Abstract:The mechanisms associated with the regulation of iron (Fe) homeostasis have been extensively examined, however, epigenetic regulation of these processes remains largely unknown. Here, we report that a naturally occurring epigenetic mutant, Colorless non-ripening (Cnr), displayed increased Fe-deficiency responses compared to its wild-type Ailsa Craig (AC). RNA-sequencing revealed that a total of 947 and 1,432 genes were up-regulated by Fe deficiency in AC and Cnr roots, respectively, while 923 and 1,432 genes were, respectively, down-regulated. Gene ontology analysis of differentially expressed genes showed that genes encoding enzymes, transporters, and transcription factors were preferentially affected by Fe deficiency. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis revealed differential metabolic responses to Fe deficiency between AC and Cnr. Based on comparative transcriptomic analyses, 24 genes were identified as potential targets of Cnr epimutation, and many of them were found to be implicated in Fe homeostasis. By developing CRISPR/Cas9 genome editing SlSPL-CNR knockout (KO) lines, we found that some Cnr-mediated Fe-deficiency responsive genes showed similar expression patterns between SlSPL-CNR KO plants and the Cnr epimutant. Moreover, both two KO lines displayed Fe-deficiency-induced chlorosis more severe than AC plants. Additionally, the Cnr mutant displayed hypermethylation in the 286-bp epi-mutated region on the SlSPL-CNR promoter, which contributes to repressed expression of SlSPL-CNR when compared with AC plants. However, Fe-deficiency induced no change in DNA methylation both at the 286-bp epi-allele region and the entire region of SlSPL-CNR gene. Taken together, using RNA-sequencing and genetic approaches, we identified Fe-deficiency responsive genes in tomato roots, and demonstrated that SlSPL-CNR is a novel regulator of Fe-deficiency responses in tomato, thereby, paving the way for further functional characterization and regulatory network dissection.

Abstract:Background The family of NAC proteins (NAM, ATAF1/2, and CUC2) represent a class of large plant-specific transcription factors. However, identification and functional surveys of NAC genes of tomato (Solanum lycopersicum) remain unstudied, despite the tomato genome being decoded for several years. This study aims to identify the NAC gene family and investigate their potential roles in responding to Al stress. Results Ninety-three NAC genes were identified and named in accordance with their chromosome location. Phylogenetic analysis found SlNACs are broadly distributed in 5 groups. Gene expression analysis showed that SlNACs had different expression levels in various tissues and at different fruit development stages. Cycloheximide treatment and qRT-PCR analysis indicated that SlNACs may aid regulation of tomato in response to Al stress, 19 of which were significantly up- or down-regulated in roots of tomato following Al stress. Conclusion This work establishes a knowledge base for further studies on biological functions of SlNACs in tomato and will aid in improving agricultural traits of tomato in the future.
The Effects of Salinity on the Anatomy and Gene Expression Patterns in Leaflets of Tomato cv. Micro-Tom

Abstract:Salinity is a form of abiotic stress that impacts growth and development in several economically relevant crops and is a top-ranking threat to agriculture, considering the average rise in the sea level caused by global warming. Tomato is moderately sensitive to salinity and shows adaptive mechanisms to this abiotic stressor. A case study on the dwarf tomato model Micro-Tom is here presented in which the response to salt stress (NaCl 200 mM) was investigated to shed light on the changes occurring at the expression level in genes involved in cell wall-related processes, phenylpropanoid pathway, stress response, volatiles' emission and secondary metabolites' production. In particular, the response was analyzed by sampling older/younger leaflets positioned at different stem heights (top and bottom of the stem) and locations along the rachis (terminal and lateral) with the goal of identifying the most responsive one(s). Tomato plants cv. Micro-Tom responded to increasing concentrations of NaCl (0-100-200-400 mM) by reducing the leaf biomass, stem diameter and height. Microscopy revealed stronger effects on leaves sampled at the bottom and the expression analysis identified clusters of genes expressed preferentially in older or younger leaflets. Stress-related genes displayed a stronger induction in lateral leaflets sampled at the bottom. In conclusion, in tomato cv. Micro-Tom subjected to salt stress, the bottom leaflets showed stronger stress signs and response, while top leaflets were less impacted by the abiotic stressor and had an increased expression of cell wall-related genes involved in expansion.
Combined salt and low nitrate stress conditions lead to morphophysiological changes and tissue-specific transcriptome reprogramming in tomato

Abstract:Despite intense research towards the understanding of abiotic stress adaptation in tomato, the physiological adjustments and transcriptome modulation induced by combined salt and low nitrate (low N) conditions remain largely unknown. Here, three traditional tomato genotypes were grown under long-term single and combined stresses throughout a complete growth cycle. Physiological, molecular, and growth measurements showed extensive morphophysiological modifications under combined stress compared to the control, and single stress conditions, resulting in the highest penalty in yield and fruit size. The mRNA sequencing performed on both roots and leaves of genotype TRPO0040 indicated that the transcriptomic signature in leaves under combined stress conditions largely overlapped that of the low N treatment, whereas root transcriptomes were highly sensitive to salt stress. Differentially expressed genes were functionally interpreted using GO and KEGG enrichment analysis, which confirmed the stress and the tissue-specific changes. We also disclosed a set of genes underlying the specific response to combined conditions, including ribosome components and nitrate transporters, in leaves, and several genes involved in transport and response to stress in roots. Altogether, our results provide a comprehensive understanding of above- and below-ground physiological and molecular responses of tomato to salt stress and low N treatment, alone or in combination.
Identification of key genes involved in the phenotypic alterations of res (restored cell structure by salinity) tomato mutant and its recovery induced by salt stress through transcriptomic analysis

Abstract:Background: The res (restored cell structure by salinity) mutant, recently identified as the first tomato mutant accumulating jasmonate in roots under non-stressful conditions, exhibits a remarkable growth inhibition and morphological alterations in roots and leaves, which are suppressed when the mutant plants are exposed to salinity. In order to understand the molecular basis of the phenotype recovery induced by salt stress in the res mutant, we carried out a comparative transcriptomic analysis in roots and leaves of wild-type and res plants in absence of stress (control) and when the phenotypic recovery of res mutant began to be observed upon salt stress (5 days of 200 mM NaCl). Results: The number of differentially expressed genes was three times greater in roots than in leaves of res vs WT plants grown in control, and included the down-regulation of growth-promoting genes and the up-regulation of genes involved in Ca2+ signalling, transcription factors and others related to stress responses. However, these expression differences were attenuated under salt stress, coinciding with the phenotypic normalisation of the mutant. Contrarily to the attenuated response observed in roots, an enhanced response was found in leaves under salt stress. This included drastic expression changes in several circadian clock genes, such as GIGANTEA1, which was down-regulated in res vs WT plants. Moreover, the higher photosynthetic efficiency of res leaves under salt stress was accompanied by specific salt-upregulation of the genes RUBISCO ACTIVASE1 and ALTERNATIVE OXIDASE1A. Very few genes were found to be differentially expressed in both tissues (root and leaf) and conditions (control and salt), but this group included SlWRKY39 and SlMYB14 transcription factors, as well as genes related to protein homeostasis, especially protease inhibitors such as METALLOCARBOXYPEPTIDASE INHIBITOR, which also seem to play a role in the phenotype recovery and salt tolerance of res mutant. Conclusions: In summary, in this study we have identified genes which seem to have a prominent role in salt tolerance. Moreover, we think this work could contribute to future breeding of tomato crops with increased stress tolerance.

Abstract:Regulation of sodium homeostasis is crucial for plant response to salinity conditions. Here we report on the genetic and physiological characterization of two tomato allelic mutants, ( ), which exhibit pronounced chlorosis and hyperhydration under salt stress. Mapping-by-sequencing revealed that mutant phenotype resulted from mutations in the gene, and CRISPR/Cas9 knockouts of this gene gave phenocopies of the mutants. Physiological analyses showed that salt hypersensitivity is linked to an increase of Na and water transport from roots to shoots, which explains their extreme chlorosis and hyperhydration under salinity conditions. At the molecular level, gene, an aquaporin down-regulated in the WT under salt stress, was overexpressed in the mutants, which could enhance water transport to the shoot. Also, mutants exhibited a significant reduction in the expression of key sodium transporters, thus modifying the normal distribution of Na in tomato plant tissues. Furthermore, treatment of WT plants with the NADPH oxidase inhibitor DPI prevented H O production in response to salinity, resulting in elevated Na accumulation in the shoot and reduced expression of the gene in root. Altogether, our results show that plays a central role in salt tolerance through ROS-mediated signaling.

Hormonal and metabolic regulation of tomato fruit sink activity and yield under salinity

Abstract:Salinization of water and soil has a negative impact on tomato (Solanum lycopersicum L.) productivity by reducing growth of sink organs and by inducing senescence in source leaves. It has been hypothesized that yield stability implies the maintenance or increase of sink activity in the reproductive structures, thus contributing to the transport of assimilates from the source leaves through changes in sucrolytic enzymes and their regulation by phytohormones. In this study, classical and functional physiological approaches have been integrated to study the influence of metabolic and hormonal factors on tomato fruit sink activity, growth, and yield: (i) exogenous hormones were applied to plants, and (ii) transgenic plants overexpressing the cell wall invertase (cwInv) gene CIN1 in the fruits and de novo cytokinin (CK) biosynthesis gene IPT in the roots were constructed. Although salinity reduces fruit growth, sink activity, and trans-zeatin (tZ) concentrations, it increases the ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC) during the actively growing period (25 days after anthesis). Indeed, exogenous application of the CK analogue kinetin to salinized actively growing fruits recovered sucrolytic activities (mainly cwInv and sucrose synthase), sink strength, and fruit weight, whereas the ethylene-releasing compound ethephon had a negative effect in equivalent non-stressed fruits. Fruit yield was increased by both the constitutive expression of CIN1 in the fruits (up to 4-fold) or IPT in the root (up to 30%), owing to an increase in the fruit number (lower flower abortion) and in fruit weight. This is possibly related to a recovery of sink activity in reproductive tissues due to both (i) increase in sucrolytic activities (cwInv, sucrose synthase, and vacuolar and cytoplasmic invertases) and tZ concentration, and (ii) a decrease in the ACC levels and the activity of the invertase inhibitor. This study provides new functional evidences about the role of metabolic and hormonal inter-regulation of local sink processes in controlling tomato fruit sink activity, growth, and yield under salinity.
Dissecting the genetic regulation of lateral root development in tomato under salt stress

Abstract:Tomatoes are crucial for global food security and agricultural economies. With over 50% of arable land projected to become saline by 2050, understanding tomato responses to saline environments is essential. Soil salinization, prevalent in arid regions, leads to salt stress. Roots are primary salinity sensors and integrate stress responses like nutrient deficiency. Understanding the molecular processes maintaining root growth is vital for crop resilience. While salt stress impacts on tomato seed germination, shoot growth, and fruit yield are documented, root development remains underexplored. Most studies have focused on a limited number of cultivars and their physiological responses, not the genetic basis of salt resilience. Wild tomato relatives, such as Solanum pimpinellifolium, offer traits that are beneficial for breeding resilient crops. However, few studies have examined the genetics of salinity tolerance in S. pimpinellifolium. This study characterizes salt stress-induced changes in root system architecture (RSA) using a natural diversity panel of 220 wild tomato accessions. We identified natural variation in lateral root development and several candidate loci through Genome-Wide Association Study (GWAS). Bulk segregant analysis (BSA) revealed 22 candidate genes overlapping with GWAS findings. Exploring root transcriptome reprogramming in two accessions with differing lateral root responses highlighted genotype-specific changes. Integrating GWAS, BSA, and RNA sequencing data identified four key genes underlying differential lateral root development under salt stress. These findings offer novel genetic targets for improving salt resilience in tomatoes, providing opportunities for future research and breeding programs to enhance crop sustainability and productivity.

Growth, yield and water relations of normal fruited and cherry tomato cultivars irrigated with saline water

Abstract:Plants of a tomato cultivar of normal fruit-size, weak vegetative development and high productivity (L. esculentum cv. New Yorker) and another of small fruit-size (cherry) and strong vegetative development (L. esculentum var. cerasiforme cv. PE-62) were grown in an unheated plastic greenhouse and irrigated with 0 and 140 mM NaCl water (control and saline treatments, respectively). The shoot and leaf dry weights and leaf area reductions induced by salinity were related to the osmotic adjustment achieved at each time and cultivar. Plants of cv. PE-62 showed a decrease in shoot to root dry-weight ratio in response to salinity. Yields of both cultivars were reduced significantly by salinity; a result of a fewer fruits and smaller fruits. Fruits from salt treated plants contained a higher concentration of reducing sugars and organic acids than those of control plants. The fruit life span was shortened in plants under saline stress. In both cultivars, the leaf osmotic potential at full turgor (Ψos) levels were reduced by salinity. The parallel pattern observed in both cultivars, between Ψos and Ψtlp, (leaf water potential at turgor loss point) could indicate that Ψos values were controlling the behaviour of Ψtlp. The bulk modulus of elasticity (E) and the relative water content at the turgor loss point (RWCtlp) were not affected by salinity. The RWCtlp values in both cultivar seems to be controlled by E values.

Abstract:Introduction Tomato ( Solanum lycopersicum L.) is a major horticultural crop that is cultivated worldwide and is characteristic of the Mediterranean agricultural system. It represents a key component of the diet of billion people and an important source of vitamins and carotenoids. Tomato cultivation in open field often experiences drought episodes, leading to severe yield losses, since most modern cultivars are sensitive to water deficit. Water stress leads to changes in the expression of stress-responsive genes in different plant tissues, and transcriptomics can support the identification of genes and pathways regulating this response. Methods Here, we performed a transcriptomic analysis of two tomato genotypes, M82 and Tondo, in response to a PEG-mediated osmotic treatment. The analysis was conducted separately on leaves and roots to characterize the specific response of these two organs. Results A total of 6,267 differentially expressed transcripts related to stress response was detected. The construction of gene co-expression networks defined the molecular pathways of the common and specific responses of leaf and root. The common response was characterized by ABA-dependent and ABA-independent signaling pathways, and by the interconnection between ABA and JA signaling. The root-specific response concerned genes involved in cell wall metabolism and remodeling, whereas the leaf-specific response was principally related to leaf senescence and ethylene signaling. The transcription factors representing the hubs of these regulatory networks were identified. Some of them have not yet been characterized and can represent novel candidates for tolerance. Discussion This work shed new light on the regulatory networks occurring in tomato leaf and root under osmotic stress and set the base for an in-depth characterization of novel stress-related genes that may represent potential candidates for improving tolerance to abiotic stress in tomato.

Responses of Tomato Plants under Saline Stress to Foliar Application of Copper Nanoparticles

Abstract:The tomato crop has great economic and nutritional importance; however, it can be adversely affected by salt stress. The objective of this research is to quantify the agronomic and biochemical responses of tomato plants developed under salt stress with the foliar application of copper nanoparticles. Four treatments were evaluated: foliar application of copper nanoparticles (250 mg L−1) with or without salt stress (50 mM NaCl), salt stress, and an absolute control. Saline stress caused severe damage to the development of tomato plants; however, the damage was mitigated by the foliar application of copper nanoparticles, which increased performance and improved the Na+/K+ ratio. The content of Cu increased in the tissues of tomato plants under salinity with the application of Cu nanoparticles, which increased the phenols (16%) in the leaves and the content of vitamin C (80%), glutathione (GSH) (81%), and phenols (7.8%) in the fruit compared with the control. Similarly, the enzyme activity of phenylalanine ammonia lyase (PAL), ascorbate peroxidase (APX), glutathione peroxidase (GPX), superoxide dismutase (SOD), and catalase (CAT) increased in leaf tissue by 104%, 140%, 26%, 8%, and 93%, respectively. Foliar spraying of copper nanoparticles on tomatoes under salinity appears to induce stress tolerance to salinity by stimulating the plant’s antioxidant mechanisms.

Abstract:Biostimulants have rapidly and widely been adopted as growth enhancers and stress protectants in agriculture, however, due to the complex nature of these products, their mechanism of action is not clearly understood. By using two algal based commercial biostimulants in combination with the Solanum lycopersicum cv. MicroTom model system, we assessed how the modulation of nitrogen metabolites and potassium levels could contribute to mediate physiological mechanisms that are known to occur in response to salt/and or osmotic stress. Here we provide evidence that the reshaping of amino acid metabolism can work as a functional effector, coordinating ion homeostasis, osmotic adjustment and scavenging of reactive oxygen species under increased osmotic stress in MicroTom plant cells. The Superfifty biostimulant is responsible for a minor amino acid rich-phenotype and could represent an interesting instrument to untangle nitrogen metabolism dynamics in response to salinity and/or osmotic stress.

Abstract:Salt stress can seriously reduce crop yield and quality, causing huge economic losses. Identification and functional of salt stress regulator could provide us a way to improve plants tolerance to salt stress. The HD-ZIP II subfamily is plant-specific transcription factors, plays a crucial role in plant development and abiotic stress. In this study, we analyzed the expression profiling of all HD-ZIP II members response to salt, drought, high temperature and cold stress using publicly accessible transcriptome data, and identified a novel salt stress negative regulator SlABIG1 in tomato. Under salt stress, knockout of SlABIG1 significantly improved the salt tolerance of tomato, with higher chlorophyll content and photosynthetic capacity, root dry weight; decreased the accumulation of ROS and MDA. Collectively, our results revealed that SlABIG1 plays a key role in tomato tolerance to salinity stress and provide a candidate novel gene to create salt-tolerant tomato varieties in future.
The differential expressions of aquaporins underline the diverse strategies of cucumber and tomato against salinity and zinc stress

Abstract:Salinity and excess zinc are two main problems that have limited agriculture in recent years. Aquaporins are crucial in regulating the passage of water and solutes through cells and may be essential for mitigating abiotic stresses. In the present study, the adaptive response to moderate salinity (60 mM NaCl) and excess Zn (1 mM ZnSO4) were compared alone and in combination in Cucumis sativus L. and Solanum lycopersicum L. Water relations, gas exchange and the differential expression of all aquaporins were analysed. The results showed that cucumber plants under salinity maintained the internal movement of water through osmotic adjustment and the overexpression of specific PIPs aquaporins, following a "conservation strategy". As tomato has a high tolerance to salinity, the physiological parameters and the expression of most aquaporins remained unchanged. ZnSO4 was shown to be stressful for both plant species. While cucumber upregulated 7 aquaporin isoforms, the expression of aquaporins increased in a generalized manner in tomato. Despite the differences, water relations and transpiration were adjusted in both plants, allowing the RWC in the shoot to be maintained. The aquaporin regulation in cucumber plants facing NaCl+ZnSO4 stress was similar in the two treatments containing NaCl, evidencing the predominance of salt in stress. However, in tomato, the induced expression of specific isoforms to deal with the combined stress differed from independent stresses. The results clarify the key role of aquaporin regulation in facing abiotic stresses and their possible use as markers of tolerance to salinity and heavy metals in plants.

Recovering Tomato Landraces to Simultaneously Improve Fruit Yield and Nutritional Quality Against Salt Stress

Abstract:Salt stress generally induces important negative effects on tomato (Solanum lycopersicum) productivity but it may also cause a positive effect improving fruit quality, one of the greatest challenges in nowadays agriculture. Because of the genetic erosion of this horticultural species, the recovery of locally adapted landraces could play a very important role in avoiding, at least partially, production losses and simultaneously improving fruit quality. Two tomato landraces endemic of the Spanish Southeast area, characterized by the harsh climatic conditions of the Mediterranean basin, have been selected: Negro Yeste (NY) characterized by its dark-red colored fruits and Verdal (V), which fruits did not achieve the characteristic red color at ripening. Here the agronomic, physiological, and metabolic responses of these landraces were compared with the reference tomato commercial cv. Moneymaker (MM), in plants grown without salt (control) and with salt stress (100 mM NaCl) for 70 days. The higher salt tolerance of both landraces was mainly reflected in the fruit number, as NY only reduced the fruit number in salt stress by 20% whereas in MM it was reduced till 43%, and in V the fruit number even showed an increase of 33% with salt stress. An important fruit quality parameter is soluble solids content, which increases induced by salinity were significantly higher in both landraces (60 and 78% in NY and V, respectively) compared with MM (34%). Although both landraces showed a similar response in relation to the high chlorophyll accumulation detected in their fruits, the fruit metabolic profiles were very different. Increased carotenoids levels were found in NY fruits, especially lycopene in ripe fruit, and this characteristic was observed in both control and salt stress. Contrarily, the carotenoid biosynthesis pathway was disrupted in V ripe fruits, but other metabolites, such as Ca2+, mannose, formate, and glutamate were accumulated. These results highlight the potential of tomato landraces to improve nutritional fruit quality and maintain fruit yield stability under salt stress.

Abstract:Abstract Soil salinity affects the plant growth and productivity detrimentally, but Solanum chilense , a wild relative of cultivated tomato ( Solanum lycopersicum L.), is known to have exceptional salt tolerance. It has precise adaptations against direct exposure to salt stress conditions. Hence, a better understanding of the mechanism to salinity stress tolerance by S. chilense can be accomplished by comprehensive gene expression studies. In this study 1-month-old seedlings of S. chilense and S. lycopersicum were subjected to salinity stress through application of sodium chloride (NaCl) solution. Through RNA-sequencing here we have studied the differences in the gene expression patterns. A total of 386 million clean reads were obtained through RNAseq analysis using the Illumina HiSeq 2000 platform. Clean reads were further assembled de novo into a transcriptome dataset comprising of 514,747 unigenes with N50 length of 578 bp and were further aligned to the public databases. Genebank non-redundant (Nr), Viridiplantae, Gene Ontology (GO), KOG, and KEGG databases classification suggested enrichment of these unigenes in 30 GO categories, 26 KOG, and 127 pathways, respectively. Out of 265,158 genes that were differentially expressed in response to salt treatment, 134,566 and 130,592 genes were significantly up and down-regulated, respectively. Upon placing all the differentially expressed genes (DEG) in known signaling pathways, it was evident that most of the DEGs involved in cytokinin, ethylene, auxin, abscisic acid, gibberellin, and Ca 2+ mediated signaling pathways were up-regulated. Furthermore, GO enrichment analysis was performed using REVIGO and up-regulation of multiple genes involved in various biological processes in chilense under salinity were identified. Through pathway analysis of DEGs, “Wnt signaling pathway” was identified as a novel pathway for the response to the salinity stress. Moreover, key genes for salinity tolerance, such as genes encoding proline and arginine metabolism, ROS scavenging system, transporters, osmotic regulation, defense and stress response, homeostasis and transcription factors were not only salt-induced but also showed higher expression in S. chilense as compared to S. lycopersicum . Thus indicating that these genes may have an important role in salinity tolerance in S. chilense . Overall, the results of this study improve our understanding on possible molecular mechanisms underlying salt tolerance in plants in general and tomato in particular.

Abstract:Polyamines (PAs) are key signaling molecules involved in plant growth and stress acclimation processes. This work investigated the effect of spermidine, spermine, and putrescine (alone and in a mixture) in tomato plants using a combined metabolomics and lipidomics approach. The experiments were carried out under non-stress and 100 mM NaCl salinity conditions. Shoot and root biomass, as well as SPAD values, were increased by the application of exogenous PAs but with differences across treatments. Similarly, root length density (F: 34, p < 0.001), average root diameter (F: 14, p < 0.001), and very fine roots (0.0-0.5 mm) increased in PA-treated plants, compared to control. Metabolomics and lipidomics indicated that, despite being salinity the hierarchically prevalent factor, the different PA treatments imposed distinct remodeling at the molecular level. Plants treated with putrescine showed the broader modulation of metabolite profile, whereas spermidine and spermine induced a comparatively milder effect. The pathway analysis from differential metabolites indicated a broad and multi-level intricate modulation of several signaling molecules together with stress-related compounds like flavonoids and alkaloids. Concerning signaling processes, the complex crosstalk between phytohormones (mainly abscisic acid, cytokinins, the ethylene precursor, and jasmonates), and the membrane lipids signaling cascade (in particular, sphingolipids as well as ceramides and other glycerophospholipids), was involved in such complex response of tomato to PAs. Interestingly, PA-specific processes could be observed, with peculiar responses under either control or salinity conditions.
The Combination of Trichoderma harzianum and Chemical Fertilization Leads to the Deregulation of Phytohormone Networking, Preventing the Adaptive Responses of Tomato Plants to Salt Stress

Abstract:Plants have evolved effective mechanisms to avoid or reduce the potential damage caused by abiotic stresses. In addition to biocontrol abilities, Trichoderma genus fungi promote growth and alleviate the adverse effects caused by saline stress in plants. Morphological, physiological, and molecular changes were analyzed in salt-stressed tomato plants grown under greenhouse conditions in order to investigate the effects of chemical and biological fertilizations. The application of Trichoderma harzianum T34 to tomato seeds had very positive effects on plant growth, independently of chemical fertilization. The application of salt stress significantly changed the parameters related to growth and gas-exchange rates in tomato plants subject to chemical fertilization. However, the gas-exchange parameters were not affected in unfertilized plants under the same moderate saline stress. The combined application of T34 and salt significantly reduced the fresh and dry weights of NPK-fertilized plants, while the opposite effects were detected when no chemical fertilization was applied. Decaying symptoms were observed in salt-stressed and chemically fertilized plants previously treated with T34. This damaged phenotype was linked to significantly higher intercellular CO2 and slight increases in stomatal conductance and transpiration, and to the deregulation of phytohormone networking in terms of significantly lower expression levels of the salt overlay sensitivity 1 (SOS1) gene, and the genes involved in signaling abscisic acid-, ethylene-, and salicylic acid-dependent pathways and ROS production, in comparison with those observed in salt-challenged NPK-fertilized plants.
Transcriptome Mechanisms of Tomato Seedlings Induced by Low-Red to Far-Red Light Ratio under Calcium Nitrate Stress

Abstract:In recent times, the excessive accumulation of nitrate has been one of the main reasons for the secondary salinization of greenhouse soils. Light plays a key role in a plant's growth, development, and response to stress. A low-red to far-red (R:FR) light ratio could enhance plant salinity tolerance, but the mechanism at a molecular level is unclear. Thus, we analyzed the transcriptome responses of tomato seedlings to calcium nitrate stress under either a low R:FR ratio (0.7) or normal light conditions. Under calcium nitrate stress, a low R:FR ratio enhanced both the antioxidant defense system and the rapid physiological accumulation of proline in tomato leaves, which promoted plant adaptability. Using weighted gene co-expression network analysis (WGCNA), three modules including 368 differentially expressed genes (DEGs) were determined to be significantly associated with these plant traits. Functional annotations showed that the responses of these DEGs to a low R:FR ratio under excessive nitrate stress were enriched in the areas of hormone signal transduction, amino acid biosynthesis, sulfide metabolism, and oxidoreductase activity. Furthermore, we identified important novel hub genes encoding certain proteins, including FBNs, SULTRs, and GATA-like transcription factor, which may play a vital role in low R:FR light-induced salt responses. These findings offer a new perspective on the mechanisms and environmental implications behind low R:FR ratio light-modulated tomato saline tolerance.

Abstract:Plants irrigated with saline solutions undergo osmotic and oxidative stresses, which affect their growth, photosynthetic activity and yield. Therefore, the use of saline water for irrigation, in addition to the increasing soil salinity, is one of the major threats to crop productivity worldwide. Plant tolerance to stressful conditions can be improved using different strategies, i.e., seed priming and acclimation, which elicit morphological and biochemical responses to overcome stress. In this work, we evaluated the combined effect of priming and acclimation on salt stress response of a tomato cultivar (Solanum lycopersicum L.), very sensitive to salinity. Chemical priming of seeds was performed by treating seeds with polyamines (PAs): 2.5 mM putrescine (PUT), 2.5 mM spermine (SPM) and 2.5 mM spermidine (SPD). Germinated seeds of primed and non-primed (controls) were sown in non-saline soil. The acclimation consisted of irrigating the seedlings for 2 weeks with tap water, followed by irrigation with saline and non-saline water for 4 weeks. At the end of the growth period, morphological, physiological and biochemical parameters were determined. The positive effects of combined treatments were evident, when primed plants were compared to non-primed, grown under the same conditions. Priming with PAs improved tolerance to salt stress, reduced the negative effects of salinity on growth, improved membrane integrity, and increased photosynthetic pigments, proline and enzymatic and non-enzymatic antioxidant responses in all salt-exposed plants. These results may open new perspectives and strategies to increase tolerance to salt stress in sensitive species, such as tomato.
