Abstract:We demonstrate three-dimensional vibrational imaging of director structures in liquid crystals using coherent anti-Stokes Raman scattering (CARS) polarized microscopy. Spatial mapping of the structures is based on sensitivity of a polarized CARS signal to orientation of anisotropic molecules in liquid crystals. As an example, we study structures in a smectic material and demonstrate that single-scan CARS and two-photon fluorescence images of molecular orientation patterns are consistent with each other and with the structure model.

Abstract:Liquid crystals are a class of industrially important materials whose optical properties make them useful particularly in display technology. Optical imaging of these materials provides information about their structure and physical properties. Coherent anti-Stokes Raman scattering (CARS) microscopy is used to provide three-dimensional chemical maps of liquid crystalline samples without the use of external labels. CARS is an optical imaging technique that derives contrast from Raman-active molecular vibrations in the sample. Compared to many other three-dimensional imaging techniques, CARS offers more rapid chemical characterization without the use of external dyes or contrast agents. The use of CARS to image chemical and orientational order in liquid crystals is demonstrated using several examples, and the limitations and benefits are discussed.

Abstract:Coherent anti-Stokes Raman scattering (CARS) microscopy permits vibrational imaging with high-sensitivity, high speed, and three-dimensional spatial resolution. We review recent advances in CARS microscopy, including experimental design, theoretical understanding of contrast mechanisms, and applications to chemical and biological systems. We also review the development of multiplex CARS microspectroscopy, which allows high-speed characterization of microscopic samples, and CARS correlation spectroscopy, which probes fast diffusion dynamics with vibrational selectivity.

Abstract:Recent advances on CARS microscopy provided exciting possibilities for imaging living cells with high sensitivity, high 3D spatial resolution and chemical selectivity based on vibrational spectroscopy. Epi-detection avoids the forward-going water signal and allows imaging of small features inside cells. Polarization-sensitive detection improves the vibrational contrast in CARS microscopy via suppression of the non-resonant background. High-speed CARS imaging of living cells is realized by raster scanning two collinearly overlapped near infrared picosecond laser beams. Vibrational mapping of lipids and proteins inside living cells is achieved. CARS imaging of unstained cells during mitosis and apoptosis is carried out. Incorporation of CARS microscopy with the photon correlation technique allows probing diffusion dynamics of 50-nm beads with chemical selectivity.

Abstract:Coherent Anti Stokes Raman Scattering (CARS) offers many advantages for nonlinear bio-imaging, thanks to its sub-cellular spatial resolution and unique chemical specificity. Its working principle requires two incident pulsed laser beams with distinct frequencies to be focused in space and time, which focus quality however rapidly deteriorates when propagating at large depths in biological tissues. The depth limits of CARS and the capability of wavefront correction to overcome these limits are currently unknown. In this work we exploit the spectral correlation properties of the transmission matrix of a scattering medium in a pulsed regime, to recover coherent focusing for two distant incident CARS wavelengths which propagation is initially uncorrelated. Using wavefront shaping with a single spatial light modulator, we recover CARS generation through thick mice spinal cord tissues where initially no signal is measurable due to scattering, and demonstrate point scanning over large field of views of tens of micrometers.

Abstract:We propose and develop a new way for nonscanning coherent anti-Stokes Raman scattering (CARS) imaging by using CARS holography, which can capture, at a single shot, both the amplitude and the phase of a coherent Raman image. It hence allows for chemically selective three-dimensional imaging at a laser pulse duration limited speed, which is not possible to achieve by using any of the existing laser scanning based Raman imaging techniques. We have demonstrated single-shot (pulse duration: approximately 5 ns) holographic CARS imaging of microspheres. Our results show that coherent anti-Stokes Raman holography can open a new path for nonscanning label-free imaging.

Abstract:We present a systematic characterization of coherent anti-Stokes Raman scattering (CARS) microscopy. CARS signal generation in a heterogeneous sample under a tight-focusing condition is formulated by the Green’s function method. The CARS radiation pattern and the forward- and backward-detected CARS signals from a three-dimensional Raman scatterer are calculated. The coherent nature of CARS image formation and its consequences for image contrast and spatial resolution are investigated. Experimental implementations of CARS microscopy with collinearly copropagating and counterpropagating excitation beams, forward and backward data collection, and polarization-sensitive detection are described. Finally, CARS images of unstained live cells with forward detection, epidetection, and polarization-sensitive detection are presented and compared.

Abstract:We have achieved rapid nonlinear vibrational imaging free of nonresonant background with heterodyne coherent anti-Stokes Raman scattering (CARS) interferometric microscopy. This technique completely separates the real and imaginary responses of nonlinear susceptibility chi(3) and yields a signal that is linear in the concentration of vibrational modes. We show that heterodyne CARS microscopy permits the detection of weak vibrational resonances that are otherwise overshadowed by the strong interference of the nonresonant background.

The theory of CARS and its integration with digital holography is explored. Abstract:We report coherent anti-Stokes Raman scattering correlation spectroscopy (CARS-CS) that measures the fluctuation of the CARS signal from scatterers in a subfemtoliter excitation volume. This method probes dynamical processes with chemical selectivity based on vibration spectroscopy. High-sensitivity CARS-CS measurements are carried out with epi-detection or polarization-sensitive detection. Theoretical expressions of CARS intensity autocorrelation functions are derived and supported by experimental data. The properties of CARS-CS are-characterized with measurements of diffusion dynamics of small polystyrene spheres.

Abstract:Laser-scanning coherent anti-Stokes Raman scattering (CARS) microscopy with fast data acquisition and high sensitivity has been developed for vibrational imaging of live cells. High three-dimensional (3D) resolution is achieved with two collinearly overlapped near infrared picosecond beams and a water objective with a high numerical aperture. Forward-detected CARS (F-CARS) and epi-detected CARS (E-CARS) images are recorded simultaneously. F-CARS is used for visualizing features comparable to or larger than the excitation wavelength, while E-CARS allows detection of smaller features with a high contrast. F-CARS and E-CARS images of live and unstained cells reveal details invisible in differential interference-contrast images. High-speed vibrational imaging of unstained cells undergoing mitosis and apoptosis has been carried out. For live NIH 3T3 cells in metaphase, 3D distribution of chromosomes is mapped at the frequency of the DNA backbone Raman band, while the vesicles surrounding the nucleus is imaged by E-CARS at the frequency of the C-H stretching Raman band. Apoptosis in NIH 3T3 cells is monitored using the CARS signal from aliphatic C-H stretching vibration.

Abstract:Coherent anti-Stokes Raman microscopy (CARS) is a quantitative, chemically specific, and label-free optical imaging technique for studying inhomogeneous systems. However, the complicating influence of the nonresonant response on the CARS signal severely limits its sensitivity and specificity and especially limits the extent to which CARS microscopy has been used as a fully quantitative imaging technique. On the basis of spectral focusing mechanism, we establish a dual-soliton Stokes based CARS microspectroscopy and microscopy scheme capable of quantifying the spatial information of densities and chemical composition within inhomogeneous samples, using a single fiber laser. Dual-soliton Stokes scheme not only removes the nonresonant background but also allows robust acquisition of multiple characteristic vibrational frequencies. This all-fiber based laser source can cover the entire fingerprint (800-2200 cm-1) region with a spectral resolution of 15 cm-1. We demonstrate that quantitative degree determination of lipid-chain unsaturation in the fatty acids mixture can be achieved by the characterization of C = C stretching and CH2 deformation vibrations. For microscopy purposes, we show that the spatially inhomogeneous distribution of lipid droplets can be further quantitatively visualized using this quantified degree of lipid unsaturation in the acyl chain for contrast in the hyperspectral CARS images. The combination of compact excitation source and background-free capability to facilitate extraction of quantitative composition information with multiplex spectral peaks will enable wider applications of quantitative chemical imaging in studying biological and material systems.

Abstract:We report polarization coherent anti-Stokes Raman scattering (P-CARS) microscopy that allows vibrational imaging with high sensitivity and spectral selectivity. The nonresonant background signals from both Raman scatterers and the solvent are efficiently suppressed in P-CARS microscopy. We demonstrate P-CARS imaging of unstained cells based on the contrast of the protein amide I band.

Abstract:CARS holography captures both the amplitude and the phase of a complex anti-Stokes field, and can perform three-dimensional imaging by digitally focusing onto different depths inside a specimen. The application of CARS holography for bio-imaging is demonstrated. It is shown that holographic CARS imaging of sub-cellular components in live HeLa cells can be achieved.

Super-resolved CARS by coherent image scanning

Abstract:We present super-resolved coherent anti-Stokes Raman scattering (CARS) microscopy by implementing phase-resolved image scanning microscopy (ISM), achieving up to two-fold resolution increase as compared with a conventional CARS microscope. Phase-sensitivity is required for the standard pixel-reassignment procedure since the scattered field is coherent, thus the point-spread function (PSF) is well-defined only for the field amplitude. We resolve the complex field by a simple add-on to the CARS setup enabling inline interferometry. Phase-sensitivity offers additional contrast which informs the spatial distribution of both resonant and nonresonant scatterers. As compared with alternative super-resolution schemes in coherent nonlinear microscopy, the proposed method is simple, requires only low-intensity excitation, and is compatible with any conventional forward-detected CARS imaging setup.

Abstract:We demonstrate a multi-color background-free coherent anti-Stokes Raman scattering (CARS) imaging system, using a robust, all-fiber, low-cost, multi-wavelength time-lens source. The time-lens source generates picosecond pulse trains at three different wavelengths. The first is 1064.3 nm, the second is tunable between 1052 nm and 1055 nm, and the third is tunable between 1040 mu and 1050 mu. When the time-lens source is synchronized with a mode-locked Ti:Sa laser, two of the three wavelengths are used to detect different Raman frequencies for two-color on-resonance imaging, whereas the third wavelength is used to obtain the off-resonance image for nonresonant background subtraction. Mixed poly(methyl methacrylate) (PMMA) and polystyrene (PS) beads are used to demonstrate two-color background-free CARS imaging. The synchronized multi-wavelength time-lens source enables pixel-to-pixel wavelength-switching. We demonstrate simultaneous two-color CARS imaging of CH2 and CH3 stretching vibration modes with real-time background subtraction in ex vivo mouse tissue.
