Abstract:Background: Enzymatic reactions during tea growth, processing, storage and post-processing conduce to the diversity in the organoleptic qualities and health benefits of tea. While chemical component transformations by enzymatic catalysis have been extensively explored, the dynamic alteration of endogenous and exogenous enzyme activities throughout manufacture chains remain in a mist.Scope and approach: This paper discusses main enzymatic reactions involved in tea growth, processing, storage and post-processing, including the dominant enzymes of specific processes, the chemical transformation driven by enzymes and the alteration of endogenous and exogenous enzymes activities. The direct and indirect regulatory factors influencing enzyme activities and their possible applications in tea production practice are also explored. Key findings and conclusion: The endogenous enzyme system of tea plants regulates secondary metabolism and stress defense, contributing to the primary accumulation of volatile and non-volatile compounds in tea plants. Endogenous enzymes activities altered unceasingly and are exposed to more substrates due to cell rupture during the withering, spread, rolling and tumbling stages, establishing the intricate substance basis for distinct organoleptic characteristics. Microbial or intentionally added exogenous enzymes, such as beta-glucosidase and tannase, dominate during fermentation, storage, and post-processing, transforming chemical profiles to a more radical extent. This review provides theoretical foundation for precise controlling and adequate utilization of enzymes throughout the tea manufacture chains.

Abstract:Enzymes are protein molecules functioning as specialized catalysts for chemical reactions. They have contributed greatly to the traditional and modern chemical industry by improving existing processes. In this article, we first give a survey of representative industrial applications of enzymes, focusing on the technical applications, feed industry, food processing and cosmetic products. The recent important developments and applications of enzymes in industry are reviewed. Then large efforts are dedicated to the worldwide enzyme market from the demand and production perspectives. Special attention is laid on the Chinese enzyme market. Although enzyme applications are being developed in full swing, breakthroughs are needed to overcome their weaknesses in maintaining activities during the catalytic processes. Strategies of metagomic analysis, cell surface display technology and cell-free system might give valuable solutions in novel enzyme exploiting and enzyme engineering.

Abstract:Extraction is the critical link during pharmaceutical process of traditional Chinese medicine (TCM), which is directly related to the quality of drugs. So the key to technology upgrading of pharmaceutical equipment in Chinese materia medica enterprise is the development of new extraction techniques, which concerns the modernization of TCM. In this paper, fundamentals, traits, and development status of new extraction technologies were firstly introduced, including ultrasound extraction, microwave extraction, super fluid extraction, semi-bionic extraction method, enzymatic treatment extraction, continuous countercurrent extraction, vacuum extraction. Then information of projects supported by the National Natural Science Foundation of China was analyzed in order to recognize the assistance and research results of new extraction techniques. The patents authorized by the State Intellectual Property Office were also summarized for the purpose of understanding the achievement transformation. The information about extraction equipments was collected and screened to acquire the characteristics and market situation. The results showed that there are still problems about new extraction technologies, such as weak basic study, hard transformation of achievements, and the disconnection between research study and practical application. It is necessary to discuss the approaches and methods for accelerating the transformation of fundamental research, which will provide references for the long-term development of new extraction techniques of TCM.

Abstract:The extraction and separation of effective components from Chinese herb and natural plant is one of the most important processes of natural medicine production. Recently, more and more advanced techniques have been used in the field of modern Chinese medicines, for example, microwave-assisted extraction (MAE). Developed on the base of traditional extracting technology using by organic solvents, MAE is of higher extraction rate and efficiency and better extract quality, as well as few investment, simple equipment, wide adaptability, high selectivity, good fidelity and no pollution. This paper mainly reviews the action principle and characteristic of MAE and its application in the extraction of natural products such as flavonoids, glycosides, polysaccharides, terpenoids and essence. Otherwise, the developing foreground of MAE is also prospected here.

Abstract:Plant natural products are a kind of active substance widely used in pharmaceuticals and foods. However, the current production mode based on plant culture and extraction suffer complex processes and severe concerns for environmental and ecological. With the increasing awareness of environmental sustainability, engineered microbial cell factories have been an alternative approach to produce natural products. Many engineering strategies have been utilized in microbial biosynthesis of complex phytochemicals such as dynamic control and substructure engineering. Meanwhile, Enzyme engineering including directed evolution and rational design has been implemented to improve enzyme catalysis efficiency and stability as well as change promiscuity to expand product spectra. In this review, we discussed recent advances in microbial biosynthesis of complex phytochemicals from the following aspects, including pathway construction, strain engineering to boost the production. Abstract:Enzyme-assisted extraction (EAE) involves the use of hydrolytic enzymes for the degradation of the cell wall or other cell components. This supports the diffusion of the solvent into the plant or fungal material, leading to easier elution of its metabolites. This technique has been gaining increasing attention, as it is considered an eco-friendly and cost-effective improvement on classical or modern extraction methods. Its promising application in improving the recovery of different classes of bioactive metabolites (e.g., polyphenols, carotenoids, polysaccharides, proteins, components of essential oil, and terpenes) has been reported by many scientific papers. This review summarises information on the theoretical aspects of EAE (e.g., the components of the cell walls and the types of enzymes used) and the most recent discoveries in the effective involvement of enzyme-assisted extraction of natural products (plants, mushrooms, and animals) for nutraceutical and pharmaceutical applications.

Abstract:Traditional Chinese medicine (TCM) is an important part of Chinese culture and the Chinese pharmaceutical industry. The adoption of stricter requirements for the quality of medicines has resulted in the loss of conformity of some traditional techniques used to prepare TCMs modern pharmaceutical methods. The TCM industry has begun to seek out novel methods to improve product quality and increase economic efficiency. Membrane technology is efficient, energy saving, and environmentally friendly. It is widely used in food processing, desalination, wastewater treatment, and in the medical industry. Membrane technology has also been adopted by the TCM industry as an alternative to traditional technology and has achieved good results. At present, membrane technology has been used in the separation, purification and concentration of TCMs. This review describes the use of microfiltration, ultrafiltration, nanofiltration, reverse osmosis, and membrane distillation, in TCM processing that have been reported. Membrane fouling is the most important obstacle in the use of membrane-based processes by the TCM industry. The causes and control of membrane fouling associated with TCM extracts are also analyzed.

Abstract:Traditional Chineae medicine (TCM) is often composed of a variety of natural medicines. Its composition is complex, and many of its components can not be analyzed and identified. The first step in the rational application of TCM is to successfully separate the effective components which is also a great inspiration for the development of new drugs. Among the many separation technologies of TCM, the traditional heating concentration separation technology has high energy consumption and low efficiency. As a new separation technology, membrane separation technology has the characteristics of simple operation, high efficiency, environment-friendly and so on. The separation effect of high molecular weight difference solution is better. The applications of several main membrane separation technologies such as microfiltration, nanofiltration, ultrafiltration and reverse osmosis are reviewed, the methods of restoring membrane flux after membrane fouling are discussed, and their large-scale industrial applications in the future are prospected and summarized.

Abstract:Natural medicines are now gaining increasing acceptance around the global scope. Due to the complexity of composition, it is imperative to adopt a unique evaluation system, which is not confined to the content of finite marker compounds. This research presents the successful application of pharmacodynamics-based optimization to the ultrasound-assisted extraction of Chansu, a widely used natural medicine with complex bioactive ingredients. The effect of solvent concentration, solvent to solid ratio, extraction time and particle size, on two response indexes (extraction yield of total content of bufalin, cinobufagin and resibufogenin and anticancer activity) were evaluated by response surface methodology (RSM). Two models were built based on two response indexes, and both of them provided accurate predictions. Compared with extraction yield-based optimization, pharmacodynamics-based optimization aligns well with the integrity and complexity of natural medicines. Moreover, the pharmacodynamics-based optimization gives a more economical and efficient optimal condition, which is specifically embodied in the coarser requirement for particle size and the saving of extraction time and solvent. The recommended extraction conditions were methanol concentration of 83.54% (v/v), solvent to solid ratio of 26.23 mL/g, extraction time of 40.43 min and particle size of 61 mesh (0.212-0.25 mm). The present study elaborated the feasibility and implication of pharmacodynamics-based optimization, which provides useful information for further research on the extraction process of natural medicines.

Abstract:Like other traditional medicine in the world, Chinese traditional medicine (CTM) has a long history, which is a treasure of the combination of medicine and Chinese classical culture even more than 5000 years. For thousands of years, CTM has made great contributions to the reproduction and health of the Chinese people. It was an efficient therapeutic tool under the guidance of Chinese traditional medical theory, its source is generally natural products, but there are also a small number of it are natural products after some processing methods. In fact, the definition of Chinese medicine (CM) includes both traditional and new CM developed by modern technology. It is well known that the chemical composition of most CM and natural products is very complex, for example, a single herb may contain hundreds of different chemicals, including active ingredients, side effects, and even toxic ingredients. Therefore, the extraction process is particularly crucial for the quality and clinical efficacy of CM and natural products. In this work, a new classification method was proposed to divide the extraction technologies of CM and natural products into 21 kinds in recent years and analyze their status, advantages, and disadvantages. Then put forward a new technical route based on ultra‐high‐pressure extraction technology for rapid extraction else while removing harmful impurities and making higher utilization of CM and natural products. It is a useful exploration for the extraction industry of medicinal materials and natural products in the world.
