Abstract:Animals use stereo sampling of odor concentration to localize sources and follow odor trails. We analyze the dynamics of a bilateral model that depends on the simultaneous comparison between odor concentrations detected by left and right sensors. The general model consists of three differential equations for the positions in the plane and the heading. When the odor landscape is an infinite trail, then we reduce the dynamics to a planar system whose dynamics have just two fixed points. Using an integrable approximation (for short sensors) we estimate the basin of attraction. In the case of a radially symmetric landscape, we again can reduce the dynamics to a planar system, but the behavior is considerably richer with multi-stability, isolas, and limit cycles. As in the linear trail case, there is also an underlying integrable system when the sensors are short. In odor landscapes that consist of multiple spots and trail segments, we find periodic and chaotic dynamics and characterize the behavior on trails with gaps and that turn corners.

Abstract:Two simple algorithms based on combining odor concentration differences across time and space along with information on the flow direction are tested for their ability to locate an odor source in four different odor landscapes. Image data taken from air plumes in three different regimes and a water plume are used as test environments for a bilateral ("stereo sampling") algorithm using concentration differences across two sensors and a "casting" algorithm that uses successive samples to decide orientation. Agents are started at random locations and orientations in the landscape and allowed to move until they reach the source of the odor (success) or leave the imaged area (failure). Parameters for the algorithm are chosen to optimize success and to minimize path length to the source. Success rates over 90% are consistently obtained with path lengths that can be as low as twice the starting distance from the source in air and four times the distance in the highly turbulent water plumes. We find that parameters that optimize success often lead to more exploratory pathways to the source. Information about the direction from which the odor is coming is necessary for successful navigation in the water plume and reduces the path length in the three tested air plumes.

Abstract:Locating the source of odor in a turbulent environment—a common behavior for living organisms—is nontrivial because of the random nature of mixing. Here we analyze the statistical physics aspects of the problem and propose an efficient strategy for olfactory search that can work in turbulent plumes. The algorithm combines the maximum likelihood inference of the source position with an active search. Our approach provides the theoretical basis for the design of olfactory robots and the quantitative tools for the analysis of the observed olfactory search behavior of living creatures (e.g., odor-modulated optomotor anemotaxis of moths).

Abstract:As an evolutionarily ancient sense, olfaction is key to learning where to find food, shelter, mates, and important landmarks in an animal's environment. Brain circuitry linking odor and navigation appears to be a well conserved multi-region system among mammals; the anterior olfactory nucleus, piriform cortex, entorhinal cortex, and hippocampus each represent different aspects of olfactory and spatial information. We review recent advances in our understanding of the neural circuits underlying odor-place associations, highlighting key choices of behavioral task design and neural circuit manipulations for investigating learning and memory.

Abstract:In olfactory search an immobile target emits chemical molecules at constant rate. The molecules are transported by the medium, which is assumed to be turbulent. Considering a searcher able to detect such chemical signals and whose motion follows the infotaxis strategy, we study the statistics of the first-passage time to the target when the searcher moves on a finite two-dimensional lattice of different geometries. Far from the target, where the concentration of chemicals is low, the direction of the searcher's first movement is determined by the geometry of the domain and the topology of the lattice, inducing strong fluctuations on the average search time with respect to the initial position of the searcher. The domain is partitioned in well-defined regions characterized by the direction of the first movement. If the search starts over the interface between two different regions, large fluctuations in the search time are observed.

Abstract:We consider the problem of olfactory searches in a turbulent environment. We focus on agents that respond solely to odor stimuli, with no access to spatial perception nor prior information about the odor location. We ask whether navigation strategies to a target can be learned robustly within a sequential decision making framework. We develop a reinforcement learning algorithm using a small set of interpretable olfactory states and train it with realistic turbulent odor cues. By introducing a temporal memory, we demonstrate that two salient features of odor traces, discretized in few olfactory states, are sufficient to learn navigation in a realistic odor plume. Performance is dictated by the sparse nature of turbulent plumes. An optimal memory exists which ignores blanks within the plume and activates a recovery strategy outside the plume. We obtain the best performance by letting agents learn their recovery strategy and show that it is mostly casting cross wind, similar to behavior observed in flying insects. The optimal strategy is robust to substantial changes in the odor plumes, suggesting minor parameter tuning may be sufficient to adapt to different environments.

Abstract:Abstract Odor attraction in walking Drosophila melanogaster is commonly used to relate neural function to behavior, but the algorithms underlying attraction are unclear. Here we develop a high-throughput assay to measure olfactory behavior in response to well-controlled sensory stimuli. We show that odor evokes two behaviors: an upwind run during odor (ON response), and a local search at odor offset (OFF response). Wind orientation requires antennal mechanoreceptors, but search is driven solely by odor. Using dynamic odor stimuli, we measure the dependence of these two behaviors on odor intensity and history. Based on these data, we develop a navigation model that recapitulates the behavior of flies in our apparatus, and generates realistic trajectories when run in a turbulent boundary layer plume. The ability to parse olfactory navigation into quantifiable elementary sensori-motor transformations provides a foundation for dissecting neural circuits that govern olfactory behavior.

Abstract:Navigating an odor landscape is a critical behavior for the survival of many species, including mice. An ethologically relevant mouse behavior is locating food using information about odor concentration. To approximate this behavior, we used an open field odor-based spot-finding task indoors with little wind, examining navigation strategies as mice search for and approach an odor source. After mice were trained to navigate to odor sources paired with food reward, we observed behavioral changes consistent with localization 10–45 cm away from the source. These behaviors included orientation toward the source, decreased velocity, and increased exploration time. We also found that the amplitude of 'casting,' which we define as lateral back and forth movement of the nose, increased with proximity to the source. Based on these observations, we created a concentration-sensitive agent-based model to simulate mouse behavior. This model provided evidence for a binaral-sniffing strategy (inter-nostril comparison of concentration in a single sniff) and a serial-sniffing strategy (sampling concentration, moving in space, and then sampling again). Serial-sniffing may be accomplished at farther distances by moving the body and at closer distances by moving the head (casting). Together, these results elucidate components of behavioral strategies for odor-based navigation.

Abstract:Animals navigating turbulent odor plumes exhibit a rich variety of behaviors, and employ efficient strategies to locate odor sources. A growing body of literature has started to probe this complex task of localizing airborne odor sources in walking mammals to further our understanding of neural encoding and decoding of naturalistic sensory stimuli. However, correlating the intermittent olfactory information with behavior has remained a long-standing challenge due to the stochastic nature of the odor stimulus. We recently reported a method to record real-time olfactory information available to freely moving mice during odor-guided navigation, hence overcoming that challenge. Here we combine our odor-recording method with head-motion tracking to establish correlations between plume encounters and head movements. We show that mice exhibit robust head-pitch motions in the 5-14Hz range during an odor-guided navigation task, and that these head motions are modulated by plume encounters. Furthermore, mice reduce their angles with respect to the source upon plume contact. Head motions may thus be an important part of the sensorimotor behavioral repertoire during naturalistic odor-source localization.

Abstract:Living systems continually respond to signals from the surrounding environment. Survival requires that their responses adapt quickly and robustly to the changes in the environment. One particularly challenging example is olfactory navigation in turbulent plumes, where animals experience highly intermittent odor signals while odor concentration varies over many length- and timescales. Here, we show theoretically that Drosophila olfactory receptor neurons (ORNs) can exploit proximity to a bifurcation point of their firing dynamics to reliably extract information about the timing and intensity of fluctuations in the odor signal, which have been shown to be critical for odor-guided navigation. Close to the bifurcation, the system is intrinsically invariant to signal variance, and information about the timing, duration, and intensity of odor fluctuations is transferred efficiently. Importantly, we find that proximity to the bifurcation is maintained by mean adaptation alone and therefore does not require any additional feedback mechanism or fine-tuning. Using a biophysical model with calcium-based feedback, we demonstrate that this mechanism can explain the measured adaptation characteristics of Drosophila ORNs.

Optimal switching strategies for navigation in stochastic settings

Abstract:Inspired by the intermittent reorientation strategy seen in the behavior of the dung beetle, we consider the problem of the navigation strategy of an active Brownian particle moving in two dimensions. We assume that the heading of the particle can be reoriented to the preferred direction by paying a fixed cost as it tries to maximize its total displacement in a fixed direction. Using optimal control theory, we derive analytically and confirm numerically the strategy that maximizes the particle speed, and show that the average time between reorientations scales inversely with the magnitude of the environmental noise. We then extend our framework to describe execution errors and sensory acquisition noise. Our approach may be amenable to other navigation problems involving multiple sensory modalities that require switching between egocentric and geocentric strategies.

Abstract:In order to survive, animals often need to navigate a complex odor landscape where odors can exist in airborne plumes. Several odor plume properties change with distance from the odor source, providing potential navigational cues to searching animals. Here we focus on odor intermittency, a temporal odor plume property that measures the fraction of time odor is above a threshold at a given point within the plume and decreases with increasing distance from the odor source. We sought to determine if mice can use changes in intermittency to locate an odor source. To do so, we trained mice on an intermittency discrimination task. We establish that mice can discriminate odor plume samples of low and high intermittency and that the neural responses in the olfactory bulb can account for task performance and support intermittency encoding. Modulation of sniffing, a behavioral parameter that is highly dynamic during odor-guided navigation, affects both behavioral outcome on the intermittency discrimination task as well as neural representation of intermittency. Together, this work demonstrates that intermittency is an odor plume property that can inform olfactory search and more broadly supports the notion that mammalian odor-based navigation can be guided by temporal odor plume properties.

Abstract:Odour plumes in the wild are spatially complex and rapidly fluctuating structures carried by turbulent airflows1-4. To successfully navigate plumes in search of food and mates, insects must extract and integrate multiple features of the odour signal, including odour identity5, intensity6 and timing6-12. Effective navigation requires balancing these multiple streams of olfactory information and integrating them with other sensory inputs, including mechanosensory and visual cues9,12,13. Studies dating back a century have indicated that, of these many sensory inputs, the wind provides the main directional cue in turbulent plumes, leading to the longstanding model of insect odour navigation as odour-elicited upwind motion6,8-12,14,15. Here we show that Drosophila melanogaster shape their navigational decisions using an additional directional cue-the direction of motion of odours-which they detect using temporal correlations in the odour signal between their two antennae. Using a high-resolution virtual-reality paradigm to deliver spatiotemporally complex fictive odours to freely walking flies, we demonstrate that such odour-direction sensing involves algorithms analogous to those in visual-direction sensing16. Combining simulations, theory and experiments, we show that odour motion contains valuable directional information that is absent from the airflow alone, and that both Drosophila and virtual agents are aided by that information in navigating naturalistic plumes. The generality of our findings suggests that odour-direction sensing may exist throughout the animal kingdom and could improve olfactory robot navigation in uncertain environments.

Abstract:The olfactory system of male moths is exquisitely sensitive to pheromones emitted by females and transported in the environment by atmospheric turbulence. Moths respond to minute amounts of pheromones and their behavior is sensitive to the fine-scale structure of turbulent plumes where pheromone concentration is detectible. The signal of pheromone whiffs is qualitatively known to be intermittent, yet quantitative characterization of its statistical properties is lacking. This challenging fluid dynamics problem is also relevant for entomology, neurobiology and the technological design of olfactory stimulators aimed at reproducing physiological odor signals in well-controlled laboratory conditions. Here, we develop a Lagrangian approach to the transport of pheromones by turbulent flows and exploit it to predict the statistics of odor detection during olfactory searches. The theory yields explicit probability distributions for the intensity and the duration of pheromone detections, as well as their spacing in time. Predictions are favorably tested by using numerical simulations, laboratory experiments and field data for the atmospheric surface layer. The resulting signal of odor detections lends to implementation with state-of-the-art technologies and quantifies the amount and the type of information that male moths can exploit during olfactory searches.

Learning to predict target location with turbulent odor plumes

Abstract:Animal behavior and neural recordings show that the brain is able to measure both the intensity of an odor and the timing of odor encounters. However, whether intensity or timing of odor detections is more informative for olfactory-driven behavior is not understood. To tackle this question, we consider the problem of locating a target using the odor it releases. We ask whether the position of a target is best predicted by measures of timing intensity of its odor, sampled for a short period of time. To answer this question, we feed data from accurate numerical simulations of odor transport to machine learning algorithms that learn how to connect odor to target location. We find that both intensity and timing can separately predict target location even from a distance of several meters; however their efficacy varies with the dilution of the odor in space. Thus organisms that use olfaction from different ranges may have to switch among different modalities. This has implications on how the brain should represent odors as the target is approached. We demonstrate simple strategies to improve accuracy and robustness of the prediction by modifying odor sampling and appropriately combining distinct measures together. To test the predictions, animal behavior and odor representation should be monitored as the animal moves relative to the target, or in virtual conditions that mimic concentrated dilute environments.

Abstract:Animals smelling in the real world use a small number of receptors to sense a vast number of natural molecular mixtures, and proceed to learn arbitrary associations between odors and valences. Here, we propose a new interpretation of how the architecture of olfactory circuits is adapted to meet these immense complementary challenges. First, the diffuse binding of receptors to many molecules compresses a vast odor space into a tiny receptor space, while preserving similarity. Next, lateral interactions "densify" and decorrelate the response, enhancing robustness to noise. Finally, disordered projections from the periphery to the central brain reconfigure the densely packed information into a format suitable for flexible learning of associations and valences. We test our theory empirically using data from Drosophila. Our theory suggests that the neural processing of olfactory information differs from the other senses in its fundamental use of disorder.

Spontaneous Rapid Odor Source Localization Behavior Requires Interhemispheric Communication

Abstract:Navigation, finding food sources, and avoiding danger critically depend on the identification and spatial localization of airborne chemicals. When monitoring the olfactory environment, rodents spontaneously engage in active olfactory sampling behavior, also referred to as exploratory sniffing [1]. Exploratory sniffing is characterized by stereotypical high-frequency respiration, which is also reliably evoked by novel odorant stimuli [2, 3]. To study novelty-induced exploratory sniffing, we developed a novel, non-contact method for measuring respiration by infrared (IR) thermography in a behavioral paradigm in which novel and familiar stimuli are presented to head-restrained mice. We validated the method by simultaneously performing nasal pressure measurements, a commonly used invasive approach [2, 4], and confirmed highly reliable detection of inhalation onsets. We further discovered that mice actively orient their nostrils toward novel, previously unexperienced, smells. In line with the remarkable speed of olfactory processing reported previously [3, 5, 6], we find that mice initiate their response already within the first sniff after odor onset. Moreover, transecting the anterior commissure (AC) disrupted orienting, indicating that the orienting response requires interhemispheric transfer of information. This suggests that mice compare odorant information obtained from the two bilaterally symmetric nostrils to locate the source of the novel odorant. We further demonstrate that asymmetric activation of the anterior olfactory nucleus (AON) is both necessary and sufficient for eliciting orienting responses. These findings support the view that the AON plays an important role in the internostril difference comparison underlying rapid odor source localization.

Collective olfactory search in a turbulent environment

Abstract:Finding the distant source of an odor dispersed by a turbulent flow is a vital task for many organisms, either for foraging or for mating purposes. At the level of individual search, animals like moths have developed effective strategies to solve this very difficult navigation problem based on the noisy detection of odor concentration and wind velocity alone. When many individuals concurrently perform the same olfactory search task, without any centralized control, sharing information about the decisions made by the members of the group can potentially increase the performance. But how much of this information is actually valuable and exploitable for the collective task ? Here we show that, in a model of a swarm of agents inspired by moth behavior, there is an optimal way to blend the private information about odor and wind detections with the publicly available information about other agents' heading direction. At optimality, the time required for the first agent to reach the source is essentially the shortest flight time from the departure point to the target. Conversely, agents who discard public information are several fold slower and groups that do not put enough weight on private information perform even worse. Our results then suggest an efficient multi-agent olfactory search algorithm that could prove useful in robotics, for instance in the identification of sources of harmful volatile compounds.

Olfactory learning alters navigation strategies and behavioral variability in C. elegans

Abstract:Animals adjust their behavioral response to sensory input adaptively depending on past experiences. The flexible brain computation is crucial for survival and is of great interest in neuroscience. The nematode C. elegans modulates its navigation behavior depending on the association of odor butanone with food (appetitive training) or starvation (aversive training), and will then climb up the butanone gradient or ignore it, respectively. However, the exact change in navigation strategy in response to learning is still unknown. Here we study the learned odor navigation in worms by combining precise experimental measurement and a novel descriptive model of navigation. Our model consists of two known navigation strategies in worms: biased random walk and weathervaning. We infer weights on these strategies by applying the model to worm navigation trajectories and the exact odor concentration it experiences. Compared to naive worms, appetitive trained worms up-regulate the biased random walk strategy, and aversive trained worms down-regulate the weathervaning strategy. The statistical model provides prediction with $>90 \%$ accuracy of the past training condition given navigation data, which outperforms the classical chemotaxis metric. We find that the behavioral variability is altered by learning, such that worms are less variable after training compared to naive ones. The model further predicts the learning-dependent response and variability under optogenetic perturbation of the olfactory neuron AWC$^\mathrm{ON}$. Lastly, we investigate neural circuits downstream from AWC$^\mathrm{ON}$ that are differentially recruited for learned odor-guided navigation. Together, we provide a new paradigm to quantify flexible navigation algorithms and pinpoint the underlying neural substrates.

Optimal policies for Bayesian olfactory search in turbulent flows

Abstract:In many practical scenarios, a flying insect must search for the source of an emitted cue which is advected by the atmospheric wind. On the macroscopic scales of interest, turbulence tends to mix the cue into patches of relatively high concentration over a background of very low concentration, so that the insect will only detect the cue intermittently and cannot rely on chemotactic strategies which simply climb the concentration gradient. In this work, we cast this search problem in the language of a partially observable Markov decision process (POMDP) and use the Perseus algorithm to compute strategies that are near-optimal with respect to the arrival time. We test the computed strategies on a large two-dimensional grid, present the resulting trajectories and arrival time statistics, and compare these to the corresponding results for several heuristic strategies, including (space-aware) infotaxis, Thompson sampling, and QMDP. We find that the near-optimal policy found by our implementation of Perseus outperforms all heuristics we test by several measures. We use the near-optimal policy to study how the search difficulty depends on the starting location. We discuss additionally the choice of initial belief and the robustness of the policies to changes in the environment. Finally, we present a detailed and pedagogical discussion about the implementation of the Perseus algorithm, including the benefits -- and pitfalls -- of employing a reward shaping function.
