Abstract:Amphiphilic cationic polymers mimicking the membrane-disrupting mechanism of natural antimicrobial peptides displays high-efficacy bactericidal activity and meanwhile good biocompatibility, while electrostatic adsorption enables bacteria to adhere to substrate surfaces, effectively lowering the antimicrobial effect, so it is worthwhile to tackle the problem of bacterial adhesion to prolong its antibacterial activity. Herein, a kind of polyquaternium latex (PQL) was synthesized to coat polypropylene nonwoven fabrics (PPNFs) with bacterial repellency and rapid biocidal activity toward both Staphylococcus aureus and Escherichia coli. Compared with polyquaternium solution coatings, latex coatings provided PPNFs with unique microscopic morphology and surface chemistry rich in polyoxyethylene moieties, achieving better resistance to bacterial adhesion, and meanwhile PQL coated PPNFs also delivered 99% killing rates against both bacteria within 2 min. After exposure to ultrasonic cleaning for 4 h, PQL modified PPNFs still demonstrated good bactericidal activity. Moreover, they also displayed low cytotoxicity. Abstract:Hydrogel materials are the preferred candidates for antimicrobial materials due to their high water absorption, superior biocompatibility and structural diversity. In this work, we present for the first time that diamine-alkyl and 1,3-dibromo-2-propanol can be rapidly cross-linked to prepare a series of new cationic polymer gels by polycondensation reactions under a strongly alkaline environment. The presence of C−N+ bonds are demonstrated in the formation of dendritic structured polymers by XPS analysis. In addition, the physicochemical properties of b-PHIs hydrogels were evaluated and it was found that the variation of diamine alkyl chain length could modulate the storage modulus, water uptake properties, thermal stability and zeta-potential of the gels. The increase in hydroxyl content and the C−N+/C-C ratio is evidenced by the elevated water absorption properties of the gels and the variations in zeta potential, respectively. In addition, the antibacterial performance evaluation showed that gels had good antibacterial activity against S. aureus and E. coli, with antimicrobial rates of b-PHOI as high as 96% and 97%, respectively. This work pioneers a brand-new idea for the simple and rapid synthesis of branched cationic polymers with low cytotoxicity in the field of antimicrobial biomaterials and proceeds a new attempt in the directional modulation of the physical properties of the materials.

Abstract:In this paper, we have designed and synthesized well-defined pegylated-polymers with tertiary amines from readily available commodity monomers 2-(dimethylamino)ethyl methacrylate (DMAEMA) and oligo(ethylene glycol) methyl ether methacrylate (OEGMA, Mn ∼ 475 Da) by reversible addition–fragmentation chain transfer (RAFT) polymerisation. By employing a simple and efficient post-polymerisation functionalization strategy, tertiary amines were quaternized to result in cationic polymers. By the careful selection of the functional halide, X–(CH2)q–R, (where in X=halide; R=the chemical functionality; q=the number of alkyl spacer between the quaternary ammonium group and R), a series of polymers with well-controlled molecular weight, different amphiphilic balance and chemical functionalities (such as alkyl, primary alcohol, primary amine and carboxylic acid) were readily synthesized. The antimicrobial activities of these cationic polymers were determined against Gram-positive bacteria Bacillus subtilis. Minimum inhibitory concentration (MIC), the polymer concentration to completely inhibit the bacterial growth, was found to be dependent both on the nature of functional group and the hydrophobicity of the polymer. Amongst the functional groups, both the alkyl and the alcohol groups were found to be effective, with MIC values in the range of 20–80mg/L. The haemolytic properties of polymers were analyzed against mouse red blood cells. The poymers with a short alkyl or hydroxyl group demonstrated little haemolysis, yet retained strong antimicrobial activity. The overall hydrophobicity of the polymer influenced its haemolytic behavior. These polymers can be promising antimicrobial agents. In addition, the approach proposed in this study to atom-efficient design and synthesis of antimicrobial polymers from the commercially available monomers can also be applied to develop well-defined functional cationic polymers for various biomedical applications.

Abstract:Diseases induced by bacterial and viral infections are common occurrences in our daily life, and the main prevention and treatment strategies are vaccination and taking antibacterial/antiviral drugs. However, vaccines can only be used for specific viral infections, and the abuse of antibacterial/antiviral drugs will create multi−drug−resistant bacteria and viruses. Therefore, it is necessary to develop more targeted prevention and treatment methods against bacteria and viruses. Proteins on the surface of bacteria and viruses can specifically bind to sugar, so glycopolymers can be used as potential antibacterial and antiviral drugs. In this review, the research of glycopolymers for bacterial/viral detection/inhibition and antibacterial/antiviral applications in recent years are summarized.

Abstract:Antimicrobial polymers have attracted substantial interest due to high demands on improving the health of human beings via reducing the infection caused by various bacteria. The review presented herein focuses on rendering polysaccharides, mainly cellulosic-based materials and starch to some extent, antimicrobial via incorporating cationic polymers, guanidine-based types in particular. Extensive review on synthetic antimicrobial materials or plastic/textile has been given in the past. However, few review reports have been presented on antimicrobial polysaccharide, cellulosic-based materials, or paper packaging, especially. The current review fills the gap between synthetic materials and natural polysaccharides (cellulose, starch, and cyclodextrin) as substrates or functional additives for different applications. Among various antimicrobial polymers, particular attention in this review is paid to guanidine-based polymers and their derivatives, including copolymers, star polymer, and nanoparticles with core-shell structures. The review has also been extended to gemini surfactants and polymers. Cationic polymers with tailored structures can be incorporated into various products via surface grafting, wet-end addition, blending, or reactive extrusion, effectively addressing the dilemma of improving substrate properties and bacterial growth. Moreover, the pre-commercial trial conducted successfully for making antimicrobial paper packaging has also been addressed.

Abstract:Poly-L-lysine (PLL) is a water-soluble biopolymer consist of repeating unit of L-lysine. It displays intrinsic non-antigenicity, antibacterial property, biocompatibility, and biodegradability, and facile synthesis procedure, therefore has attracted particular attention in various biomedical and pharmaceutical applications. Following a brief introduction to PLL synthesis, this review focuses on the significant advances of PLL and PLL-derived complexes utilized in cargo delivery, tissue adhesives, and antibacterial materials. Meanwhile, this review summarized the modification and optimization of PLL complexes for further clinical translational studies. The final section discusses the challenges and opportunities of PLL-based complexes in the biomedical areas.

Abstract:A novel geminized amphiphilic polymer containing double cationic head groups and hydrophobic tails in each structural unit (denoted as PAGBn), has been prepared in our study. The self-assembly behavior on the solid/liquid interface was investigated by quartz crystal microbalance with dissipation (QCM-D), steady-state fluorescence, dynamic light scattering (DLS) and transmission electron microscope (TEM). A two-regime buildup can be observed, where the first regime is attributed to the rapid adsorption of polymers, while a rearrangements of pre-adsorbed molecules and their aggregation on the surface dominate the second regime, exhibiting a formation of aggregates in large sizes due to the effect of the charge density and hydrophobes on the self-assembly process. It exhibits a drastically enhanced killing efficiency of ∼99.9% against both of S. aureus and E. coli bacteria for PAGBn compared with other traditional single-chained polymers, through the reinforced synergistic effect of electrostatic and hydrophobic interactions, and the formation of large-sized polymer aggregates, encompassing and killing the bacteria more efficiency. Abstract:Antibacterial materials have always suffered from high toxicity. In this study, we fabricated injectable and biodegradable hydrogels utilizing poly(hexamethylene guanidine) hydrochloride and poly (ethylene glycol) as the backbone through a thiol-ene "click" reaction under physiological conditions, and they showed remarkably low toxicity and high and broad-spectrum antibacterial activity.

Abstract:Glycopolymer has been developed in bactericidal applications, but the traditional synthesis, such as click chemistry, needs poly-step reactions and complicated processes. Herein, incorporating the glyco-units into the polymers by quaternization has been exploited to give a facile strategy by Copper-mediated reversible deactivation radical polymerization (Cu(0)-RDRP) to prepare quaternized poly(ionic liquid)s (PILs) with pendant saccharide. The obtained PILs have the suitable property of killing typical Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus. Simultaneously, they perform the lower hemolytic rate of blood cells, higher selectivity index against bacteria, and higher affinity with Concanavalin A (ConA) than those quaternized by bromohydrin at the same quaternization degree. This result demonstrated that the introduced saccharide units, such as glucose and mannose, could maintain the electrostatic interaction to kill bacteria, perform lectin recognition behavior and interaction with ConA, and relatively harmlessness against red blood cells. Moreover, the sugar-containing PILs became insoluble and still antibacterial after anion exchange reaction. These new findings illuminated that such quaternized antibacterial agents with saccharide units that are easy-to-prepare, low cytotoxic could pave the way for further water disinfectant and pathogen control application.

Abstract:The emergence of various resistant bacteria and overuse of antibiotics have led to severe side effects. Therefore, developing efficient and safe antibacterial systems is important. Herein, well-defined antimicrobial material–helical poly(phenyl guanidinium isocyanide) block copolymers with different conformations (L-P3-van, D-P3-van, and DL-P3-van) that connect vancomycin (van) into the polymer through a disulfide bond were synthesized. The prepared antimicrobial materials exhibit broad-spectrum antimicrobial activity, low bacterial resistance, and good proteolytic stability. They also overcome the intrinsic resistance of Gram-negative bacteria to van with a 100-fold increase in antimicrobial activity. Interestingly, the conformation of the material promotes its antimicrobial activity. The left-handed helix conformation shows fivefold more antimicrobial active than the right-handed helical conformation, thereby opening a path for the application of nanochirality in the field of antibiotics.

Abstract:Antimicrobial peptides (AMPs) are expected to be an alternative promising solution to the global public health problem of antibiotic resistance due to their unique antimicrobial mechanism. However, extensive efforts are still needed to improve the shortcomings of traditional AMPs, such as rapid proteolysis, hemolysis, slow response, toxicity, etc., by exploring AMP-based new antimicrobial strategies. Here, we develop cationic peptide bundles into novel antimicrobial architectures that can rapidly kill multiple types of bacteria including drug-resistant bacteria. Remarkably, cationic peptide bundles can be used as polymerization units to cross-link with other polymers through simple two-component polymerization to produce diverse antimicrobial materials. For the proof of concept, three materials were fabricated and investigated, including an antimicrobial hydrogel that can significantly accelerate the healing of infected wounds, a multifunctional antimicrobial bioadhesive that shows promise in antimicrobial coatings for medical devices, and a photo-cross-linked antimicrobial gelatin hydrogel with broad application potential. The integration of antimicrobial units into the materials' backbone endows their biocompatibility. Cationic peptide bundles not only represent a new antimicrobial strategy but also provide a versatile and promising processing method to create diversified, multifunctional, and biocompatible antimicrobial materials.

Abstract:Poly(vinyl alcohol) (PVA) hydrogels are widely employed in various biomedical applications due to their excellent biocompatibility, high water solubility, low protein adsorption, and chemical stability. In this work, two types of PVA-based hydrogels were synthesized by copolymerization of cationic [2-(methacryloyloxy) ethyl]trimethyl ammonium chloride (DMC) or zwitterionic sulfobetaine methacrylate (SBMA) with acrylated PVA (Acr-PVA), prepared through modification of the hydroxyl groups of PVA with glycidyl methacrylate (GMA). The antibacterial rates of Acr-PVA-g-PDMC hydrogels can reach 99.88% against Escherichia. coli and 99.99% against Staphylococcus. aureus. By comparison, the antibacterial rates of the Acr-PVA-g-PSBMA hydrogels can reach 88.40% against E. coli and 96% against S. aureus. Moreover, the Acr-PVA-g-PSBMA hydrogels possess better cell anti-adhesive properties than the Acr-PVA-g-PDMC hydrogels. The two types of hydrogels similarly possess macroporous structures, suitable swelling capacity, good mechanical properties and favorable cytocompatibility toward L929 cells. All results imply that charged group modified PVA hydrogels not only maintain the inherent properties of PVA but also possess excellent antibacterial and anti-adhesive properties. As such, the hydrogels show potential applications in biological materials fields.

Abstract:A novel class of antimicrobial cationic polycarbonate/PEG hydrogels are designed and synthesized by Michael addition chemistry. These hydrogels demonstrate strong broad-spectrum antimicrobial activities against various clinically isolated multidrug-resistant microbes. Moreover, they exhibit nonfouling properties and prevent the substrate from microbial adhesion. These antimicrobial and antifouling gels are promising materials as catheter coatings and wound dressings to prevent infections.

Abstract:Antimicrobial peptide-mimicking antibacterial polymers represent a practical strategy to conquer the ever-growing threat of antimicrobial resistance. Herein, we report the syntheses and antibacterial performance of degradable amphiphilic cationic polyesters containing pendent quaternary ammonium motif and hydrophobic alkyl or fluoroalkyl groups. These polyesters were conveniently prepared from poly(3-methylene-1,5-dioxepan-2-one) by the highly efficient one-pot successive thiol-Michael addition reactions. The antibacterial activity of these polyesters against S. aureus and E. coli and their hemolytic activity toward red blood cells were evaluated, some of them showed moderate antibacterial activity and selectivity against Gram-positive S. aureus. The membrane disruption mechanism of these cationic polyesters was briefly explored by monitoring the bacteria killing kinetics and SEM observations. Moreover, the effects of cationic/hydrophobic ratio and the incorporation of fluoroalkyl group on the antibacterial activity and selectivity of the polyesters were demonstrated.

Abstract:With the continuous emergence of drug-resistant pathogens, new strategies with high antibacterial efficacy are urgently needed. Herein, five cationic nano-sized hyperbranched polymers (CNHBPs) with cationic functional groups have been constructed, and their antibacterial mechanism has been studied in detail. CNHBPs bearing secondary ammonium salt groups and long alkyl chains (S12-CNHBP) exhibited weak antibacterial and antibiofilm ability, while CNHBPs bearing quaternary ammonium salt groups and long alkyl chains (Q12-CNHBP) showed highest antimicrobial and stronger antibiofilm activities. ζ potential and isothermal titration microcalorimetry (ITC) results suggested that negatively surface charged of bacteria cells provided Q12-CNHBP with a higher intrinsic electrostatic driving force for bacterial killing than S12-CNHBP. Fluorescent tracing and morphology observations indicated that the bacterial genome might be another antibacterial target for S12-CNHBP in addition to the cell wall and membrane, which were mainly antibacterial targets for Q12-CNHBP, which made it less likely induced bacterial resistance. Surprisingly, Q12-CNHBP exhibited superior in vivo therapeutic efficacy in a mouse wound model of methicillin-resistant Staphylococcus aureus (MRSA) infection with low toxicity during treatment. These advantages and ease of preparation will undoubtedly distinguish Q12-CNHBP as a new class of suitable candidates to combat multidrug-resistant pathogen infections. This study opens up a new avenue for exploiting antibacterial biomaterials to treat infections caused by drug-resistant bacteria.

Abstract:The emergence of antibiotic-resistant "superbugs" poses a serious threat to human health. Nanomaterials and cationic polymers have shown unprecedented advantages as effective antimicrobial therapies due to their flexibility and ability to interact with biological macromolecules. They can incorporate a variety of antimicrobial substances, achieving multifunctional effects without easily developing drug resistance. Herein, this article discusses recent advances in cationic polymers and nano-antibacterial materials, including material options, fabrication techniques, structural characteristics, and activity performance, with a focus on their fundamental active elements.

Antibacterial modification of an injectable, biodegradable, non-cytotoxic block copolymer-based physical gel with body temperature-stimulated sol-gel transition and controlled drug release

Abstract:Biomaterials are being extensively used in various biomedical fields; however, they are readily infected with microorganisms, thus posing a serious threat to the public health care. We herein presented a facile route to the antibacterial modification of an important A-B-A type biomaterial using poly (ethylene glycol) methyl ether (mPEG)- poly(ε-caprolactone) (PCL)-mPEG as a typical model. Inexpensive, commercial bis(2-hydroxyethyl) methylammonium chloride (DMA) was adopted as an antibacterial unit. The effective synthesis of the antibacterial copolymer mPEG-PCL-∼∼∼-PCL-mPEG (where ∼∼∼ denotes the segment with DMA units) was well confirmed by FTIR and (1)H NMR spectra. At an appropriate modification extent, the DMA unit could render the copolymer mPEG-PCL-∼∼∼-PCL-mPEG highly antibacterial, but did not largely alter its fascinating intrinsic properties including the thermosensitivity (e.g., the body temperature-induced sol-gel transition), non-cytotoxicity, and controlled drug release. A detailed study on the sol-gel-sol transition behavior of different copolymers showed that an appropriate extent of modification with DMA retained a sol-gel-sol transition, despite the fact that a too high extent caused a loss of sol-gel-sol transition. The hydrophilic and hydrophobic balance between mPEG and PCL was most likely broken upon a high extent of quaternization due to a large disturbance effect of DMA units at a large quantity (as evidenced by the heavily depressed PCL segment crystallinity), and thus the micelle aggregation mechanism for the gel formation could not work anymore, along with the loss of the thermosensitivity. The work presented here is highly expected to be generalized for synthesis of various block copolymers with immunity to microorganisms. Light may also be shed on understanding the phase transition behavior of various multiblock copolymers.
Preparation and characterization of carboxymethyl chitosan/polyvinyl alcohol composite hydrogel with improved mechanical and antibacterial properties

Abstract:Biopolymer-based antibacterial hydrogels have attracted considerable interest in biomedical field. Herein, we report on an effective strategy for the preparation of carboxymethyl chitosan/polyvinyl alcohol (CMCh/PVA) composite hydrogel with improved mechanical and antibacterial properties by using poly(ethylene glycol) diglycidyl ether (PEGDE) as crosslinking agent and zinc acetate (ZnAc 2 ) as catalyst, respectively. FT-IR and gelation time measurements confirm that the introduction of both PVA and ZnAc 2 can accelerate the formation of ester bonds between CMCh and PEGDE. SEM images reveal the interconnected macroporous inner architecture in the current CMCh/PVA composite. While the hydrogels exhibit pH-sensitive swelling behaviors in the pH range from 3 to 11, they remain relatively stable in phosphate buffered saline buffer. Compression mechanical tests show that, when increasing the PVA concentration to 1.0%, the resultant CMCh/PVA composite hydrogel shows a compressive strength of 1.96 MPa. The dried composite sample prepared with 1.0% PVA has a water absorption capacity of 1860%. The antibacterial activities of the CMCh/PVA hydrogel are studied against both Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus by inhibition zone method, indicating that the composite hydrogel exhibits improved antibacterial activity after the adsorption of zinc ions.

Abstract:Polymeric materials with antibacterial properties hold great promise for combating multidrug-resistant bacteria, which pose a significant threat to public health. However, the synthesis of most antibacterial polymers typically involves complicated and time-consuming procedures. In this study, we demonstrate a simple and efficient strategy for synthesizing functional poly(vinylpyridinium salt)s via pyridinium-yne click polymerization. This click polymerization could proceed with high atom economy under mild conditions without any external catalyst, yielding soluble and thermally stable poly(vinylpyridinium salt)s with satisfactory molecular weights and well-defined structures in excellent yields. Additionally, the incorporation of luminescent units such as fluorene, tetraphenylethylene, and triphenylamine into the polymer backbone confers excellent aggregation-enhanced emission properties upon the resulting polymers, rendering them suitable for bacterial staining. Moreover, the existence of pyridinium salt imparts intrinsic antibacterial activity against multidrug-resistant bacteria to the polymers, enabling them to effectively inhibit wound bacterial infection and significantly expedite the healing process. This work not only provides an efficient method to prepare antibacterial polymers, but also opens up the possibility of various applications of polymers in healthcare and other antibacterial fields.
