Abstract:A series of phosphorescent zwitterionic iridium(III) complexes, with 4-carboxy-2, 2'-bipyridine-4'-carboxylate (Hdcbpy) as ancillary ligand, Ir(C(wedge)N)(2)(Hdcbpy) (C(wedge)N = 1-phenylpyrazole (ppz), 1-phenyl-pyridine (ppy), 2-(4',6'-difluoro-phenyl)pyridine (dfppy), 1-phenyl-isoquinoline (piq), dibenzo[f,h]quinoxaline (dbq)), were prepared and characterized. Their photophysical properties were studied, and intense luminescence emissions were observed based on metal-to-ligand-charge-transfer ((3)MLCT), ligand-to-ligand charge-transfer ((3)LLCT), ligand-centered transitions ((3)LC, i.e., (3)pi --> pi*), or intraligand-charge-transfer ((3)ILCT) excited states, which were confirmed by theoretical calculations. The quantum yield of Ir(dfppy)(2)(Hdcbpy) is as high as 0.106 in aqueous solution. With Hdcbpy as a hydrophilic part, their amphiphilic structures as further confirmed by X-ray single crystal data endow them with different solubilities in phosphate buffer solution (PBS, pH 7.0). The compounds were successfully applied as luminescent dyes for cell imaging in aqueous solution. Their different stain ability in cell imaging was fairly well supported by the experimental data based on the measurement of oil/water partition coefficients and encapsulation/release with liposomes.

Abstract:Detecting viscosity is vital for pathological investigation since subtle changes in cellular microviscosity are related to pathologies and diseases. In this work, five cationic Ir(III) complexes Ir1–Ir5 with the formulas as [Ir(ppy)2L]PF6 have been synthesized, where L1-L5 are a family of coplanar bithiazole derivatives having versatile donor/acceptor tails (proton, bromo, tetraphenylethylene, benzothiophene, ethyl benzoate). Fluorescent experiment shows that L1-L5 have a fluorescent activity with high quantum yields (Ф > 33%). However, the quantum yields of Ir1–Ir5 are weak in the range from 2.7% to 7.7%. There is a linear relationship between Log(I620 nm) and Log(viscosity), which can quantitatively measure viscosity in the range of 1.36–669 cP. Furthermore, a cell colocalization experiment has been performed using commercial nuclear dye (DAPI), lysosomal dye (LysoGreen), and mitochondrial dye (MitoGreen) to explore the subcellular localization ability of Ir1 in living cells, indicating that the probe Ir1 can easily pass through the cell membrane, and can accurately target lysosomes in living cells. In addition, complex Ir1 can quantitatively detect viscosity and target lysosomes, possessing the potential for practical application in intracellular viscosity detection.

Abstract:We report herein the design, synthesis and characterisation of a series of luminescent iridium(III) porphyrin complexes [Ir(ttp)(CH 2 CH 2 OH)] (H 2 ttp = 5,10,15,20-tetra-4-tolylporphyrin) ( 1 ), [Ir(tpp-Ph-NO 2 )(CO)Cl] (H 2 tpp-Ph-NO 2 = 5-(4-((4-nitrophenoxy)carbonyloxymethyl)phenyl)-10,15,20-triphenylporphyrin) ( 2 ), [Ir(tpp-COOMe)(Py) 2 ](Cl) (H 2 tpp-COOMe = 5-(4-methoxycarbonylphenyl)-10,15,20-triphenylporphyrin; Py = pyridine) ( 3 ) and [Ir(tpp-COOH)(Py) 2 ](Cl) (H 2 tpp-COOH = 5-(4-carboxylphenyl)-10,15,20-triphenylporphyrin) ( 4 ). All the complexes displayed long-lived near-infrared (NIR) emission attributed to an excited state of mixed triplet intraligand ( 3 IL) (π →π*) (porphyrin) and triplet metal-to-ligand charge transfer ( 3 MLCT) (dπ(Ir) → π*(porphyrin)) character. The cytotoxicity of the complexes toward HeLa cells was examined by the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium bromide (MTT) assay. The cationic complexes 3 and 4 exhibited higher cytotoxic activity toward HeLa cells than their neutral counterparts 1 and 2 . Cellular uptake studies by inductively coupled plasma-mass spectrometry (ICP-MS) and laser-scanning confocal microscopy (LSCM) indicated that complexes 3 and 4 showed higher cellular uptake efficiencies than complexes 1 and 2 due to their cationic charge, and they were enriched in the perinuclear region of the cells with negligible nucelar uptake. Additionally, the carboxyl complex 4 was used to label a model protein bovine serum albumin (BSA) via an amidation reaction. The resultant luminescent protein conjugate 4-BSA displayed similar photophysical properties and intracellular localisation behaviour to its parent complex. The results of this work will contribute to the development of luminescent iridium(III) porphyrin complexes and related bioconjugates as NIR-emissive probes for bioimaging applications.

Abstract:Luminescent iridium(III) complexes exhibit high phosphorescence quantum yields and photostability, good cell membrane permeability, rich and sensitive photophysical properties, and thus have been extensively studied as intracellular sensors for photoluminescence microscopy imaging. The microenvironmental parameters and interactions of the complexes with cellular biological species can be readily reflected by changes in luminescence properties. In many studies, such changes are limited to responses in the phosphorescence intensity and wavelength; the phosphorescence lifetime is often ignored because it is invisible and difficult to analysis in cellular conditions. In recent decades, photoluminescence lifetime imaging microscopy (PLIM) has been emerging and fast developed, which allows visualizing and mapping the decays of luminescent dyes in the time domain during cellular imaging. In this review article, we focus on the design and applications of lifetime-responsive phosphorescent iridium(III) complexes as imaging reagents for mapping environmental parameters and sensing analytes of interest in intracellular conditions. These parameters and analytes either interact with the probes in the excited state to affect their decay rates or directly react with the probes yielding new products. The utilization of the phosphorescent complexes to visualize intracellular labeling of biomolecules via PLIM are also discussed.

Abstract:Many luminescent probes have been developed for intracellular imaging and sensing. During cellular luminescence sensing, it is difficult to distinguish species generated inside cells from those internalized from extracellular environments since they are chemically the same and lead to the same luminescence response of the probes. Considering that endogenous species usually give more information about the physiological and pathological parameters of the cells while internalized species often reflect the extracellular environmental conditions, we herein reported a series of cyclometalated iridium(iii) complexes as phosphorescent probes that are partially retained in the cell membrane during their cellular uptake. The utilization of the probes for sensing and distinguishing between exogenous and endogenous analytes has been demonstrated using hypoxia and hypochlorite as two examples of target analytes. The endogenous analytes lead to the luminescence response of the intracellular probes while the exogenous analytes are reported by the probes retained in the cell membrane during their internalization.

Abstract:Improvement of cellular uptake and subcellular resolution remains a major obstacle in the successful and broad application of cellular optical probes. In this context, we design and synthesize seven non-emissive cyclometalated iridium(III) solvent complexes [Ir(C-N)(2)(solv)(2)]L-+(-) (LIr2-LIr8, in which C-N = 2-phenylpyridine (ppy) or its derivative; solv = DMSO, H2O or CH3CN: L- = PF6- or OTf-) applicable in live cell imaging to facilitate selective visualization of cellular structures. Based on the above variations (including different counter ions, solvent ligands, and C-N ligands), structure-activity relationship analyses reveal a number of clear correlations: (1) variations in counter anions and solvent ligands of iridium(III) complexes do not affect cellular imaging behavior, and (2) length of the side carbon chain in C-N ligands has significant effects on cellular uptake and localization/accumulation of iridium complexes in living cells. Moreover, investigation of the uptake mechanism via low-temperature and metabolism inhibitor assays reveal that [Ir(4-Meppy)(2)(CH3CN)(2)]+OTf- (LIr5) with 2-phenylpyridine derivative with side-chain of methyl group at the 4-position as C-N ligand permeates the outer and nuclear membranes of living cells through an energy-dependent, non-endocytic entry pathway, and translocation of the complex from the cell periphery towards the perinuclear region possibly occurs through a microtubule-dependent transport pathway. Nuclear pore complexes (NPCs) appear to selectively control the transport of iridium(III) complexes between the cytoplasm and nucleus. A generalization of trends in behavior and structure-activity relationships is presented, which should provide further insights into the design and optimization of future probes. Abstract:Phosphorescent heavy-metal complexes exhibit relatively long lifetimes and significant Stokes Shifts for easy separation of excitation and emission and could be used as promising candidates for fluorescence probes. To date, however, very limited examples of file use of phosphorescent heavy-metal complexes in bioimaging have been reported. Herein, a series of cationic indium(III) complexes, [Ir(dfpy)(2)L]+PF6- (dfpy = 2-(2,4-difluorophenyl)pyridine; L denotes a series of N boolean AND N ligands with different conjugated lengths), have been synthesized. By variation of the N boolean AND N ligands, file emission colors of these complexes can be tuned from blue to red, while high quantum efficiencies are retained. Further investigation reveals that these complexes exhibit different excited-state properties, which arc responsible for the significant emission color tuning, Moreover, the exclusive staining of cytoplasm and low cytotoxicity were observed for these cationic iridium(III) complexes, as well as good cell membrane permeability, which makes them promising candidates as fluorescent probes for living cell imaging. Furthermore, in order to tune the excitation wavelength to the longer region, another cationic iridium(III) complex with lower singlet and triplet energy levels was also investigated to be used as living cell imaging material. achieving a longer excitation wavelength of 488 nm.

Abstract:Eight neutral iridium(III) complexes (Ir1–Ir8) containing an electron-withdrawing pentafluorosulfanyl (−SF5) group on cyclometalated ligands with different ancillary ligands were synthesized and investigated. 2-(3/4-(Pentafluorosulfanyl)phenyl)pyrimidine (for Ir1 and Ir2), 1-(3/4-(pentafluorosulfanyl)phenyl)isoquinoline (for Ir3 and Ir4), and 2-(3/4-(pentafluorosulfanyl)phenyl)pyridine (for Ir5–Ir8) were chosen as cyclometalated ligands and 2,2,6,6-tetramethylheptane-3,5-dione (for Ir1–Ir4), tetraphenylimidodiphosphinate (for Ir5 and Ir6), and bis(diphenylphorothioyl)amide (for Ir7 and Ir8) were used as ancillary ligands. The crystal structures of Ir1, Ir3, Ir4, Ir6, and Ir8 also confirmed the identities of these complexes. Complexes Ir1, Ir2, and Ir5–Ir8 exhibit sky blue to green emissions (480–533 nm) with high photoluminescence quantum efficiency yields up to 94.7% in CH2Cl2 solution at room temperature. Complexes Ir3 and Ir4 are red phosphors (λmax = 607 nm, Φ = 76.8% for Ir3 and λmax = 627 nm, Φ = 49.2% for Ir4, respectively). Theoretical calculations were carried out to provide a further study of the orbital distributions and electronic states of eight Ir(III) complexes. Additionally, ion detection studies reveal that Ir2 has the potential for detecting Hg2+ ion.

Abstract:Two new iridium complexes,（dfppy） 2 Ir（L-alanine）（dfppy = 2-（2,4-difluorophenyl）pyridine） and（piq） 2 Ir（L-alanine）（piq = 1-phenylisoquinoline） were prepared with L-alanine as ancillary ligand.The two complexes show bright greenish-blue and red emission respectively.Theoretic study demonstrated that the emission nature of these complexes is mainly determined by the main ligand.And their improved aqueous solubility and the retained quantum yield favor their application in cell imaging.Intracellular imaging suggested that these two complexes have fine cell membrane permeability and is mainly distributed in cytoplasm.This study displayed a new strategy to design aqueous soluble phosphorescent cyclometallated Ir（III） complex via introducing amino acid as ancillary ligand.

Abstract:In the present study, we explored a novel design strategy of heteronuclear phosphorescent iridium(III) complexes chelated by BF2 moiety with 3-hydroxypicolinic acid as the chelate ligand and synthesized a new series of iridium(III) complexes [Ir(dfppy)(2)(hpa)BF2] (1b), [Ir(ppy)(2)(hpa)BF2] (2b) and [Ir(tpq)(2)(hpa)BF2] (3b) (hpa = 3-hydroxypicolinic acid, dfppy = 2-(2,4-difluorophenyl) pyridine, ppy = 2-phenylpyridine, tpq = 2-(thiophen-2-yl) quinoline) under mild conditions. The emission colors and wavelengths of iridium(III) complexes can be affected evidently by chelating BF2 moiety into iridium(III) complexes, and this effect will be changed with the difference of cyclometalating C boolean AND N ligands. A combination of UV-vis absorption, photoluminescence, excited-state lifetime measurements and theoretical calculations has provided the significant insight into the nature of the excited state and photophysical properties of these interesting iridium(III) complexes. Moreover, the exclusive staining of cytoplasm and low cytotoxicity were demonstrated for these new iridium(III) complexes, which made them promising candidates as multi-color phosphorescent dyes for living cell imaging.

Abstract:Iridium(III) complexes with intense phosphorescence in solution have been widely applied in organic light-emitting diodes, chemosensors and bioimaging. However, little attention has been paid to iridium(III) complexes showing weak phosphorescence in solution and enhanced phosphorescence emission in the solid state (EPESS). In the present study, two beta-diketonate ligands with different degrees of conjugation, 1-phenyl-3-methyl-4-benzoyl-5-pyrazolone (HL1) and 1-phenyl-3-methyl-4-phenylacetyl-5-pyrazolone (HL2), have been synthesized to be used as ancillary ligands for two iridium(III) complexes, Ir(ppy)(2)(L1) and Ir(ppy)(2)(L2) (Hppy = 2-phenylpyridine). The two complexes have been characterized by single-crystal X-ray crystallography, H-1 NMR and elemental analysis. Interestingly, Ir(ppy)(2)(L1) is EPESS-active whereas Ir(ppy)(2)(L2) exhibits moderately intense emission both in solution and as a neat film, indicating that the degree of conjugation of the beta-diketone ligands determines the EPESS-activity. The single-crystal X-ray analysis has indicated that there are pi-pi interactions between the adjacent ppy ligands in Ir(ppy)(2)(L1) but not in Ir(ppy)(2)(L2). Finally, EPESS-active Ir(ppy)(2)(L1) has been successfully embedded in polymer nanoparticles and used as a luminescent label in bioimaging.

Abstract:Phosphorescent iridium(III) complexes are widely recognized for their unique properties in the excited triplet state, making them crucial for various applications including biological sensing and imaging. Most of these complexes display single phosphorescence emission from the lowest‐lying triplet state after undergoing highly efficient intersystem crossing (ISC) and ultrafast internal conversion (IC) processes. However, in cases where these excited‐state processes are restricted, the less common phenomenon of dual emission has been observed. This dual emission phenomenon presents an opportunity for developing biological probes and imaging agents with multiple emission bands of different wavelengths. Compared to intensity‐based biosensing, where the existence and concentration of an analyte are indicated by the brightness of the probe, the emission profile response involves modifications in emission color. This enables quantification by utilizing the intensity ratio of different wavelengths, which is self‐calibrating and unaffected by the probe concentration and excitation laser power. Moreover, dual‐emissive probes have the potential to demonstrate distinct responses to multiple analytes at separate wavelengths, providing orthogonal detection capabilities. In this concept, we focus on iridium(III) complexes displaying fluorescence‐phosphorescence or phosphorescence‐phosphorescence dual emission, along with their applications as biological probes for sensing and imaging.

Abstract:The three near-infrared-emitting cationic indium(III) complexes [Ir(pbq-g)(2)((NN)-N-boolean AND)]+PF6- (pbq-g = phenylbenzo[g]quinoline; (NN)-N-boolean AND = bipyridine (1), 1,10-phenanthroline (2), 4,7-diphenyl-1,10-phenanthroline (3)) have been demonstrated as phosphorescent dyes in live cell imaging. These complexes with different ancillary ligands show similar near-infrared (NIR) emission With lambda(max,peak) at 698 nm and lambda(max;shoulder) at 760 nm in CH2Cl2 solutions, with a moderate quantum yield of around 3%. However, these complexes behave quite differently as NIR dyes for live cell imaging. Complexes 1 and 2 exhibit exclusive staining in the cytoplasm with good cell membrane permeability under excitation at 488 nm, while 3 gives almost no cell uptake, as further determined by flow cytometry. Although the lipophilicities of these complexes follow the order 1 < 2 < 3, their cytotoxicities are in the reverse order. The exceptionally low cytotoxicity of 3 could be attributed to its poor solubility in aqueous buffer and thus substantially low exposure dose. This comparative study suggested that the ancillary ligands could fine-tune the amphiphilicity and cytotoxicity of the cyclometalated iridium(III) complexes and thus might play a key role in the design of NIR-emitting iridium(III) complexes for practical applications in bioimaging.

Abstract:Live cell actin visualization is fundamental for exploring cellular motility, cytokinesis, intracellular transport, and other correlated functions. The current imaging techniques that allow imaging of actin in its native environment are optical and electron microscopy . Such imaging techniques offer high enough resolution to investigate the ultrastructure of actin however they come at the expense of actin integrity. Inspired by the lack of suitable probes that preserve actin's integrity, we designed a cyclometalated Ir (III) complex that interacts with live cells and displays light switch behaviour upon specific actin binding. The exceptional photophysical properties of the proposed probe allow unprecedented resolution of cytoskeleton ultrastructures under stimulated emission depletion (STED) super-resolution nanoscopy. Moreover, the Ir complex enables the capability of visualizing actin polymers and periodicity under correlative light electron microscopy (CLEM) and liquid-phase electron microscopy (LPEM) at ∼ 8 nm resolution.

Abstract:Homocysteine (Hcy) and cysteine (Cys) are crucial to the physiological balance in living systems. Specific detection of intracellular Hcy and Cys is of growing importance. Herein, we demonstrated phosphorescence imaging of intracellular Hcy and Cys using a cationic iridium(III) complex Ir(pba)(2)(bpy)(+)center dot PF(6)(-) [pba = 4-(2-pyridyl)benzaldehyde, bpy = bipyridine] containing aldehyde groups as a luminescent probe. Upon addition of Hcy or Cys to a semiaqueous solution of Ir(pba)2(bpy)+, a change in luminescence from yellow to red was visible to the naked eye. The successful chemical reaction of the aldehyde in Ir(pba)(2)(bpy)(+) with Hcy and Cys to form thiazinane and thiazolidine was confirmed by (1)H NMR. Moreover, complexation with Hcy and Cys disturbed the p-pi conjugation between the aldehyde group and the bpy moiety, and led to the excited states switching to [d pi(Ir)-pi(star)(N boolean AND N)] (3)MLCT and [pi(C boolean AND N)-pi(star)(N boolean AND N)] (3)LLCT from (pi-pi(star))(pba(-)) (3)IL. Furthermore, the MTT assay was used to determine that the probe has low cytotoxicity. Importantly, cell imaging experiments demonstrated that the probe is membrane permeable and can monitor the changes of Hcy/Cys within living cells in a ratiometric mode.

Abstract:Abnormal homocysteine (Hcy) levels in human serum have been associated with serious or vital diseases, making the reliable and easy detection of Hcy important to clinical analysis and biological study. In this work, five phosphorescent Ir(C^N)2(N^N) complexes (Irn) having aldehyde group were synthesized as probes (C^N and N^N denoted ligands). A discussion was conducted on their molecular structure, electronic structure, photophysical parameters, and Hcy sensing ability, revealing the correlations between their molecular structures and performances. Irn emission was enhanced (by ∼ two folds) and blue-shifted (by 100 nm) after meeting Hcy (free state), via a cyclization reaction between the -CHO group (from Irn) and Hcy. In addition, using RE(BTC) as a supporting material (RE = Tb and Eu), the Ir(III) probe was loaded onto a supporting material of RE(BTC) (H3BTC = 1, 3, 5-benzenetricarboxylic acid). The emission color was changed by increasing Hcy concentration. Straight working curves were obtained with LOD (limit of detection) of 1.9 μM and a response time of ∼200 s. The novelty of this work was the combination of Irn with RE(BTC), which offered enhanced and blue-shifted emission upon Hcy via a cyclization reaction. This demonstrated a high level of sensitivity towards homocysteine detection.

Abstract:Herein, we developed an ER-targeting NIR probe HL-Cys for Cys which could fluctuate ratiometrically in the UV-vis absorption and fluorescent spectra with large stokes shift, desirable selectivity and sensitivity. Biological imaging demonstrated that HL-Cys could locate in ER and image the Cys level during ER oxidative stress in HeLa cells.
