Abstract:Lone-pair−π interactions, which form a stabilizing association between a lone pair of electrons and a π system, are widely recognized as supramolecular bonds. However, such noncovalent interactions rarely serve as a key stereocontrolling element in chemical reactions. Herein, we report a well-engineered lone-pair−π interaction-directed catalytic system for asymmetric dihydroxylation (AD)-based desymmetrization. The lone-pair−π interaction between the C9 carbonyl in mono-cinchona alkaloids and the electron-deficient phthalazine in the symmetric alkene substrates was exploited to realize a highly efficient AD-based desymmetrization, which was difficult to execute following the traditional Sharpless asymmetric dihydroxylation method.

Abstract:Herein we report a catalytic asymmetric inverse-electron-demand Diels-Alder reaction between alkylidene pyrazolones and allyl ketones. Allyl ketone gets activated by a bifunctional thiourea catalyst and acts as a dienolate in this reaction. The trisubstituted tetrahydropyrano[2,3-c]pyrazoles were obtained in moderate to good yields with high diastereo- and enantioselectivities. Few applications, including a decarbonylation reaction, have been demonstrated.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:The organopalladium complex containing ortho-metalated (S)-[1-(dimethylamino)ethyl]naphthalene as the chiral auxiliary has been successfully employed to promote the asymmetric Diels–Alder reaction between 3,4-dimethyl-1-phenylphosphole and (Z)-diphenyl-1-styrylphosphine or (E)-diphenyl-1-styrylphosphine. In the intramolecular cycloaddition reaction between (E)-diphenyl-1-styrylphosphine and 3,4-dimethyl-1-phenylphosphole, a pair of diastereomeric endo-cycloadducts were obtained in the ratio of 1:1. However, the asymmetric reaction between (Z)-diphenyl-1-styrylphosphine and 3,4-dimethyl-1-phenylphosphole also produced the same endo-product in the same stereoselectivity unexpectedly. The chiral naphthylamine auxiliary could be removed chemoselectively from the template by treatment with concentrated hydrochloric acid to generate the neutral stable dichloro complex [(P–P)PdCl2]. Treatment of the dichloro complex with KCN gave the pure bis-phosphine ligand in quantitative yield.

Abstract:The catalytic asymmetric dearomatization (CADA) reaction has proved to be a powerful protocol for rapid assembly of valuable three-dimensional cyclic compounds from readily available planar aromatics. In contrast to the well-studied indoles and naphthols, phenols have been considered challenging substrates for intermolecular CADA reactions due to the combination of strong aromaticity and potential regioselectivity issue over the multiple nucleophilic sites (O, C2 as well as C4). Reported herein are the chiral phosphoric acid-catalyzed divergent intermolecular CADA reactions of common phenols with azoalkenes, which deliver the tetrahydroindolone and cyclohexadienone products bearing an all-carbon quaternary stereogenic center in good yields with excellent ee values. Notably, simply adjusting the reaction temperature leads to the chemo-divergent intermolecular (3 + 2) and alkylation dearomatization reactions. Moreover, the stereo-divergent synthesis of four possible stereoisomers in a kind has been achieved via changing the sequence of catalyst enantiomers.

Abstract:The dehydro-Diels–Alder (DDA) reaction is a powerful method for the construction of aromatic compounds. However, the enantioselective DDA reaction has been rarely developed, probably due to the competitive thermal reaction. Herein, we report a copper-catalyzed enantioselective DDA reaction through vinyl cation pathway. The reaction leads to the atom-economical synthesis of axially chiral phenyl and indolyl carbazoles in generally excellent yields with good to excellent atroposelectivities. This methodology represents the first example of non-noble metal-catalyzed enantioselective DDA reaction. Notably, new chiral ligand and organocatalyst derived from the constructed axially chiral carbazole are demonstrated to be useful in asymmetric catalysis.

Abstract:A series of C2-symmetric and asymmetric chiral thiourea derivatives were synthesized from commercial l-phenylalanine. All of the new compounds have been fully characterized by IR, 1H NMR, 13C NMR, MS spectra and elemental analyses. The chiral thioureas were used as chiral ligands in the catalytic enantioselective ethylation of aldehydes with diethylzinc, the corresponding sec-alcohols were gained with excellent enantioselectivities (up to 87.1% ee) and high yields (up to 76.7%) after the conditions were optimized.

Abstract:A remote cascade asymmetric inverse‐electron‐demand oxa‐Diels−Alder reaction of allyl ketones with isatin‐derived β,γ‐unsaturated α‐keto esters has been developed in the presence of a chiral bifunctional squaramide catalyst. Taking advantage of the secondary amide activating group, a series of enantioenriched 3,4'‐pyranyl spirooxindole derivatives bearing three contiguous chiral centers were attained in high yields (84 to >99%) with excellent enantioselectivity (96−99% ee). Moreover, the gram‐scale synthesis and the construction of 1‐benzazepine scaffold by the product were also demonstrated.

Abstract:Peptide‐phosphonium salt (PPS) catalysts, which contain multiple hydrogen‐bonding donors, have displayed remarkable phase‐transfer catalytic efficiency in recent years. Abstract:AbstractThe enantioselective aza‐Diels–Alder reaction is efficient for constructing chiral tetrahydropyridines, but the catalytic asymmetric aza‐Diels–Alder reaction of ketimines with unactivated dienes is still a challenging topic. Herein, guided by computational screening, a highly enantioselective aza‐Diels–Alder reaction of 2‐aryl‐3H‐indol‐3‐ones with unactivated dienes was realized by using a B(C6F5)3/chiral phosphoric acid catalyst system under mild conditions. The reaction has a broad scope with respect to both aza‐Diels–Alder reaction partners and hence offers rapid access to an array of tetrahydropyridine derivatives with pretty outcomes (up to 99 % yield, >20:1 dr and 98:2 er). The reaction is very efficient: lowering catalyst loadings for the model reaction to 0.1 mol %, enantioselectivity is still maintained. The synthetic utility was confirmed by transformations of the products. DFT calculations provide convincing evidence for the interpretation of stereoselection.

Abstract:A novel proline derivative, (4R)-4-(beta-Naphthalenyl)methoxy-(S)-proline, was conveniently prepared from the naturally occurring (4S)-hydroxy-(S)-proline; 5 mol % of this compound efficiently catalyzes the asymmetric aldol reactions of various benzaldehydes with acetone in excess of acetone as the solvent, giving the aldol adducts in good yields with ee up to 89.8%.

Abstract:A palladium -catalyzed asymmetric 1,n-remote aminoacetoxylation of cis-alkenes has been developed using PhI(OAc)2 as oxidant, providing the acetoxylated lactams with excellent enantioselectivities under mild reaction conditions. The sterically hindered pyridine-oxazoline (Pyox) L3 with a tert-butyl group in oxazoline ring and propyl group in C6 position of pyridinyl is vital for the reaction, where the former is good for asymmetric aminopalladation step and the latter for the chain walking process. The enantioenriched lactam products were proven to be good building blocks for the synthesis of azabicycles.

Abstract:We report a Cu(I)-Ph-BPE-catalyzed asymmetric vinylogous Mannich reaction of β,γ-alkynyl-α-ketimino esters with β,γ-unsaturated N-acylpyrazoles. In this process, the Cu(I)-Ph-BPE catalyst activates the β,γ-alkynyl-α-ketimino ester through N,O-coordination, enabling the subsequent nucleophilic addition of a dienolate generated from the β,γ-unsaturated N-acylpyrazole via α-position deprotonation with a catalytic amount of tertiary amine. The reactions gave useful products with very high enantioselectivities. A broad range of substrates with various substituents are tolerated in this reaction. The versatility of this method was demonstrated by a gram-scale reaction, and subsequent elaboration of the Mannich adducts was also provided.
