Abstract:Quick suberin-based healing after wounding played a protective role for plant to prevent further damage. In this study, the stimulative effect of exogenous abscisic acid (ABA) on wound suberization in postharvest kiwifruit was evaluated through suberin staining with toluidine blue O as well as the determination of suberin phenolics and aliphatics in wound tissue. Furthermore, to reveal the regulatory involvement of ABA in wound suberization, comparative quantitative proteomics and transcriptomics analyses based on iTRAQ and qRT-PCR technique were performed. In proteomics levels, a total of 95 protein species consistently showed differential abundance between ABA and control, including 29 down-regulated and 66 up-regulated protein species. The Kyoto Encyclopedia of Genes and Genomes (KEGG) with protein-protein interaction analyses revealed that ABA mainly affected the antioxidant system, phenylpropanoid metabolism and lipid metabolism associated with wound suberization. Based on the data of proteomics analysis, the differential expressions of genes encoding 11 selected protein species were confirmed by qRT-PCR analyses. GSH-Px, MDHAR, SOD, APX, POD, PAL, CCR, PPO, CYP86B1, DGGT and KCS11 were likely to be the key enzymes that involved the response of ABA to stimulate wound suberization by mediating the antioxidant system, phenylpropanoid metabolism and lipid metabolism.
Mechanism of Methyl Jasmonate-Induced Resistance to Soft Rot Disease in Kiwifruit Based on Transcriptomics Analysis

Abstract:[Objective]Methyl jasmonate(MeJA),a biological signaling molecule in plants,can stimulate fruit resistance to biotic or abiotic factors stress and plays an important role in storage.Transcriptome technology has been widely used to explore the regulatory mechanisms associated with postharvest disease control,and it can elucidate the mechanisms of pathogen-plant interactions at the molecular level,which is conducive to the identification and mining of stress-responsive genes.Previous research has demonstrated that by regulating the defense system's improvement,(MeJA)can cause kiwifruit fruit to become resistant to postharvest soft rot.MeJA-induced kiwifruit resistance to soft rot has been linked to the phenylpropane pathway,membrane lipid metabolism,and energy metabolism.A preliminary description of the physiological and molecular mechanisms underlying MeJA-mediated kiwifruit resistance to soft rot has been made.This study aims to analyze MeJA-induced transcriptome data on kiwifruit resistance to Botryosphaeria dothidea and the expression of disease-resistance-related genes in response to pathogen infestation,thus providing a theoretical reference for studying the molecular mechanism of MeJA-mediated resistance to soft rot in kiwifruit.[Method]'Hongyang'kiwifruit was used as the test material.The inoculated B.dothidea group and the MeJA-treated,inoculated group were selected,and 0,1 and 5 d kiwifruit samples were subjected to transcriptome sequencing.Differentially expressed genes(DEGs)were screened by log2FC≥2,Qvalue≤0.05 and FPKM>2,validated by real-time fluorescence quantitative PCR,and analysed by Gene ontology(GO)functional annotation and KEGG pathway enrichment.[Result]About 114.97 GB of clean data were obtained from the transcriptome sequencing,and the clean data of each sample was about 6.39 GB.The percentage of Q30 base was 90.41%or above.Statistics on the number of differentially expressed genes in each period revealed that kiwifruit fruit DEGs gradually increased with the prolongation of B.dothidea infestation;whereas the number of DEGs induced in the main base of DEGs in response to MeJA was gradually reduced at the time of pretreatment,and after inoculation with the pathogen,suggesting that the exogenous MeJA induced a large number of genes involved in fruit resistance at an early stage.The un-treated inoculation group and the MeJA-treated inoculation group shared 164 and 314 DEGs at 0,1 and 5 d,respectively.GO enrichment results indicated that DEGs were mainly associated with cellular processes,metabolic processes,response to stimuli and biological regulation in biological processes;cellular anatomical entities and intracellular in cellular components;and binding,catalytic activity and transcriptional regulatory activity in molecular functions.The results of KEGG pathway enrichment showed that kiwifruit fruits inoculated with B.dothidea after MeJA treatment were specifically enriched in the plant-pathogen interaction pathway,alpha-Linolenic acid metabolism,glutathione metabolism,benzoxazinoid biosynthesis,ascorbate and aldarate metabolism;phenylpropanoid biosynthesis,stilbenoid,diarylheptanoid and gingerol biosynthesis,flavonoid biosynthesis,MAPK signaling pathway-plant were jointly enriched in both treatment groups.A total of 31 DEGs responding to both MeJA and B.dothidea were screened,and it is hypothesized that these genes are associated with MeJA-mediated disease resistance processes.[Conclusion]In this study,RNA-Seq analysis of externally assisted MeJA-treated kiwifruit fruits at different periods(0 d,1 d and 5 d)after inoculation with B.dothidea shows that MeJA induces a large number of genes of physiological metabolic pathways to change during the process of kiwifruit fruits'resistance to soft rot.This change helps to enhance the initiation of the fruit signalling molecules to the defence system,and increase the synthesis of antimicrobial substances and antioxidant capacity,which in turn improves the disease resistance of kiwifruit fruits and prolongs their storage time.
The Mechanism of Gibberellins Treatment Suppressing Kiwifruit Postharvest Ripening Processes by Transcriptome Analysis

Abstract:Gibberellins (GAs) control numerous processes in plants, including fruit ripening. However, the role of GAs in kiwifruit ripening is not well characterized. Here, we noticed that GA3 treatment in postharvest kiwifruit delayed the ripening process and many physiological traits were detected such as firmness, starch/total soluble solid content, cell-wall components (pectin, cellulose and hemicellulose), ethylene and ester production, and ascorbic acid (vitamin C) concentration. Transcriptome analysis supplied a molecular basis for investigating the inhibiting mechanism of GA3 and metabolic variations during kiwifruit ripening. A total of 3116 differently expressed genes (DEGs) were identified, including 51 genes involved in plant hormone signal transduction pathways and 36 structural genes related to these physiological traits. 16 major DEGs were analyzed by real-time PCR during the whole storage period. The expression of most of them was significantly suppressed by GA3 treatment, such as two ethylene biosynthesis genes (ACOs ), two β-amylase (BAMs), five cell-wall modification genes, one GA2ox, and one GA receptor GID1, whereas the expression of Actinidia29942 (JAZ) was enhanced by GA3 treatment. Correlation analysis between multiple physiological indicators and candidate genes showed that GA3 could regulate kiwifruit postharvest ripening effectively and comprehensively. Together, these GA-responsive genes may play important roles in controlling kiwifruit ripening, and GA treatment could be a promising approach for the preservation of postharvest kiwifruit.
Methyl jasmonate promotes wound healing by activation of phenylpropanoid metabolism in harvested kiwifruit

Abstract:A rapid wound healing in vegetables and fruit is instrumental in maintaining fruit quality and extension of shelf life. In this study, wounded kiwifruit were separately treated with methyl jasmonate (MeJA) and diethyldithiocarbamic acid (DIECA, an inhibitor of jasmonate biosynthesis) to evaluate the effect of MeJA on wound healing of kiwifruit. The results showed that MeJA treatment leads to increases of endogenous JA biosynthesis, lipoxygenase (LOX) and 12-oxo-phytodienoic acid reductase (OPR) activities, and JA synthesis genes expression, while DIECA represses these processes. Meanwhile, MeJA promoted the autofluorescence of suberin polyphenolic (SPP) and the accumulation of SPP monomers. MeJA-treated group demonstrated significantly enhanced total phenols, flavonoids, and lignin contents. Besides, MeJA promoted the activities of phenylalanine ammonia-lyase (PAL), cinnamic acid-4-hydroxylase (C4H), 4-COumaric-COA ligase (4CL), peroxidase (POD), polyphenol oxidase (PPO), and the respective gene expression levels. Results suggest that wound healing in kiwifruit could be promoted by MeJA via activation of phenylpropanoid metabolism.

Abstract:Propyl 4-[2-(4,6-dimethoxy-2-pyrimidinyloxy) benzylamino] benzoate(ZJ0273) is a highly active herbicide with novel molecule structure with China's independent intellectual property rights.In order to provide a theoretical base for ZJ0273 application in orchard,the effects of ZJ0273 on seedlings of strawberry,grape and kiwifruit,and the activity of ace-tolactate synthase(ALS) were examined.The results showed that:after the application of ZJ0273,the severe injury appeared in seedlings of grape,while nearly no symptom was observed in strawberry and kiwifruit.In the seedlings treated with ZJ0273,ALS from grape was significantly inhibited,but not from strawberry and kiwifruit.An addition of ZJ0273 at higher concentration to extraction solutions of Acetolactate synthase could significantly inhibit the activity of ALS in three species.
Identification and Functional Analyses of the Transcription Factors AcWRKY117 and AcWRKY29 Involved in Waterlogging Response in Kiwifruit Plant

Abstract:Waterlogging is becoming an increasingly significant problem for crop production and food security. As one of the largest transcription factor (TFs) families in plant crops, WRKYs are reported to be involved in various abiotic stress responses, including waterlogging resistance. However, little is known about the molecular regulation of WRKYs responding to waterlogging stress in kiwifruit plant (Actinidia chinensis). Based on the transcriptome and genome database, a total of 112 AcWRKYs, which were divided into seven subgroups, had been identified in A. chinensis cv. Donghong. Transcript levels of 7 out of these 112 AcWRKYs showed closely correlation with increasing of acetaldehyde and ethanol contents resulted from the high expression of AcPDCs and AcADHs during waterlogging response of the roots. AcWRKY117 and AcWRKY29, which were clustered into group I and II-e respectively, had been verified to trans-activate the expression of AcPDC2 based on the results of dual lucif-erase assays. Both AcWRKY117 and AcWRKY29 could bind to the w-box (5 '-TGACC (A/T)-3 ') motif located in target promoter of AcPDC2. Furthermore, contents of acetaldehyde and ethanol in both 35S::AcWRKY117 and 35S::AcWRKY29 overexpression kiwifruit calli had been significantly increased. These were consisted with the significant higher transcript levels of AcPDCs and AcADHs, as well as AcERF74 and AcERF75 which trans-activate the activity of AcADH1 promoter, in the transgenic kiwifruit calli. Moreover, the expression pattern of AcWRKY117 had been dramatically induced in 35S::AcWRKY29 transgenic lines, and vice verse. Taken together, our results provide molecular evidence for WRKYs regulation on waterlogging response in kiwifruit.
Two Ethylene-Responsive Transcription Factors, AdVAL2 and AdKAN2, Regulate Early Steps in Kiwifruit Starch Degradation

Abstract:Starch degradation is considered as an early step in the initiation of kiwifruit postharvest ripening and softening after ethylene treatment, which is driven by various transcription factors (TFs) and genes in the carbohydrate pathway. Studies regarding starch degradation have been conducted after ethylene treatment then removal, but early ethylene responsive genes and their function in triggering starch degradation remains unclear. In this study, a continuous sampling approach was conducted with 2 h intervals throughout 24 h treatment. As a result, starch degradation and accumulation of total soluble solid were accelerated after 10 h ethylene treatment. Based on RNA-seq and Real-time quantitative PCR (RT-qPCR) data, three beta-Amylase genes, AdBAM3, AdBAM3.1 and AdBAM3L, were significantly up-regulated in ethylene treated samples. Weighted Gene Co-expression Network Analysis (WGCNA) identified TFs that are highly correlated with these three BAM genes, based on gene expression patterns. AdVAL2 (a B3 domain-containing transcription factor) and AdKAN2 (a MYB-like domaincontaining transcription factor) were found to transcriptionally repress the promoter activity of AdBAM3 and AdBAM3L, respectively. Additionally, the expression of both AdVAL2 and AdKAN2 was observed to be repressed by ethylene treatment. Transient overexpression analysis confirmed the inhibitory effect of AdVAL2 and AdKAN2 on the expression of BAM genes, resulting in starch accumulation in kiwifruit. Altogether, continuous sampling during ethylene exposure has revealed that these early ethylene responsive TFs could be the potential triggers for starch degradation.
Ozone treatment modulates reactive oxygen species levels in kiwifruit through the antioxidant system: Insights from transcriptomic analysis

Abstract:Owing to its easy decomposition and residue-free properties, ozone has been used as an effective and environmentally friendly physical preservation method for maintaining the post-harvest quality of fruits. This study aimed to investigate the effects of ozone treatment on the levels of oxidative stress markers and the status of the antioxidant defense system in refrigerated kiwifruit. Additionally, the study aimed to identify the differences in gene expression levels and potential regulatory effects from the transcriptional level. The results showed that ozone treatment reduced the respiration rate, maintained the fruit hardness and storage quality, and inhibited the ripening and senescence of kiwifruit. Ozone treatment activated antioxidant enzymes (superoxide dismutase, peroxidase, and catalase) and ascorbate-glutathione cycle to prevent the increase of reactive oxygen species levels (H2O2, O2-•) and malonaldehyde content, maintaining lower membrane lipid peroxidation and reactive oxygen species (ROS) accumulation than the control treatment. Further analysis showed that the regulatory ability of ROS in kiwifruit treated with ozone was not only related to the synergistic effect of enzyme activity and gene expression related to the antioxidant oxidase system and the ascorbate-glutathione (ASA-GSH) cycle but also related to downstream hormone signaling. This study provides a foundation for understanding the potential effects of ozone treatment on the antioxidant cycle of kiwifruit and provides valuable insights into the molecular basis and related key genes involved in regulating ROS to delay aging in kiwifruit.
Transcriptome Analysis Reveals the Roles of Chlorophylla/b-Binding Proteins (cabs) and Stay-Green (SGR) in Chlorophyll Degradation During Fruit Development in Kiwifruit

Abstract:Commercial cultivars of kiwifruit (Actinidiaspp.) can have green-fleshed or yellow-fleshed fruit depending on their chlorophyll degradation ability. In recent years, an increasing number of yellow-fleshed cultivars have been bred in China. However, the mechanisms underlying the flesh colouration of these new cultivars remain elusive. In this study, the pigment contents of the yellow-fleshed cultivar 'Jinshi NO.1' were determined during fruit development, and RNA sequencing was conducted to screen for differentially expressed genes and identify potential candidates for pigment metabolism. The chlorophyll content continuously decreased from 0 days after full bloom (DAFB) (T1) to 189 DAFB (T4), resulting in a more than 30-fold change, while the contents of anthocyanins and carotenoids showed sharp decreases from T1 to 44 DAFB (T2) and remained stable thereafter. Weighted gene co-expression network analysis (WGCNA) indicated that the chlorophyll contents were highly correlated with chlorophyll-degradation-related genes, especially three chlorophylla/b-binding protein (CAB) genes and one stay-green gene (AcSGR1), but no carotenoid-related genes. Furthermore, these genes were found to be highly correlated with 15 transcription factors and 11 hub genes. These results indicate the important role of chlorophyll degradation in the colouration of 'Jinshi NO.1' and provide candidates for further functional characterisation and transcriptional regulation.
Transcriptome Analysis Identifies a Zinc Finger Protein Regulating Starch Degradation in Kiwifruit

Abstract:Ripening, including softening, is a critical factor in determining the postharvest shelf-life of fruit and is controlled by enzymes involved in cell wall metabolism, starch degradation, and hormone metabolism. Here, we used a transcriptomics-based approach to identify transcriptional regulatory components associated with texture, ethylene, and starch degradation in ripening kiwifruit (Actinidia deliciosa). Twelve differentially expressed structural genes, including seven involved in cell wall metabolism, four in ethylene biosynthesis, and one in starch degradation, and 14 transcription factors (TFs) induced by exogenous ethylene treatment and inhibited by the ethylene signaling inhibitor 1-methylcyclopropene were identified as changing in transcript levels during ripening. Moreover, analysis of the regulatory effects of differentially expressed genes identified a zinc finger TF, DNA BINDING WITH ONE FINGER (AdDof3), which showed significant transactivation on the AdBAM3L (β-amylase) promoter. AdDof3 interacted physically with the AdBAM3L promoter, and stable overexpression of AdBAM3L resulted in lower starch content in transgenic kiwifruit leaves, suggesting that AdBAM3L is a key gene for starch degradation. Moreover, transient overexpression analysis showed that AdDof3 up-regulated AdBAM3L expression in kiwifruit. Thus, transcriptomics analysis not only allowed the prediction of some ripening-regulating genes but also facilitated the characterization of a TF, AdDof3, and a key structural gene, AdBAM3L, in starch degradation.
Integrated metabolomic and transcriptomic analyses provide comprehensive new insights into the mechanism of chitosan delay of kiwifruit postharvest ripening

Abstract:Chitosan (CTS) plays an important role in delaying fruit ripening and extending fruit shelf life when used as an eco-friendly and edible coating. However, there is still limited understanding about the molecular mechanisms and effects of chitosan on the quality of postharvest kiwifruit . In this study, firmness, total soluble solid, acid, phenolic content, flavonoid content, starch and ascorbic acid concentration, ethylene production, and cell-wall components were determined after CTS treatment. Fruit treated with CTS maintained higher firmness, starch and flavonoids (3.85-, 1.78-and 2.08-fold higher, respectively, after 6d compared to the control). Widely targeted metabolome analysis revealed flavonoids (dihydrokaempferol-7- O -glucoside, eriodictyol-3′- O -glucoside) and lipids (LysoPC 16:0 (2 n isomer)), and punicic acid (9 Z ,11 E ,13 Z -octadecatrienoic acid) were the main differential metabolites. KEGG pathway enrichment analysis showed 'metabolic pathways (ko01100)' and 'biosynthesis of secondary metabolites (ko01110)' were the main KEGG pathways. Integrated metabolomic and transcriptomic analyses revealed that the expression of five key structural genes, including three starch degradation genes ( AcBAM3L , AcBAM3.1 , Acc31818 ( PHS )), one cell-wall modification gene ( AcPG1 ), and one flavonoids biosynthesis gene ( Acc18331 ( F3 ' H )), and 12 transcription factors ( AcNAC083 , AcRAP2–10 , AcERF14 , AcERF64 , Acc27131 ( bZIP ), AcHSFB2a , Acc12589 ( IAA ), AcMYB13 , Acc20159 ( bHLH ), AcBEL1 , AcbHLH149 , AcWRKY75 ) were different. Real-time PCR analyses verified that the expression of AcBAM3L , AcBAM3.1 , AcPG1 and most of the 12 transcription factors were suppressed by CTS treatment, while the expression of Acc31818 ( PHS ), Acc18331 ( F3'H ) and AcBEL1 were enhanced by CTS treatment. Together, these CTS-responsive genes may play critical roles in determining the rate of ripening and quality change of postharvest kiwifruit.

Abstract:Postharvest ripening and processing of fresh-cut (FC) kiwifruit brings quality deterioration and spoilage, including substantial changes in fruit color, texture and flavor. Hence, a series of preservation techniques have emerged to reduce postharvest loss and maintain FC kiwifruit quality. In this study, we evaluated the efficacy of acetylsalicylic acid (ASA), a derivative of plant endogenous hormone salicylic acid, in preserving the quality and delaying the ripening process of FC kiwifruit. Compared to the control fruit, ASA treatment maintained a better fruit appearance and higher fruit firmness, as well as reducing mass loss, total soluble solids and ethylene production. Based on the RNA-seq data and weighted gene co-expression network analysis (WGCNA), eight biosynthetic genes were identified as involved in fruit quality change, which participates in cell wall metabolism, starch degradation/sugar accumulation and ethylene biosynthesis. Moreover, the analysis showed that four transcription factors were significantly induced by exogenous ASA treatment. Dual-luciferase assays proved that ASA-responsive transcription factors regulated fruit quality-related genes, resulting in delaying ripening process and extending the shelf-life of FC kiwifruit. This study showed that ASA effectively delayed the ripening process, and AcWRKY7/AcDOF21/AcC2H2–151/AcbHLH123 are associated with the ASA-mediated fruit quality maintenance.

Here, we performed a transcriptome analysis by combining single-molecule real-time (SMRT) sequencing and Illumina RNA sequencing and investigated the physiological responses of the roots of KR5 (A. valvata, a tolerant genotype) after 0, 12, 24 and 72 h of waterlogging stress. KR5 roots responded to waterlogging stress mainly via carbohydrate and free amino acids metabolism and reactive oxygen species (ROS) scavenging pathways. Trehalose-6-phosphate synthase (TPS) activity, alcohol dehydrogenase (ADH) activity and the total free amino acid content increased significantly under waterlogging stress. The nicotinamide adenine dinucleotide-dependent glutamate synthase/alanine aminotransferase (NADH-GOGAT/AlaAT) cycle was correlated with alanine accumulation. Levels of genes encoding peroxidase (POD) and catalase (CAT) decreased and enzyme activity increased under waterlogging stress. Members of the LATERAL ORGAN BOUNDARIES (LOB), AP2/ERF-ERF, Trihelix and C3H transcription factor families were identified as potential regulators of the transcriptional response. Several hub genes were identified as key factors in the response to waterlogging stress by a weighted gene co-expression network analysis (WGCNA). Our results provide insights into the factors contributing to waterlogging tolerance in kiwifruit, providing a basis for further studies of interspecific differences in an important plant trait and for molecular breeding.

Abstract:Waterlogging stress is one of the major natural issues resulting in stunted growth and loss of agricultural productivity. Cultivated kiwifruits are popular for their rich vitamin C content and unique flavor among consumers, while commonly sensitive to waterlogging stress. The wild kiwifruit plants are usually obliged to survive in harsh environments. Here, we carried out a transcriptome analysis by high-throughput RNA sequencing using the root tissues of Actinidia deliciosa (a wild resource with stress-tolerant phenotype) after waterlogging for 0 d, 3 d, and 7 d. Based on the RNA sequencing data, a high number of differentially expressed genes (DEGs) were identified in roots under waterlogging treatment, which were significantly enriched into four biological processes, including stress response, metabolic processes, molecular transport, and mitotic organization, by gene ontology (GO) simplify enrichment analysis. Among these DEGs, the hypoxia-related genes AdADH1 and AdADH2 were correlated well with the contents of acetaldehyde and ethanol, and three transcription factors Acc26216, Acc08443, and Acc16908 were highly correlated with both AdADH1/2 genes and contents of acetaldehyde and ethanol. In addition, we found that there might be an evident difference among the promoter sequences of ADH genes from A. deliciosa and A. chinensis. Taken together, our results provide additional information on the waterlogging response in wild kiwifruit plants.

Abstract:Fruit is constantly challenged by wounding events, inducing accelerated ripening and irreversible metabolic changes. However, cognate mechanisms that regulate this process are little known. To expand our knowledge of ripening metabolism induced by wounding, an artificial-wound global transcriptome investigation combined with metabolite profiling study was conducted in postharvest kiwifruit (Actinidia chinensis var. deliciosa (A. Chev.) A. Chev. 'Hayward'). Wounding treatment promoted fruit ripening, as demonstrated by changes in fruit firmness, ethylene production and respiration activity determined periodically during a ripening period of 8 d at room temperature. Calcium imaging using fluorescent probe Fluo-3 AM revealed spatial dynamics of Ca2+ signaling in the wounding area following 8d ripening. Several sugars including fructose, glucose, and sucrose as well as organic acids such as citric, succinic and galacturonic acid were increased by wounding. Changes of various amino acids in wounded-treated fruit, especially 5-oxoproline and valine along with alternations of soluble alcohols, like myo-inositol were detected. Gene expression analysis of the wounded fruit showed increased expression of genes that are mainly involved in defense response (e.g., AdTLP.1-3, AdPP2C.1-2, AdMALD1), calcium ion binding (e.g., AdCbEFh, AdCLR, AdANX), TCA cycle (e.g., AdMDH.1, AdMDH.2, AdCS), sugars (e.g., AdSUSA.1, AdSPS4, AdABFr), secondary metabolism (e.g., AdPAL.1-3, AdCCR, AdHCT.1-2), lipid processing (e.g., AdGELP.1-4, AdGELP) and pectin degradation (e.g., AdPE.1-2, AdPAE.1-2, AdPG.1-2) as well as in ethylene (AdERF7, AdERF1B, AdACO.1-4) and auxin (AdICE, AdAEFc, AdASII) synthesis and perception. Moreover, genes related to aquaporins, such as AdAQP2, AdAQP4 and AdAQP7 were down-regulated in fruit exposed to wounding. These results demonstrate multiple metabolic points of wounding regulatory control during kiwifruit ripening and provide insights into the molecular basis of wounding-mediated ripening.

Abstract:Brassinosteroids (BRs) play pivotal roles in improving plant stress tolerance. To investigate the mechanism of BR regulation of salt tolerance in kiwifruit, we used 'Hongyang' kiwifruit as the test material. We exposed the plants to 150 mmol/L NaCl stress and irrigated them with exogenous BR (2,4-epibrassinolide). The phenotypic analysis showed that salt stress significantly inhibited photosynthesis in kiwifruit, leading to a significant increase in the H2O2 content of leaves and roots and a significant increase in Na+/K+, resulting in oxidative damage and an ion imbalance. BR treatment resulted in enhanced photosynthesis, reduced H2O2 content, and reduced Na+/K+ in leaves, alleviating the salt stress injury. Furthermore, transcriptome enrichment analysis showed that the differentially expressed genes (DEGs) related to BR treatment are involved in pathways such as starch and sucrose metabolism, pentose and glucuronate interconversions, and plant hormone signal transduction, among others. Among the DEGs involved in plant hormone signal transduction, those with the highest expression were involved in abscisic acid signal transduction. Moreover, there was a significant increase in the expression of the AcHKT1 gene, which regulates ion transduction, and the antioxidant enzyme AcFSD2 gene, which is a key gene for improving salt tolerance. The data suggest that BRs can improve salt tolerance by regulating ion homeostasis and reducing oxidative stress.

Abstract:Kiwifruit are climacteric fruit, so they must be harvested before they are fully ripe, allowing for the extension of their shelf-life via cold storage. Therefore, an adequate knowledge about how ethylene-induced fruit senescence is required to avoid significant economic losses. The main goal of the present study was to investigate the kiwifruit ripening process at the physiological and molecular levels by RNA-seq after 1-methylcyclopropene (1-MCP, ethylene inhibitor) and Ethrel® (ethylene stimulator) treatments. The results showed that Ethrel® (ethephon) treatment induced more accelerated fruit ripening, leading to rapid fruit senescence, meanwhile 1-MCP caused a slowing flesh softening, and thus a longer shelf-life period. The RNA-seq was carried out on the fruit after 4 and 13 days, considering day 4 as the most determinant in terms of differentially expressed genes (DEGs). The sequencing achieved 70.7% alignment with the 'Hongyang' genome, obtaining 18,036 DEGs. The protein-protein interaction (PPI) network shows the interaction between different pathways in two main clusters: (1) pentose and glucoronate interconversions, citrate cycle, glycolysis and gluconeogenesis or starch, and sucrose metabolism and (2) porphyrin and chlorophyll metabolism. The first cluster is mainly interconnected by G6PD1 (pentose pathway); E1 ALPHA and ACLB-2 (citrate cycle); Achn209711 (pentose and glucoronate); LOS2 (glycolysis); HKL1 and HXK1 (glycolysis—starch and sucrose); and PHS2 (starch and sucrose). In the second cluster, GUN5 through PORA is interacting with CRD1 and NYC1 which were overexpressed by 1-MCP in the porphyrin and chlorophyll metabolism. In addition, genes linked to PSBY and PSBP photosynthesis-linked proteins in photosystem 2 were overexpressed by 1-MCP which is undoubtedly related to chlorophyll degradation and fruit senescence. These results suggest that in kiwifruit, the main pathways that are regulated by ethylene-induced senescence comprise sugar catabolism and chlorophyll degradation.

Abstract:Starch is one of the most important storage carbohydrates in plants. Kiwifruit typically accumulate large amounts of starch during development. The fruit retain starch until commercial maturity, and its postharvest degradation is essential for consumer acceptance. The activity of genes related to starch degradation has, however, rarely been investigated. Based on the kiwifruit genome sequence and previously reported starch degradation-related genes, 17 novel genes were isolated and the relationship between their expression and starch degradation was examined using two sets of materials: ethylene-treated (100 mu L/L, 20 degrees C; ETH) vs. control (20 degrees C; CK) and controlled atmosphere stored (CA, 5% CO2 + 2% O-2, 0 degrees C) vs. normal atmosphere in cold storage (NA, 0 degrees C). Physiological analysis indicated that ETH accelerated starch degradation and increased soluble solids content (SSC) and soluble sugars (glucose, fructose and sucrose), while CA inhibited starch reduction compared with NA. Using these materials, expression patterns of 24 genes that may contribute to starch degradation (seven previously reported and 17 newly isolated) were analyzed. Among the 24 genes, AdAMY1, AdAGL3 and AdBAM3.1/3L/9 were significantly induced by ETH and positively correlated with starch degradation. Furthermore, these five genes were also inhibited by CA, conforming the likely involvement of these genes in starch degradation. Thus, the present study has identified the genes with potential for involvement in starch degradation in postharvest kiwifruit, which will be useful for understanding the regulation of kiwifruit starch content and metabolism.

Abstract:So far, a variety of metabolite components of kiwifruit have been elucidated. However, the identification and analysis of flavonoids in different tissues of kiwifruit are rarely carried out. In this study, we performed transcriptome and metabolome analyses of roots (Gkf_R), stems (Gkf_T), leaves (Gkf_L), and fruits (Gkf_F) to provide insights into the differential accumulation and regulation mechanisms of flavonoids in kiwifruit. Results showed that a total of 301 flavonoids were identified, in four tissues with different accumulation trends, and a large proportion of flavonoids had high accumulation in Gkf_L and Gkf_R. A total of 84 genes have been identified involved in the flavonoid biosynthesis pathway, and the expression levels of five LAR, two DFR, and one HCT were significantly correlated with the accumulation of 16 flavonoids and co-localized in the flavonoid biosynthesis pathway. In addition, a total of 2362 transcription factor genes were identified, mainly MYBs, bHLHs, ERFs, bZIPs and WRKYs, among which the expression level of bHLH74, RAP2.3L/4L/10L, MYB1R1, and WRKY33 were significantly correlated with 25, 56, 43, and 24 kinds of flavonoids. Our research will enrich the metabolomic data and provide useful information for the directed genetic improvement and application in the pharmaceutical industry of kiwifruit.
