Creating peroxidase-mimetic clusterzymes for efficient and selectively enzymatic diagnosis of biomarkers

Abstract:Developing visual, portable, versatile, and highly sensitive colorimetric biosensors are of great significance for future disease diagnoses and therapies. To overcome the challenges in creating enzymatic nanomaterials that present integrated advantages of high efficiency, selectivity, stability, and biocompatibility for visual colorimetric biosensors, here, we have constructed a new nanosized Ru clusterzyme supported by TiO 2 () via a facile two-step method to serve as peroxidase (POD)-like enzymatic nanomaterials for colorimetrical biomarker detection. Our morphology and chemical/electronic structure analysis prove that the consists of abundant surface-decorated Ru clusterzyme centers via Ru-O coordination. Benefiting from the unique enzymatic properties of Ru clusterzymes, the displays the highest V max (135.14 M s −1 ) and turnover number (210.8 × 10 -3 s −1 ) for H 2 O 2 among the state-of-the-art POD-mimics, manifesting the exceptional biosensing properties of compared to recently reported POD-like nanomaterials. Colorimetric studies of have displayed a relatively larger range and lower detection limit for the H 2 O 2 , L-cysteine, and glucose, thus exhibiting comparable detecting sensitivity with previous reports. Moreover, the presents extraordinary resistance to extreme environments, including low pH and high temperature. This work will create a new path to design efficient colorimetrical biosensors and offer tremendous potential for future biocatalysis, bioassays, and therapeutic applications.
