Abstract:Synthesis and application of azacycloalk-1-enefused oxazol-3-ium salts are summarized. The microreview covers all synthetic methods for obtaining 6,7-dihydro-5 H -pyrrolo[2,1- b ]oxazol-4-ium and 5,6,7,8-tetrahydrooxazolo[3,2- a ]-pyridin-4-ium derivatives that have been classified in two distinct categories: cyclization leading to the formation of azacycloalk-1-ene ring and cyclization leading to the formation of oxazole ring. Application of these azacycloalk-1-enefused oxazol-3-ium salts has been discussed.

Abstract:Abstract: Oxadiazole is an organic compound featuring a heterocyclic ring housing carbon, oxygen, and nitrogen atoms. Due to their heightened stability in biological environments, oxadiazole rings exhibit significant biological activities, effectively addressing health challenges like infectious diseases and chronic conditions in medicinal chemistry. The main objective of this review is to discuss various synthetic approaches related to 1,3,4-oxadiazole and its derivatives. The diverse reactivity positions oxadiazole as a valuable building block in organic synthesis, with derivatives exhibiting promising pharmacological activities. It involves a systematic literature review, critical analysis, and synthesis of existing research. This review comprises the ever-expanding chemical knowledge and has significant implications for drug development. Synthetic approaches to synthesizing oxadiazole through different schemes and methods have been discussed thoroughly. This review also concisely associated the pharmacological activities of oxadiazole and its derivatives. This review highlights the importance of continued research into the structure-activity relationships of oxadiazole derivatives, paving the way for developing novel and more potent therapeutic agents.

Abstract:Substrate‐based synthetic strategies and biological activities of 1,3,4‐oxadiazole. The five‐membered 1,3,4‐oxadiazole heterocyclic ring has received considerable attention because of its unique bio‐isosteric properties and an unusually wide spectrum of biological activities. After a century since 1,3,4‐oxadiazole was discovered, its uncommon potential attracted medicinal chemist's attention, leading to the discovery of a few presently accessible drugs containing 1,3,4‐oxadiazole units, and a large number of patents have been granted on research related to 1,3,4‐oxadiazole. It is worth noting that interest in 1,3,4‐oxadiazoles' biological applications has doubled in the last few years. Herein, this review presents a comprehensive overview of the recent achievements in the synthesis of 1,3,4‐oxadiazole‐based compounds and highlights the major advances in their biological applications in the last 10 years, as well as brief remarks on prospects for further development. We hope that researchers across the scientific streams will benefit from the presented review articles for designing their work related to 1,3,4‐oxadiazoles.

Abstract:Abstract: Oxazolines are important heterocyclic systems due to their biological activities, such as antibacterial, antimalarial, anticancer, antiviral, anti-inflammatory, antifungal, antipyretic, and antileishmanial. They have been widely applied as chiral auxiliaries, polymers, catalysts, protecting groups, building blocks, and ligands in asymmetric synthesis. Due to their importance, many synthetic routes to prepare oxazoline moieties have been investigated and developed by researchers around the world. In this review, we summarized several synthetic methodologies published in the literature. The main substrates are nitriles, carboxylic acids, and acid derivatives, which react with a variety of reactants under conventional heating, microwave irradiation or ultrasound irradiation conditions. Syntheses via intramolecular cyclisation from amides have also been reported. Many publications have highlighted procedures based on solvent-free conditions using eco-friendly, reusable, and easy-availability catalysts.

Abstract:An efficient method for the synthesis of isoquinolinone derivatives via photopromoted carboamination of alkynes is developed. Starting from the readily available propargyl alcohol derivatives, the polycyclic isoquinolinone derivatives could be obtained with good aryl and heterocycle tolerance. Both terminal and alkyl substituted alkynes could be employed. This protocol is operationally easy, and easily conducted on a gram-scale. A possible mechanism involving radical addition and cyclization following aromatization was proposed.

Abstract:In this microreview, the application of noncatalyzed hetero-Diels–Alder reaction (formally [4+2] cycloaddition) with participation of conjugated nitroalkenes and their analogs for easy and selective preparation of 1,2-oxazine N -oxides was reviewed on the basis of recent publications. It was found that the best described processes are realized with full regioselectivity as well as at mild temperatures. It is interesting, that in some cases of the title reactions, the stepwise mechanism of the formation of the six-membered heterocyclic ring is more possible than classic one-step mechanism.

This short review illustrates the significant advances in the construction of isoquinolone ring with atom- and step-economy, focusing on the intermolecular annulation protocols and intramolecular cyclization in the presence of a variety of catalyst systems. The syntheses of isoquinolone-fused rings are also included.

Abstract:Three-membered rings, such as epoxides, aziridines, oxaziridines, cyclopropenes, vinyloxaziridines, and azirines, are recognized as crucial pharmacophores and building blocks in organic chemistry and drug discovery. Despite the significant advances in the synthesis of these rings through photo/electrochemical methods over the past decade, there has currently been no focused discussion and updated overviews on this topic. Therefore, we presented this review article on the efficient synthesis of three-membered rings using photo- and electrochemical strategies, covering the literature since 2015. In this study, a conceptual overview and detailed discussions were provided to illustrate the advancement of this field. Moreover, a brief discussion outlines the current challenges and opportunities in synthesizing the three-membered rings using these strategies.

Abstract:Benzoisoxazoles, with a distinctive 10‐π‐electron system and significant polarity, readily trigger N‐O bond cleavage, enabling multifaceted applications in organic synthesis. Over the past decade, the exponentially increased use of benzoisoxazoles towards the synthesis of highly valuable N‐containing scaffolds, especially N‐heterocycles, has garnered substantial attention. In this review, we summarize recent developments in the transformation of 2,1‐ and 1,2‐benzoisoxazoles since 2015, highlighting the neglected resemblances of these two nitroxy heterocycles. Key processes in the assorted reactions of benzoisoxazoles, including mechanisms, scope, catalyst‐controlled chemodivergence, current limitations, and developmental prospects, are also introduced and discussed.
