Abstract:Homocysteine (Hcy) and cysteine (Cys) are crucial to the physiological balance in living systems. Specific detection of intracellular Hcy and Cys is of growing importance. Herein, we demonstrated phosphorescence imaging of intracellular Hcy and Cys using a cationic iridium(III) complex Ir(pba)(2)(bpy)(+)center dot PF(6)(-) [pba = 4-(2-pyridyl)benzaldehyde, bpy = bipyridine] containing aldehyde groups as a luminescent probe. Upon addition of Hcy or Cys to a semiaqueous solution of Ir(pba)2(bpy)+, a change in luminescence from yellow to red was visible to the naked eye. The successful chemical reaction of the aldehyde in Ir(pba)(2)(bpy)(+) with Hcy and Cys to form thiazinane and thiazolidine was confirmed by (1)H NMR. Moreover, complexation with Hcy and Cys disturbed the p-pi conjugation between the aldehyde group and the bpy moiety, and led to the excited states switching to [d pi(Ir)-pi(star)(N boolean AND N)] (3)MLCT and [pi(C boolean AND N)-pi(star)(N boolean AND N)] (3)LLCT from (pi-pi(star))(pba(-)) (3)IL. Furthermore, the MTT assay was used to determine that the probe has low cytotoxicity. Importantly, cell imaging experiments demonstrated that the probe is membrane permeable and can monitor the changes of Hcy/Cys within living cells in a ratiometric mode.

Abstract:Fluorescent imaging of cysteine (Cys) and homocysteine (Hcy) is crucial to understand sulphur metabolism in living cells. Whereas two-photon active cyclometalated Ir(III) complex for detecting Cys/Hcy has been rarely reported. Herein, two novel cyclometalated iridium (III) complexes Ir1-1r2 with different electron-donor were designed, synthesized and fully characterized as well as confirmed by single-crystal X-ray diffraction analysis, which could selectively react with Cys/Hcy, accompanying with an obvious one- and two-photon fluorescence enhancement in vitro. The mechanism for sensing Cys/Hcy was further analyzed in detail by mass spectra and time-dependent density functional theory calculations. It was found that Ir2, bearing ester group, exhibited higher performance on the selective detection of Cys and Hcy compared to In. Notably, the two-photon action cross-section value in the near-infrared region (similar to 800 nm) of Ir2 is significantly enhanced upon the addition of Cys/Hcy. Further application to cellular imaging indicated that Ir2 is membrane permeable and capable of monitoring the changes of intracellular mitochondrial Cys/Hcy. These results support that such two-photon active complexes might provide a promising platform for the detection of mitochondrial Cys/Hcy in living biological systems. Water-soluble Phosphorescent Iridium(iii) Complexes As Multicolor Probes for Imaging of Homocysteine and Cysteine in Living Cells

Abstract:With the emergence of phosphorescent heavy-metal complexes as a class of attractive probes for bioimaging, there is a parallel need to develop new phosphorescent probes with complete solubility in pure water for phosphorescent staining in living cells. Herein, a convenient and general design strategy for realizing phosphorescent heavy-metal complexes with complete water-solubility is provided and a series of cationic iridium(III) complexes [Ir(C^N)2(N^N)]+PF6− (C^N = 2-(2,4-difluorophenyl)pyridine (dfppy), 2-(4-(tert-butyl)phenyl)pyridine(t-buppy), 2-(thiophen-2-yl)quinoline) (thq), 4-(pyridin-2-yl)benzaldehyde (pba)) are prepared. The water-solubility of the complexes was successfully realized through the quaternization of the tertiary amino group in the N^N ligand. By changing the C^N ligands, the luminescent emission colors of these complexes can be tuned from green to red. These cationic iridium(III) complexes are membrane-permeable and can be applied as phosphorescent dyes for cell imaging in phosphate buffer solution (PBS). Complexes Ir1–Ir3 displayed specific staining of the cytoplasm and complex Ir4 containing two aldehyde groups could detect the changes of cysteine/homocysteine concentration in living cells. These results demonstrated that our design strategy offers an effective way to develop excellent phosphorescent cellular probes for real applications.

Abstract:In this work, tetrahydro-acridine salt hybrid coumarin dyes was designed and developed as a Near-infrared (NIR) fluorescent probe CM for the detection and fluorescent bioimaging of Cys/Hcy. The activated chlorine atom in probe facilitates more efficient nucleophilic substitution reaction, resulting in fast responsive time (3 min toward 3 eq. Cys) and high sensitivity toward Cys with detection limit of 46 nM. More importantly, probe displays "Turn -on" near-infrared fluorescence emission (Emmax: 674 nm) with Large Strokes shift (about 200 nm) toward Cys/ Hcy. Furthermore, probe could be used as efficient tool for imaging and detection of Cys/Hcy in mitochondria of living cells and in vivo. This work innovatively proposed a new design strategy for NIR fluorescent probe platform for the detection and fluorescence imaging of bio-thiols.

Abstract:Homocysteine (Hcy) and cysteine (Cys) are two important kinds of amino acids in human bodies. Herein, we synthesized an iridium(III) complex-functionalized poly(N-isopropylacrylamide) and its hydrogel, which could be used as the excellent phosphorescent bioprobe for sensing Hcy and Cys. Their detection can be realized in aqueous system through the variations in absorption and photoluminescence spectra. Furthermore, living cell imaging experiments demonstrate that the phosphorescent bioprobe is membrane permeable and can monitor the changes of Hcy and Cys within living cells. In addition, the probe is also thermoresponsive, and its photoluminescence intensified with increasing temperature. These results suggests that this bioprobe has promising application in biomedical fields.

Abstract:A series of phosphorescent zwitterionic iridium(III) complexes, with 4-carboxy-2, 2'-bipyridine-4'-carboxylate (Hdcbpy) as ancillary ligand, Ir(C(wedge)N)(2)(Hdcbpy) (C(wedge)N = 1-phenylpyrazole (ppz), 1-phenyl-pyridine (ppy), 2-(4',6'-difluoro-phenyl)pyridine (dfppy), 1-phenyl-isoquinoline (piq), dibenzo[f,h]quinoxaline (dbq)), were prepared and characterized. Their photophysical properties were studied, and intense luminescence emissions were observed based on metal-to-ligand-charge-transfer ((3)MLCT), ligand-to-ligand charge-transfer ((3)LLCT), ligand-centered transitions ((3)LC, i.e., (3)pi --> pi*), or intraligand-charge-transfer ((3)ILCT) excited states, which were confirmed by theoretical calculations. The quantum yield of Ir(dfppy)(2)(Hdcbpy) is as high as 0.106 in aqueous solution. With Hdcbpy as a hydrophilic part, their amphiphilic structures as further confirmed by X-ray single crystal data endow them with different solubilities in phosphate buffer solution (PBS, pH 7.0). The compounds were successfully applied as luminescent dyes for cell imaging in aqueous solution. Their different stain ability in cell imaging was fairly well supported by the experimental data based on the measurement of oil/water partition coefficients and encapsulation/release with liposomes.
An Ir(III) Complex Capable of Discriminating Homocysteine from Cysteine and Glutathione with Luminescent Signal and Imaging Studies.

Abstract:A new cationic Ir(III) complex with aldehyde and amino groups was synthesized and characterized. The Ir(III) complex has rich photophysical properties. The reaction of the aldehyde group in the Ir(III) complex with homocysteine (Hcy) afforded thiazinane derivatives which resulted in obvious changes in the luminescence spectra. After addition of Hcy to the Ir(III) complex containing 4,4'-diamino-2,2'-bipyridine, the luminescence intensity at ca. 580-610 nm decreased, and a new band at ca.490-520 nm appeared and enhanced strongly with a large blue shift of ca.90 nm, and the luminescent color changed from orange red to green. Based on this ratiometric probe, it can sensitively and selectively recognize Hcy by the ratio of emission intensity at two wavelengths to the concentrations of Hcy. While after addition of cysteine (Cys) or glutathione (GSH), the luminescence band showed a mild decrease in intensity with an unnoticeable shift. These different phenomena make it capable of discriminating homocysteine from cysteine and glutathione. The cytotoxicity and imaging of the complex were also studied in this work. The complex exhibited very low cytotoxicity on HeLa cells and showed sensitivity toward Hcy in living cells. These advantages provide it a good candidate for the application in the analytical and bioanalytical field.

Abstract:An activatable iridium(III) complex probe for phosphorescence/time-gated luminescence detection of cysteine (Cys) in vitro and in vivo is reported. The probe, [Ir(ppy)2(NTY-bpy)][(PF6)] features large Stokes shift, long-lived emission, good biocompatibility, high sensitivity and selectivity for Cys detection. Compound [Ir(ppy)2(NTY-bpy)][(PF6)] is mitochondria targetable, which makes it convenient for the determination of Cys-mediated redox activities in mitochondria. Abstract:Cysteine (Cys) takes part in redox balance in cells as an antioxidant, and imbalance of Cys content in the body can cause a variety of diseases. It is very important to develop a new fluorescent chemosensor to specifically detect Cys intracellular. In this work, a novel NIR fluorescent probe was constructed based on 3-ethyl-1,1,2-trimethyl-1H-benzo[e]indol-3-ium iodide and 5-(4-hydroxyphenyl)furan-2-carbaldehyde. The probe could selectively detect Cys in the presence of homocysteine (Hcy), glutathione (GSH), and other interferences. It also had a number of advantages, including nucleolus-targeting ability, long fluorescence emission wavelength (685 nm), low detection limit (56 nM), and large Stokes shift (172 nm). The probe was employed to enable visualization of Cys in HepG2 cells, and due to its good response in viscous environment, the probe could also locate nucleoli intracellular.

Abstract:We designed and synthesized a water-soluble near-infrared (NIR) fluorescent probe with the recognition unit of the cyanine-like structure and acrylate group. Through an aromatic ring nucleophilic substitution reaction based on sulfhydryl moiety, an off-on fluorescence response toward cysteine (Cys) was realized. The probe exhibited excellent spectral performance with an emission wavelength of 720nm and a detection limit of 0.20μM. The spectral properties, selectivity and anti-interference performance of the probe were systematically investigated. Density functional theory (DFT) calculations were conducted to clarify the luminescence mechanism of the probe. Furthermore, the probe was successfully applied to the detection of free Cys in human serum and the NIR imaging of endogenous Cys in living cells. Thus, the probe has a promising application prospect in clinical diagnosis and fluorescence imaging.

Abstract:The three near-infrared-emitting cationic indium(III) complexes [Ir(pbq-g)(2)((NN)-N-boolean AND)]+PF6- (pbq-g = phenylbenzo[g]quinoline; (NN)-N-boolean AND = bipyridine (1), 1,10-phenanthroline (2), 4,7-diphenyl-1,10-phenanthroline (3)) have been demonstrated as phosphorescent dyes in live cell imaging. These complexes with different ancillary ligands show similar near-infrared (NIR) emission With lambda(max,peak) at 698 nm and lambda(max;shoulder) at 760 nm in CH2Cl2 solutions, with a moderate quantum yield of around 3%. However, these complexes behave quite differently as NIR dyes for live cell imaging. Complexes 1 and 2 exhibit exclusive staining in the cytoplasm with good cell membrane permeability under excitation at 488 nm, while 3 gives almost no cell uptake, as further determined by flow cytometry. Although the lipophilicities of these complexes follow the order 1 < 2 < 3, their cytotoxicities are in the reverse order. The exceptionally low cytotoxicity of 3 could be attributed to its poor solubility in aqueous buffer and thus substantially low exposure dose. This comparative study suggested that the ancillary ligands could fine-tune the amphiphilicity and cytotoxicity of the cyclometalated iridium(III) complexes and thus might play a key role in the design of NIR-emitting iridium(III) complexes for practical applications in bioimaging.

Abstract:Chemiluminescence (CL) probes that possess near-infrared (NIR) emission are highly desirable for in vivo imaging due to their deeper tissue penetration ability and intrinsically high sensitivity. Herein, a novel iridium-based CL probe () with direct NIR emission was reported as the result of hypochlorous acid (HClO)-initiated oxidative deoximation. To improve its biocompatibility and extend the CL time for in vivo imaging applications, this was prepared as a CL nanoparticle probe (s) through encapsulation by an amphiphilic polymer Pluronic F127 (F127). All results demonstrate that the have good selectivity and sensitivity for visualization of HClO even at the depth of 1.2 cm. Owing to these advantages, the CL imaging of exogenous and endogenous HClO in mice was achieved. This study could provide new insights into the construction of new NIR emission CL probes and expand their applications in biomedical imaging.
Aldehyde Bearing Bis-Cyclometalated Ir(Iii) Complex As Selective Photoluminescence Turn-On Probe for Imaging Intracellular Homocysteine

Abstract:A new photoluminescence (PL) turn-on probe based on aldehyde bearing bis-cyclometalated Ir(III) complex, [Ir(bt)(2)phen-CHO](+)PF6(-) (1), is synthesized and applied to imaging homocysteine (Hcy) in living cells. The probe 1 is readily prepared by using 2-phenylbenzothiazole (bt) as main ligand and 1,10-phenanthroline-5-carboxaldehyde (phen-CHO) as ancillary ligand. Probe 1 displayed excellent selective PL enhancement in response to Hcy in neutral buffer solution based on aldehyde group cyclization reaction. The increased PL intensity of probe 1 is directly proportional to the concentration of Hcy in the range of 10 mu M to 500 mu M with a detection limit of 4.6 mu M. More importantly, probe 1 can highly discriminate Hcy from cysteine (Cys), which is agreed with the higher luminescent quantum yield of the adduct of probe 1 and Hcy (1-Hcy, 0.46) compared with that of probe 1 and Cys (1-Cys, 0.05) in 10 mM phosphate buffer solution (pH 7.4). Furthermore, the probe I has appreciable cell permeability and low cytotoxicity and has been successfully applied to selectively imaging Hcy in living cells. The superior properties of the probe 1 make it of great potential use for studying the effects of Hcy in other biosystems. (C) 2017 Elsevier B.V. All rights reserved.
An AIE and ICT Based NIR Florescent Probe for Cysteine and Homocysteine

Abstract:A combination of aggregation-induced emission and intramolecular charge transfer was achieved by using a triphenylamine analogue and a dicyanovinyl moiety as the electron donating and accepting units, respectively. Hence, we designed and synthesized a probe with a D-pi-A framework as a near-infrared fluorescence turn-on probe for biothiols (cysteine and homocysteine). Owing to the remarkable intramolecular charge transfer effect as well as intramolecular rotations associated with the donor moiety, the probe exhibits extremely weak fluorescence; which becomes a good starting point for developing fluorescence "turn-on" probes. Upon reaction with cysteine or homocysteine utilizing the dicyanovinyl moiety, the intramolecular charge transfer character was weakened, and the reacting products were observed to aggregate in aqueous solutions, resulting in the aggregation-induced emission effect with red fluorescence at 651 and 656 nm, respectively. Hence, the probe could be used as a fluorescence "turn-on" sensor for cysteine and homocysteine, with the sensing time of less than 4 min and the detection limits of 8.4 mu M and 5.7 mu M towards cysteine and homocysteine, respectively. The probe could distinguish cysteine and homocysteine from glutathione. The sensing mechanism was systematically investigated by employing high resolution mass spectrometry, H-1 NMR and density functional theory calculations as well as checking the solvent viscosity dependent fluorescence, and thus the nucleophilic addition products, the intramolecular charge transfer character, and the aggregation-induced emission behaviour were clearly elucidated. It is noteworthy that the low cytotoxicity, the intrinsic aggregation induced emission nature and near-infrared emissions enable the application of the probe in living cell imaging. Through hydrophobic-hydrophilic interactions with Pluronic F127, A1-F127 based fluorescent organic nanoparticles (FONs) was prepared, which was intrinsically non-fluorescent due to presence of nitroolefin moiety. However, after being reacted with Cys, Michael addition of thiols to nitroolefin elicited a significant NIR fluorescent turn-on response at 654 nm. A1-F127 FONs can selectively discriminate Cys from GSH and Hcy with a detection limit of 5.31x10(-8) M. Its biocompatibility and cell imaging application were further investigated. The work could provide a general, simple and efficient strategy for the application of hydrophobic fluorescent molecules to sensing analytes in a totally aqueous system.

Abstract:In this paper, by modifying the thioxanthene-benzothiozolium fluorophore, BCy-Cys, a lysosome-targeted near-infrared (NIR) fluorescent probe was synthesized for the detection of cysteine (Cys) from homocysteine (Hcy)/glutathione (GSH). As expected, BCy-Cys exhibited high selectivity and high sensitivity for detection of Cys over Hcy/GSH, with an extremely low limit of detection at 0.31 μM, marked by obvious color changes. HRMS was conducted to confirm that the fluorescence intensity at 795 nm was significantly enhanced by the enhancement of intramolecular charge transfer (ICT). Importantly, BCy-Cys could be used to visualize both exogenous and endogenous lysosomal Cys, signifying its potential application in complex organismal systems.
