Abstract:Direct-flow filtration is a common technique for filtering impurities from a fluid using a porous-walled channel or a pipe, one end of which is closed off with a cap. Pure fluid flows out of the porous walls, while impurities are left in the channel. Such systems are composed of a series of individual porous channels or pipes stacked in close proximity. We develop a mathematical model for the flow in a 2D filtration channel and a 3D pipe, with a capped end, to describe the behaviour within a direct-flow device. We study the axial dependence of the transmembrane pressure (TMP) across the membrane walls on the imposed flux, wall permeability and the proximity of the neighbouring fibres. The mathematical models derived are used to predict the operating regimes of the device that maximize the spatial uniformity in the TMP and thus optimize the use of the entire membrane area. We show how a large portion of the available membrane area is not used when the fibres are packed too closely together, with the majority of the filtration behaviour being localized near to the impermeable capped end; this leads to inefficient filtration. We quantify the device performance by examining the uniformity of the filtration across the length of the device and the output of the filtered fluid for a given operating pressure.

Abstract:Size-based particle filtration has become indispensable in numerous biomedical and environmental applications. In this study, bioinspired by the filter-feeding mechanism (lobe filtration) of manta rays, we designed a U-shaped biomimetic gill rake filter that combined lobe filtration and Dean flow to filter monodisperse suspensions, bi-disperse suspensions and yeast cells. Compared with other equipment using the inertial focusing technology, our equipment can perform high-throughput (up to 8 mL min-1) and high-efficiency filtration of particles (maximum filtration efficiencies of 96.08% and 97.14% for 10 and 15 mu m monodisperse suspensions at the optimum flow rate of 6 mL min-1). The complex velocity field of the micro-fluidic flow within the filter is numerically simulated, and in combination with experiments, a threshold for the flow rate is identified. When the inlet flow rate exceeds the threshold value, the efficiency of particle filtration is increased rapidly. Afterwards, by analysing the filtration mechanism, we develop three novel filtration processes. The equilibrium positions of the particles and yeast cells in the main channel are close to the outer wall at high flow rate, which diminishes the likelihood of particles and yeast cells entering the side channel. This configuration establishes a self-cleaning mechanism, ensuring prolonged and efficient operation of the filter with high-throughput processing. Furthermore, the influence of the filter lobe angle and channel width on the filtration efficiency and outlet flow rate ratio are explored, and an optimisation plan is prepared. Size-based particle filtration has become indispensable in numerous biomedical and environmental applications.
Flow and fouling in membrane filters: effects of membrane morphology

Abstract:Membrane filters are used extensively in microfiltration applications. The type of membrane used can vary widely depending on the particular application, but broadly speaking the requirements are to achieve fine control of separation, with low power consumption. The solution to this challenge might seem obvious: select the membrane with the largest pore size and void fraction consistent with the separation requirements. However, membrane fouling (an inevitable consequence of successful filtration) is a complicated process, which depends on many parameters other than membrane-pore size and void fraction; and which itself greatly affects the filtration process and membrane functionality. In this work we formulate mathematical models that can (i) account for the membrane internal morphology (internal structure, pore size and shape, etc.); (ii) describe fouling of membranes with specific morphology; and (iii) make some predictions as to what type of membrane morphology might offer optimum filtration performance.

Nanostructured micrometric-pore membranes for nanofiltration: Micrometric geometry may optimize performance, energy efficiency and operational lifetime

Abstract:Media and membranes composed of micrometric-diameter pores are well known in academia and industry to be capable of efficacious nanofiltration of fluids once the pore inner surfaces are coated with nanostructures. Given the large mismatch between the two very different scales of these hybrid systems, it could be expected that trapping of nanoimpurities would almost entirely depend on the characteristics of the nanostructures. However, we show here that the micrometric-scale nominal geometry does have noticeable impact on the nanofiltration performance, on its evolution with time, and on the energy spent per trapped impurity. For that, we apply stochastic calculations customized to combine the cumulative probabilities of wall-impurity attraction and binding, supplemented with continuity equations as the fluid flows; this allows tracking the nanofiltration without a many-particle simulation, prohibitive in such multi-scale system. We focus on the influence of the micrometric nominal geometry over filtration features like logarithmic removal value LRV, operational lifetimes, energy balances, or spatial profiles of the trapped-impurity layer in the pore inner walls. Our results identify some pore geometries (e.g., decreasing-conical and sinusoidal-corrugated) with about 4-fold larger initial nanofiltering performance and operational lifetime than simple cylinders of the same average diameter. The optimal geometry is also shown to depend on the LRV value acceptable for each specific application.

Optimizing Flux Capacity of Dead-end Filtration Membranes by Controlling Flow with Pulse Width Modulated Periodic Backflush

Abstract:Abstract Standard dead-end sample filtration is used to improve sample purity, but is limited as particle build-up fouls the filter, leading to reduced recovery. The fouling layer can be periodically cleared with backflush algorithms applied through a customized fluidic actuator using variable duty cycles, significantly improving particulate recovery percentage. We show a Pulse Width Modulation (PWM) process can periodically backflush the filter membrane to repeatedly interrupt cake formation and reintegrate the fouling layer into the sample, improving net permeate flux per unit volume of sample by partially restoring filter flux capacity. PWM flow for 2.19 um (targeted) and 7.32 um (untargeted) polystyrene microbeads produced 18-fold higher permeate concentration, higher recovery up to 68.5%, and an 8-fold enrichment increase, compared to a uniform flow. As the duty cycle approaches 50%, the recovery percentage monotonically increases after a critical threshold. Further, we developed and validated a mathematical model to determine that fast, small-volume backflush pulses near 50% duty cycle yield higher recovery by decreasing fouling associated with the cake layer. Optimized PWM flow was then used to purify custom particles for immune activation, achieving 3-fold higher recovery percentage and providing a new route to improve purification yields for diagnostic and cellular applications.

Abstract:The local transmembrane pressure (TMP) distribution plays a crucial role in the performance of hollow-fiber microfiltration and ultrafiltration membrane devices. A direct-flow filter comprises an array of hollow fibers encased within a single module. The TMP in each hollow fiber is dependent on the spacing between neighboring fibers, and evolves with time due to pore clogging by contaminants (fouling). We consider an idealized set-up in which the fibers are undeformable and equally spaced within the device, and study the impact of the pore-blocking phenomena on the TMP during the filtration process. The model is used to evaluate the optimum inter-fiber spacing that maximizes the fluid processed either after a prescribed time or before the filter blocks and its dependence on the membrane permeability and the fouling rate. We show that significant improvements can be made on the operating efficiency of a direct-flow device through careful choice in the fiber spacing during fabrication.

Abstract:Pleated membrane filters are widely used in many applications, and offer significantly better surface area to volume ratios than equal-area unpleated membrane filters. However, their filtration characteristics are markedly inferior to those of equivalent unpleated membrane filters in dead-end filtration. While several hypotheses have been advanced for this, one possibility is that the flow field induced by the pleating leads to spatially non-uniform fouling of the filter, which in turn degrades performance. In this paper we investigate this hypothesis by developing a simplified model for the flow and fouling within a pleated membrane filter. Our model accounts for the pleated membrane geometry (which affects the flow), for porous support layers surrounding the membrane, and for two membrane fouling mechanisms: (i) adsorption of very small particles within membrane pores; and (ii) blocking of entire pores by large particles. We use asymptotic techniques based on the small pleat aspect ratio to solve the model, and we compare solutions to those for the closest-equivalent unpleated filter.

Abstract:<p>Fluid flow rate and total throughput are the major controlling parameters to calculate the required size of membrane-based filter equipment for manufacturing of pharmaceuticals. Filtration equipment comprises several resistances to flow such as pipes, connectors and the filter construction itself. The incorporated membrane is a main factor that determines the flow rate through the filter element. With larger membrane area, its resistance to flow declines and total filter throughput increases. Yet, additional hydrodynamic resistances in the filter device lead to lower flow rates than expected from the hydrodynamic resistances of the membrane. Especially the membrane pleats and the spacer material in-between can cause additional flow restrictions. This study investigates the causes of these pleat resistances in manufacturing scale filters. First, manufacturing scale filter flow rates were metered to quantify the effects of pleat geometry, filtration pressure and liquid viscosity on pleat resistance. Subsequent computed tomography (CT) scans of filter devices, performed under simulated operating conditions, reveal so far unreported pleat compressions that rise with increasing differential pressure up to 50% at 1.5 bar. In-plane flow resistances of the nonwoven spacer material between the pleats were determined. Finally, these pleat geometries, measured under pressure, and the in-plane nonwoven resistances were implemented into CFD simulations. These simulations show that reduced fluid flow in the nonwoven due to the compression of pleats can explain the previously observed hydrodynamic pleat resistances.</p>

Abstract:Configurations of crossflow filtration cells are important to determine membrane separation performance in laboratory studies. We systematically investigated the effects of crossflow filtration cell designs, including sealing approaches and back support configurations, on the determination of membrane water flux, salts rejection, and fouling behaviour. In particular, the definition of effective filtration area in the O-ring sealed cell could lead to an overestimated (defined as flow channel area) or underestimated (defined as O-ring enclosed area) membrane water flux compared to the benchmark data acquired from a silicone gasket sealed cell. In addition, it was found that the highly porous back support could yield higher water flux and salt rejection as well as better antifouling performance than that of the perforated plate support. The porous back support could significantly reduce the total resistance to water permeation thanks to the minimized average path length and increased hydraulic conductance for water transport. Moreover, the porous back support enabled better control of concentration polarization as it avoided local flux ‘hot spots’ that would occur above local perforations, and thus benefits membrane rejections. These findings provide important insights into the design of crossflow filtration cells for a more reliable and representative determination of membrane separation performance.

Abstract:A pleated membrane filter consists of a porous membrane layer, which is surrounded by two supporting layers, and the whole structure is pleated and placed into a cylindrical cartridge. Pleated membrane filters are used in a variety of industrial applications, since they offer more surface area to volume ratio that is not found in equivalent flat filters. In this work, we introduce a novel three-dimensional model of a pleated membrane filter that consists of an empty region, a pleated region, and a hollow region. The advection diffusion equation is used to model contaminant concentration in the membrane pores along with Darcy's law to model the flow within the membrane and support layers, while the Stokes equation is used for the flow in the empty region and the hollow region. We further use the key assumptions of our model based on small aspect ratios of the filter cartridge and the pleated membrane to simplify the governing equations, which can be easily solved by numerical methods. By performing these steps, we seek to discover an optimal pleat packing density to find the optimum filter performance, while not exceeding a threshold for the particle concentration at the filter outlet.

High throughput clogging free microfluidic particle filter by femtosecond laser micromachining

Abstract:In recent decades, driven by the needs of industry and medicine, researchers have been investigating how to remove carefully from the main flow microscopic particles or clusters of them. Among all the approaches proposed, crossflow filtration is one of the most attractive as it provides a non‐destructive, label‐free and in‐flow sorting method. In general, the separation performance shows capture and separation efficiencies ranging from 70% up to 100%. However, the maximum flow rate achievable (μL/min) is still orders of magnitude away from those suitable for clinical or industrial applications mainly due to the low stiffness of the materials typically used. In this work, we propose an innovative hydrodynamic‐crossflow hybrid filter geometry, buried in a fused silica substrate by means of the femtosecond laser irradiation followed by chemical etching technique. The material high stiffness combined with the accuracy of our manufacturing technique allows the 3D fabrication of non‐deformable channels with higher aspect ratio posts, while keeping the overall device dimensions compact. The filter performance has been validated through experiments with both Newtonian (water‐based solution of microbeads) and non‐Newtonian fluids (blood), achieving separation efficiencies of up to 94% and large particles recovery rates of 100%, even at very high flow rates (mL/h).

Abstract:Previous work has suggested that membrane patterning is a promising approach to fouling mitigation. In this systematic study, we describe performance metrics for the cross-flow filtration of colloidal suspensions through ultrafiltration membranes with lithographically patterned surfaces. The effects of particle size, cross-flow velocity, pattern size, and flow configuration on membrane performance are specifically considered. Using an unpatterned membrane as a reference, results show that the presence of surface patterns increases the critical flux associated with filtration of colloidal feed solutions. For the patterned membranes, the critical flux increases with particle size, cross-flow velocity, the angle between the feed flow and the pattern lines, and the pattern height. These experimental findings can be correlated with a back-transport mechanism such as shear-induced diffusion, which ultimately increases the threshold permeation flux associated with the onset of particle deposition.

Abstract:Membrane filtration processes are best known for their application in the water, oil, and gas sectors, but also in food production they play an eminent role. Filtration processes are known to suffer from a decrease in efficiency in time due to e.g., particle deposition, also known as fouling and pore blocking. Although these processes are not very well understood at a small scale, smart engineering approaches have been used to keep membrane processes running. Microfluidic devices have been increasingly applied to study membrane filtration processes and accommodate observation and understanding of the filtration process at different scales, from nanometer to millimeter and more. In combination with microscopes and high-speed imaging, microfluidic devices allow real time observation of filtration processes. In this review we will give a general introduction on microfluidic devices used to study membrane filtration behavior, followed by a discussion of how microfluidic devices can be used to understand current challenges. We will then discuss how increased knowledge on fundamental aspects of membrane filtration can help optimize existing processes, before wrapping up with an outlook on future prospects on the use of microfluidics within the field of membrane separation.

Optimizing virus filtration for continuous processing using serial filtration at high area ratio

Abstract:Compared to batch operation, continuous bioprocessing can offer numerous advantages, including increased productivity, improved process control, reduced footprint, and increased flexibility. However, integration of traditional batch operations into a connected process can be challenging. In contrast to batch operations run at constant pressure or high flux, virus filtration in continuous processes may be operated at very low flux. This change in operating conditions may reduce the viral retention performance of the filter which has inhibited adoption of truly continuous virus filtration. To overcome this limitation, a novel approach is described that utilizes serial virus filtration, with a high area ratio between first to second stage filters, to achieve virus retention targets. In this study, virus filters were operated continuously (except for planned process interruptions) for 200 h in a serial configuration at a first to second stage filter area ratio of 13:1 and at a first stage flux of 5 L/m2/h. While the minute virus of mice (MVM) retention performance of the first stage filter was about 4 log reduction value (LRV), there was no virus detected in the second stage filtrate, translating to an MVM LRV across the filtration train of ≥6.7. The second stage filter was the dominant flow resistance at the start of the run but, as it was protected from foulants by the first stage filter, it suffered minimal fouling and the life of the filter train was controlled by the first stage. A theoretical case study projected that continuous virus filtration using serial configuration at high area ratio would have about 30% longer filter changeout time, 14% higher productivity, and virus retention nearly six LRV greater than single stage operation. The findings of this research are expected to provide valuable insights into optimizing virus filtration in continuous bioprocessing.

Abstract:Sterile filters incorporating endotoxin adsorption function emerged recently to pretreat dialysate liquids fed to a hemodialysis filtration process. Their application significantly enhances the survival rate during dialysis treatment as they function as steril filters as well as an endotoxin adsorber. Little is known about the fluid flow distribution in such single use membrane modules. We report a detailed analysis of the local 3D flow field distribution in such membrane modules using magnetic resonance flow imaging. Next to pure water filtration representing the application case of endotoxin adsorption from an already pure liquid, we also used the module as a filtration device rejecting for instance colloidal silica. Such experiments performed in-situ allow the quantification of cake layer development and its concomitant redistribution of the flow field. Particularly novel is the quantification of the time evolution of local permeate flux distribution. These detailed insights of this study encourage the use of flow-MRI when designing and applying new membrane module configurations.

Abstract:The hollow fiber filter is the primary cell‐retention device used in high‐density perfusion cell culture and often used in an alternating tangential flow (ATF) configuration. The limited commercially available diaphragm pumps for ATF prevent utilization of vertical space when scaling beyond 500 L. Stacking hollow fiber filters coupled with viscous cell culture imposes vacuum pressure exceeding facility capabilities. Additionally, the longer filter assembly increases the hold‐up volume and exceeds the diaphragm pump's fluid exchange capacity. The conventional tangential flow filtration (TFF) configuration circumvents this issue by exchanging culture from the bioreactor and cell‐retention device in a unidirectional recirculation loop; however, the increased filter length when scaled up exacerbates the TFF's inherent issue with product retention from Starling flow. Stacking commercially available 20 cm TFF filters to make up the similar single‐module length TFF used for the platform 3 and 50 L perfusion process at 41.5 and 65 cm, respectively, attempts to reduce fouling caused by Starling flow. The permeate of a single‐module filter is partitioned into short independent segments through serially stacked filters, each harvested separately. By partitioning the permeate, the sieving coefficient increased for both 3 and 50 L scales. Reduction of Starling flow was confirmed with lower total hydraulic membrane resistance throughout the culture. This work demonstrates a method for increasing sieving coefficient and filter capacity by stacking TFF filters with independent permeate streams.
