Abstract:MicroRNAs (miRNAs) are evolutionarily conserved, small ( approximately 20-25 nucleotides), single-stranded molecules that suppress the expression of protein-coding genes by translational repression, messenger RNA degradation, or both. More than 700 miRNAs have been identified in the human genome. Amazingly, a single miRNA can regulate the expression of hundreds of mRNAs or proteins within a cell. The small RNAs are fast emerging as master regulators of innate and adaptive immunity and likely to play a pivotal role in transplantation. The clinical application of RNA sequencing ("next-generation sequencing") should facilitate transcriptome profiling at an unprecedented resolution. We provide an overview of miRNA biology and their hypothesized roles in transplantation.

Abstract:A recent Keystone Symposium on `MicroRNAs and siRNAs: Biological Functions and Mechanisms' was organized by David Bartel and Shiv Grewal (and was held in conjunction with 'RNAi for Target Validation and as a Therapeutic', organized by Stephen Friend and John Maraganore). The `MicroRNAs and siRNAs' meeting brought together scientists working on diverse biological aspects of small regulatory RNAs, including microRNAs, small interfering RNAs (siRNAs) and Piwi-interacting RNAs (piRNAs and rasiRNAs). Among the themes discussed were the diversity of small regulatory RNAs and their developmental functions, their biogenesis, the identification of their regulatory targets, their mechanisms of action, and their roles in the elaboration of multicellular complexity.

Abstract:Key PointsMicroRNAs (miRNAs) are a family of ∼21–25-nucleotide small RNAs that negatively regulate gene expression at the post-transcriptional level.The founding members of the miRNA family, lin-4 and let-7, were identified through genetic screens for defects in the temporal regulation of Caenorhabditis elegans larval development.Owing to genome-wide cloning efforts, hundreds of miRNAs have now been identified in almost all metazoans, including flies, plants and mammals.MiRNAs exhibit temporally and spatially regulated expression patterns during diverse developmental and physiological processes.Most of the miRNAs that have been characterized so far seem to regulate aspects of development, including larval developmental transitions and neuronal development in C. elegans, growth control and apoptosis in Drosophila melanogaster, haematopoietic differentiation in mammals, and leaf development, flower development and embryogenesis in Arabidopsis thaliana.The majority of the animal miRNAs that have been characterized so far affect protein synthesis from their target mRNAs. During the past ten years these tiny non-coding RNAs have been linked to virtually all known physiological and pathological processes, including cancer. In the same way as certain key protein-coding genes, miRNAs can be deregulated in cancer, in which they can function as a group to mark differentiation states or individually as bona fide oncogenes or tumour suppressors. Importantly, miRNA biology can be harnessed experimentally to investigate cancer phenotypes or used therapeutically as a target for drugs or as the drug itself.

Abstract:miRNAs acting naturallyThe discovery of microRNAs (miRNAs), the non-coding RNAs thought to be involved in many biological processes, is changing our perception of gene regulation. Little is known about their function in mammalian systems in vivo, but a newly developed group of compounds that silences miRNAs in mice should provide a powerful tool for the study of their function — and a potential therapeutic strategy for silencing miRNAs in disease. These ‘antagomirs’ are chemically engineered oligonucleotides with sequences that complement natural miRNAs. Intravenous administration of antagomirs to miR-16, -122, -192 and -194 in mice caused a marked reduction of corresponding miRNA expression in liver, lung, kidney, heart, muscle, intestine, fat, skin, bone marrow, ovaries and adrenals. In worms and flies, miRNAs play important developmental roles in the embryo. In vertebrates, various developmental genes have been shown to be targets of miRNA regulation, but there were no examples of miRNAs playing specific roles in known developmental processes. Now one such example has been found: miR-196 acts in mouse embryos as a mechanism to ensure accurate expression of genes primarily regulated by Hoxb8 and Shh transcription factors. This supports the idea that vertebrate miRNAs may function as a secondary level of gene regulation.

Abstract:Ever since microRNAs (miRNAs) were first recognized as an extensive gene family >20 years ago, a broad community of researchers was drawn to investigate the universe of small regulatory RNAs. Although core features of miRNA biogenesis and function were revealed early on, recent years continue to uncover fundamental information on the structural and molecular dynamics of core miRNA machinery, how miRNA substrates and targets are selected from the transcriptome, new avenues for multilevel regulation of miRNA biogenesis and mechanisms for miRNA turnover. Many of these latest insights were enabled by recent technological advances, including massively parallel assays, cryogenic electron microscopy, single-molecule imaging and CRISPR–Cas9 screening. Here, we summarize the current understanding of miRNA biogenesis, function and regulation, and outline challenges to address in the future.

Abstract:The Nobel Prize in Chemistry 2006 was awarded to Roger D. Kornberg for his fundamental study of the molecular basis of eukaryotic transcription. Roger D. Kornberg spent ten years in developing an in vitro yeast transcription system, and depicted pictures of RNA polymerase II and other complexes containing general transcription factors, Mediator, DNA and RNA by combining crystallography with biochemical knowledge. Furthermore, he firstly illustrated the whole process of how transcription works at a molecular level in a eukaryotic cell. Accumulative evidence demonstrates that the study of molecular mechanisms of eukaryotic transcription plays important roles in the treatment of diseases and the regulation of stem-cell development.

Abstract:MicroRNAs, identified only relatively recently, are regulators of gene expression with potential medical benefits. The combination of microRNAs and regenerative medicine is an emerging interdisciplinary medical field that can yield exciting new possibilities for clinical medicine. In this paper, we review the prospects of microRNAs as future therapies in regenerative medicine. Recently, researchers have demonstrated the crucial roles of microRNAs, not only in the differentiation and proliferation of stem cells, which have a key function in the regeneration and transplantation of organs, but also in oncogenesis. Several lines of indirect evidence show that the initiation and maintenance of cancer stem cells might also be under the control of microRNAs. Further, microRNAs have been indicated to be involved in diverse biological processes, suggesting the potential role of these molecules in the treatment of diseases.

Abstract:MicroRNAs (miRNAs) maintain cellular homeostasis by blocking mRNAs by binding with them to fine-tune the expression of genes across numerous biological pathways. The 2024 Nobel Prize in Medicine and Physiology for discovering miRNAs was long overdue. We anticipate a deluge of research work involving miRNAs to repeat the history of prizes awarded for research on other RNAs. Although miRNA therapies are included for several complex diseases, the realization that miRNAs regulate genes and their roles in addressing therapies for hundreds of diseases are expected; but with advancement in drug discovery tools, we anticipate even faster entry of new drugs. To promote this, we provide details of the current science, logic, intellectual property, formulations, and regulatory process with anticipation that many more researchers will introduce novel therapies based on the discussion and advice provided in this paper.

Abstract:Communication between cells ensures coordination of behavior. In prokaryotes, this signaling is usually referred to as quorum sensing, while eukaryotic cells communicate through hormones. In recent years, a growing number of reports have shown that small signaling molecules produced by organisms from different kingdoms of nature can facilitate cross‐talk, communication, or signal interference. This trans‐kingdom communication (also termed as trans‐kingdom signaling or inter‐kingdom signaling) mediates symbiotic and pathogenic relationships between various organisms (e.g., microorganisms and their hosts). Strikingly, it has been discovered that microRNAs (miRNAs)—single‐stranded noncoding RNAs with an average length of 22 nt—can be transmitted from one species to another, inducing posttranscriptional gene silencing in distant species, even in a cross‐kingdom fashion.
