Magnetic iron oxide platforms associated with phytochemicals for applications in MRI and magnetic hyperthermia: A comprehensive review

Abstract:Phytochemicals derived from natural products have been widely studied due to their potential in several pathophysiological contexts, including cancer. Although more than 60 % of chemotherapeutic compounds approved for pharmaceutical use are derived from natural products, their clinical application limitations, mainly due to their low solubility and bioavailability. A growing approach to overcome these limitations involves associating these compounds with magnetic iron oxide nanoparticles (MIONPs), explored for more than 50 years in the biomedical field. Several reports have been published on MIONPs, focusing on their synthesis and biomedical applications associated with conventional drugs. However, this study specifically focuses on the exploration of magnetic nanocarriers associated with phytochemicals, with an emphasis on cancer treatment. A total of 90 articles published between 2018 and 2024 were reviewed, including applications in diagnostics through magnetic resonance imaging (MRI) and therapy through the delivery of anticancer phytocompounds and magnetic hyperthermia (MH). This review addresses the use of MIONPs in cancer therapy, the main classes of phytochemicals and their mechanisms of action, the functionalizing agents of MIONPs and the types of nanocarriers most commonly used for drug delivery. In addition, we discuss in detail several in vitro and in vivo studies that employed MRI and MH techniques as tools for nanomedicine. Finally, we performed a comprehensive statistical analysis, considering pertinent inferences on all data collected in the articles, in order to provide an overview of recent advances in the respective field and barriers that still need to be overcome so that phytochemical therapy combined with MIONPs can achieve effective clinical application in the future.
