Abstract:The catalytic activity of novel carbohydrate-derived prolinamide in the enantioselective aldol reactions has been examined for the first time. The organocatalyst 2b was found to be an efficient organocatalyst for the asymmetric aldol reaction in water. This organocatalyst is applicable to the reactions of a wide range of aromatic aldehydes with cyclic and acyclic ketones in high yields (up to 98%). These reactions exhibit high anti/syn ratios (up to 99%) and excellent enantioselectivities (up to 99%). Importantly, the catalyst can be easily recovered and recycled with only a slight decrease in enantioselectivity after five cycles.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:A series of new N-terminal prolyl-dipeptide derivatives have been synthesized and evaluated as organocatalysts for the direct asymmetric aldol reaction of acetone with electron-deficient aromatic aldehydes. At room temperature, the presence of 10 mol % of catalysts 2 and 5 efficiently catalyzes the direct asymmetric aldol reaction to give the aldol adducts with modest to excellent enantiomeric excesses (ee) values, which are up to 96%. Abstract:A novel proline derivative, (4R)-4-(beta-Naphthalenyl)methoxy-(S)-proline, was conveniently prepared from the naturally occurring (4S)-hydroxy-(S)-proline; 5 mol % of this compound efficiently catalyzes the asymmetric aldol reactions of various benzaldehydes with acetone in excess of acetone as the solvent, giving the aldol adducts in good yields with ee up to 89.8%.

Abstract:A novel hyperbranched polyethylene (HBPE)-supported L-proline has been developed via a bottom-up copolymerization strategy. Copolymerization of ethylene and a protected proline acrylate comonomer with cationic Pd-diimine catalysts was conducted, followed by de-protection of the proline groups. Well-defined HBPE copolymers having molecular weights (MWs) of 10.3-50.3 kDa and 3.2-15.6 L-proline molecules per HBPE polymer chain were synthesized. The effects of the L-proline amount and HBPE MW on the catalyst performance were studied. The HBPE catalysts were efficient in asymmetric aldol reactions of p-nitrobenzaldehyde (p-NBA) or benzaldehyde derivatives with cyclohexanone. High p-NBA conversions of up to 98% and excellent product selectivities with anti/syn = 98/2 and ee > 99% were achieved. Moreover, the HBPE catalysts could be easily recovered by adding water into the product. The recovered catalysts could be reused multiple times with only a slight decline in reactivity and selectivity.

Abstract:L-proline amides derived from various chiral beta-amino alcohols that bear substituents with various electron natures at their stereogenic centers are prepared and evaluated for catalyzing the direct Aldol reaction of 4-nitrobenzaldehyde with acetone. Catalysts with strong electron-withdrawing groups are found to exhibit higher catalytic activity and enantioselectivity than their analogues with electron-donating groups. The presence of 2 mol % catalyst 4g significantly catalyzes the direct Aldol reactions of a wide range of aldehydes with acetone and butanone, to give the beta-hydroxy ketones with very high enantioselectivities ranging from 96% to >99% ee. High diastereoselectivity of 95/5 was observed for the anti Aldol product from the reaction of cyclohexanone, and excellent enantioselectivity of 93% ee was provided for anti Aldol product from the reaction of cyclopentanone.

Abstract:The Aldol reaction is recognized as one of the most powerful methods for the formation of carbon-carbon bonds in modern organic synthesis.[1] It provides an efficient approach to synthesis β-hydroxy carbonyl compounds,at the same time,producing two new stereocenters.Since the first intermolecule proline-catalyzed direct aldol reaction was described by List and co-workers in 2000,[2] organocatalytic stereoselective methods for asymmetric direct aldol reactions have obtained intensive research.Here we synthesized a new class of chiral bifunctional primary amines which could catalyze direct aldol reaction of cyclohexanone and p-nitrobenzaldehyde in the presence of 10 mol % of catalyst1,5 mol % of 2-naphthol as cocatalyst and iso-propanol as solvent,affording the products with moderate to high yield,enantioselectivity and diastereoselectivity(up to 98% yield,84 %ee.,82: 18dr).The mechanism of reaction is still on-going.

Abstract:An easily available and efficient chiral N,N'-dioxide-nickel(II) complex catalyst has been developed for the direct catalytic asymmetric aldol reaction of alpha-isothiocyanato imide with aldehydes which produces the products in morderate to high yields (up to 98 %) with excellent diastereo- (up to >99:1 d.r.) and enantioselectivities (up to >99 % ee). A variety of aromatic, heteroaromatic, alpha,beta-unsaturated, and aliphatic aldehydes were found to be suitable substrates in the presence of 2.5 mol % L-proline-derived N,N'-dioxide L5-nickel(II) complex. This process was air-tolerant and easily manipulated with available reagents. Based on experimental investigations, a possible transition state has been proposed to explain the origin of reactivity and asymmetric inductivity.

Abstract:Sterically and electronically tunable and bifunctional organocatalysts have been developed and evaluated in the direct aldol reaction of heterocyclic ketones. Catalysts with different substituents showed variable catalytic efficiency for analogous substrates, indicating the importance of fine-tuning the strength of the hydrogen bonding in the two NH groups. The reactions all proceeded in good to high yield and with excellent enantioselectivities ranging from 90% to > 99% ee. In most cases, high diastereoselectivities ranging from 96/4 to 99/1 were obtained for the anti aldol adduct.

Abstract:A new, simple bifunctional, recoverable and reusable L-prolinamide organocatalyst that promotes aldol reactions while achieving a respectable level of enantioselectivity is reported. This organocatalyst is applicable to the reactions of a wide range of aromatic and heteroaromatic aldehydes with cyclic and acyclic ketones, and the anti-aldol products could be obtained with up to 99:1 anti/syn ratio and 98% ee. The catalyst can be easily recovered and reused, and only a slight decrease of enantioselectivity was observed for five cycles. This novel catalyst can be efficiently used in large-scale reactions with the enantioselectivity being maintained at the same level, which offers a great possibility for application in industry.

Abstract:Direct asymmetric aldol additions of acetone/and butanone to alpha-ketoesters were achieved using L-proline as a chiral catalyst. Various optically active alpha-hydroxyesters were obtained in the yields of 43 % - 93 % with enantioselectivities up to 81 % e.e. The steric effect seems to be an important factor which influences the activity and the selectivity of the reaction. Acetone showed higher reactivity than butanone while the latter provided better enantioselectivity in some cases.

Abstract:L-Prolinethioamide derivative 1c, prepared from the readily available natural amino acids L-proline and L-valine, was studied for the direct asymmetric aldol reaction of acetone with various aromatic aldehydes at 0 degrees C or room temperature. A loading of only 0.1-0.2 mol-% of derivative 1c was employed in this catalytic system, and excellent enantio-selectivities and yields (up to 98% yield, >99 % ee) could be achieved in aqueous media.

Abstract:A series of chiral prolinamide compounds with pyridine-2, 6-dicarboxylic acid moieties derived from L-proline have been designed and synthesized, their catalytic properties for direct asymmetric aldol reactions were also studied in this article. These catalysts gave the aldol product in high yield (87%) and high enantioselectivity, up to 85%, of the anti-structure at room temperature but gave disappointing results at a lower temperature or when additive was added. Conditions, including solvents, temperature and additives were screened for the reactions. Moreover, the influence of presence of water on yield and stereoselectivity was also discussed.

Abstract:l-Prolinamides 2, prepared froml-proline and simple aliphatic and aromatic amines, have been found to be active catalysts for the direct aldol reaction of 4-nitrobenzaldehyde with neat acetone at room temperature. They give moderate enantioselectivities of up to 46% enantiomeric excess (ee). The enantioselectivity increases as the amide N—H becomes a better hydrogen bond donor.l-Prolinamides 3, derived from the reaction ofl-proline with α,β-hydroxyamines such that there is a terminal hydroxyl group, show more efficient catalysis and higher enantioselectivities. In particular, catalyst 3h, prepared froml-proline and (1S,2S)-diphenyl-2-aminoethanol, exhibits high enantioselectivities of up to 93% ee for aromatic aldehydes and up to >99% ee for aliphatic aldehydes under –25°C. Model reactions of benzaldehyde with three enamines derived from the condensation of prolinamides with acetone have been studied by quantum mechanics calculations. The calculations reveal that the amide N—H and the terminal hydroxyl groups form hydrogen bonds with the benzaldehyde substrate. These hydrogen bonds reduce the activation energy and cause high enantioselectivity. Our results suggest a new strategy in the design of new organic catalysts for direct asymmetric aldol reactions and related transformations.

Abstract:Compared with the well-explored enamine catalysis with secondary amines, the development of efficient enamine-based primary amine catalysts has remained as an elusive goal until recently. We present herein that a simple chiral primary-tertiary diamine 1d in combination with TfOH acts as an efficient enamine-based primary amine catalyst. The 1d-TfOH catalytic system could effectively catalyze the asymmetric direct aldol reaction with broad substrate scope as well as high regio- and diastereoselectivity and enantioselectivity under ambient temperature. Significantly, the reactions accommodate the synthetically important but challenging substrates, such as linear aliphatic ketones, with high regioselectivity and unprecedented syn diastereoselectivity. These results are in sharp contrast with the secondary amine mediated similar reactions wherein anti diastereoselectivity was normally observed.

Abstract:Enantioselective aldol reaction of ketones with aldehydes catalyzed by (2S,5S)-pyrrolidine-2,5-dicarboxylic acid in the presence of an equal molar amount of Et3N was described. By using the new chiral organocatalyst, the direct aldol condensation products were obtained in reasonable yields and up to 90% ee.

Abstract:A series of novel proline-based organocatalysts with amide and thiourea-amine units (7a–7f) were developed and evaluated in the asymmetric aldol reaction of 4-nitrobenzaldehyde with cyclohexanone. The organocatalyst (7c or 7d, 5 mol%) exhibited efficient catalytic activity to afford aldol products in high diastereoselectivity (up to >99:1), enantioselectivity (up to >99%), and yield (up to >96%) at 0 °C in saturated brine without adding an acid. Aldol products of benzaldehyde derivatives almost universally provide high diastereoselectivity and enantioselectivity.
