Abstract:Many efforts have been made to prevent interferences in enzymatic electrochemical biosensors by permselective membranes or mediators with low redox potential. However, it is difficult to completely eliminate interferents without compromised sensitivity by these traditional procedures. We propose here a method based on an origami paper device that separates the electrochemical reactions of interferents and substrates for complete depletion of interferents and precise analysis of substrates. Interferents such as ascorbate, urate and paracetamol were completely consumed by a simple electrolysis step, while substrates were quantitatively analyzed by coulometry. With GOx as a model enzyme, an interference-free and calibration-free coulometric glucose biosensor has been demonstrated successfully. The proposed origami paper device provides a facile and easy-controlled approach to eliminate the electroactive interferents completely for enzymatic electrochemical biosensors.
Formation of PtCuCo Trimetallic Nanostructures with Enhanced Catalytic and Enzyme-like Activities for Biodetection

Abstract:The unique enzyme-like properties of metal nanoparticles (NPs) hold great promise for chemical and biomedical applications; implementation relies on improvements in their catalytic efficiency. We have constructed PtCuCo nanostructures using hydrothermal treatment of Co3O4, Cu2+, and Pt2+ in aqueous solutions, resulting in PtCuCo trimetallic alloy NPs having hollow structures, as confirmed by TEM, XRD, XPS, and EDS analyses. These PtCuCo NPs are capable of oxidizing 3,3',5,5'-tetramethyl-benzidine (TMB) in either the absence or presence of hydrogen peroxide, suggesting efficient peroxidase- and oxidase-like activity. These NPs could also catalyze the reduction of 4-nitrophenol by NaBH4. We believe that the combination of Pt and oxidized Cu and Co species results in surfaces with rich active sites possessing a wide range of electronic variation, which promotes both generation of hydroxyl radicals and electron transfer. Consequently, the catalytic capabilities of our PtCuCo NPs are greater than those of either Pt or PtCu NPs. Building upon the peroxidase-like activity of PtCuCo NPs, we developed a colorimetric method not only capable of specifically detecting glucose, but also able to detect rongalite and HS-, on the basis of the inhibitory effect of these substances upon PtCuCo nanoperoxidase.

Abstract:Nanozymes are becoming substitutes of some natural enzymes due to their excellent properties, however, little attention has been paid to the catalytic mechanisms and especially inhibition mechanism of nanozymes, which impeded advancement in nanozymes and their wide application including bioanalysis. Herein, non-toxic filamentous virus, M13 phage templated Co 3 O 4 nanowires (NWs) was facilely prepared, which exhibits excellent oxidase (OXD)-like activity and catalytic efficiency. The catalytic mechanism of OXD-like is oxygen vacancies, confirmed by high resolution X-ray photoelectron spectrum of O 1 s, electron paramagnetic resonance spectra and reactive oxygen species scavenging experiments. The substrate of 3,3′,5,5′-tetramethylbenzidine (TMB) for OXD-like is oxidized into blue ox-TMB, and then turned to yellow diimine, depending on TMB concentration and incubation time. Then the mechanism of ferrous ions (Fe 2+ ) inhibition of the oxidase-like activity of Co 3 O 4 NWs was uncovered that Fe 2+ competes with TMB for oxygen vacancies. The inhibition is reversible with the type of hybrid of competitive inhibition and non-competitive inhibition with the inhibition constants (K I and K I ') of 0.134 mM and 1.012 mM, respectively. Finally, on the basis of the inhibitory effect, a fast, selective and sensitive Fe 2+ colorimetric analysis was proposed with two linear parts and a limit of detection of 0.21 μM Fe 2+ (S/N = 3), which is applicable to detect Fe 2+ in horticultural fertilizers. This study may offer a new strategy for the in-depth study of inhibition mechanism of other nanozymes, which would extend the scope of nanozyme-based biosensing.

Abstract:The poor activity and specificity of nanozymes limit their applications in many fields owing to their low density of active sites, inhomogeneous atomic composition, and complex structure. To overcome these issues, a single iron-anchored carbon material, Fe/CN-800-A, with FeN2 sites has been fabricated, which exhibited exclusive oxidase-like activity. The mechanisms of N-coordination-number-regulated enzymatic activity and specificity of the composite were also explored to elaborate the working principle. The FeN2 sites in Fe/CN-800-A exhibited higher oxidase-like activity and specificity than the FeN4 sites in Fe/CN-700, owing to their lower energy barriers for O2 absorption and the decomposition of H2O2 into O2. These findings provide a theoretical basis for the rational design of nanozymes with high oxidase-like activity and specificity. The fabricated nanocomposites have commercial potential for effective chlorpyrifos detection and antibacterial applications due to their unique structural traits and high efficiency.

Abstract:Single-atom catalysts with atomically dispersed sites have been widely used as nanozymes for colorimetric sensing because their tunable M–N x active centers are similar to those of natural enzymes. However, their low metal atom loading leads to insufficient catalytic activity and affects the sensitivity of colorimetric sensing, which limits their further applications. Herein, multi-walled carbon nanotubes (MWCNs) are selected as carriers to reduce the aggregation of ZIF-8 and improve the electron transfer efficiency of nanomaterials. Meanwhile, MWCN/FeZn-NC single-atom nanozymes with excellent peroxidase-like activity were prepared by pyrolysis on ZIF-8 doped with a second metal Fe. Based on the excellent peroxidase activity of MWCN/FeZn-NCs, a dual-functional colorimetric sensing platform for Cr( VI ) and 8-hydroxyquinoline was established. The detection limits of the dual-function platform are 40 nM for Cr( VI ) and 55 nM for 8-hydroxyquinoline. This work provides a highly sensitive and selective strategy for the detection of Cr( VI ) and 8-hydroxyquinoline in hair care products, which has great application prospects in the field of pollutant detection and control.

Abstract:Artificial enzyme mimics have attracted considerable interest due to easy denaturation and leakage of enzymes during their storage and immobilization procedure. Herein we describe the design of a novel mimic peroxidase, a nanostructure of sheet-like FeS prepared by a simple micelle-assisted synthetic method. Such a nanostructure has a large specific surface area and high peroxidase-like activity, and was thus further used as a mimic enzyme for the development of biocatalysts and amperometric biosensors. The sheet-like FeS nanostructure showed typical Michaelis-Menten kinetics and good affinity to both H2O2 and 3,3',5,5'-tetramethyl benzidine. At pH 7.0 the constructed amperometric sensor showed a linear range for the detection of H2O2 from 0.5 to 150 mu M with a correlation coefficient of 0.9998 without any electron transfer mediator. ne H2O2 sensor based on the sheet-like FeS showed more sensitive response than those based on spherical FeS nanostructure, and resulted in a better stability than horseradish peroxidase when they were exposed to solutions with different pH values and temperatures. These excellent properties made the sheet-like nanostructured FeS powerful tools for a wide range of potential applications as ail "artificial peroxidase" as biosensors and biotechnology.

Abstract:The advent of dual-atom nanozymes (DAzymes) featuring distinctive bimetallic active sites garnered significant attention, representing enhanced iterations of conventional single-atom nanozymes. The quest for an effective and universal strategy to modulate the catalytic activity of DAzymes posed a formidable challenge, yet few published reports addressed this. Herein, we designed and synthesized a S-doped Fe/Co DAzymes (S-FeCo-NC) under theoretical guidance and revealed its excellent oxidase-like activity. Experimental and theoretical calculations indicated that the superior oxidase-like activity exhibited by S-FeCo-NC was attributed to the S-doping, which modulated the local electronic structure of the dual-atom active site. This modulation of the local electronic structure significantly optimizes oxygen adsorption energy, thereby accelerating the rate of enzyme-catalyzed reactions. As a proof-of-concept, this study integrated S-FeCo-NC with the cascade inhibition reaction of acetylcholinesterase (AChE) to devise a sensitive analytical platform for detecting organophosphorus pesticides. This study paved the way for elucidating the correlation between the local electronic structure of the active site and enzyme activity, offering novel methodologies and insights for the rational design of DAzymes.

Reversible Inhibition of Iron Oxide Nanozyme by Guanidine Chloride

Abstract:Nanozymes have been widely applied in bio-assays in the field of biotechnology and biomedicines. However, the physicochemical basis of nanozyme catalytic activity remains elusive. To test whether nanozymes exhibit an inactivation effect similar to that of natural enzymes, we used guanidine chloride (GuHCl) to disturb the iron oxide nanozyme (IONzyme) and observed that GuHCl induced IONzyme aggregation and that the peroxidase-like activity of IONzyme significantly decreased in the presence of GuHCl. However, the aggregation appeared to be unrelated to the quick process of inactivation, as GuHCl acted as a reversible inhibitor of IONzyme instead of a solo denaturant. Inhibition kinetic analysis showed that GuHCl binds to IONzyme competitively with H2O2 but non-competitively with tetramethylbenzidine. In addition, electron spin resonance spectroscopy showed that increasing GuHCl level of GuHCl induced a correlated pattern of changes in the activity and the state of the unpaired electrons of the IONzymes. This result indicates that GuHCl probably directly interacts with the iron atoms of IONzyme and affects the electron density of iron, which may then induce IONzyme inactivation. These findings not only contribute to understanding the essence of nanozyme catalytic activity but also suggest a practically feasible method to regulate the catalytic activity of IONzyme.

Abstract:Replacement of enzymes with nanomaterials such as atomically dispersed metal catalysts is one of the most crucial steps in addressing the challenges in biocatalysis. Despite the breakthroughs of single-atom catalysts in enzyme-mimicking, a fundamental investigation on the development of an instructional strategy is still required for mimicking biatomic/multiatomic active sites in natural enzymes and constructing synergistically enhanced metal atom active sites. Herein, Fe2NC catalysts with atomically dispersed Fe-Fe dual-sites supported by the metal-organic frameworks-derived nitrogen-doped carbon are employed as biomimetic catalysts to perform proof-of-concept investigation. The effect of Fe atom number toward typical oxidase (cytochrome C oxidase, NADH oxidase, and ascorbic acid oxidase) and peroxidase (NADH peroxidase and ascorbic acid peroxidase) activities is systematically evaluated by experimental and theoretical investigations. A peroxo-like O2 adsorption in Fe2NC nanozymes could accelerate the O-O activation and thus achieve the enhanced enzyme-like activities. This work achieves the vivid simulation of the enzyme active sites and provides the theoretical basis for the design of high-performance nanozymes. As a concept application, a colorimetric biosensor for the detection of S2− in tap water is established based on the inhibition of enzyme-like activity of Fe2NC nanozymes.

Abstract:Nanoparticles are massively discharged into the aquatic environment leading to adverse health effects. Considering their adsorption of small molecules and polymers, it is important to study the effect of aqueous system composition on NPs’ behavior and toxicity. Nanomaterials with enzyme-like activities have caught a lot of recent research interest. Since nanozyme reactions take place on the surface of nanomaterials, their catalytic activities often show significant influence by adsorption of small molecules and polymers. CeO2 and Fe3O4 nanoparticles are two classic nanozymes, showing oxidase and peroxidase like activities, respectively. Inorganic arsenic species are critical for environmental and catalytic research. In this study, we used 3,3′,5,5′-tetramethylbenzidine (TMB) as a model substrate to examine the effect of inorganic arsenic species (As(III) and As(V)) on the activity of CeO2 and Fe3O4. The oxidation capacity of nanoceria was decreased to one third with the addition of 1 mM As(III), but was improved by 3-fold in the presence of 0.1 mM As(V). The former is mainly due to the reducing power of As(III), while the latter is attributable to the adsorption of As(V) on the nanoceria surface decreasing the electrostatic repulsion between nanoceria and TMB. For Fe3O4, its activity was first promoted and later inhibited by both As(III) and As(V). Therefore, nanomaterials not only affect arsenic species through redox chemistry and adsorption, but in turn arsenic can affect the catalytic activity of nanomaterials.

Single-atom Iron Containing Nanozyme with Peroxidase-Like Activity and Copper Nanoclusters Based Ratio Fluorescent Strategy for Acetylcholinesterase Activity Sensing

Abstract:Single-atom nanozymes (SAzymes) have attracted extensive interests in bio-catalysis filed because of their robustness, excellent catalytic activity and unique feature of maximum atomic utilization. Herein, the peroxidase-like activity of single-atom iron anchored on N-doped porous carbon (Fe-SAs/NC) SAzyme was successfully demonstrated and a ratio fluorescence method for acetylcholinesterase (AChE) activity sensing was constructed based on Fe-SAs/NC and polyvinylpyrrolidone protected copper nanoclusters (PVP-CuNCs) for the first time. With H2O2 presence, Fe-SAs/NC with peroxidase-like activity catalyzed oxidation of nonfluorescent o-phenylenediamine (OPD) to fluorescent 2,3-diaminophenazine (DAP) with maximum fluorescence emission peak at 566 nm. Meanwhile, the generated DAP efficiently quenched PVP-CuNCs fluorescence at 438 nm through fluorescence resonance energy transfer (FRET). However, AChE possessed function of catalyzing the hydrolysis of acetylthiocholine (ATCh) to generate thiocholine (TCh), which can inhibit the Fe-SAs/NC peroxidase-like activity to block DAP generation and preserve PVP-CuNCs fluorescence. Therefore, the fluorescence intensity ratio F-566/F-438 of the system can serve as signal output for AChE activity sensing ranging from 2 to 70 U L-1 with a detection limit of 0.56 U L-1. Moreover, Fe-SAs/NC and PVP-CuNCs based system was utilized for AChE determination in human serum sample.

Abstract:Nanozymes as promising alternatives to natural enzymes have attracted enormous attentions for their high robustness, low cost, and tunable activity. Among various candidates, carbonaceous nanomaterials possess extra advantage features of designable surface properties and good biocompatibility. However, their peroxidase-like activity is typically moderate owing to the short half-life of radical hydroxyl (·OH), the poor catalytic activity and substrate affinity. Herein, we synthesize Fe,N-doped carbon (Fe,N-C-800) via pyrolyzing Fe-doped ZIF-8 (ZnFe-ZIF) in NH3 at 800 °C. Fe,N-C-800 shows excellent peroxidase mimic with outstanding substrate affinity and catalytic velocity superior to the advanced Fe single-atom catalyst. The excellent enzyme-like performance is ascribed to the synergistic contributions of Fe- and N-doping. Fe functions as catalytic center while pyrrolic N absorbs TMB (3,3',5,5'-Tetramethylbenzidine) substrate and enhances the electron density of Fe sites, thereby shortening the migration distance of ·OH and further improving the catalytic activity. Based on Fe,N-C-800, disposable paper bioassays are fabricated and exhibit ultrasensitive detection of H2O2.

Abstract:Despite the extensive investigation of the nanozymes exhibit their favorable performance compared to natural enzymes, nevertheless, the highly specific nanozyme still needs to be developed so that it can meet the requirements of exploring the mechanism as well as administration of related diseases and selective monitoring in biological system. In this study, self-assembled glutathione-Cu/Cu 2 O nanoparticles (GSH-Cu/Cu 2 O NPs) that exhibits specific ascorbic acid (AA) oxidase-like catalytic activity were constructed for AA-activated and H 2 O 2 -reinforced cancer cell proliferation inhibition and selective neurochemical monitoring. Cu/Cu 2 O NPs demonstrates effective AA oxidase-like activity and no common characteristics of other redox mimic enzymes often present in nanozyme. In particular, we found that the AA oxidase-like activity of GSH-Cu/Cu 2 O nanozyme was significantly improved by about 40% by improving the activation ability toward oxygen. The synthesized nanozyme can induce the generation of active oxygen by accelerating the oxidation of AA, which effectively suppresses the proliferation of cancer cells. We constructed an online electrochemical system (OECS) though loading nanozyme with enhanced ascorbate oxidase activity into a microreactor and setting it in the upstream of the detector. This GSH-Cu/Cu 2 O NPs-integrated microreactor can completely eliminate AA interference of the physical level toward 3,4-dihydroxy phenylacetic acid (DOPAC) electrochemical measurement, and the nanozyme-based OECS is able to continuously capture DOPAC alteration in rat brain acidosis model. Our findings may inspire rational design of nanozymes with high specificity as well as nanozyme-based selectivity solution for in vivo detection and show promising opportunities for their involvement in neurochemistry investigation.

Abstract:Single-atom nanozymes (SAzymes) have drawn ever-increasing attention due to their maximum atom utilization efficiency and enhanced enzyme-like activity. Herein, a facile pyrolysis strategy is reported for the synthesis of the iron–nitrogen–carbon (Fe-N-C) SAzyme using ferrocene trapped within porous zeolitic imidazolate framework-8 (ZIF-8@Fc) as a precursor. The as-prepared Fe-N-C SAzyme exhibited exceptional oxidase-mimicking activity, catalytically oxidizing 3,3′,5,5′-tetramethylbenzidine (TMB) with high affinity (Km) and fast reaction rate (Vmax). Taking advantage of this property, we designed two colorimetric sensing assays based on different interaction modes between small molecules and Fe active sites. Firstly, utilizing the reduction activity of ascorbic acid (AA) toward oxidized TMB (TMBox), a colorimetric bioassay for AA detection was established, which exhibited a good linear range of detection from 0.1 to 2 μM and a detection limit as low as 0.1 μM. Additionally, based on the inhibition of nanozyme activity by the thiols of glutathione (GSH), a colorimetric biosensor for GSH detection was constructed, showing a linear response over a concentration range of 1–10 μM, with a detection limit of 1.3 μM. This work provides a promising strategy for rationally designing oxidase-like SAzymes and broadening their application in biosensing.

Size-controllable Fe-N/C single-atom nanozyme with exceptional oxidase-like activity for sensitive detection of alkaline phosphatase

Abstract:Nanozymes become currently a frontier of chemical research. However, exploiting a novel nanozyme with high activity, good stability and reproducibility is challenging. Here, size-controllable Fe-N/C nanozymes containing exclusive single Fe atoms coordinated Fe-Nx sites were succesfully prepared through a facile pyrolysis of size controllable Fe-Zn ZIFs precursors. The Fe-N/C nanozymes exhibit exceptional high oxidase-mimicking activity able to catalyze oxidation of colorless 3,3',5,5'-tetramethylbenzidine (TMB) by dissolved oxygen to generate blue product. Their catalytic activities can be regulated by modulating the molar ratios of methanol to metal salts (e.g., Fe and Zn) through which the size controllable Fe-Zn ZIFs precursors are obtained. Upon introduction of ascorbic acid (AA) into Fe-N/C/TMB system, complete inhibition of TMB oxidation was observed, resulting in significant decline in absorbance with a clear color change. In the presence of alkaline phosphatase (ALP), ascorbic acid 2-phosphate (AAP) is hydrolyzed to produce ascorbic acid (AA). When coupled with AAP, a novel colorimetric biosensor platform was fabricated for ALP activity screening in the range of 0.05 U/L-100 U/L (four orders of magnitude) with an ultra-low limit of detection of 0.02 U/L. The work provides a promising strategy to rationally design the transition metal-N/C single-atom nanozyme with high oxidase-like activity.

Abstract:MoS2 nanosheets were doped with Fe elements simply via the one-pot hydrothermal route resulting in the peroxidase-like nanozymes with dramatically enhanced catalysis. It was discovered that the resulted Fe-MoS2 could display controllable catalysis activities depending on the Mo-to-Fe molar ratios, among which the highest one was achieved at 3/1 with catalysis efficiency over three folds higher than that of pristine MoS2. The greatly improved catalysis performances of Fe-MoS2 nanozyme are thought to be resulted from the synergistic effects of the doped Fe atom and Mo-S sites. On the one hand, herein, the Fe sites might serve as the electron transferring mediators to facilitate the peroxidase-like catalytic reactions, as confirmed by the cytochrome C-based electron transportation experiments. On the other hand, the Mo-S sites might conduct the catalytic behavior of Fenton-like reactions, as revealed by the hydroxyl radicals-capturing experiments. Furthermore, the feasibility of practical applications for the developed Fe-MoS2 nanozyme based colorimetric methods was demonstrated for glucose in blood sample. Importantly, an insight can be thereby obtained into the design of different nanozymes with enhanced catalysis designed by doping heteroatoms like Fe, promising for the catalysis-based applications in the biomedical analysis, food safety, and environmental monitoring fields.

Abstract:Peroxidase-mimicking nanozymes offer unique advantages in terms of high stability and low cost over natural peroxidase for applications in bioanalysis, biomedicine, and the treatment of pollution. However, the design of high-efficiency peroxidase-mimicking nanozymes remains a great challenge. In this study, we adopted a structural-design approach through hybridization of cube-CeO2 and Pt nanoparticles to create a new peroxidase-mimicking nanozyme with high efficiency and excellent stability. Relative to pure cube-CeO2 and Pt nanoparticles, the as-hybridized Pt/cube-CeO2 nanocomposites display much improved activities because of the strong metal-support interaction. Meanwhile, the nanocomposites also maintain high catalytic activity after long-term storage and multiple recycling. Based on their excellent properties, Pt/cube-CeO2 nanocomposites were used to construct high-performance colorimetric biosensors for the sensitive detection of metabolites, including H2 O2 and glucose. Our findings highlight opportunities for the development of high-efficiency peroxidase-mimicking nanozymes with potential applications such as diagnostics, biomedicine, and the treatment of pollution.
