Abstract:Two series of N-containing heterocycles were synthesized by one-step intramolecular or intermolecular annulations of N-substituted pyrrole-2-carbaldehydes and pyrrole-2-carbonitriles. These facile and atom-economic synthetic routes involved the inherent three potential electrophilic and nucleophilic reactive sites of the synthons and the reaction solvent plays a crucial role leading to these different types of products. Additionally, plausible mechanisms are proposed.

Abstract:A selective synthesis of either 2H-pyran-2-ones (4) or chiral dihydropyranones (6) from the same substrates of 3-bromoenals and 1,3-dicarbonyl compounds upon oxidative N-heterocyclic carbene catalysis is presented. It is shown that the oxidative transformation of 3-bromoenals under NHC catalyst can be well controlled to proceed through two pathways, i.e., elimination of reducible beta-bromide or by an external oxidant 3, allowing the selective generation two sorts of unsaturated acyl azoliums, respectively.

Abstract:An efficient and highly regioselective Pd-catalyzed direct arene C(sp(2))-H acyloxylation of pyrrolo[2,3-d]pyrimidine derivatives is reported. The key strategy involves the utilization of the unique reactivity of pyrrolo[2,3-d]pyrimidine and the employment of pyrrolo[2,3-d]pyrimidine as the directing group. A variety of monoacyloxylated pyrrolo[2,3-d]pyrimidine derivatives can be achieved by switching the solvents under mild conditions, and they can be further modified and exhibit various biological activities.

Abstract:Arynes are known to serve as highly reactive benzene-based synthons, which have gained numerous successes in preparing functionalized arenes. Due to the superb electrophilic nature of these fleeting species, however, it is challenging to modulate the designated aryne transformation chemoselectively, when substrates possess multiple competing reaction sites. Here, we showcase our effort to manipulate chemoselective control between two major types of aryne transformations using either 3-methoxybenzyne or 3-silylbenzyne, where nucleophilic addition-triggered reactions and non-polar pericyclic reactions could be differentiated. This orthogonal chemoselective protocol is found to be applicable between various nucleophiles, i.e., imidazole, N-tosylated/N-alkyl aniline, phenol, and alcohol, and an array of pericyclic reaction partners, i.e., furan, cyclopentadiene, pyrrole, cycloheptatrienone, and cyclohexene. Beyond arylation reactions, C-N bond insertion, Truce-Smiles rearrangement, and nucleophilic annulation are appropriate reaction modes as well. Moreover, this chemoselective protocol can find potential synthetic application.

Abstract:Herein, we describe a high yielding and switchable synthesis of a range of functionalized pyridines and pyrroles via Cu-catalyzed cascade reactions with alkynes and N-sulfonyl azadienes which are readily available from saccharin. Both unsymmetrical 2,4,6-trisubstituted pyridines and multi-substituted pyrroles could be transformed into some bioactive compounds and chiral ligands.

Abstract:1H‐Pyrroles can be directly functionalized by means of the incorporation of carbene groups from diazocompounds, in a process catalyzed by TpxCu complexes (Tpx = hydrotrispyrazolylborate ligand). The reactions take place with a complete selectivity toward the formal insertion of the carbene into the Cα‐H bond, leading to alkylated pyrroles, with no modification of the Cβ‐H, N‐H or C=C bonds of the pyrrole unit. Alkyl substituents at C‐ring as well as alkyl, aryl, allyl or alkyne substitution at Natom are tolerated, the strategy affording 20 new pyrrole derivatives. The observance of partial deuteration at the methylene group when the reaction is carried out with added D2O serves to discard the direct insertion of the carbene group into the Csp2‐H bond, the alternative electrophilic attack to the pyrrole ring being feasible.

Abstract:A diastereoselective approach to access cis-pyrido and pyrrolo[1,2-c][1,3]oxazin-1-ones has been developed through a one-pot (BF3Et2O)-Et-.-catalyzed [4+2] process starting with N,O-acetals and terminal alkynes. In addition, the utility of this transformation is demonstrated by the scalable synthesis of (+)-Febrifugine in 7 steps from the N,O-acetal (15% overall yield).

Abstract:C2-arylation of N-acyl pyrroles with aryl halides is developed for the first time using Pd(PPh3)4 as a catalyst in combination with Ag2CO3 under air, which allowed the application of a good compatibility catalytic system. This protocol provides a straightforward method for the preparation of valuable arylated pyrroles in moderate to good yields under the standard conditions with good substrate tolerance. Interestingly, while N-benzoyl pyrroles reacted well, the use of substrates with a thiophene or furan ring indicated that the thiophene and furan rings are more reactive than pyrrole for the present catalytic system.

Abstract:We present herein an unprecedented stereoselective synthesis of triaryl-2-pyrones with monoaxial or contiguous diaxes from readily available starting materials. This N-heterocyclic carbene catalysis method adopts an atroposelective annulation of 2-aryketones with ynals under oxidative conditions. The annulation includes the construction of one or two axes in a single operation, achieves step economy, and affords axially chiral triaryl-2-pyrones in moderate to good yields, with high to excellent enantioselectivities. DFT calculations of the relative energies of stereoisomers and rotational barriers were performed.

Abstract:A highly enantioselective allylic C-H alkylation reaction of allylarenes with pyrazol-5-ones has been established by the cooperative catalysis of a chiral palladium complex and chiral Bronsted acid to afford a wide spectrum of functionalized chiral N-heterocycles with an all-carbon quaternary stereogenic center in high yields and with high levels of enantioselectivity (up to 96% ee), wherein the chiral ligand and phosphoric acid showed synergistic effect on the control of stereoselectivity. In addition, a palladium-catalyzed asymmetric allylic C-H alkylation of 1,4-pentadienes with pyrazol-5-ones has been realized to furnish highly functionalized pyrazol-5-ones in high enantioselectivities. In this case, the chiral ligand controls the stereoselectivity while the achiral Bronsted acid, 2-fluorobenzoic acid, turns out to be a better cocatalyst than the chiral phosphoric acid. The installation of electron-deficient substituents at 3,3'-positions of binaphthyl backbone of chiral phosphoramidites is actually beneficial to the allylic C-H oxidation due to their survival in the presence of quinone derivative oxidants. These allylic C-H alkylation reactions undergo smoothly under mild conditions and tolerate a wide range of substrates. The resultant highly functionalized chiral pyrazol-5-ones have been applied to the preparation of more structurally diverse heterocycles by classical transformations.

Abstract:A transformation of fluxional into configurationally stable axially chiral N‐arylpyrroles was achieved with a highly atroposelective electrophilic aromatic substitution catalyzed by a chiral‐at‐metal rhodium Lewis acid. Specifically, N‐arylpyrroles were alkylated with N‐acryloyl‐1H ‐pyrazole electrophiles in up to 93 % yield and with up to >99.5 % ee , and follow‐up conversions reveal the synthetic utility of this new method. DFT calculations elucidate the origins of the observed excellent atroposelectivity.

Abstract:The apparent and the real: What looks like a Friedel-Crafts alkylation reaction of electron-deficient pyrroles is actually a Pd(II)-catalyzed, norbornene-mediated C-H activation reaction, in which the alkylation of the pyrrole core occurs by reductive elimination. As well as ethyl-1H-pyrrole-2-carboxylate, several other 2,3-disubstituted pyrroles underwent the selective C5 alkylation in good yield.

Abstract:Here, we developed an expedient access route to highly functionalized pyrroles from readily available α-amino acid ester hydrochlorides and alkynals via a cascade condensation/intramolecular cyclization followed by a unique C-N ester migration process. A variety of 1,2,3-trisubstituted pyrroles, which were difficult to acquire with the common methodologies, were successfully prepared in good yields under mild conditions.

Abstract:4-Functionalized pyridines are privileged scaffolds in various fields. The C-4 selective functionalization of pyridines is perhaps the most convenient protocol to pyridine derivatives. However, this reaction is still unexplored because of the inert property of pyridine and the challenge in the regioselectivity. Herein, we report an aromatic C-4 selective functionalization of pyridine N-oxides with DABCO and Tf2O as the regioselective controller. The commercial availability of reagents, good tolerance of functional groups, and simple operability made it a promising approach to 4-functionalized pyridines. The mechanism studies revealed that the N-(4-pyridyl)-DABCO salt was the key intermediate, and the bulky steric hindrance of the CF3SO2 group and DABCO was the origin of C-4 selectivity. This intermediate can serve as a generic handle to realize C–X, C–N, C–O, C–S, and C–P bond formation.

Abstract:Pyridines and related N-heteroarenes are commonly found in pharmaceuticals, agrochemicals and other biologically active compounds1,2. Site-selective C–H functionalization would provide a direct way of making these medicinally active products3–5. For example, nicotinic acid derivatives could be made by C–H carboxylation, but this remains an elusive transformation6–8. Here we describe the development of an electrochemical strategy for the direct carboxylation of pyridines using CO2. The choice of the electrolysis setup gives rise to divergent site selectivity: a divided electrochemical cell leads to C5 carboxylation, whereas an undivided cell promotes C4 carboxylation. The undivided-cell reaction is proposed to operate through a paired-electrolysis mechanism9,10, in which both cathodic and anodic events play critical roles in altering the site selectivity. Specifically, anodically generated iodine preferentially reacts with a key radical anion intermediate in the C4-carboxylation pathway through hydrogen-atom transfer, thus diverting the reaction selectivity by means of the Curtin–Hammett principle11. The scope of the transformation was expanded to a wide range of N-heteroarenes, including bipyridines and terpyridines, pyrimidines, pyrazines and quinolines. An electrochemical strategy is described in which the direct carboxylation of pyridines and related N-heteroarenes with CO2 shows divergent site selectivity depending on the type of reactor used.

Abstract:AbstractA novel rhodium‐catalyzed dearomatization of O‐substituted pyridines to access N‐substituted 2‐pyridones has been developed. A computational study suggests a mechanism involving the formation of a pyridinium ylide followed by an unprecedented 1,4‐acyl migratory rearrangement from O to C. Furthermore, the chiral dirhodium complexes serve as the catalyst for the asymmetric transformation with excellent enantioselective control. DFT calculations indicate the chirality is transferred from axial chirality to the central stereogenic centre. The stronger π–π interaction and CH–π interaction account for the high enantioselectivity.
