antibiotic, neuritogenic, cytotoxic, and antiviral activities. Therefore, 3‐acyl‐2‐pyridone scaffold offers a versatile platform for the development of new molecules with potential applications in medicinal chemistry. The recent exploration on synthesis and bioevaluation of 3‐acyl‐2‐pyridones has yielded promising results, making them attractive targets for further optimization and development of new therapeutic agents. This review summarizes the available information on total synthesis of 3‐acyl‐2‐pyridone containing natural products, and the other synthetic strategies developed for accessing the 3‐acyl pyridones such as multicomponent synthesis, condensation reaction, microwave‐assisted synthesis, catalytic hydrogenation reactions, metalation reaction, Michael addition, and formylation reactions. The presented information may serve as a valuable resource for researchers working in the field of medicinal chemistry and drug discovery, stimulating further exploration and development of this intriguing scaffold.

Abstract:This feature article aims to review the evolution and recent advances in different protocols for synthesizing fused hybrids of N‐heterocycles, specifically [1,2,3]triazolo[1,5‐a] quinoline. Approaches to synthesize this unit will be discussed, including dehydrogenative cyclization, N−N oxidative coupling, Dieckmann condensation, intramolecular Heck reactions, (3+2) cycloaddition, Ullmann‐type coupling, and direct intramolecular arylation reactions. This report outlines the evolution and recent progress about the different protocols to synthesize the N‐heterocycles fused hybrids, specifically [1,2,3]triazolo[1,5‐a]quinoline. This review encompasses a broad range of approaches, describing several reactions for obtaining this since, such as dehydrogenative cyclization, oxidative N−N coupling, Dieckmann condensation, intramolecular Heck, (3+2)‐cycloaddition, Ullman‐type coupling and direct intramolecular arylation reactions. We divided this review in three section based in the starting materials to synthesize the target [1,2,3]triazolo[1,5‐a]quinolines. Starting materials containing quinoline or triazole units previously formed, as well as starting materials which both quinoline and triazole units are formed in situ. Different methods of obtaining are described, such as metal‐free or catalyzed conditions, azide‐free, using conventional heating or alternative energy sources, such as electrochemical and photochemical methods. Mechanistic insights underlying the reported reactions were also described in this comprehensive review.

Synthesis of Some New 1, 3, 4-Oxadiazole, Pyrazole, and Pyrimidine Bearing Thienopyrazole Moieties

Abstract:Aim and objective: According to the literature survey, pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the present work, pyrazol- 3-one 1 was reacted with cyanoacetic acid hydrazide and elemental sulfur to afford the corresponding thieno[3,2-c]pyrazol-6-carbohydrazide 3 derivatives. The latter compound reacted with some electrophilic reagents such as DMF-DMA, triethylorthoformate, arylidenemalononitriles and chalcones under neat conditions to give substituted oxadiazole and pyrazole, respectively. The treatment of compound 3 with active methylene reagents such as acetylacetone, diethylmalonate, ethyl acetoacetate and ethyl cyanoacetate under suitable conditions afforded pyrazole derivatives 10, 11, 13, and 15, respectively. Novel pyrazolothienopyrimidine 27 and 30 were prepared from precursor 26 with carbon disulfide and triethylorthoformate, respectively. The chemical structures of the newly synthesized compounds were established by elemental and spectral analyses including IR, and 1HNMR in addition to 13C-NMR and mass spectra. Materials and methods: A novel substituted pyrazole, pyrimidine and pyrazolothienopyrimidine were obtained via Gewald synthesis of thiophene and fused thiophene and Mannich reactions of 5-amino-3-phenyl-1Hthieno[ 3,2-c]pyrazole-6-carbohydrazide. Results and discussion: A series of some newly azoles and azines were prepared via reaction of thieno[3,2- c]pyrazol-6-carbohydrazide derivative 3 as starting material with some electrophilic and nucleophilic reagents. The structures of target compounds were established by elemental analyses and spectral data. Conclusion: Pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the current investigation, we have developed new and efficient methods for the synthesis of thieno[3,2-c]pyrazol-6-carbohydrazide derivatives. In addition, we have explored the preparative potential of these substances as intermediates for the synthesis of substituted pyrazoles and fused pyrazoles 10-30, respectively.

Abstract:The focus of this review is on the synthetic routes available for different types of pyrazoloquinoline derivatives. There are three types of synthetic methods: i) from pyrazole derivatives; ii) from quinoline derivatives; and iii) miscellaneous methods. The position of the linkage between pyrazole and quinoline rings determines the seven isomers of pyrazoloquinolines. The purpose of this review is to provide a guide for both synthetic and medicinal chemists to discover and design new pyrazoloquinolines for medical purposes.

Abstract:This article presents an efficient one-step synthesis of tetrahydropyrazolo[1,5- a ]pyrimidines through the recyclization of N -arylitaconimides with aminopyrazoles. The heterocyclic system of pyrazolo[3,4- b ]pyrimidine is known for its diverse biological properties, making its derivatives significant in pharmaceutical and medicinal chemistry. The study focuses on the regio- and chemoselective cascade reaction of N -arylitaconimides with 5-aminopyrazoles, demonstrating a new approach to synthesizing pyrazolo[1,5- a ]pyrimidines and pyrazolo[3,4- b ]pyridines. The methodology, involving boiling in isopropyl alcohol with acetic acid, yields selectively either pyrazolo[3,4- b ]pyridines or pyrazolo[1,5- a ]pyrimidines based on the substituents in the aminopyrazoles. The study elucidates the reaction mechanism, structural characterization using NMR spectroscopy, and confirms the structures via high-performance liquid chromatography and mass spectrometry. The simplicity and synthetic potential of this approach make it a valuable method for the preparation of these heterocyclic frameworks. Abstract:Schematic reperesentation of fused pyrimidine derivatives. Pyrimidine and its derivatives play a paramount role in drug discovery as privileged pharmacophores with considerable chemical and biological significance and its presence in genes. This review aims to assemble a systematic evaluation of synthetic tactics of various fused pyrimidine derivatives containing nitrogen heterocycles such as pyridopyridines, pyridopyrimidines, and pyrimidopyrimidine from a pharmacological point of view and deliver an overview of methodologies presenting the chemistry of fused pyrimidine derivatives. The review details the importance of various catalysts and ring substitution using various electrophilic and nucleophilic reagents. These synthetic strategies were elaborated based on the different synthetic routes that lead to the specific type of pyrimidine and pyridine fused derivatives. The literature accumulates various developments in one‐pot condensation, the Knoevenagel–Michael addition mechanism, microwave and ultrasound irradiation, intramolecular cyclization, nano‐catalytic reactions, and so forth. Short reaction times, catalyst reusability, solvent‐free conditions, excellent yields, and stereo‐selectivity are some of the benefits of certain synthetic approaches.

Abstract:Pyrimidine is a pharmacologically important moiety that exhibits diverse biological activities. This review reflects the growing significance of transition metal-catalyzed reactions for the synthesis of pyrimidines (with no discussion being made on the transition metal-catalyzed functionalization of pyrimidines). The effect of different catalysts on the selectivity/yields of pyrimidines and catalyst recyclability (wherever applicable) are described, together with attempts to illustrate the role of the catalyst through mechanisms. Although several methods have been researched for synthesizing this privileged scaffold, there has been a considerable push to expand transition metal-catalyzed, sustainable, efficient and selective synthetic strategies leading to pyrimidines. The aim of the authors with this update (2017–2023) is to drive the designing of new transition metal-mediated protocols for pyrimidine synthesis.
