Abstract:Poly(dimethylsiloxane) (PDMS) is usually considered as a dielectric material and the PDMS microchannel wall can be treated as an electrically insulated boundary in an applied electric field. However, in certain layouts of microfluidic networks, electrical leakage through the PDMS microfluidic channel walls may not be negligible, which must be carefully considered in the microfluidic circuit design. In this paper, we report on the experimental characterization of the electrical leakage current through PDMS microfluidic channel walls of different configurations. Our numerical and experimental studies indicate that for tens of microns thick PDMS channel walls, electrical leakage through the PDMS wall could significantly alter the electrical field in the main channel. We further show that we can use the electrical leakage through the PDMS microfluidic channel wall to control the electrolyte flow inside the microfluidic channel and manipulate the particle motion inside the microfluidic channel. More specifically, we can trap individual particles at different locations inside the microfluidic channel by balancing the electroosmotic flow and the electrophoretic migration of the particle.

Abstract:Deterministic lateral displacement (DLD) is a microfluidic passive method for bioparticle separation and isolation, based on particle size and shape with high sample volume and low latency. Despite widespread application and research, experts report various fundamental, practical, and commercial challenges faced in the field of DLD represented in the literature. One of the fundamental challenges is the fluid's flow behaviour and the fluid–structure interaction inside the DLD array. To address this problem, this paper investigates the modelling of a three-dimensional DLD array constructed by considering lateral displacements and wall boundaries. Nine different geometric shapes of the symmetrical pillars are reported in the literature opted for the study which are examined with different velocities to analyse how pillar shape impacts the flow patterns inside the DLD array. Also, an investigation of the two-way interaction between the fluid and the pillar structure is carried out. The response of the fluid due to the interaction with the pillar and the pillar response due to the interaction with the fluids is reported. Additionally, the realistic influence of the fluid flow and the surrounding pillars on the middle pillar in the array are examined in 3D simulations. The simulation was carried out using the finite element software COMSOL Multiphysics.

Abstract:Microfluidic deivices have increasing application as analytical system s, biomedical devices, tools for chemistry and biochemistry, and systems for fun damental research. Conventional methods of fabricating microfluidic devices have focused on etching in glass and silicon. Fabrication of microfluidic devices in poly(dimethylsiloxane)(PDMS) by soft lithography provides faster, less expensiv e routes than these conventional methods. These soft-lithographic methods are b ased on rapid prototyping and replica molding. At the same time, they are also m ore accessible for chemists and biologists to work under benchtop conditions bec ause devices do not need to be fabricated in a cleanroom using soft lithography. This paper describes devices fabricated in PDMS for separations and patterning of biological material.
Effects of the particle deformability on the critical separation diameter in the deterministic lateral displacement device

Abstract:Deterministic lateral displacement (DLD) technology is a newly developed method which can separate microscale and nanoscale particles continuously and efficiently. In this paper, a direct numerical simulation method (i.e. a fictitious domain method) is used to simulate the motion of an elastic particle (modelled as homogeneously elastic body) in the DLD device. The effects of the particle deformability on the critical separation diameter are investigated. Our results indicate that there exists a critical deformability, below which the critical diameter decreases with increasing deformability, whereas beyond which the critical diameter increases with increasing deformability. The reasons are discussed via the consideration of the effects of the particle deformation and the lubrication force on the lateral position of the particle centre point. In addition, our results show that the increase in the gap distance between adjacent posts in both directions or in the longitudinal direction alone leads to the increase in the critical particle size with respect to the gap size, which can be explained by the lateral position of the separation streamline of the undisturbed flow.

Abstract:In order to solve the problems of the accuracy of numerical simulation of deterministic lateral displacement(DLD) device and study of particle separation,a direct numerical simulation method(i.e.fictitious domain method) was used to simulate the motion of rigid particle in DLD.After the analyzing the effects of lateral boundary condition,the relationship of internal flow field tilt angle and boundary conditions were established.The periodic boundary conditions were presented to simulate DLD with a huge number of lateral arrays.The numerical method and boundary condition were evaluated on a characteristic of the internal flow field and the separation diameter.The critical diameter of a rigid particle in DLD was tested.The experimental results show that for the practical DLD devices with a huge number of lateral arrays,the periodic boundary condition with fewer arrays can be adopted to reduce the computational cost.

Abstract:We proposed an innovative method to achieve dynamic control of particle separation by employing viscoelastic fluids in deterministic lateral displacement (DLD) arrays. The effects of shear-thinning and elasticity of working fluids on the critical separation size in DLD arrays are investigated. It is observed that each effect can lead to the variation of the critical separation size by approximately 40%. Since the elasticity strength of the fluid is related to the shear rate, the dynamic control can for the first time be easily realized through tuning the flow rate in microchannels.

Abstract:This paper reports a microfluidic reactor for synthesizing micro- and nano-droplets using fluid shearing instability of immiscible flow in microchannels. Three factors, flow rate between the two inputs, spout geometry and surface tension are numerically studied. Based on the simulation, a device with the optimal parameters was fabricated and tested. Both computational fluid dynamics (CFD) simulation results and experimental results suggested that this technique is capable of conveniently controlling the droplets sizes from hundreds of micrometers down to several micrometers or even nanometers. This pilot research offers a proof-of-concept demonstration using instabilities in the microchannels to synthesize large quantity of microdroplets. Although the microdroplets in this study were formed by DI water containing Tween20 and silicon oil, the design with modification could be applied for synthesis of other types of micro- and nano-droplets.

Abstract:Deterministic lateral displacement (DLD) is a microfluidic method for accurately separating particles by size or deformability. Recent efforts to operate DLD devices in the inertial, rather than in the Stokes flow regime, have been hindered by a loss of separation efficiency and difficulty predicting the separation behavior. One factor contributing to these problems is the onset of inertia-induced flow anisotropy where the average flow direction does not align with the direction of the pressure gradient in the device. We use the lattice-Boltzmann method to simulate two-dimensional flow through a rotated-square DLD geometry with circular pillars at a Reynolds number up to 100 for different gap sizes and rotation angles. We find that anisotropy in this geometry is a nonmonotonous function of Reynolds number and can be positive or negative. This finding is in contradiction to the naive expectation that inertia would always drive flow along the principal direction of the pillar array. Anisotropy tends to increase in magnitude with gap size and rotation angle. By analyzing the traction distribution along the pillar surface, we explain how the change of the flow field upon increasing inertia leads to the observed trends of anisotropy. Abstract:To non-invasively monitor personal biological and environmental samples in Internet of Things (IoT)-based wearable microfluidic sensing applications, the particle size could be key to sensing, which emphasizes the need for particle size fractionation. Deterministic lateral displacement (DLD) is a microfluidic structure that has shown great potential for the size fractionation of micro- and nano-sized particles. This paper introduces a new externally balanced multi-section cascade DLD approach with a section-scaling technique aimed at expanding the dynamic range of particle size separation. To analyze the design tradeoffs of this new approach, a robust model that also accounts for practical fabrication limits is presented, enabling designers to visualize compromises between the overall device size and the achievement of various performance goals. Furthermore, results show that a wide variety of size fractionation ranges and size separation resolutions can be achieved by cascading multiple sections of an increasingly smaller gap size and critical separation dimension. Model results based on DLD theoretical equations are first presented, followed by model results that apply the scaling restrictions associated with the second order of effects, including practical fabrication limits, the gap/pillar size ratio, and pillar shape.

Abstract:Microfluidic devices with open lattice structures, equivalent to a type of porous media, allow for the manipulation of fluid transport processes while having distinct structural, mechanical, and thermal properties. However, a fundamental understanding of the design principles for the solid structure in order to achieve consistent and desired flow patterns remains a challenge, preventing its further development and wider applications. Here, through quantitative and mechanistic analyses of the behavior of multi-phase phenomena that involve gas-liquid-solid interfaces, we present a design framework for microfluidic devices containing porous architectures (referred to as poroFluidics) for deterministic control of multi-phase fluid transport processes. We show that the essential properties of the fluids and solid, including viscosity, interfacial tension, wettability, as well as solid manufacture resolution, can be incorporated into the design to achieve consistent flow in porous media, where the desired spatial and temporal fluid invasion sequence can be realized. Experiments and numerical simulations reveal that different preferential flow pathways can be controlled by solid geometry, flow conditions, or fluid/solid properties. Our design framework enables precise, multifunctional, and dynamic control of multi-phase transport within engineered porous media.

Abstract:In order to fulfill flow control of liquid sample caused by the microfluidic biochip, realize the miniaturization and integration of the microfluidic biochip, this paper proposed a research on the micro-liquid flow sensor. Based on the principle of thermal distribution, the micro flow sensor fabricated in this paper was composed of a sensor chip and a micro-channel chip. The sensor chip consisted of a micro heater made by the Pt metallic film and 10 micro ambient sensors, two of which were used to detect the working temperature and the other eight were used to measure flow; the microchannel chip was made of PDMS by casting. ANSYS was employed to simulate and optimize the structural design of the micro-liquid flow sensor.

In contrast to the typical approach that alters the angle between the pillar array and fluid streamlines by modifying the geometrical parameters, this study introduces a novel perspective that focuses on changing the direction of the streamlines. The proposed technique offers a tunable DLD chip featuring a straightforward design that allows for easy fabrication. This chip features one completely horizontal pillar array with two bypass channels on the top and bottom of the DLD chamber. The width of these bypass channels changes linearly from their inlet to their outlet. Two design configurations are suggested for this chip, characterized by either parallel or unparallel slopes of the bypass channels. This chip is capable of generating a wide range of Dc values by manipulating two distinct control parameters. The first control parameter involves adjusting the flow rates in the two bypass channels. The second control parameter entails controlling the slopes of these bypass channels. Both of these parameters influence the direction of particle-carrying streamlines resulting in a change in the path-line of the particles. By changing the angle of streamlines with pillar array, the Dc can be tuned. Prior to determining the Dc for each case, an initial estimation was made using a Python script that utilized the streamline coordinates. Subsequently, through FEM modeling of the particle trajectories, precise Dc values were ascertained and compared with the estimated values, revealing minimal disparities. By adjusting the flow rate and slope of the bypass channels, maximum Dc ranges of 4-10 μm and 8-13 μm can be achieved, respectively. This innovative chip enables the attainment of Dc values spanning from 0.5 to 14 μm.

Abstract:Over the last decade, researchers have developed a variety of new analytical and clinical diagnostic devices. These devices are predominantly based on microfluidic technologies, where biological samples can be processed and manipulated for the collection and detection of important biomolecules. Polydimethylsiloxane (PDMS) is the most commonly used material in the fabrication of these microfluidic devices. However, it has a hydrophobic nature (contact angle with water of 110°), leading to poor wetting behavior and issues related to the mixing of fluids, difficulties in obtaining uniform coatings, and reduced efficiency in processes such as plasma separation and molecule detection (protein adsorption). This work aimed to consider the fabrication aspects of PDMS microfluidic devices for biological applications, such as surface modification methods. Therefore, we studied and characterized two methods for obtaining hydrophilic PDMS surfaces: surface modification by bulk mixture and the surface immersion method. To modify the PDMS surface properties, three different surfactants were used in both methods (Pluronic® F127, polyethylene glycol (PEG), and polyethylene oxide (PEO)) at different percentages. Water contact angle (WCA) measurements were performed to evaluate the surface wettability. Additionally, capillary flow studies were performed with microchannel molds, which were produced using stereolithography combined with PDMS double casting and replica molding procedures. A PDMS microfluidic device for blood plasma separation was also fabricated by soft lithography with PDMS modified by PEO surfactant at 2.5% (v/v), which proved to be the best method for making the PDMS hydrophilic, as the WCA was lower than 50° for several days without compromising the PDMS's optical properties. Thus, this study indicates that PDMS surface modification shows great potential for enhancing blood plasma separation efficiency in microfluidic devices, as it facilitates fluid flow, reduces cell aggregations and the trapping of air bubbles, and achieves higher levels of sample purity.

Accelerated Particle Separation in a DLD Device at Re > 1 Investigated by Means of µPIV

Abstract:A pressure resistant and optically accessible deterministic lateral displacement (DLD) device was designed and microfabricated from silicon and glass for high-throughput fractionation of particles between 3.0 and 7.0 µm comprising array segments of varying tilt angles with a post size of 5 µm. The design was supported by computational fluid dynamic (CFD) simulations using OpenFOAM software. Simulations indicated a change in the critical particle diameter for fractionation at higher Reynolds numbers. This was experimentally confirmed by microparticle image velocimetry (µPIV) in the DLD device with tracer particles of 0.86 µm. At Reynolds numbers above 8 an asymmetric flow field pattern between posts could be observed. Furthermore, the new DLD device allowed successful fractionation of 2 µm and 5 µm fluorescent polystyrene particles at Re = 0.5-25.

Abstract:This paper presents a highly reliable macro to micro domain interconnection technology for microfluidic applications using Polydimethylsiloxane (PDMS) casting techniques. Characteristic to the interconnectors are long flanges fabricated in the PDMS film; therefore the contact area between PDMS and tubes is considerably increased compared to other interconnection technologies. Thus, both glass capillaries and Polytetrafluoroethylene (PTFE) tubes can be held in position very reliably and rigidly. To test the reliability of the interconnectors, PTFE tubes were successfully connected to microfluidic chips without the aid of any liquid adhesives. Both leakage and pull-out tests demonstrated the functionality and reliability of the PDMS interconnectors; no leakage was detected under a working pressure up to 400 kPa. A pull-out test yielded a pull-out force of 22.45 N. Furthermore, once a casting mould is fabricated, it can be re-used as a template repeatedly achieving a low cost technology and making it suitable for batch production.

Abstract:Droplet-based microfluidics exhibit numerous benefits leading to relevant innovations and many applications in various fields. The precise handling of droplets in capillaries, including droplet formation, manipulation, and separation, is essential for successful operation. Only a few reports are known concerning the separation of segmented flows, particularly the continuous separation of droplets, which is of high interest regarding the control of biochemical and chemical reactions or other applications where the contact time of the involved phases is crucial. Here, the separation must be flexible and adjusted to different flow parameters, such as the surface tension, the volumetric flow rates, and their ratios. This contribution presents two novel open-source approaches based on additive manufacturing and mechanical deforming for continuous liquid–liquid separation under various flow conditions. The Laplace pressure is the driving force for the separation, which is adjusted to the flow conditions by adapting the distance of pinning points provided by the design of the devices. Details of the device design and experimental setup are shown along with limitations to promote further development and to increase availability for researchers. With the right parameters, sophisticated separations can be realized by inexpensive laboratory equipment and simple control of them. It was found that the distance between the pinning points needs to enlarged for increasing volumetric flow rates and reduced for higher viscosities of the continuous phase respectively higher amounts of the dispersed phase. The open source approach of this article expands the exploration space in addition to commercially available phase separators only available to a selected group of people.

Abstract:Particle sorting methods in microfluidic platforms are gaining momentum for various biomedical applications. Bioparticles are found in different shapes and sizes. However, conventional separation techniques are mainly designed for separation of spherical particles. Thus, there is a need to develop new methods for effective separation of spherical and non-spherical bioparticles for various applications. Deterministic lateral displacement (DLD) microfluidic methods have become popular for high separation resolution, simplicity, and predictability. However, shape sorting in the DLD separation methods is not well researched. Recently, we explored this area and found that pillar shapes in DLD significantly affect bioparticle separation. In this work, we designed a group of different pillar shapes with protrusions and groove structures with the hypothesis that pillar protrusions will induce particle rotation while pillar grooves will confine the particle rotational movement in a directed path for effective separation in a DLD pillar array. Using combinations of protrusions and grooves, 3-dimensional spherical particles, 2-dimensional planar disc-shaped red blood cells and 1-dimensional rod-shaped bacteria were separated and two interesting phenomena were observed. Firstly, the arrangement of pillar protrusions and grooves induces inertial movements, enhancing the separation of spherical particles. Secondly, non-spherical particles experience dominant rotational movements due to the protrusions and grooves which help in changing their orientations. This gives an opportunity to perform efficient separation based on the desired orientation (the longest dimension of the particles) by restricting or containing their movement within a specific DLD path.
A General Approach for Fluid Patterning and Application in Fabricating Microdevices.

Abstract:Engineering the fluid interface such as the gasliquid interface is of great significance for solvent processing applications including functional material assembly, inkjet printing, and high-performance device fabrication. However, precisely controlling the fluid interface remains a great challenge owing to its flexibility and fluidity. Here, a general method to manipulate the fluid interface for fluid patterning using micropillars in the microchannel is reported. The principle of fluid patterning for immiscible fluid pairs including air, water, and oils is proposed. This understanding enables the preparation of programmable multiphase fluid patterns and assembly of multilayer functional materials to fabricate micro-optoelectronic devices. This general strategy of fluid patterning provides a promising platform to study the fundamental processes occurring on the fluid interface, and benefits applications in many subjects, such as microfluidics, microbiology, chemical analysis and detection, material synthesis and assembly, device fabrication, etc.
