Abstract:The Nobel Prize originated in 1895 when Alfred Nobel signed his will, leaving a large part of his wealth to the creation of the Nobel Foundation and the establishment of this prize, with the vision that people could help improve humanity through knowledge, science, and humanism. The Nobel Prize has been awarded in areas such as physics, physiology or medicine, chemistry, literature, peace, and economics. 943 people and 25 organizations have been awarded since 1901 to this day. The history and knowledge commemorated in the Nobel Prize have allowed an integral development in the understanding, diagnosis, therapy, and scientific progress in different types of cancer, laying the foundations and being the inspiration for thousands of scientists worldwide who work hard in the area of oncology. GLOBOCAN estimates indicated that there were approximately 19 million new cancer cases globally and almost 10 million cancer deaths in 2020 alone; hence, this study reviews and brings together the main scientific discoveries awarded with the Nobel Prize in the area of physiology or medicine and chemistry, which contributed to the knowledge, diagnosis and/or treatment of cancer from 1901 to 2021.
New principles of cancer therapy give new hope for oncological patients

Abstract:The Nobel Prize in Physiology or Medicine 2018 was awarded jointly to James P. Allison and Tasuku Honjo "for their discovery of cancer therapy by inhibition of negative immune regulation". A number of therapeutic approaches are available for cancer treatment, including surgery, radiation, and other strategies, some of which have been awarded previous Nobel Prizes. These include methods for hormone treatment for prostate cancer (Huggins, 1966), chemotherapy (Elion and Hitchins, 1988), and bone marrow transplantation for leukemia (Thomas, 1990). Many scientists engaged in intense basic research and uncovered fundamental mechanisms regulating immunity and also showed how the immune system can recognize cancer cells. T-cells were shown to have receptors that bind to structures recognized as non-self and such interactions trigger the immune system to engage in defense. However, additional proteins acting as T-cell accelerators are also required to trigger a full-blown immune response. Many scientists contributed to this important basic research and identified other proteins that function as brakes on the T-cells, inhibiting immune activation. This intricate balance between the accelerators and inhibitors is essential for a tight control. New strategy was developed into a therapy for humans. Promising results soon emerged from several groups, and in 2010 an important clinical study showed striking effects in patients with advanced melanoma. In several patients, signs of remaining cancer disappeared. The results were dramatic, leading to long-term remission and possible cure in several patients with metastatic cancer, a condition that had previously been considered essentially untreatable. Abstract:Precision medicine emphasizes patient-specific formulation for treatment of diseases, especially cancer. However, in targeted cancer treatment, because the expression level of tumor receptors in each patient varies even for the same type of cancer, the ligand/receptor-mediated approach does not seem promising for precision medicine. In this work, we demonstrated our strategy of using a phage display technique for breast cancer precision medicine. Using in vivo biopanning, we first selected an MCF-7 breast tumor-targeting peptide, then tested the effectiveness of the as-selected peptide in tumor homing and finally conjugated the peptide to a model photothermal drug, namely, gold nanorods, to achieve enhanced cancer killing efficacy. The peptides identified by the phage display technique can guide the drug to the tumors without the need to know the exact receptors on the tumor. This approach requires significantly less effort to explore patient-specific targeting molecules for precision medicine.

Abstract:Toll,a protein involved in the dorsoventral development of fruit flies,was first discovered by the German Scientist Christiane Nüsslein-Volhard and his colleagues in 1985.In 1996,Jules Hoffmann at the National Center for Scientific Research in Strasbourg,France,discovered that the product of Toll was involved in the sensing of pathogenic infections in fruit flies,and its activation was necessary for the initiation of effective defense;In 1998,Bruce A.Beutler at the Scripps Research Institute discovered that the lipopolysaccharide(LPS,a microbial pathogenic product) resistant mouse had a mutation in the receptor gene very similar to the Toll in fruit flies,and proved the Toll-like receptor was the sensor of LPS.These discoveries showed that mammals and fruit flies adopt the similar molecules to activate innate immunity.Therefore,they shared the half of the 2011 Nobel Prize in Physiology or Medicine for their discoveries concerning the sensors for the activation of innate immunity.Another half of the prize was awarded to Ralph M.Steinman at the Rockefeller University in New York.In 1973,Steinman discovered the Dendritic cells(DCs) and proved that it could activate the T cells and initiate the adaptive immune response.Further studies showed that the DCs could sense the signals arising from the innate immune response and control the T cell activation.This makes it possible for the immune system to react towards pathogenic microorganisms while avoiding attacks on own body's endogenous molecules.These discoveries provide us further insights into the mechanism of activation and regulation of the immune system,and will facilitate the development of novel methods in disease prevention and treatment.
The Era of Immune Checkpoint Therapy: from Cancer to Viral Infection—A Mini Comment on the 2018 Medicine Nobel Prize

Abstract:The 2018 Medicine Nobel Prize was awarded jointly to two immunologists, James P. Allison at the University of Texas MD Anderson Cancer Center in Houston and Tasuku Honjo at Kyoto University in Japan, who pioneered a new way to treat cancers ( Ledford et al. 2018 ). Both Laureates have shown how so called checkpoints on T cells can be used to manipulate the immune responses so that T cells can efficiently attack cancer cells. Using the immune system to fight cancers has been investigated for more than a 100 years. Recent advances in cancer immunotherapy, particularly immune checkpoint blockade therapy have dramatically changed the therapeutic strategy against advanced cancers. Through inhibiting negative immune regulation, these approaches have demonstrated improved overall survival for patients with advanced cancers. Importantly, for some of the patients treated with such strategies, their tumors seem to totally disappear.

Abstract:The Nobel Prize in Chemistry 2012 was awarded jointly to Robert J. Lefkowitz and Brian K. Kobilka based on their remarkable contributions to the studies of G-protein-coupled receptors (GPCRs). Robert J. Lefkowitz is the Godfather of the modern GPCRs studies. He developed the radioligand binding assay to study the receptors, verified the existence of the receptors, cloned the first GPCR gene, found G protein-coupled receptor kinases (GRKs) and β-arrestins which mediate the desensitization and endocytosis of the receptors, and found the siganling pathways mediated by β-arrestins. Brian K. Kobilka developed several breakthrough methods in GPCRs crystallography. His team first solved the high-resolution structure of β2-adrenergic receptor. His findings led rapid development of GPCRs crystallography. Amazingly, he and his research team got a snapshot of the β-adrenergic receptor at the exact moment when it is activated. The present review summarizes the GPCRs discovery experiences of these two outstanding scientists.

Abstract:Recent Advances: The 2019 Nobel Prize awarded to the mechanisms for oxygen sensing and adaptation according to oxygen availability, highlighting the fundamental importance of gaseous molecules. Gaseous molecules including reactive oxygen species (ROS) can interact with different cations generated during metabolic and redox dysregulation in cancer cells. Crosstalk between calcium signaling and metabolic/redox pathways leads to network-based dyregulation in cancer. Significance: Recent discovery on using small molecules targeting the ion channels, redox signaling, and protein modification on metabolic enzymes can effectively inhibit cancer growth. Several FDA-approved drugs and clinical trials are ongoing to target the calcium channels, such as TRPV6 and TRPM8. Multiple small molecules from natural products targets on metablic and redox enzymes to exert anti-cancer effect. Critical Issues: Small molecules targeting key ion channels, metabolic enzyme that control key aspects of metabolism and redox proteins are promising, but their action mechanisms of the target are needed to be elucidated with advanced -omic technologies which can give network-based and highly-dimensioal data. In addition, small molecules that can directly modify the protein residues has emerged as a novel anti-cancer strategy. Future Directions: Advanced technology accelerates the detection of ions, metabolic, and redox changes in clinical samples for diagnosis and informs the decision of cancer treatment. The improvement of ROS detection, ROS target identification, and computational-aid drug discovery also improve clincal outcome.Overall, network-based or holistic regulation of cancer via ion therapy, metabolic and redox intervention are promising as new anti-cancer strategies.

Abstract:Human beings have highly developed sense of touch. Through touching we can perceive heat, pain, mechanical stimuli, recognize features of an object such as texture and hardness, and even understand emotion of a person. Hence, human touch is one of the most important ways to sense and adapt to the world around us. However, we had little understanding of how cutaneous perception was formed in our nervous system, until the groundbreaking discoveries were made by this year's Nobel Prize laureates in Physiology or Medicine, David Julius from University of California, San Francisco, and Ardem Patapoutian from Scripps Research. They found that heat and mechanical stimuli can initiate specific nerve impulses for temperature, pain, and touch sensations. The quest to understand temperature sensation and pain spans millennia. In the early 20th century, the Greek physician Galen proposed that sensations were transmitted along pathways that connect the organs to the brain. It was not until 1997 that the true underlying molecular mechanism was revealed by Julius in a breakthrough work. Julius and his team found that a receptor on the cell's membrane arranges itself from one side to the other, so that it creates a passage called a transient receptor potential (TRP) channel. This ion channel opens or closes in response to heat. Inspired by Julius's work, Patapoutian in the early 21st century explored a mechanosensitive ion channel, called the Piezo channel, and identified that it is essential for the sense of touch. Patapoutian and his team also found that the Piezo channel is responsible for the sensing of position and motion of our body, which explains why we can balance on a steep stair. Not only did these discoveries elucidate the physiological understanding of human somatosensory, they also enlightened the researchers who work on reproducing such human functions in robotics and prosthetics. The molecular mechanisms of thermo-sensation and mechano-sensation provided a new paradigm for designing artificial cutaneous perception and constructing electronic skin for robots. Once fully armed by tactile perception and haptic feedback, smart robots and artificial intelligence may start to have consciousness and feel emotions like we humans do. With the awarding of the 2021 Nobel Prize in Physiology or Medicine to research in how humans perceive touch and pain, the research in tactile intelligence now has a clearer roadmap and gains a greater momentum. We foresee many exciting breakthroughs in this field in the near future.

Abstract:Professors James P. Allison and Tasuku Honjo were awarded with the 2018 Nobel Prize in Medicine for their contributions in cancer immunotherapy. The latter is a breakthrough in cancer therapy, aimed to overcome tumor-induced immunosuppression, leading to the reactivation of the immune system against cancer cells. Under physiological conditions, the CTLA-4 and PD-1 proteins expressed on T-cells and discovered by the awarded scientists, lead to immune tolerance. Cancer cells exploit these control points to enhance the inhibition of T-cells. The expression of PD ligands (PD-L1) in tumor cells and CTLA-4 ligands in antigen presenting cells, which bind the PD-1 receptor and CTLA-4 respectively, block anti-tumor immunity. This situation led to a biotechnological race focused on the development of effective antibodies able to "turn-on" the immune system cheated by the tumor. Anti-CTLA-4 and anti-PD-1 antibodies improve life-expectancy in cancer patients. In this review, we perform an historical overview of Professors Allison and Honjo contribution, as well as the immunological basis of this new and powerful therapeutic strategy, highlighting the clinical benefits of such intervention.
