Abstract:A new catalytic enantioselective approach for the formation of allyl alpha-amino acid derivatives by reaction of N-tosyl alpha-imino esters with allyl stannanes and silanes catalyzed by chiral copper(I) complexes has been developed. A series of different BINAP and phosphine-oxazoline (P,N) ligands have, in combination with various Lewis acids, been tested as chiral catalysts for allylation of N-tosyl alpha-imino esters. It has been found that both type of ligands, in combination with copper(I) salts, give highly valuable unsaturated alpha-amino acid derivatives. The reaction has been investigated for different allyl stannanes and silanes, and it has been found that tri-n-butyl allyl stannane gives the best results of the simple allyl compounds tested, leading to gamma,delta-unsaturated alpha-amino acid derivatives in up to 94% yield and with up to 83% ee, which can be improved to be >95% ee by recrystallization. The reaction has also been investigated using different acyclic and cyclic allyl stannanes leading to various types of unsaturated alpha-amino acid derivatives in very high yield (up to 95%) and with up to 98% ee. The stereochemistry and absolute configurations of the allyl alpha-amino acid derivatives have been determined by X-ray analysis, and it is suggested that the reaction takes place as an ene-like reaction.

Abstract:A three-component reaction of one molecule of imine and two molecules of alkynoates is realized with the catalysis of Cu(I)–Fe(III) in a sequential manner to allow the direct synthesis of functionalized pyrroles, during which, one C–N bond and two C–C bonds are formed with high atom economy. This method benefits from easily available starting materials, low-cost catalysts, and convenient operations.

Abstract:AbstractDirect α‐functionalization of NH2‐free glycinates with relatively weak electrophiles such as α,β‐unsaturated esters still remains a big challenge in organic synthesis. With chiral pyridoxal 5 d as a carbonyl catalyst, direct asymmetric conjugated addition at the α‐C of glycinate 1 a with α,β‐unsaturated esters 2 has been successfully realized, to produce various chiral pyroglutamic acid esters 4 in 14–96 % yields with 81–97 % ee's after in situ lactamization. The trans and cis diastereomers can be obtained at the same time by chromatography and both of them can be easily converted into chiral 4‐substituted pyrrolidin‐2‐ones such as Alzheimer's drug Rolipram (11) with the same absolute configuration via tert‐butyl group removal and subsequent Barton decarboxylation.

Abstract:A simple and efficient methodology was developed for the direct C(sp(2))-H acetoxylation of pyrrolo[2,3-d]pyrimidine derivatives. This highly regioselective one-pot acetoxylation was carried out under transition-metal-free conditions in the presence of NaI. This protocol provides a variety of acetoxylated pyrrolo[2,3-d]pyrimidines in good to excellent yields, which are potentially of great importance in medicinal chemistry. And this methodology was also proved to be practical by the gram-scale synthesis.

Abstract:In the last 20 years, efficient transition metal catalysts for the α‐arylation of enolates have been introduced. Despite the popularity and utility of these reactions, there remains room for improvement (reduced costs, elimination of transition metals and specialized ligands). Herein is reported a general, scalable and green method for aroylation of simple diarylmethane pronucleophiles through direct acyl C–N cleavage of N‐Bn‐N‐Boc arylamides and N‐acylpyrroles under transition metal‐free conditions. Importantly, a 1 : 1 ratio of the amide to the pronucleophile is employed. Unlike. use of Weinreb amides, this method avoids preformed organometallics (organolithium and Grignard reagents) and does not employ cryogenic temperatures, which are difficult and costly to achieve on scale. The operationally simple protocol provides straightforward access to a variety of sterically and electronically diverse 1,2,2‐triarylethanones, a group of compounds with high‐value in medicinal chemistry.

Abstract:Although nucleophilic benzylation-type reaction to introduce various aromatic systems into molecules has been widely explored, the related pyrrolylmethylation version remains to be disclosed. Reported herein is a palladium-catalysed multiple auto-tandem reaction between N-Ts propargylamines, allyl carbonates and aldimines in the presence of an acid, proceeding through sequential allylic amination, cycloisomerisation, vinylogous addition and aromatisation steps. A diversity of formal pyrrolylmethylated amine products were finally furnished efficiently. In addition, switchable regiodivergent 3-pyrrolylmethylation and 4-pyrrolylmethylation were realised by tuning catalytic conditions. Moreover, remote chirality transfer with readily available enantioenriched starting materials was well achieved with an achiral ligand, relying on diastereoselective generation of η2-Pd(0) complexes between Pd(0) and chiral 1,3-diene intermediates in the key vinylogous addition step. A few control experiments was conducted to elucidate the palladium-involved tandem reaction and regiodivergent synthesis.

Abstract:A diastereoselective approach to access cis-pyrido and pyrrolo[1,2-c][1,3]oxazin-1-ones has been developed through a one-pot (BF3Et2O)-Et-.-catalyzed [4+2] process starting with N,O-acetals and terminal alkynes. In addition, the utility of this transformation is demonstrated by the scalable synthesis of (+)-Febrifugine in 7 steps from the N,O-acetal (15% overall yield).

Abstract:The asymmetric allylation of 1-pyrroline-5-carboxylic esters has been accomplished through a synergistic Pd/Cu catalyst system under mild reaction conditions. The mechanistic studies suggested that (1) nucleophilic attack is the enantiodiscriminating step; (2) the cooperative action of two chiral reactive species, N-metalated azomethine ylides and π-allylpalladium, is most likely responsible for its high reactivity and excellent enantioselectivity (up to >99% ee); and (3) the steric hindrance and electronic factors of the allylic electrophiles and imino ester substrates are crucial for the formation of the linear products. A series of 3,4-2 H-pyrrole derivatives bearing a quaternary stereogenic center were easily synthesized in high yields and with high to excellent regioselectivity and enantioselectivity.

Abstract:Transition-metal-free synthesis of α-aryl esters and nitriles using arylboronic acids with α-aminoesters and α-aminoacetonitriles, respectively, as the starting materials has been developed. The reaction represents a rare case of converting C(sp(3))-N bonds into C(sp(3))-C(sp(2)) bonds. The reaction conditions are mild, demonstrate good functional-group tolerance, and can be scaled up.

Abstract:Here, we developed an expedient access route to highly functionalized pyrroles from readily available α-amino acid ester hydrochlorides and alkynals via a cascade condensation/intramolecular cyclization followed by a unique C-N ester migration process. Abstract:Chiral acetylenic derivatives are found in many bioactive compounds and are versatile functional groups in organic chemistry. Here, we describe an enantioselective nickel/Lewis acid-catalyzed asymmetric propargylic substitution reaction from simple achiral materials under mild condition. The introduction of a Lewis acid cocatalyst is crucial to the efficiency of the transformation. Notably, we investigate this asymmetric propargylic substitution reaction for the development of a range of structurally diverse natural products. The power of this strategy is highlighted by the collective synthesis of seven biologically active compounds: (−)-Thiohexital, (+)-Thiopental, (+)-Pentobarbital, (−)-AMG 837, (+)-Phenoxanol, (+)-Citralis, and (−)-Citralis Nitrile.
